Discovery of lipid biomarkers for idiopathic Parkinson's disease in red blood cells and plasma.

Parkinson's disease (PD) is a neurodegenerative disorder characterized by dopaminergic neuron loss and lipid-rich Lewy bodies containing misfolded alpha-synuclein, implicating lipid dysregulation in pathogenesis. We performed comprehensive lipidomic profiling using liquid chromatography-ion mobility mass spectrometry on paired red blood cell (RBC) and plasma samples from idiopathic PD (iPD) patients (n = 156) and cognitively normal controls (n = 155). We identified significant alterations in 240 RBC and 113 plasma lipids, with elevated sphingomyelins, ceramides, and phosphatidylcholines in RBC

Curcumin from Turmeric Alleviates Lipid Metabolism: Network Pharmacology and Cellular Validation.

Curcumin, the primary active compound in turmeric, a widely used medicinal and edible herb in traditional Chinese medicine, exhibits beneficial effects against lipid metabolic dysfunction (LMD). Nevertheless, its comprehensive regulatory network and precise mechanisms remain unclear. In this study, network pharmacology, molecular docking, and in vitro cellular assays were integrated to elucidate the targets and mechanisms by which curcumin acts against LMD. Seven hub targets were identified: AKT1, ALB, INS, IL6, TP53, TNF, and PPARG. Kyoto Encyclopedia of Genes and Genomes analysis revealed si

Exploration of biodistribution of mRNA-delivering lipid nanoparticles in the neonatal mouse inner ear.

Hearing loss affects millions worldwide, yet hearing aids and cochlear implants have major limitations. mRNA-based gene therapy could address the underlying molecular causes, but lipid nanoparticle (LNP) delivery in the inner ear remains underexplored and its biodistribution has not been systematically characterized. Here we establish and scale up an mRNA-encapsulated LNP formulation using a design-of-experiments (DoE) approach built on predefined critical quality attributes, and map the route-dependent distribution of reporter mRNA expression in the neonatal mouse cochlea. In HEI-OC1 auditory

Optimization of lipid nanoparticles reduces hepatic accumulation and enhances local delivery of mRNA.

Lipid nanoparticles (LNPs) have emerged as the most widely used non-viral gene delivery system, particularly for mRNA therapeutics. However, predominant hepatic accumulation and poor local delivery remain major barriers to their broader application. To address this challenge, we employed a design of experiments (DOE) strategy to systematically refine LNP composition and identified an optimized formulation, SM102M. Compared with the parental SM102 formulation, SM102M markedly reduced liver-associated signal after both intramuscular and intravenous administration, while preserving or enhancing l

Nanostructured Quercetin-Lipid Carrier on 3D Printed Scaffolds for Improved Bone Regeneration.

Musculoskeletal defects remain difficult to treat because current grafts often have limited supply, donor-site morbidity, risk of immune reaction, or insufficient local biological stimulation. To address this problem, this study developed 3D-printed β-tricalcium phosphate (TCP) scaffolds loaded with quercetin-loaded nanostructured lipid carriers (Que-NLCs) to combine an osteoconductive ceramic structure with local delivery of a natural osteogenic and anti-resorptive compound. Que-NLCs provided controlled release for up to 21 days and reduced the initial burst release compared with free Que at

Loss of SREBF1 disrupts lipid signaling and causes slow muscle fiber degeneration.

SREBF1, encoding the lipogenic transcription factor SREBP1, has been linked to lean mass by human genomics. We used a CRISPR-Cas9 srebf1 zebrafish knockout combined with LC-MS/MS lipidomics, RNA-seq, Lugol-enhanced micro-CT imaging, and functional assays. Srebp1 loss disrupted slow-fiber organization, increased locomotion in larvae, and reduced muscle mass in adults. This knockout muscle showed altered fatty acid composition, reduced arachidonoylethanolamide (AEA), elevated palmitoylethanolamide (PEA), broad remodeling of polyunsaturated fatty (PUFA) acid-derived mediators, and differential ex

IMiD/CELMoD resistance in myeloma is associated with changes in lipid metabolism that may be targetable.

