To understand how adaptive immune responses are generated against bacteria that avoid being delivered to lysosomes, interactions between professional antigen-presenting cells (APCs) and the intracellular pathogen Legionella pneumophila were examined. In contrast to murine bone marrow-derived macrophages (BMMs), we show that dendritic cells (DCs) restrict the growth of intracellular Legionella. Similar to what has been reported in BMMs, phagosomes containing Legionella matured into endoplasmic reticulum (ER)-derived organelles after DC internalization. Biogenesis of an ER-derived vacuole did no

Here we present data showing oral immunogenicity of recombinant hepatitis B surface antigen (HBsAg) in preclinical animal trials. Mice fed transgenic HBsAg potato tubers showed a primary immune response (increases in HBsAg-specific serum antibody) that could be greatly boosted by intraperitoneal delivery of a single subimmunogenic dose of commercial HBsAg vaccine, indicating that plants expressing HBsAg in edible tissues may be a new means for oral hepatitis B immunization. However, attainment of such a goal will require higher HBsAg expression than was observed for the potatoes used in this s

Quantitative measurements of blood flow (BF) to skin and several other tissues were made using radioactive microspheres in conscious sheep. The sheep were from established flocks that had been selectively bred for greater (Fleece plus) or lesser (Fleece minus) wool production. The BF rate per unit area of wool-bearing skin was significantly greater in the Fleece plus (n = 9) than in the Fleece minus (n = 6) group, but the correlation between skin BF and the wool growth rate in individual animals was modest (r = .581). There was a strong, positive correlation (r = .813) between wool production

A 28-year-old woman developed leukopenia and slight cervical lymphadenopathy. Bone marrow aspiration and special stains established the diagnosis of acute monocytic leukemia. Following chemotherapy a complete hematologic remission was elicited. Seven months later, she consulted an ophthalmologist because of bilateral conjunctival lesions. Ophthalmologic examination showed subconjunctival, perilimbal grayish-pink infiltrates. A conjunctival biopsy disclosed sheets of mononuclear cells consistent with acute monocytic leukemia. Four months later, she developed cutaneous lesions in the face and ch

The sensing of molecular oxygen is an essential component of development and environmental response in both animals and plants. Few substrates of the plant oxygen-sensing PLANT CYSTEINE OXIDASE (PCO) N-degron pathway have been identified. Here we show that HYPOXIA TRANSDUCER1 (HYT1, previously named HYPOXIA-RESPONSIVE UNKNOWN PROTEIN, HUP40), an angiosperm-specific tail anchored-domain protein, is a substrate of this pathway through a Cys-2 dependent N-degron. HYT1 is stabilised in response to acute hypoxia in root tissues and is associated with the endoplasmic reticulum and nucleus. Genetic e

Lysosomal and endoplasmic reticulum (ER) Ca2+ release mutually influence each other's functions. Recent work revealed that ER-located ryanodine receptor(s) (RyR(s)) Ca2+ release channels suppress autophagosome turnover by the lysosomes. In familial Alzheimer's disease, inhibiting RyR hyperactivity restored autophagic flux by normalizing lysosomal vacuolar H+-ATPase (vATPase) levels. However, the mechanisms by which RyRs control lysosomal function and how this involves the vATPase remain unknown. Here, we show that RyRs interact with the ATP6v0a1 subunit of the vATPase, contributing to ER-lysos

Tumor-associated macrophages (TAMs), essential components of the tumor immune microenvironment (TIME), undergo metabolic reprogramming as part of functional adaptation. Tumor cells modulate TAMs through multiple mechanisms, including metabolic cross-feeding, cytokine production, extracellular vesicles, tumor-derived proteins (such as GRP78) and pathogen-associated patterns (such as Lipopolysaccharide) signaling mediators. In turn, metabolic alterations in TAMs fine-tune TAM function via intricate signaling networks with outcomes that vary across cancer types. These functional and phenotypic sh

In Parkinson's disease and other synucleinopathies, αSynuclein (αSyn) misfolds and forms Ser129-phosphorylated aggregates (pSyn129) with the factors controlling this process largely unknown. Here, we used arrayed CRISPR-mediated gene activation and ablation to discover new pSyn129 modulators. Using quadruple-guide RNAs (qgRNAs) and Cas9, or an inactive Cas9 fused to a synthetic transactivator, we ablated 2304 and activated 2428 human genes related to mitochondria, trafficking, and motility functions in HEK293 cells. After exposure of cells to αSyn fibrils, pSyn129 signals were recorded by high

Fibroblast growth factor receptor 1 (FGFR1) is a cell surface receptor tyrosine kinase implicated in cellular signaling and homeostasis. Several reports indicate that N-glycosylation of FGFR1 is critical for the FGFR1 trafficking to the cell surface, as glycosylation-deficient mutant of FGFR1 (FGFR1.GF) is trapped inside the cell, in the endoplasmic reticulum (ER), and in the nuclear envelope. Our recent mass spectrometry analyses revealed dehydrogenase/reductase 2 (DHRS2) as a putative binding partner of the intracellular FGFR1.GF. Here, we identified a peroxisomal targeting signal 1 (PTS1) a

BACKGROUND: T-2 toxin, a prevalent mycotoxin in feed, poses severe health risks to poultry. While its systemic toxicity is recognized, its neurotoxic effects in birds, and effective countermeasures, remain underexplored. Sodium butyrate (NaB), a green feed additive, has shown broad biological benefits, but its potential to alleviate T-2-induced neurotoxicity is unclear.
OBJECTIVE: This study aimed to investigate the neurotoxic mechanisms of T-2 toxin in quails and evaluate the protective role of sodium butyrate.
METHODS: Two-hundred-and-forty 10-day-old quails were randomly assigned to Contr

