Evaluation of the efficacy of the combination of oral bexarotene and methotrexate for the treatment of early stage treatment-refractory cutaneous T-cell lymphoma.

BACKGROUND: Various combination therapies are used in refractory cutaneous T-cell lymphoma (CTCL). Although bexarotene has been studied in combination with psoralen photochemotherapy (PUVA), IFN-alpha and denileukin difitox, there have been no published data assessing the efficacy of the combination of bexarotene and methotrexate. OBJECTIVE: To evaluate the efficacy and safety of an oral bexarotene and methotrexate combination for the treatment of refractory CTCL. METHOD: A retrospective study was carried out of 12 patients with refractory stage CTCL treated with an oral combination of bexar

Mesenchymal Stem Cell Therapy for Systemic Sclerosis: Current Evidence and Clinical Applications.

Systemic sclerosis is a multisystem autoimmune disease characterized by immune dysfunction, vasculopathy, and progressive fibrosis, yet current therapies rarely address these pathogenic axes simultaneously. Mesenchymal stem cell therapy has gained increasing interest as a disease-modifying strategy because its therapeutic activity appears to depend largely on paracrine signaling rather than durable engraftment. Through these mechanisms, mesenchymal stem cells may suppress profibrotic fibroblast activation, recalibrate innate and adaptive immune responses, and enhance vascular repair. Cell-free

Hyaluronic acid-based reduction responsive nanoparticles for Improved anti-psoriasis effects of traditional Chinese herb monomer oleanolic acid via blocking YAP-AREG axis.

BACKGROUND: Oleanolic acid (OA), a pentacyclic triterpenoid abundantly present in various traditional Chinese herbs, exhibits promising anti-psoriatic potential owing to its broad pharmacological activities. Nevertheless, its clinical translation has been hindered by challenges including poor aqueous solubility, limited cutaneous permeation, and rapid systemic clearance, all of which compromise bioavailability when administered topically. To address these limitations, we designed and developed encapsulated and disulfide bonded OA-hyaluronic acid nanoprodrugs (OA-NPs@OA) for topical treatment o

Atypical Vesicular Skin Eruption Following a Low-Dose Methotrexate Dosing Error: A Case Report.

Methotrexate is widely used to manage various autoimmune conditions, including psoriasis and psoriatic arthritis. Although weekly low-dose regimens are generally well-tolerated, dosing errors, particularly daily instead of weekly administration, can result in acute methotrexate toxicity with mucocutaneous and hematologic complications. We present a case of clinically significant methotrexate toxicity manifesting with painful oral mucositis and vesicular cutaneous lesions on the right lateral abdomen and right supraclavicular region in a 68-year-old Caucasian female patient with psoriasis and p

Computed Tomographic Findings and Clinical Manifestations of Acute Severe Hypersensitivity to a Non-Ionic Iodinated Contrast Agent in a Castrated Male Dog: A Case Report.

Severe adverse reactions induced by non-ionic contrast media remain an uncommon complication in veterinary medicine. This report describes the clinical manifestations and sequential computed tomography alterations indicative of acute shock captured during 2 min and 4 min delayed phases in a 10-year-old, castrated male Maltese dog. The patient, previously diagnosed with a cutaneous mast cell tumor on the lip exhibiting incomplete histopathological margins, underwent CT for metastatic evaluation. The patient had no clinical evidence of systemic disease, and no anesthetic adverse events were obse

FRONT block: a cadaveric study of a dual-plane injection block targeting femoral rami and obturator nerve trunk for anterior hip joint analgesia.

BACKGROUND: The quest for a single nerve block to anesthetize all anterior hip articular nerve branches has long been a challenge for anesthetists, particularly in targeting the obturator nerve (ON). The FRONT block (Femoral Rami Obturator Nerve Trunk) is a newly developed dual-injection technique providing comprehensive anesthetic coverage of the anterior hip joint by targeting both the sensory femoral rami and the ON trunk. METHODS: 15 intact cadaveric sides from eight embalmed cadavers were dissected after FRONT block administration with one injection and two dye deposits in the iliopsoas

Relapse of minimal change nephrotic syndrome after initiation of sulfamethoxazole-trimethoprim combination therapy: a case report.

