Root exudates drive metabolic remodeling of microbial and resistance to facilitate sulfonamide biodegradation in karst soil-carbonate rock interface.

Sulfonamide antibiotics have become pervasive contaminants in karst systems, where low carbon and rapid infiltration hinder natural attenuation, leading to persistent groundwater pollution and elevated antibiotic resistance risks. Although root exudates shape soil microbial metabolism and resistance, their role at the karst soil-carbonate rock interface remains unclear. This study demonstrates that artificial root exudates (ARE) significantly boost sulfamethazine (SMZ) removal at the soil-carbonate rock interface, achieving up to 92.0 % (35.1 % higher than control). Analyses of 16S rRNA gene s

Exploring the composition of placental microbiome and its potential origin in preterm birth.

INTRODUCTION: For years, the placenta was believed to be sterile, but recent studies reveal it hosts a unique microbiome. Despite these findings, significant questions remain about the origins of the placental microbiome and its effects on pregnancy and fetal health. Some studies suggest it may originate from the vaginal tract, while others indicate that oral bacteria can enter the maternal bloodstream and seed the placenta. However, research analyzing the vaginal, oral, and placental microbiomes within the same cohort is lacking. Additionally, it's unclear whether the placental microbiome dif

Testing the Luedemann hypothesis: the discovery of novel antimicrobials from slow-growing microbes from nutrient-limited environments.

George Luedemann is known throughout the antimicrobial community as one of the discoverers of the natural product antibiotic gentamicin. He subsequently hypothesized that slow-growing organisms inhabiting inhospitable, nutrient-limited environments may represent an enriched source of previously undescribed microbes that produce novel antimicrobials to create a competitive advantage over faster-growing rival organisms. Accordingly, 750 slow-growing microorganisms were isolated from desert rock surfaces and archived prior to Dr. Luedemann's passing in 2000. Here, we describe the characterization

Bacterial blood microbiome of Mastomys rodents: implications for disease spill-over at the animal-human interface within the Bushbuckridge-East community, South Africa.

The Bushbuckridge-East community in Mpumalanga Province, South Africa is bordered by nature reserves, including the Manyeleti Game Reserve. Murid rodents are prevalent in both Manyeleti and communal rangelands adjoining the community households. Although rodents are reservoir hosts for a broad range of viral, bacterial and parasitic pathogens, the rodent microbial diversity and transmission of zoonotic agents to humans in the community is understudied. In this study we investigated bacterial diversity in wild and commensal rodents sampled from different habitats. The 16S rRNA gene was amplifie

Development of a multiplex PCR assay for detection of Riemerella anatipestifer serotype 1 and serotype 2 strains.

Riemerella anatipestifer infection is one of the main infectious diseases that threaten the poultry industry at present. Serotypes 1 and 2 are the main serotypes causing global outbreaks of R. anatipestifer. In this study, we designed the primers specific for R. anatipestifer serotypes 1 and serotype 2 strains, and for R. anatipestifer 16S rRNA (for the species identification) to establish a multiplex PCR assay for rapid detection of the serotype 1 and serotype 2 strains. The specificity test showed that a 505 bp fragment was amplified from serotype 1 strains, and a 1125 bp fragment was amplif

Water chlorination increases the relative abundance of an antibiotic resistance marker in developing sourdough starters.

UNLABELLED: Multiple factors explain the proper development of sourdough starters. Although the role of raw ingredients and geography, among other things, have been widely studied recently, the possible effect of air quality and water chlorination on the overall bacterial communities associated with sourdough remains to be explored. Here, using 16S rRNA amplicon sequencing, we show that clean, filtered-air severely limited the presence of lactic acid bacteria in sourdough starters, suggesting that surrounding air is an important source of microorganisms necessary for the development of sourdou

DNA metabarcoding analyses reveal fine-scale microbiome structures on Western Canadian bat wings.

UNLABELLED: Healthy wings are vital for the survival and reproduction of bats, and wing microbiome is a key component of bat wing health. However, relatively little is known about the wing microbiome of bats in western Canada where the white nose syndrome has become an increasing threat. Here, we used DNA metabarcoding to investigate the bacterial and fungal communities on the wings of three bat species: the big brown bat (Eptesicus fuscus), the Yuma myotis (Myotis yumanensis), and the little brown myotis (M. lucifugus) from four field sites in Lillooet, British Columbia, Canada. The bacterial

Upper respiratory tract detection of Mycoplasma ovipneumoniae employing nasopharyngeal swabs.

