Immunological aspects of B-cell derived tumors in humans and rodents.

Translocation of the c-myc protooncogene to an immunoglobulin locus is a rate-limiting step in the genesis of three B-cell derived tumors: Burkitt lymphoma (BL) in humans, mouse plasmocytoma (MPC) and rat immunocytoma (RIC). Its consequences have been best analysed in BL. They involve a non-immunological and an immunological component. The former acts by preventing the B-cell from leaving the cycling compartment and entering the resting stage when programmed to do so. The latter acts by the down-regulation of certain HLA class I polymorphic specificities, leukocyte adhesion molecules and Epste

Establishment of a peripheral T-cell lymphoma cell line showing amplification of the c-myc oncogene.

A new T-cell lymphoma cell line, designated T34, was established from freshly isolated lymph node tumor cells of a patient with non-Hodgkin's diffuse large cell lymphoma. The T34 cells, as well as the parental lymphoma cells, showed mature helper/inducer immunophenotypes in that they formed spontaneous sheep erythrocyte rosettes and reacted with OKT-3 and OKT-4 monoclonal antibodies. They were negative for OKT-6, OKT-8, terminal deoxynucleotidyl transferase, WT-1, and HLA-DR antigens. Molecular analysis revealed that the T34 cells contained 8- to 16-fold amplified c-myc DNA. The same genetic c

In situ c-myc expression and genomic status of the c-myc locus in infiltrating ductal carcinomas of the breast.

We have studied the expression of the c-myc protooncogene and the cycle-dependent histone 4 gene at the cellular level by RNA:RNA in situ hybridization in 18 primary breast ductal adenocarcinomas. These tumors have previously been examined by Southern and Northern blot analysis for the genomic status of c-myc and its expression, respectively (Escot et al., Proc. Natl. Acad. Sci. USA, 83: 4834-4838, 1986). Positive c-myc hybridization signals were associated with carcinoma cells in all cases, including tumors which had no apparent alterations of the c-myc locus. Steady-state levels of c-myc mRN

Expression of the c-raf protooncogene in human hematopoietic cells and cell lines.

The murine sarcoma virus 3611 contains the transforming v-raf gene that has partial nucleotide homology with the src family of tyrosine kinase-encoding oncogenes. Although this virus induces fibrosarcomas in mice, a recombinant murine retrovirus carrying both the raf and myc oncogenes induces immunoblastic lymphomas and immortalizes mouse macrophages in vitro. The present study has thus monitored the expression of c-raf in human hematopoietic cells. The results demonstrate the presence of a 3.6-kb c-raf transcript in HL-60 promyelocytic leukemic cells. The induction of HL-60 cell differentiati

Bruce suppresses autophagy-regulated caspase activity and wing tissue growth in Drosophila.

Caspases are cysteine-aspartic proteases that mediate both lethal and non-lethal cellular outcomes, including the promotion of tissue growth. However, the mechanisms underlying the differential regulation of these activities remain unclear. We have previously shown that among the two Drosophila executioner caspases, Dcp-1 and Drice, Dcp-1 promotes tissue growth in a non-lethal manner, independent of canonical apoptotic signaling. Herein, we demonstrated that overexpressed Dcp-1, but not Drice, was activated without canonical apoptosome components. TurboID-based proximity labeling revealed dist

KRAS mutation-driven O-GlcNAcylation of CLDN18.2 enhances the progression of pancreatic cancer and reduces the efficacy of CLDN18.2-targeted therapy.

BACKGROUND: CLDN18.2 has emerged as a promising therapeutic target in gastric and gastro-oesophageal junction cancers. However, its clinical efficacy in pancreatic ductal adenocarcinoma (PDAC) has been modest, suggesting the presence of regulatory mechanisms impairing its efficacy. OBJECTIVE: We aim to investigate how O-linked N-acetylglucosaminylation (O-GlcNAcylation) affects CLDN18.2 subcellular localisation, tumour progression and therapeutic resistance in PDAC, while exploring strategies to restore treatment sensitivity. DESIGN: This study used samples from patients with PDAC, along wit

Multi-omics and in silico evidence reveal perospirone-induced developmental neurotoxicity in zebrafish via AKT/FOXO3a-mediated apoptosis.