Minimal change nephrotic syndrome (MCNS) is characterized by podocyte injury leading to severe proteinuria, mainly mediated by T-cell activation and cytokine imbalance. Relapses are often triggered by immunological stimuli such as infections, vaccinations, or drugs; however, relapse following administration of sulfamethoxazole-trimethoprim (ST) combination therapy has not been reported previously. We report an extremely rare case of MCNS relapse triggered by ST combination therapy.A 55-year-old woman with a history of breast cancer treated with tamoxifen developed nephrotic syndrome and was di

From the laboratory to the skin: translational potential of nanofibers in the treatment of cutaneous leishmaniasis - an overview and systematic approach.

Cutaneous leishmaniasis (CL) remains a major global health challenge, particularly in low- and middle-income countries, due to the limitations of current therapies, including toxicity, high cost, invasive administration routes, and poor patient adherence. In this context, nanotechnology-based drug delivery systems, such as nanofibers, have emerged as promising alternatives to improve therapeutic outcomes. This study provides a comprehensive overview of CL, its epidemiology, clinical manifestations, and current pharmacological treatments, followed by a systematic and critical analysis of nanofi

Antimelanoma potential of natural compounds derived from plants: a systematic review of in vivo studies of B16 melanoma and its sublines.

Cutaneous melanoma, a lethal neoplasm originating from epidermal melanocytes, stands out for its resistance to conventional therapies and high metastatic rate. Given the urgent need for new therapeutic approaches, this study focuses on the antitumor potential of natural compounds derived from plants, recognized as primary sources of antineoplastic chemotherapeutics. In this systematic review, we selected studies that evaluated the efficacy of such compounds in vivo, using the murine melanoma B16 model. The research was registered at the International Platform of Registered Systematic Review an

NPD1/GPR37 signaling protects against painful traumatic brain injury and comorbidities by regulating demyelination, glial responses, and neuroinflammation in the mouse brain.

Traumatic brain injury (TBI) often leads to neuropathic pain and a range of comorbidities, including post-traumatic stress disorder (PTSD), cognitive decline and depression. Neuroprotectin D1 (NPD1), a lipid mediator derived from the omega-3 fatty acid docosahexaenoic acid (DHA), exhibits neuroprotective properties; however, the distinct roles of NPD1 and DHA in mitigating TBI-induced deficits remain unclear. In a mouse model of closed-head TBI, transient neuropathic pain lasting less than two weeks was observed, characterized by periorbital and cutaneous mechanical allodynia/hyperalgesia, mot

Validation of the Clinical Performance and Reproducibility of the Savanna HSV 1+2/VZV Assay.

Herpes simplex virus 1 (HSV-1), HSV 2 (HSV-2), and varicella-zoster virus (VZV) cause nondescript cutaneous and mucocutaneous lesions requiring rapid, differential identification for appropriate diagnosis and patient counseling. Decentralized multiplex molecular assays may provide more rapid results than existing methodologies but require clinical validation. This multicenter study evaluated the clinical performance of the Savanna HSV 1+2/VZV Assay against the high-complexity Lyra Direct HSV 1+2/VZV real-time PCR nucleic acid test for the detection of HSV-1, HSV-2, and VZV from clinical specim

Hair Thickness Growth Effect of Adenosine Complex in Male-/Female-Patterned Hair Loss via Inhibition of Androgen Receptor Signaling.

Aging (senescence) is an unavoidable biological process that results in visible manifestations in all cutaneous tissues, including scalp skin and hair follicles. Previously, we evaluated the molecular function of adenosine in promoting alopecia treatment in vitro. To elucidate the differences in the molecular mechanisms between minoxidil (MNX) and adenosine, gene expression changes in dermal papilla cells were examined. The androgen receptor (AR) pathway was identified as a candidate target of adenosine for hair growth, and the anti-androgenic activity of adenosine was examined in vitro. In ad

The alleviative effects of canagliflozin on imiquimod-induced mouse model of psoriasis-like inflammation.