Invertebrates rely on post-translational modifications (PTMs) of immune effectors to regulate innate immunity, yet how hemocyanin fucosylation mediates intracellular antimicrobial signaling remains a critical knowledge gap in crustacean immunology. Although fucosylation enhances the in vitro antibacterial activity of Penaeus vannamei hemocyanin (PvHMC), its function in regulating intracellular stress-immune crosstalk remains unclear. Here, we identify a novel axis linking PvHMC fucosylation, endoplasmic reticulum stress (ERS), and antimicrobial peptide (AMP) production to bolster shrimp defens

CD320, also known as the transcobalamin receptor, is a key receptor that mediates the cellular uptake of circulating vitamin B12 bound to transcobalamin. In humans, CD320 abnormalities cause metabolic and neurological disorders. Previous biochemical studies indicate that the CD320 is a monomeric receptor. In this study, we conducted molecular and cellular experiments in human embryonic kidney 293 cells to examine biosynthesis and molecular forms of human CD320. We found that CD320 was present on the cell surface in an oligomeric form, which consisted of five homomers interconnected via disulfi

The tumor suppressor p53 coordinates cellular stress responses, but underlying mechanisms remain incompletely understood. We previously demonstrated that, in response to metabolic stress, C16-ceramide produced by ceramide synthase 6 (CerS6) directly binds to p53, thus preventing its MDM2-mediated degradation and promoting p53 activation. Here, we investigated the structural requirements and functional consequences of ceramide binding to p53. Using a panel of p53 mutants, including naturally occurring oncogenic variants, we characterized the ceramide-binding interface of p53 and the role of ami

Endoplasmic reticulum (ER) stress is triggered by several cellular perturbations causing protein misfolding, and activates the unfolded protein response (UPR), an initially adaptive signaling network that aims to restore ER and cellular homeostasis. Growing evidence indicates that UPR signaling extends beyond ER proteostasis, influencing mitochondrial function and bioenergetics through ER-mitochondria contact sites (ERMCs). The CHOP-ERO1A-IP3R axis has a primary role in recruiting mitochondria to adaptive UPR. However, its sustained activation renders UPR signaling maladaptive, leading to mito

Prostate cancer (PCa) is the second most common and a leading cause of cancer-related deaths among men. Current screening methods lack precision in distinguishing aggressive cases, emphasizing a need for tissue-based biomarkers. Although Golgi disorganization, ER stress, and elevated high-mannose (Man) glycoproteins (e.g., Integrin αv, key metastatic player) are recognized features of metastatic prostate tumors, their interrelationships remain unexplored. It is observed that the growth of primary prostate tumors is linked to an increase in endoplasmic reticulum (ER)-plasma membrane (PM) juncti

Rheumatoid arthritis (RA) is a chronic autoimmune disease characterized by synovium infiltration of immune cells, leading to inflammatory activation of fibroblast-like synoviocyte (FLS). Neutrophils infiltrate the synovium early in RA and exacerbate disease by releasing neutrophil extracellular traps (NETs). CCDC25 has been identified as a sensor for NETs-DNA, but its role in FLS activation remains to be investigated. Here, CCDC25 was found upregulated in RA-FLS, with reduced dimeric and increased monomeric forms relative to controls, and only monomeric CCDC25 bound to NETs-DNA. NETs-DNA bindi

Osteoarthritis (OA) is the most common age-related and degenerative joint disease. Proteostasis and protein quality control (autophagy, unfolded protein response, and the ubiquitin-proteasome system) are pivotal for cellular homeostasis and their impairment leads to protein misfolding and amyloid deposition in aged tissues. We here investigated amyloid deposition in OA. Amyloid fibrils were observed in chondrocytes in ex vivo cartilage samples. The underlying mechanisms were assessed in vitro: chondrocytes and cartilage organ cultures were treated with chloroquine and/or lipopolysaccharide for

Lysosomes have emerged as central hubs in the regulation of the endomembrane system, extending beyond degradation to coordinate organelle communication. Central to this regulatory role is vacuolar-type H+-ATPase (V-ATPase), a proton pump that acidifies the lysosomal lumen to enable hydrolase activity and support proteostasis. In addition to its lysosomal functions, V-ATPase influences the physiology of other organelles, including the endoplasmic reticulum (ER), Golgi apparatus and mitochondria, through both direct and indirect mechanisms involving acidification-dependent processes, such as pro

It is known that Hepatocellular carcinoma (HCC) accounts for the highest number of deaths due to primary liver cancer and is one of the leading global oncology cases. Currently, in spite of many other efforts to improve the treatment, the prognosis is rather poor with HCC patients, indicating the necessity of new biomarkers and therapeutic targets. In this study, we undertook a multi-omics approach including exome sequencing, GO analysis, PPI network analysis, bulk RNA-seq, single-cell RNA-seq, spatial transcriptomics, and Venn and survival analysis to search decisively for drivers of liver ca

Although environmental monitoring data for 4-phenylbutyric acid (4-PBA) remain limited, its potential co-occurrence with triphenyl phosphate (TPhP) in aquatic systems cannot be excluded, given its applications in agriculture and pharmaceuticals. The combined effects of these compounds on early neurodevelopment in organisms such as zebrafish remain largely unexplored. Here, zebrafish embryos were exposed from 2 h post-fertilisation (hpf) to TPhP, then co exposure to environmentally plausible 4-PBA (0.5 μM). Light-dark locomotion, high-light avoidance, and light-dark preference assays revealed t