Perospirone, an atypical antipsychotic increasingly detected in aquatic environments, raises concerns regarding potential developmental hazards in early vertebrates. As an emerging personal care product (PPCP) contaminant, its developmental neurotoxicity remains poorly characterized. To evaluate its biological effects, zebrafish embryos were exposed to perospirone (nominal 0.1, 1, 10 mg/L) from 5 to 120 hpf. At 5 dpf, larvae exposed to 1 mg/L displayed increased locomotion with anxiety-like features, whereas exposure to 10 mg/L was associated with reduced body length and hyperactivity suppress

IL7R depletion mitigates neuroinflammation and ischemic stroke via AKT dephosphorylation dependent LCP1 suppression in mice.

Stroke remains a leading cause of death and long-term disability worldwide. After reperfusion following ischemic stroke, peripheral immune cells infiltrate the injured brain hemisphere and exacerbate secondary damage. Among these cells, monocytes are among the predominant population. In this study, we performed single-cell RNA sequencing of immune cells isolated from the brain 72 h after infarction and characterized the heterogeneity of brain infiltrating monocyte-derived macrophages. We identified an IL7R+ macrophage subset with high fatty-acid metabolism scores 3 days after ischemic stroke i

Molecular Profiling and Survival Outcomes in Pancreatic Ductal Adenocarcinoma: A Multicenter Real-World Study from Turkey.

Background: Pancreatic ductal adenocarcinoma (PDAC) is a highly lethal malignancy. Next-generation sequencing (NGS) enables molecular characterization and may identify clinically actionable alterations; however, real-world multicenter data linking genomic subgroups to survival outcomes remain limited. We aimed to characterize the molecular landscape of NGS-tested PDAC in a Turkish multicenter cohort and evaluate the association of key molecular alterations, including KRAS status and KRAS variant subgroups, with survival outcomes. Methods: We conducted a multicenter retrospective cohort study i

Emerging landscape of KRAS inhibitors in cancer treatment.

Alterations in KRAS, NRAS, and HRAS occur in roughly 20% of patients with cancer, making RAS one of the most intensively studied oncogenic targets. The discovery of mutant-selective KRASG12C inhibitors has provided a proof-of-concept for RAS-directed therapies, heralding a new era in the treatment of RAS-driven cancers. Yet, the efficacy of first-generation KRASG12C inhibitors is limited by the rapid emergence of resistance. Novel classes of (K)RAS inhibitors with distinct mechanisms of action and broader target coverage hold promise to overcome resistance and extend the benefits of RAS-target

Kras G12C- and G12D-driven lung cancers differ in oncogenic potency, immunogenicity, and relapse after Kras inhibition in mouse models.

The development of allele-specific KRAS inhibitors underscores the importance of understanding the distinct tumor biology associated with common KRAS mutations, G12D and G12C, in genetically engineered mouse models (GEMMs) of non-small cell lung cancer (NSCLC) and patient samples. Lung tumors driven by the most common KRAS mutation, G12C, show delayed onset and slower progression compared with those driven by KRAS G12D in patients and mice. G12C tumors display lower proliferation and increased immune cell engagement, the latter of which is consistent with observations in patient tumors. Allele

Impaired nitrogenous waste clearance promotes hepatocellular carcinoma.

In mammals, hepatic urea cycle enzymes (UCEs) convert ammonia, the toxic nitrogenous waste, into urea for excretion. In hepatocellular carcinoma (HCC), UCE expression is often heterogeneously repressed, but its role in tumorigenesis is unclear. We show that, as in patients, UCE expression is markedly reduced in multiple HCC mouse models, including those driven by oncogenic c-MET/β-catenin, leading to impaired ammonia clearance, altered amino acid metabolism, and increased pyrimidine synthesis. In contrast, UCE expression is largely preserved in c-MET/sgAxin1 tumors, allowing assessment of the

Precision management of medullary thyroid carcinoma: a dynamic framework integrating biomarkers, genotyping, and risk stratification.

Medullary thyroid carcinoma (MTC) is a heterogeneous neuroendocrine malignancy in which outcomes are shaped by tumor burden, locoregional spread, and molecular context. Precision management therefore requires explicit separation of hereditary MTC driven by germline RET variants from presumed sporadic disease, and a structured integration of serum biomarkers, imaging, pathology, and genotype. This review synthesizes actionable evidence on calcitonin (Ctn) and carcinoembryonic antigen (CEA) baseline values and kinetics, universal germline RET testing, and tumor somatic profiling in advanced or p

Nuclear transcriptional condensates as drivers and therapeutic targets in NPM1-mutated AML.