Psoriasis is a life-long immune-mediated dermatosis with thickened, reddish, and flaky skin patches. Canagliflozin is a gliflozin antidiabetic with non-classical remarkable antioxidative, anti-inflammatory, anti-proliferative, and immune-modulating effects. The aim of this study is to examine the probable effects of topical canagliflozin on a mouse model of imiquimod-provoked psoriasis-like dermatitis. The study evaluated 20 Swiss white mice, sorted haphazardly into 4 groups of 5 animals each. Every mouse, with the exception of the control group, had imiquimod applied topically to their shaved

Pustular Eruption following COVID-19 Vaccination: A Narrative Case-Based Review.

From the beginning of public vaccinations until the relaxation of COVID-19 measures, many case reports, case series and case-control studies have been published indicating cutaneous side effects of COVID-19 vaccination. Post-vaccination pustular eruption was reported as well, with a challenging differential diagnosis between pustular psoriasis, AGEP (acute generalized exanthematous pustulosis) and neutrophil pustular eruptions. We report a case of 56-year-old woman presented with acute generalized pustular flare up culminated 5 days after the second dose of BNT162b2(Pfizer) vaccination. She wa

Characterization of trigeminal C-fiber reactivity through capsaicin-induced release of calcitonin gene-related peptide.

OBJECTIVE: We hypothesized that the response of trigeminal dermal blood flow (DBF) in the trigeminal system and consecutive expansion of flare response to capsaicin would differ from the somatosensory system (arm). We also investigated whether there are differences between patients with migraine and healthy controls (HC). BACKGROUND: Functional differences between the trigeminal and extracephalic somatosensory systems may partly explain the susceptibility for headaches in patients with migraine. Capsaicin-induced activation of nociceptive C-fibers in the skin is mainly mediated by calcitonin

Combinational photodynamic and photothermal - based therapies for melanoma in mouse models.

BACKGROUND: Melanoma is a highly metastatic skin cancer with limited response to current therapies in advanced patients. To overcome resistance, novel treatments based on photodynamic and photothermal therapies (PDT and PTT, respectively) have been developed to treat melanoma in preclinical murine models. Despite success inhibiting implanted tumors' growth, there has been limited evaluation of their long-term effectiveness in preventing metastasis, recurrence, or improving survival rates. METHODS: Combined and multidrug therapies based on PDT and/or PTT to treat cutaneous malignant melanoma i

Amphotericin B-loaded natural latex dressing for treating Candida albicans wound infections using Galleria mellonella model.

Amphotericin B (AmB) is the gold standard for antifungal drugs. However, AmB systemic administration is restricted because of its side effects. Here, we report AmB loaded in natural rubber latex (NRL), a sustained delivery system with low toxicity, which stimulates angiogenesis, cell adhesion and accelerates wound healing. Physicochemical characterizations showed that AmB did not bind chemically to the polymeric matrix. Electronic and topographical images showed small crystalline aggregates from AmB crystals on the polymer surface. About 56.6% of AmB was released by the NRL in 120 h. However,

Hard-to-heal peripheral wounds infected with Corynebacterium striatum: a prospective study.

OBJECTIVE: To investigate Corynebacterium striatum as a nosocomial pathogen infecting hard-to-heal peripheral wounds, such as skin wounds, soft tissue abscesses and osteomyelitis. As of 2023, the medical community were alerted against the risk of emerging systemic and central infections; on the other hand literature on peripheral cutaneous regions is still scarce. METHOD: In this study, two groups of patients with similar lesions which were infected were compared: one group with the presence of the coryneform rod, the other without. RESULTS: In total, Corynebacterium striatum was cultured fr
