Uveitis with occult choroiditis due to Mycobacterium kansasii: limitations of interferon-gamma release assay (IGRA) tests (case report and mini-review on ocular non-tuberculous mycobacteria and IGRA cross-reactivity).

Ocular tuberculosis is difficult to diagnose but should be suspected when uveitis fails to respond to inflammation suppressive therapy. Interferon-gamma release assays (IGRAs) represent a substantial help to diagnose suspected ocular tuberculosis especially in non-endemic areas. Indocyanine green angiography (ICGA) is able to detect clinically silent choroiditis that, when associated with a positive IGRA test, should lead the clinician to suspect ocular tuberculosis, warranting specific therapy. The fact that IGRA tests can also react with some atypical strains of mycobacteria is not always kn

The metallophosphoesterase Rv0805 regulates carbon flux and cell envelope homeostasis during growth of mycobacteria in propionate.

Mycobacterium tuberculosis relies on host-derived lipids, including cholesterol, for intracellular survival, generating propionyl-CoA-a metabolite that must be efficiently assimilated to prevent toxicity. The metallophosphoesterase Rv0805 is required for optimal growth on cholesterol, and an Rv0805 knockout strain exhibits impaired ability to colonize the murine lung. However, the mechanisms underlying the essential role of Rv0805 under host-relevant conditions remain unclear. The deletion of the rv0805 ortholog (bcg_0857) in Mycobacterium bovis BCG reveals that both its catalytic activity and

Synthesis, computational and in vitro anti-tubercular activity studies of thiadiazole-linked pyridazinone derivatives.

Mycobacterium tuberculosis (Mtb) is an infectious bacterium that causes pulmonary tuberculosis (TB). The emergence of drug-resistant Mtb strains necessitates the need for the identification of novel antitubercular agents. In this study, we have repurposed pyridazinone-1,3,4-thiadiazole hybrids for their in-vitro antitubercular activity against Mtb. We have designed and synthesised a series of pyridazinone-1,3,4-thiadiazole hybrids (7a-r). Among these, several compounds (especially 7a, 7c, 7d, 7e, 7n, 7p, 7q, and 7r) inhibited Mtb in vitro, while compound 7d showed moderate antimycobacterial ac

Activity Profiling of Mycobacterial L,D-Transpeptidases.

Antimicrobial resistance poses major therapeutic challenges, particularly for multidrug-resistant mycobacterial infections caused by Mycobacterium tuberculosis (Mtb) and non-tuberculous mycobacteria (NTM). l,d-Transpeptidases (Ldts) are attractive drug targets due to their essential role in peptidoglycan cell wall crosslinking, yet existing assays suffer from low throughput and limited sensitivity. We report a versatile, bead-based platform for high-throughput analysis of Ldt activity and inhibitor discovery. We incubated peptidoglycan stem peptides, either naturally harvested or synthetically

Engineering an Artificial Myxopyronin Derivative with Enhanced Metabolic Stability via Mutasynthesis.

The rise of multidrug-resistant pathogens, such as Staphylococcus aureus and Mycobacterium tuberculosis, underscores an urgent need for therapeutic innovation. The antibiotic development pipeline targeting these bacteria is critically limited, with most discovered candidates exhibiting structurally similar features of prominent chemical entities and with well-established molecular targets or binding modes. The myxobacterial α-pyrone antibiotics, myxopyronins, represent a highly promising compound class due to their ability to inhibit RNA polymerase by binding to the "switch region", a distinct

Isoleucyl-tRNA synthetase depletion reveals vulnerabilities in Mycobacterium abscessus and Mycobacterium marinum.

Mycobacterium abscessus and Mycobacterium marinum are nontuberculous mycobacteria that pose significant challenges due to their high drug resistance and persistence in hostile host environments. Aminoacyl-tRNA synthetases, such as isoleucyl-tRNA synthetase (IleRS), are crucial for protein synthesis and represent promising targets for antimicrobial development. This study investigates the role of IleRS in mycobacterial growth, metabolism, and pathogenesis using conditional gene silencing combined with microbiological, metabolomic, and transcriptomic analyses. Our findings indicate that IleRS is

Mycobacterium-Induced Reactive Arthritis in an Older Adult With Rheumatoid Arthritis.

The use of immunosuppressive medications to treat rheumatoid arthritis may trigger the activation of latent mycobacteria, leading to infection. These infections can lead to reactive arthritis. Conversely, both reactive and rheumatoid arthritis may be encountered in the geriatric population. When such complications arise, the treatment process becomes more complicated, necessitating careful consideration of elaborate therapeutic approaches. An 83-year-old man presented to our hospital with subacute back pain and arthralgia of the extremities. The patient was diagnosed with rheumatoid arthritis

Tuberculous Fibrosis Enhances Tumorigenic Potential via the NOX4-Autophagy Axis.

While a higher incidence of lung cancer in subjects with previous tuberculous infection has been reported in epidemiologic data, the mechanism by which previous tuberculosis affects lung cancer remains unclear. We investigated the role of NOX4 in tuberculous pleurisy-assisted tumorigenicity both in vitro and in vivo.Heat-killed Mycobacterium tuberculosis-stimulated mesothelial cells augmented the migrationand invasive potential of lung cancer cells in a NOX4-dependent manner. Mice with Mycobacterium bovis bacillus Calmette-Guérin (BCG) pleural infection exhibited increased expression of NOX4 a

Accelerating the secondary immune response by inactivating CD4(+)CD25(+) T regulatory cells prior to BCG vaccination does not enhance protection against tuberculosis.

