Leptin-induced autophagy regulates the immunomodulatory potential of adipose-derived mesenchymal stem cells through down regulating the expression of TSG-6.

Adipose-derived mesenchymal stem cells (ADMSCs) possess modulatory functions in adipose tissue, which could help combat the development of obesity. It has been reported that the functions of ADMSCs are impaired by obese microenvironment. However, the mechanisms are not clearly understood. The study investigates the role of leptin on immunoinhibitory functions of ADMSCs in obesity, and primarily explored its possible mechanisms. ADMSCs were isolated and identified by morphological observation, flow cytometry and differentiation potential. The lipopolysaccharides (LPS)-stimulated macrophages wer

Alpha-ketoglutarate Potentiates IL-1β Production and Suppressive Mechanisms of Myeloid-Derived Suppressor Cells by Altering Redox Metabolism and Inducing Autophagy.

Alpha-ketoglutarate (αKG) has shown promise in cancer immunotherapy due to its profound impact on cancer cell metabolism and gene expression. However, its effects on myeloid-derived suppressor cells (MDSCs), critical immunosuppressive components of the tumour microenvironment, remain poorly understood. Using a model of GM-CSF/IL-6-induced monocyte-derived (mo)MDSCs we found that non-toxic doses of non-esterified αKG expanded CD14+HLA-DR-/low moMDSCs, unlike esterified dimethyl-αKG. The αKG-moMDSCs displayed an enhanced suppressive phenotype, upregulated ILT-4 and IL-10, and showed an increased

LAMP2A-dependent chaperone-mediated autophagy enhances oxidative stress resistance in gastric cancer cells through selective degradation of accumulated oxidized DJ-1.

Chaperone-mediated autophagy (CMA) promotes cancer cell survival by selectively removing oxidatively damaged proteins, yet its precise molecular mechanisms and role in redox adaptation remain incompletely understood. This study aimed to elucidate the function of CMA in regulating oxidative stress resistance in gastric cancer (GC) cells, focusing on the LAMP2A-DJ-1 regulatory axis. LAMP2A expression was assessed in GC tissues and cell lines via immunohistochemistry, qPCR, and western blot. Oxidative stress models were established using hydrogen peroxide (H₂O₂). Genetic manipulation of LAMP2A wa

Machine learning-based prediction model for drug target identification and MASH improvement: a comprehensive analysis of biochemical and ferroptosis/autophagy biomarkers.

Metabolic dysfunction-associated steatotic liver disease (MASLD), marked by excess fat in the liver, has become the most prevalent chronic liver disease worldwide, affecting over 30% of adults. Its advanced form, metabolic dysfunction associated steatohepatitis (MASH), includes liver ballooning, and inflammation, and can progress to cirrhosis and hepatocellular carcinoma (HCC). Despite the increasing burden, effective pharmacotherapies for MASLD/MASH are still lacking. Programmed cell death mechanisms, such as autophagy and ferroptosis, are critical in the pathology of MASLD, influencing liver

O-GlcNAcylation of YAP1 promotes lung transplant ischemia-reperfusion injury via binding to HIF1α transcription factor and activating autophagy and mitophagy.

Lung transplant ischemia-reperfusion injury poses a significant challenge in transplantation medicine, often causing severe complications and poor patient outcomes. Our study focused on the role of O-GlcNAcylation of Yes-associated protein 1 (YAP1) in exacerbating this injury by regulating autophagy and mitochondrial autophagy pathways. We found that hypoxia-reoxygenation robustly activated the Hippo-YAP1 signaling pathway, leading to increased damage in lung epithelial cells. Concurrently, autophagy and mitochondrial autophagy levels were significantly upregulated, indicating cellular stress

Anlotinib inhibits glioma cell proliferation, migration, and invasion by suppressing c-MET/AKT/mTOR signaling and altering autophagy-associated protein expression.

Glioma remains difficult to treat because malignant cells show rapid proliferation, diffuse invasion, and frequent recurrence. This study investigated the direct antitumor effects of anlotinib in human glioma cells and examined whether these effects were associated with hepatocyte growth factor receptor/protein kinase B/mechanistic target of rapamycin (c-MET/AKT/mTOR) signaling, apoptosis-associated responses, mesenchymal-transition-related proteins, and autophagy-associated proteins. U87 and U251 cells were treated with anlotinib in vitro. Cell viability was assessed using Cell Counting Kit-8

METTL3-regulated m6A modification modulates autophagy of placental mesenchymal stem/stromal cell by YTHDF3-mediated degradation of LncRNA GAS5 in preeclampsia.