BACKGROUND: Flock-level prevalence and characterization of Mycoplasma ovipneumoniae is determined almost exclusively using nasal swabbing followed by molecular detection with either quantitative PCR or multi-locus sequence typing. However, the diagnostic performance and efficiency of swabbing the nasal passage compared to other anatomical locations has not been determined within sheep populations. The goal of this research was to assess the diagnostic capability of nasopharyngeal swabs in comparison to nasal swabs for the detection of Mycoplasma ovipneumoniae. RESULTS: Nasal and nasopharyngea

Comparison of the bacterial microbiome in the pharynx and nasal cavity of persistent, intermittent carriers and non-carriers of Staphylococcus aureus.

Introduction. Staphylococcus aureus is a bacterium that colonizes various human sites. The pharynx has been considered as a site of little clinical relevance and little studied. Recently, it has been reported that S. aureus can colonize more the pharynx than the nose. In addition, S. aureus can persist in these sites for prolonged periods of time.Hypothesis. The composition of the pharyngeal and nasal microbiome will differ between persistent, intermittent carriers and non-carriers of S. aureus.Aim. Determine whether the pharyngeal and nasal microbiome is different between carriers and non-car

The impact of anti-tuberculosis treatment on respiratory tract microbiome in pulmonary tuberculosis.

The growing evidence has underscored the significance of interactions between the host and microbiota in respiratory health, presenting a novel perspective on disease management. Yet, comprehension of the respiratory microbiome shifts before and after anti-tuberculosis treatment is limited. This study compares respiratory microbiome profiles in untreated tuberculosis (UTB) and completed TB treatment (CTB) cases with healthy controls, using 16S rRNA sequencing on sputum samples. Significant reduction in sputum microbial alpha diversity was observed in both TB groups when compared to healthy con

Janus kinase 1 in Megalobrama amblycephala: Identification, phylogenetic analysis and expression profiling after Aeromonas hydrophila infection.

Janus kinase 1 (JAK1), a member of the JAK family, plays an essential and non-redundant role in the mammalian immune system. However, the potential role of JAK1 in fish immune response remains largely unclear. In the present study, the JAK1 gene of Megalobrama amblycephala (MamJAK1) was identified and characterized. The open reading frame (ORF) of MamJAK1 was 3462 bp, encoding 1153 amino acids. MamJAK1 consists of four common domains of the JAK family, including B41, SH2, STyrKc (a pseudo kinase domain), and TyrKc (a kinase domain). Phylogenetic analysis showed that JAK1s are divided into two

Loss of tRNA methyltransferase 9 and DNA damage response genes in yeast confers sensitivity to aminoglycosides.

tRNA methyltransferase 9 (Trm9)-catalysed tRNA modifications have been shown to translationally enhance the DNA damage response (DDR). Here, we show that Saccharomyces cerevisiae trm9Δ, distinct DNA repair and spindle assembly checkpoint (SAC) mutants are differentially sensitive to the aminoglycosides tobramycin, gentamicin and amikacin, indicating DDR and SAC activation might rely on translation fidelity, under aminoglycoside stress. Further, we report that the DNA damage induced by aminoglycosides in the base excision repair mutants ogg1Δ and apn1Δ is mediated by reactive oxygen species, wh

Isolation and screening of antimicrobial biosurfactants obtained from mangrove plant root-associated bacteria.

The unique properties of biosurfactants obtained from microbes, including their activity at extreme temperatures, make them more attractive than synthetic alternatives. Henceforth, the principle objective is to isolate and detect the antibacterial and antifungal activities of the biosurfactants produced from bacteria of the economically competitive mangrove ecosystem. Using the serial dilution method, 53 bacterial strains were recovered from the Manakudy mangrove forest in Kanyakumari, India, for the investigation. Different biosurfactant screening methods and morphological and biochemical tes

Bacterial Amyloid Curli Associated Gut Epithelial Neuroendocrine Activation Predominantly Observed in Alzheimer's Disease Mice with Central Amyloid-β Pathology.

BACKGROUND: Substantial evidence from recent research suggests an influential and underappreciated force in Alzheimer's disease (AD) pathogenesis: the pathological signals originate from outside the brain. Pathogenic bacteria produce amyloid-like proteins "curli" that form biofilms and show functional similarities to human amyloid-β (Aβ). These proteins may contribute to neurological disease progression via signaling cascade from the gut to the brain. OBJECTIVE: We propose that curli causes neuroendocrine activation from the gut to brain that promotes central Aβ pathology. METHODS: PGP9.5 an
