The causal relationship between the palmitoylation gene ZDHHC13 and ulcerative colitis: A Mendelian randomization study implicating CD62L-monocyte AC.

This study explores the potential mechanisms between palmitoylation and ulcerative colitis (UC) through Mendelian randomization, bioinformatics analysis combined with eQTL and protein quantitative trait loci. This study selected a dataset of 5034 UC patients from the Finnish database and conducted Mendelian randomization analysis with palmitoylation target genes to explore the causal relationships between the two. Sensitivity and pleiotropy tests were performed alongside bioinformatics analysis to screen for the best target genes, ensuring the robustness of the results. Finally, immune cells w

Early Post-Transplant Changes in Lipoprotein(a), Autotaxin Activity, and Lipid Profile: A Prospective Observational Study of Tacrolimus-Treated Kidney Transplant Recipients in Poland.

Kidney transplantation (KTx) corrects many uremia-related metabolic disturbances; however, dyslipidemia remains common in kidney transplant recipients and contributes to persistent cardiovascular risk. Lipoprotein(a) [Lp(a)] is a largely genetically determined proatherogenic lipoprotein that increases in advanced chronic kidney disease (CKD) and may decrease after restoration of renal function. Autotaxin (ATX), an enzyme involved in proinflammatory lipid signaling through the ATX-lysophosphatidic acid axis, has also been implicated in cardiovascular pathology, but its early post-transplant dyn

Pilot-scale inoculum-free valorization of raw chicken feathers: ammonium recovery, keratinase production and community dynamics.

Chicken feathers constitute a major keratin-rich agro-industrial residue whose sustainable valorization remains challenging due to the recalcitrant structure of keratin and the energy-intensive nature of conventional treatments. In this study, we investigated the feasibility of converting raw, mechanically untreated chicken feathers into soluble nitrogen compounds and extracellular keratinases through an inoculum-free microbial process operated at pilot scale. A 50 L horizontal bioreactor was run using only tap water and the indigenous feather-associated microbiota, without sterilization, nutr

Simultaneous overexpression of three enzymes of chloroplast metabolism fails to improve CO2 assimilation or biomass.

Ensuring an adequate food supply amidst a growing global population and climate change challenges necessitates innovative strategies to enhance crop productivity. Previous studies have demonstrated that the simultaneous stimulation of different photosynthesis-related processes can increase the rate of photosynthetic carbon assimilation and plant biomass. This study evaluates an approach based on modelling aimed at simultaneously increasing photosynthetic and sink capacities in Nicotiana tabacum by overexpressing three key enzymes: sedoheptulose-1,7-bisphosphatase (SBPase), fructose-1,6-bisphos

Enzymatically synthesized kaempferol fatty acid esters enhance glycolipid metabolism regulation and α-glucosidase inhibition.

Kaempferol (KAE) is a dietary flavonoid with recognized effects on glucolipid metabolism and oxidative stress, but its poor lipophilicity and low bioavailability limit its application. In this study, KAE derivatives with different fatty acid chain lengths (C2-C12) were synthesized using acylation catalyzed by lipozyme TL IM, and their biological activities were then systematically evaluated. In vivo assays in hyperglycemic zebrafish (Danio rerio) demonstrated that two medium-chain derivatives, 4'-O-octanoyl-kaempferol (C8-KAE) and 4'-O-decanoyl-kaempferol (C10-KAE), exerted superior effects on

Screening of a feruloyl esterase-producing lactobacillus strain and its application in citrus whole-fruit pulp fermentation: organic acids, volatile compounds, and sensory evaluation.

Feruloyl esterase (FAE)-producing lactobacillus strains were selected from the citrus whole-fruit pulp after 48 h of fermentation. Among them, Lactiplantibacillus plantarum K33 showed the highest ferulic acid (FA) producing ability. Using citrus whole-fruit pulp as the fermentation matrix, physicochemical, compositional, and sensory outcomes were compared between pulps fermented by K33 and by the commercial strain L. plantarum N13 (Lp-N13). After fermentation, K33 increased total acidity, lowered pH, and markedly raised FA concentration and total phenolic content (TPC). Fermentation also alter

Effective high-dose methotrexate toxicity reversal using fixed-dose glucarpidase in obese patients: a case series.

BACKGROUND: This case report and literature review present two instances of delayed methotrexate clearance in individuals who were successfully managed using fixed dosing versus weight-based dosing of glucarpidase. These cases, along with existing literature, support the use of fixed-dose glucarpidase as an alternative to weight-based dosing for delayed methotrexate clearance. CASE PRESENTATION: Patient 1: a 26-year-old obese African American male with newly diagnosed osteosarcoma of the right distal fibula underwent his first cycle of neoadjuvant chemotherapy with MAP. Extracellular vesicle engineering using a small scaffold protein.