Immunomodulatory drugs (IMiDs) are a cornerstone of multiple myeloma treatment, and newer cereblon E3 ligase modulatory drugs (CELMoDs) are in clinical trials. However, a major barrier to improving patient outcomes is the inevitable development of resistance to these agents. To explore the mechanisms underlying IMiD/CELMoD resistance and identify novel targets for treatment, human cell line models of acquired IMiD/CELMoD resistance were generated. A quantitative proteome analysis of these models identified common changes in lipid synthesis proteins, and glucose-labeling experiments confirmed a

AMPK-Mediated Lipid Metabolic Reprogramming in Cancer: From Mechanisms to Targeted Therapies.

AMP-activated protein kinase (AMPK), a key regulator of cellular energy metabolism, is activated under conditions of energy stress to restore metabolic homeostasis by promoting catabolic pathways and inhibiting anabolic processes such as lipid synthesis. Multiple key molecules involved in lipid metabolism are regulated by AMPK, and its role in cancer progression through lipid metabolism is dual-edged. On the one hand, AMPK suppresses lipogenesis by inhibiting key regulators such as SREBP, ACC, and HMGCR, thereby limiting the biosynthetic substrates and membrane lipid availability required for

Cucurbita pepo var. styriaca Seeds: Deep Insights into Polar Lipid Profile.

The edible seeds of pumpkin plants (genus Cucurbita) are becoming increasingly appreciated as functional foods for their nutritional benefits, medicinal properties, and bioactive compounds, including lipids, proteins, and antioxidants. Particularly, the naked seeds of Cucurbita pepo var. styriaca have proved to yield both an edible oil showing anti-inflammatory properties in treating skin disorders and hydro-alcoholic extracts effective in inhibiting the growth of cancer cells. In this study, a detailed and extensive analysis of the eco-friendly alcoholic extract of the seeds of this variety w

HlncRNA-1 integrates the hepatic circadian clock with lipid metabolism by targeting Hdlbp.

The circadian rhythm and lipid metabolism in organisms are tightly coupled, yet the role of lncRNAs in the integration of these two important physiological processes remains unclear. In this study, we identified a liver-specific lncRNA named HlncRNA-1, which regulates lipid metabolism while responding to circadian rhythms. Mice overexpressing HlncRNA-1 exhibited suppressed liver fat accumulation under conditions of both nutrient surplus and deficiency, along with improved insulin sensitivity. At the molecular level, ChIP assays demonstrate that Dhx9 directly occupies the HlncRNA-1 promoter, te

Plasma lipid remodeling during the early recovery phase after myocardial infarction.

Acute myocardial infarction (MI) induces extensive disturbances in lipid homeostasis; however, most lipidomic studies have focused on the acute hours-to-days phase or the chronic months-to-years period. To the best of our knowledge, this is the first study to characterize plasma lipidomic patterns specifically during the early recovery window (3–6 weeks post-MI), a clinically stable yet biochemically active stage of healing. We compared plasma lipid profiles—including phospholipids, sphingomyelins (SMs), and ceramides (Cers)—in acute MI patients, early-recovery patients, and healthy controls.

Population-based genome-wide association study of plasma complex lipid species.

The human lipidome comprises numerous complex lipids, dysregulation of which can contribute to the pathogenesis of a wide range of diseases. Despite the high heritability of parts of the lipidome, the genetic architecture of many circulating lipid species and their structure remains mostly unknown. Thus, we perform genome-wide association studies on 970 lipid species and 267 fatty acid composite measures using samples from the population-based Rhineland Study (n = 6096). We validate our findings using corresponding data from two other independent cohorts, including FinnGen (n = 7266) and EPIC-

Fatty acid metabolism and lipid channeling in macrophages: mechanisms of inflammation, resolution, and lipotoxicity.