NPM 1-mutated acute myeloid leukemia (AML) is driven by aberrant HOX/MEIS1 expression, whose mechanistic basis remained unresolved for years. Recent paradigm-shifting studies show that mutant NPM1 organizes phase-separated nuclear condensates that concentrate transcriptional regulators at active chromatin, directly sustaining the pathogenic HOX/MEIS1 transcriptional program. This framework explains the activity of menin-KMT2A inhibitors, recently approved by the US Food and Drug Administration, in this AML subtype and positions disruption of these assemblies as a precision strategy to eliminat

Molecular Mechanisms of Diffuse Large B-Cell Lymphoma and the Complexities of Tumor Development.

Diffuse large B-cell lymphoma (DLBCL), the most prevalent aggressive non-Hodgkin lymphoma, exhibits substantial molecular heterogeneity and arises from germinal center or post-germinal center B cells through recurrent genetic and epigenetic dysregulation of critical signaling pathways. Aberrations involving BCL6, MYC, MYD88, and components of B cell receptor and nuclear factor kappa-light-chain-enhancer of activated B cells (NF-κB) signaling play central roles in lymphomagenesis, treatment resistance, and clinical outcomes. Gene expression profiling has defined major molecular subgroups in DLB

Glucose Metabolic Reprogramming in Thyroid Cancer: Underlying Molecular Mechanisms and Links to Progression and Treatment.

Thyroid cancer (TC) is a common endocrine malignancy with rapidly increasing global incidence. Clinical management remains challenging for advanced and radioiodine-refractory thyroid cancer (RAIR-TC). Metabolic reprogramming, especially enhanced aerobic glycolysis (the Warburg effect), is a core hallmark of TC that drives tumor progression and therapeutic resistance. This review systematically summarizes the molecular mechanisms of glucose metabolic reprogramming in TC, focusing on key oncogenic drivers including BRAF V600E mutation, RAS mutation, and RET/PTC rearrangement, as well as their do

First published report of the FLCN c.1222 C > T (p.Gln408Ter) variant in a Chinese family with Birt-Hogg-Dubé syndrome and literature review.

BACKGROUND: Birt–Hogg–Dubé syndrome (BHDS) is a rare inherited disorder defined by skin lesions, pulmonary cysts, spontaneous pneumothorax, and renal neoplasia. Mutations in the FLCN gene are known causes, and identifying specific variants in different populations is essential for elucidating genotype-phenotype correlations. METHODS: We investigated a Chinese family with suspected BHDS. The proband was admitted to the Affiliated Cangnan Hospital of Wenzhou Medical University in October 2023. Comprehensive clinical evaluations and imaging studies were performed. Peripheral blood samples were co

NPM1-rearranged AML: clinical features, molecular pathogenesis, and therapeutic perspectives.

Nucleophosmin 1 (NPM1) gene aberrations are among the most common genetic alterations in acute myeloid leukemia (AML). NPM1 mutations (NPM1c), detected in 30-40% of adult AML cases, are well-characterized and associated with distinct clinical and molecular features. In contrast, NPM1 rearrangements (NPM1-r), involving rare fusion partners such as MLF1, CCDC28A, HAUS1, and RARA, are much less common and have historically been poorly understood. However, recent studies have begun to shed light on the molecular mechanisms and clinical characteristics of NPM1-r AML, revealing overlapping features

Reversible RNA ADP-ribosylation on uracil bases.

ADP-ribosylation is a conserved modification that uses NAD+ as a co-substrate to regulate essential cellular processes, such as genome stability and transcription, with Poly(ADP-ribose) Polymerases (PARPs) serving as the major catalyzing enzymes in humans. Historically defined as a protein post-translational modification, ADP-ribosylation on nucleic acids has been increasingly recognized in recent years, particularly in bacterial systems, but remains poorly understood in higher organisms. Here, we identify human PARP10 as a candidate enzyme that ADP-ribosylates nucleic acid bases, showing appa

Accelerated Sampling of Protein Dynamics Using BioEmu-Augmented Molecular Simulation.

Generative models, such as BioEmu, can produce diverse conformational ensembles of proteins; however, determining which predicted states are physiologically relevant and how they are populated remains a major challenge. In particular, it is unclear which conformations correspond to functional states and whether intermediate states and transitions between them can be reliably identified. Here, we investigate these questions using the serine-threonine kinases BRAF and CDK2. We examine how the disease-associated BRAF mutation V600E alters the populations of metastable conformational states relati