CD4(+)CD25(+) natural T regulatory cells (Tregs) have been shown to suppress protective immune responses in several different vaccination models. Since the effect of Tregs on vaccination against tuberculosis (Tb) was unknown, we used a murine model to investigate whether natural Tregs suppress the development of protective immunity following Mycobacterium bovis bacille Calmette-Guérin (BCG) vaccination. Using a monoclonal antibody against CD25, natural Tregs were inactivated prior to vaccination with BCG. The primary immune response was evaluated after BCG vaccination and the secondary immune

Influence of bovine lactoferrin on expression of presentation molecules on BCG-infected bone marrow derived macrophages.

The current vaccine for tuberculosis (TB), caused by Mycobacterium tuberculosis (MTB), is an attenuated strain of Mycobacterium bovis bacillus Calmette-Guerin (BCG). BCG has proven to be effective in children, however, efficacy wanes in adulthood. Lactoferrin, a natural protein with immunomodulatory properties, is a potential adjuvant candidate to enhance efficacy of BCG. These studies define bovine lactoferrin as an enhancer of the BCG vaccine, functioning in part by modulating macrophage ability to present antigen and stimulate T-cells. BCG-infected bone marrow derived macrophages (BMMs) cul

Disseminated bovine tuberculosis in a wild red fox (Vulpes vulpes) in southern Spain.

A wild-caught, adult red fox (Vulpes vulpes) from Doñana National Park (southern Spain), in very poor condition, died during anesthesia. At necropsy, the submandibular, retropharyngeal, mediastinal, axillary, mesenteric, and popliteal lymph nodes were enlarged, and the right submandibular and mesenteric lymph nodes had hard, white-colored concretions (calcifications). Multiple white pinpoint foci were observed in the lungs, and abscesses were present in the left dorsal lung lobes. No lesions were seen in tonsils, liver, or spleen. On histopathology multiple tuberculous granulomas, with Ziehl-N

Genetic and immunologic determinants of BCG disease: from mechanism to prevention.

BACKGROUND: Bacillus Calmette-Guérin (BCG) infection, a disseminated infection triggered in immunocompromised hosts by the administration of the attenuated BCG vaccine, is characterized by a pathogenic mechanism that involves the synergistic interaction between host immune deficiencies and strain-specific genetic mutations. METHODS: This review synthesizes published multi-omics and proteomics findings from the past decade, along with reported results from in vitro drug susceptibility assays and molecular diagnostics, to explore key pathogenic genes in BCG strains. We compare differences in pa

Optimization of a molecularly defined tuberculin formulation: recombinant fusion proteins and epitope surgery.

UNLABELLED: Bovine tuberculosis is an infectious livestock disease of global economic and zoonotic importance. Surveillance programs are largely dependent on the skin test that utilizes purified protein derivatives (PPDs) of tuberculin. PPDs suffer from a number of limitations regarding their production, characterization, and performance. Recently, we developed a molecularly defined tuberculin (MDT) that overcame these limitations. The MDT formulation comprises eight antigens, initially presented as seven recombinant proteins (ESAT6, CFP10, Rv3615c, Rv3020c, Rv1789, Rv3478, and Rv3810) and one

Mandibular Nodule Caused by Mycobacterium marinum with False Positive Interferon-γ Release Assay.

Mycobacterium marinum is a ubiquitous organism inhabiting both fresh and salt water. It can cause human diseases such as skin and soft tissue infection. The organism is also known to cause a false positive reaction to interferon-γ release assay, the test to diagnose latent tuberculosis infection. Here, we present a case of submandibular nodule caused by M. marinum with positive T-SPOT®.TB test, which was likely to be false positive.

[A case of spinal osteomyelitis due to Mycobacterium intracellulare with pulmonary Mycobacterium avium complex].

A 76-year-old woman was admitted to our hospital in October 2007 because of fever and anorexia. Chest computed tomography demonstrated consolidation and reticular shadows in the right middle lobe and left lingula. Pulmonary nontuberculous mycobacteriosis was diagnosed based on isolation of Mycobacterium intracellulare from her sputum. Her symptoms, which included fever, were improved by chemotherapy with clarithromycin and ethambutol, but the treatment was halted due to adverse effects. Subsequently, she complained of lumbago from April 2008, and spinal osteomyelitis from the T-4 through T-5 v

Identification of proteins from tuberculin purified protein derivative (PPD) by LC-MS/MS.

The tuberculin purified protein derivative (PPD) is a widely used diagnostic antigen for tuberculosis, however it is poorly defined. Most mycobacterial proteins are extensively denatured by the procedure employed in its preparation, which explains previous difficulties in identifying constituents from PPD to characterize their behaviour in B- and T-cell reactions. We here described a proteomics-based characterization of PPD from several different sources by LC-MS/MS, which combines the solute separation power of HPLC, with the detection power of a mass spectrometer. The technique is able to id
