Decoding the RNA Shield: HNRNPC and YTHDF2 Act as Guardians of Neurons in Parkinson's Disease.

Parkinson’s disease (PD) is a progressive neurodegenerative disorder characterized by the selective loss of dopaminergic neurons in the substantia nigra, resulting in hallmark motor symptoms such as tremor, bradykinesia, rigidity, and postural instability. Beyond these classical features, emerging evidence highlights the critical role of RNA modifications, particularly N6-methyladenosine (m6A), in regulating neuronal health and disease. The m6A modification is dynamically controlled by writers (METTL3, METTL14, WTAP), erasers (FTO, ALKBH5), and readers (e.g., YTHDF2, HNRNPC), which together in

Downregulation of CLU mediates CART-induced apoptosis and suppressed proliferation in bovine granulosa cells†.

This study aimed to elucidate the molecular mechanisms by which cocaine- and amphetamine-regulated transcript peptide (CART) regulates granulosa cell (GC) fate in Bos taurus follicles. Bovine GCs were used as an in vitro model, and tandem mass tag-based quantitative proteomics was performed to identify differentially expressed proteins (DEPs) following CART treatment. CART significantly downregulated clusterin (CLU), the most altered DEP (P < 0.05). Functional studies demonstrated that CLU knockdown, mimicking CART action, significantly promoted GC apoptosis, as evidenced by increased Annexin

PRRT2 as an auxiliary regulator of Nav channel slow inactivation.

During sustained activity, voltage-gated sodium (Nav) channels enter a slow-inactivated state to limit cellular hyperexcitability. Disruption of this regulatory process has been implicated in skeletal, cardiac, and neurological disorders. While the kinetics of this process are well characterized, its endogenous modulators remain unclear. Here, we identify Proline-Rich Transmembrane Protein 2 (PRRT2) as a native regulator of Nav channel slow inactivation. We show that PRRT2 facilitates the entry of Nav channels into the slow-inactivated state and delays their recovery, a regulatory effect conse

Autoimmune nodopathy associated with contactin-2 antibodies manifesting as Guillain-Barré syndrome: a case report.

BACKGROUND: The autoimmune nodopathy affecting the node of Ranvier was formerly classified within the spectrum of chronic inflammatory demyelinating polyradiculoneuropathy. However, as a result of comprehensive pathological and immunological investigations conducted in recent years, it has increasingly been recognized as a distinct clinical entity. To date, there have been no reported cases linking autoimmune nodopathy at the node of Ranvier with anti-CNTN2 antibodies. This paper presents a case study demonstrating such an association, detailing the clinical and electrophysiological features,

CV2/CRMP5 antibodies are specifically associated with parenchymal neurosyphilis: a retrospective cohort study.

BACKGROUND: Neurosyphilis can trigger neuro-specific immune responses, but evidence remains scarce. This study investigates the prevalence and clinical significance of neural antibodies in neurosyphilis, with a focus on CV2/CRMP5 antibodies. METHODS: We retrospectively analyzed 1149 patients with syphilitic neurological symptoms. Of 247 diagnosed with neurosyphilis, 215 met inclusion criteria. Using CBA and line blot, 97 patients suspected of autoimmune encephalitis were tested for neural antibodies, with positives confirmed by in-house CBA. Complete medical records were collected. Myristoylation of TMEM106B by NMT1/2 regulates TMEM106B trafficking and turnover.

TMEM106B, a type II transmembrane protein localized on the lysosomal membrane, has been identified as a central player in neurodegeneration and brain aging during the past decade. TMEM106B variants that increase TMEM106B expression levels are linked to several neurodegenerative diseases, including frontotemporal lobar degeneration (FTLD). Additionally, the C-terminal lumenal fragment of TMEM106B was recently shown to form amyloid fibrils during aging and neurodegeneration. However, the mechanisms regulating TMEM106B levels are not well understood. Here we show that TMEM106B is myristoylated by

The role of endolysosomal progranulin and TMEM106B in neurodegenerative diseases.

Although different neurodegenerative diseases are defined by distinct pathological proteins, they share many common features including protein aggregation. Despite this commonality, most current therapeutic approaches in the field, such as anti-aggregate antibodies, are focused on individual diseases or single neuropathologies with only limited success. The endolysosomal proteins progranulin and TMEM106B were both initially associated with frontotemporal lobar degeneration but have subsequently also been linked to other neurodegenerative diseases. Thus, these proteins are predicted to particip

A potential drug target in epilepsy and its role in other neurological disorders.

Epilepsy (EP) has an increased incidence due to several diseases, including neurodegenerative diseases and hemorrhagic stroke. The proteome-based research on developing suitable therapeutic interventions and disease prevention is beneficial. Cis-acting protein quantitative trait loci from 885 plasma and CSF proteins were used to explore potential drug targets for EP. After we got the preliminary association of TMEM106B with EP by drug target Mendelian randomization (MR), co-localization results identified TMEM106B as a potential drug target for EP further. The protein-protein interaction netwo

A Systematic Review and Meta-Analysis of Treatment Outcomes Following Tongue Reconstruction With Neurotized Free Flaps.