Macrophages integrate metabolic signals with immune activation, and their ability to handle fatty acids is central to preventing lipotoxicity and to sustaining effector functions. In cardiometabolic settings such as obesity, metabolic dysfunction-associated steatotic liver disease, and atherosclerosis, chronic exposure to excess free fatty acids and toxic lipid species, such as ceramides, disrupts organelle integrity, impairs efferocytosis, and skews macrophages toward pro-inflammatory phenotypes. This review examines how macrophages channel fatty acids into β-oxidation, glycerolipid synthesis

Paternal obesity programs adverse serum lipid and lipoprotein profiles in rat offspring.

While maternal obesity is a well-established determinant of offspring cardiometabolic health, the contribution of paternal obesity to metabolic programming remains less well defined. This study investigated the impact of paternal obesity on obesity and metabolic health outcomes in male and female offspring using a Sprague Dawley rat model. Male rats were fed either a low-calorie control or high-calorie diet for a 6-week pre-conception period prior to mating with lean, control-fed females. Male and female offspring were assessed at weaning and in adulthood for growth parameters and cardiometabo

Lipid metabolism in hepatopancreas and ovaries of the mud crab Scylla paramamosain during vitellogenesis.

Ovarian lipids are essential nutrients that fundamentally determine oocyte and offspring quality, and ultimately affect the reproductive performance of decapod crustaceans. Lipids accumulated in the ovaries during vitellogenesis are mainly derived from the hepatopancreas. However, the molecular mechanisms underlying lipid metabolism in the hepatopancreas and ovaries during vitellogenesis remain largely unclear. In this study, comparative transcriptomic and lipidomic analyses were conducted on the hepatopancreas and ovaries of the mud crab Scylla paramamosain. From early to late vitellogenic st

Personalized polygenic profiling based on the genetic architecture of lipid metabolism in the Russian population.

OBJECTIVE: Elevated cholesterol levels are associated with the risk of the most socially significant cardiovascular diseases, such as atherosclerosis, ischemic heart disease, and myocardial infarction. METHODS: In this study, we sought to study the genetic architecture of lipid metabolism by conducting genome-wide association studies of total cholesterol, LDL-C, and HDL-C levels in a sample of the Russian population (n = 8,732) who were not carriers of variants linked to familial hypercholesterolemia and did not take lipid-regulating agents. Based on the detected associations and machine lear

Optimizing nucleic acid delivery using PF14 peptide and lipid nanoparticle systems.

Nucleic acid-based therapeutics are a rapidly expanding class of precision medicines capable of directly modulating gene expression. However, their clinical application is limited by challenges in cellular delivery, including large molecular size, hydrophilicity, and susceptibility to enzymatic degradation. To address these barriers, advanced delivery platforms such as cell-penetrating peptides and lipid nanoparticles (LNPs) have been developed. This study evaluates two delivery systems: the cell-penetrating peptide PepFect-14 (PF14) and LNP formulations, for enhancing nucleic acid delivery an

Local Organization of Biological Membranes Modulates Tau-Lipid Interactions and Fibril formation.

The formation of Tau amyloids is a hallmark of several neurodegenerative diseases, called Tauopathies, including Alzheimer's disease. In different Tauopathies, Tau amyloid filaments adopt a distinct structure, highlighting the existence of disease specific pathways. In this pathological context, lipid metabolism is heavily disrupted, leading to a perturbation of membrane composition. Lipid membranes have been shown to nucleate tau aggregation under some conditions. However, no general model has been established to explain how the organization of the lipid membrane modulates Tau aggregation. He

Membrane curvature enhances oxidation within lipid bilayers in a composition-dependent manner.

UNLABELLED: Excessive production of reactive oxygen species (ROS) in cells results in oxidative stress, which can promote lipid oxidation in cellular membranes. This oxidation of membrane lipids accompanies various diseases and can even result in cell death through processes such as ferroptosis. The complex compositions and diverse morphologies of cellular membranes make understanding the mechanisms of lipid oxidation challenging, especially when attempting to investigate membrane composition and curvature simultaneously. Here, we utilize reconstituted lipid membranes and the fluorescent oxida