BACKGROUND: Preeclampsia (PE) causes maternal and perinatal morbidity through defective placental vascularization. Autophagy enhances placental mesenchymal stem/stromal cells (PMSC) proangiogenic properties while autophagy is reduced in PE. Long non-coding RNA GAS5, elevated in PE placenta, suppresses endothelial angiogenesis. N6-methyladenosine (m6A) modification through METTL3 regulates RNA stability and autophagy. However, regulatory mechanisms governing PMSC autophagy in PE remain unclear. METHOD: Placenta specimens and PMSC from the normal and PE were compared. Molecular analyses include

ROS-Responsive Eye Drop for Controlled Release of 20-Deoxyingenol Improves Diabetic Corneal Healing by Activating Macrophage Autophagy and Metabolic Reprogramming.

Impaired corneal wound healing in diabetes remains a major clinical challenge, driven by defective autophagy, metabolic dysregulation, and sustained inflammation within a high-reactive oxygen species (ROS) microenvironment. Here, we engineered a ROS-responsive ocular delivery system based on N1-(4-boronobenzyl)-N3-(4-boronophenyl)-N1, N1, N3, N3-tetramethylpropane-1,3-diaminium (TSPBA)/poly(vinyl alcohol) (PVA) hydrogel microspheres (TP hydrogel microspheres) encapsulating 20-deoxyingenol (20-DOI), which enables sustained and ROS-triggered drug release. Under high-glucose conditions in vitro,

Helicobacter pylori-secreted outer membrane vesicles induce bone loss and accelerate osteoporosis pathogenesis in mice by regulating gut microbiota and autophagy.

The gut microbiota profoundly influences systemic health, with emerging evidence linking bacterial pathogens to bone metabolism disorders. Helicobacter pylori (H. pylori), a prevalent gastric pathogen, has been epidemiologically associated with osteoporosis, though the mechanisms remain elusive. Bacterial outer membrane vesicles (OMVs) are key mediators of pathogen-host interactions, delivering virulence factors to target cells. This study aimed to explore the potential role of H. pylori OMVs on the bone metabolic disorder and osteoporotic pathology. Results showed that H. pylori OMVs are inte

Quercetin targets the TNF-a/TNF pathway to activate mitochondrial autophagy to improve nucleus pulposus cell function and delay disc degeneration.

BACKGROUND: The primary pathogenic mechanism of lower back pain is intervertebral disc degeneration (IVDD), and the phenotypic change of nucleus pulposus cells and matrix degradation are caused by an imbalance in the "inflammation - autophagy - fibrosis" axis. Despite Tong'an decoction's obvious therapeutic benefits, it is unclear if its principal ingredient, quercetin, controls mitochondrial autophagy and postpones IVDD by interfering with TNF signaling. METHODS: Tong'an decoction's various components were screened using the traditional Chinese medicine database, and quercetin's main targets

YTH N6-methyladenosine RNA binding protein 2 mediated m6A modification of circHIPK2 promotes cellular senescence and osteoarthritis progression by inhibiting autophagy.

N6-methyladenosine (m6A) modification has emerged as a critical post-transcriptional regulatory mechanism in osteoarthritis (OA). However, the contribution of m6A-dependent regulation of circular RNAs (circRNAs) to chondrocyte senescence and OA progression remains poorly understood. We aimed to elucidate whether m6A-mediated control of circRNAs regulates chondrocyte senescence and to define the underlying molecular mechanisms contributing to OA progression. Here, we identified an OA-associated circRNA, circHIPK2, and demonstrated that its abundance and function are regulated by an m6A reader-d

Stage- and compartment-specific remodeling of autophagy and selective mitophagy in glaucoma: from aqueous outflow dysfunction to retinal ganglion cell neurodegeneration.

BACKGROUND: Glaucoma is a leading cause of irreversible blindness and is increasingly understood as a chronic neurodegenerative disorder rather than a disease explained solely by elevated intraocular pressure (IOP). Although IOP lowering remains the cornerstone of treatment, many patients continue to progress despite apparently adequate pressure control, indicating that additional mechanisms shape retinal ganglion cell (RGC) vulnerability and disease course. Among these, autophagy and mitophagy have emerged as central regulators of cellular stress adaptation in both anterior and posterior ocul