BACKGROUND AND OBJECTIVES: The tongue is the most common site of oral malignancy, and surgical treatment may result in impaired feeding, swallowing, and speech. Free tissue transfer is the preferred method for reconstructing complex defects. Evidence remains unclear on whether sensory reinnervation improves function. The objective of this review is to evaluate the outcome of patients undergoing innervated versus noninnervated free flap reconstruction following tongue cancer surgery. METHODS: A systematic review was performed to determine the role of sensory neurotization in lingual reconstruc

Neuroanatomical distribution of endogenous huntingtin and its immunohistochemical relationships with STB/HAP1 in the adult mouse brain and spinal cord.

Huntingtin-associated protein 1 (HAP1) is an essential constituent of the stigmoid body (STB) and is known as a neuroprotective interactor with causal agents for several neurodegenerative disorders, including huntingtin (HTT) in Huntington's disease. Previous in vitro studies showed that compared to normal HTT, STB/HAP1 exhibited a higher binding affinity for mutant HTT. The detailed in vivo relationships of STB/HAP1 with endogenous HTT, however, have not been clarified yet. This study examined the distribution of endogenous HTT and its relationships with STB/HAP1 in the adult mouse brain and

Expression of GDAP1 Gene Correlates With Alcohol Deprivation Effect.

Alcohol addiction is a widespread disease, and the exact causes and consequences are still not fully determined. Neurotransmitters and neuronal circuits are not only the target structure of alcohol and responsible for its direct effects but also play a central role in the development of addiction. A gene that has been linked to alcohol use disorder in recent studies is the ganglioside-induced differentiation-associated protein 1 (GDAP1) gene. The present study focuses on the hippocampus, a brain region particularly vulnerable to alcohol and rich in Gdap1 gene expression. Using an established d

Placental RTN3L-dependent ER-Phagy Contributes to Fetal Testicular Dysplasia Upon Environmental Stress.

Prenatal environmental stress damages fetal testicular development, leading to male infertility. However, the precise mechanisms underlying the impact of gestational environmental stress on fetal testicular development require further investigation. This study demonstrates that gestational environmental stressor cadmium exposure caused placental estradiol synthesis inhibition and fetal testicular dysplasia. Gestational estradiol supplementation restores fetal testicular dysplasia caused by environmental stress-induced placental estradiol synthesis inhibition. Analysis of human placentae and ca

A triple A syndrome with neurological findings; c464G>A mutation in the AAAS gene.

BACKGROUND AND PURPOSE: Allgrove syndrome or triple A (3A) syndrome is a multisystem disorder classically defined as the triad of esophageal achalasia, alacrimia and adrenal insufficiency due to adrenocorticotropic hormone insensitivity. Approximately one third of patients experience neurological dysfunction, including peripheral and autonomic nervous system dysfunction, leading some authors to use the term 4A syndrome (achalasia, alacrimia, adrenal insufficiency and autonomic abnormalities). Since its first description in 1978, knowledge of its clinical and genetic features has increased; how

Dual diagnosis at the neuro-immune interface: a case report of neuronal intranuclear inclusion disease with acute anti-CASPR2 encephalitis.

Neuronal intranuclear inclusion disease (NIID) is a rare autosomal-dominant, progressive neurodegenerative condition characterized by complex and variable clinical manifestations that can affect multiple neurological domains. This report describes the case of a 49-year-old female patient with a 10-year history of headaches, whose older sister had been diagnosed with NIID 1 year earlier through genetic testing and a skin biopsy. Recently, the patient developed dizziness and vomiting. Although symptomatic treatment reduced incidents of vomiting, her dizziness progressively worsened. It was accom

Immunohistochemical relationships of huntingtin-associated protein 1 with choline acetyltransferase in the forebrain cholinergic nuclei of adult mice.

Huntingtin-associated protein 1 (HAP1) is a core component of the stigmoid body (STB) and a neuroprotective interactor with the causative agents of several neurodegenerative diseases (NDs). The cholinergic system is often affected by NDs. Our previous studies suggest that cholinergic brainstem/spinal cord motoneurons are more vulnerable to neurodegeneration due to a lack of STB/HAP1 protectivity. The forebrain cholinergic nuclei are also major neurodegenerative/psychotic targets; however, the relationships of HAP1 with choline acetyltransferase (ChAT) have yet to be determined there. This stud

De novo GRIN variants in M3 helix associated with neurological disorders control channel gating of NMDA receptor.

N-methyl-D-aspartate receptors (NMDARs) are members of the glutamate receptor family and participate in excitatory postsynaptic transmission throughout the central nervous system. Genetic variants in GRIN genes encoding NMDAR subunits are associated with a spectrum of neurological disorders. The M3 transmembrane helices of the NMDAR couple directly to the agonist-binding domains and form a helical bundle crossing in the closed receptors that occludes the pore. The M3 functions as a transduction element whose conformational change couples ligand binding to opening of an ion conducting pore. In