Extracellular vesicles (EVs) are promising drug-delivery vehicles owing to their biocompatibility and low immunogenicity. Genetic engineering of a membrane-bound EV-sorting scaffold protein empowers EVs by installing targeting moieties on the surface and enriching therapeutic cargo in the lumen. However, the choice of scaffold proteins with simple structures and short sequences is limited. Here, we conduct mass spectrometry-based proteomic studies and identify ENPP1 as a superior scaffold protein. Furthermore, we show that a truncated 144-amino acid variant, EN144, efficiently loads diverse th

ENPP1-dependent USP2 ubiquitination governs SQSTM1-mediated autophagy-dependent ferroptosis in trophoblast cells and exacerbates placental dysfunction in gestational diabetes mellitus.

Downregulation of ectonucleotide pyrophosphatase/phosphodiesterase 1 (ENPP1) has been implicated in autophagic cell death. However, how ENPP1 regulates the interplay between autophagy and ferroptosis to maintain trophoblast homeostasis in the context of gestational diabetes mellitus (GDM) remains unclear. To determine ENPP1's role in autophagy-dependent ferroptosis and its contribution to GDM-related placental injury, clinical placental tissues, hyperglycemia-treated HTR8/SVneo trophoblasts, and streptozotocin-induced GDM mice were analyzed. Ubiquitination assays, co-immunoprecipitation, funct

Protease-serpin-microbiome interactions in inflammatory bowel disease: toward integrated biomarkers of gut health.

Proteolytic imbalance involving host and microbial serine proteases and their inhibitors (serpins) contributes to intestinal barrier disruption and chronic inflammation in inflammatory bowel disease (IBD). However, the interplay among these components remains insufficiently characterized. Here, we survey 13,304 publications over five decades, to map protease-serpin-microbiome literature in IBD. Our analysis reveals a fragmented literature structure, with uneven coverage and limited cross-domain integration among host proteases, microbial proteases, and inhibitory pathways. We synthesize eviden

An ultra-sensitive liquid chromatography-fluorescence detection method for determining hCE1 activity and evaluation of hCE1 as a biomarker of liver injury.

Human carboxylesterase 1 (hCE1) is highly expressed in the liver and can be released into the bloodstream upon hepatocellular injury, suggesting its potential as a biomarker for liver damage. However, current methods for measuring hCE1 activity suffer from poor selectivity and insufficient sensitivity and are easily interfered with by serum albumin (HSA), butyrocholinesterase (BChE), and paraoxonase 1 (PON1). In this study, a novel fluorescent probe, NCMNe, was designed and synthesized, and an ultrasensitive liquid chromatography-fluorescence detection (LC-FD) method was established for quanti

Hepatic sEH drives stress-induced neuroinflammation via IL-6 and liver-brain axis.

INTRODUCTION: Restraint stress is known to induce damage to vital organs. Previous work indicates that acute restraint stress (ARS) causes significant hepatic injury in mice. Interestingly, while the liver gradually recovered following removal of the stressor, stress-induced behavioral abnormalities persisted for up to 7 days after stress cessation. Soluble epoxide hydrolase (sEH) plays a key regulatory role in neuroinflammation and mood disorders. However, whether and how hepatic sEH upregulation contributes to ARS-induced neurological damage remains unknown. METHODS: In a mouse model of ARS

PIK3CA Mutations Downregulate PPT1 to Promote Adipogenesis by Suppressing P300 Depalmitoylation and Phase Separation.

PIK3CA mutations drive benign adipose overgrowth in facial infiltrating lipomatosis (FIL), but the downstream molecular mechanisms remain incompletely understood. This study investigated the role of palmitoyl-protein thioesterase 1 (PPT1)-mediated depalmitoylation in regulating aberrant adipogenesis induced by mutant PIK3CA. Using single-cell RNA-seq, molecular dynamics simulations, and functional assays in primary human FIL adipose-derived stem and progenitor cells (ASPCs), immortalized cell lines, and mouse models, we dissected the signaling pathway linking PIK3CA mutation to adipogenesis. Anchored kinase and phosphatase signaling networks in amyloid β-induced synaptic dysfunction.

Impaired excitatory synaptic function and synapse loss are early hallmarks of Alzheimer's disease (AD). There is strong biochemical, genetic, physiological, and anatomical evidence that the accumulation of soluble amyloid beta (Aβ) oligomers in the brain leads to AD-related synapse dysfunction and cognitive impairment. Long-term potentiation (LTP), a key form of synaptic plasticity for learning and memory, is disrupted in several mouse models harboring familial early-onset AD-linked mutations that lead to Aβ accumulation, and even acute applications of Aβ oligomers block LTP within minutes and

Quality Maintenance and Increased Firmness of Fresh-Cut Strawberries Using Vacuum Infusion of Calcium Lactate and Pectin Methylesterase.

The demand for healthy foods has led to an increase in the consumption of fresh fruits, especially fresh-cut (FC) fruits. However, FC processing negatively affects fruit firmness, an important parameter for consumer acceptance. The objective of this work was to evaluate the effectiveness of the combined use of vacuum, calcium lactate, and pectin methylesterase (PME) to increase the shelf life and quality of FC strawberries. FC strawberry halves were subjected to vacuum infusion with water, PME (10UI), PME plus 1% calcium lactate, or 1% calcium lactate alone; FC strawberries without infusion we
