Mechanisms of vasorelaxation induced by eicosapentaenoic acid (20:5n-3) in WKY rat aorta.

The vasorelaxant activity of eicosapentaenoic acid (EPA, 20:5n-3), the omega-3 polyunsaturated fatty acid, was investigated in isolated Wistar Kyoto (WKY) rat aortae by measuring isometric tension. Eicosapentaenoic acid (1 - 100 microM) relaxed rat aortae contracted with high K(+) (80 mM) or noradrenaline (NA, 1 microM) in a concentration-dependent manner. Contractions induced by Bay K 8644 or increasing concentrations of calcium were unaffected by EPA. The relaxant effect of EPA (3 - 100 microM) was significantly inhibited by indomethacin (10 microM), the cyclo-oxygenase inhibitor, but not by

Flecainide sensitizes conduction to hyponatremia through an ephaptic mechanism.

BACKGROUND: Studies suggest that voltage-gated sodium channel (SC) loss-of-function (LoF), often through the use of SC blockers, such as tricyclic anti-depressants, some recreational drugs, and importantly, class 1c anti-arrhythmics, sensitizes cardiac conduction to hyponatremia. However, the mechanism driving conduction velocity (CV) sensitivity to sodium ion (Na+) concentration ([Na+]) is unknown. We recently demonstrated CV-[Na+] sensitivity in haploinsufficient Scn5a+/- mouse and reduced CV-[Na+] sensitivity when ephaptic coupling (extracellular conduction by electric fields) is also reduc

Redox status of patients before cardiac surgery.

OBJECTIVES: Redox regulation plays a crucial role in balancing the cardiovascular system. In this prospective study we aimed to identify cur … trite levels. Notably, significant interactions were found between medication and redox parameters. Calcium channel blockers (CCBs) were positive predictors of total glutathione levels, whereas angiotensin-converting enzyme inhibitors and CCBs were negative predictors of NOx levels. Age showed the highest correlation with the duration of the intensive care stay, followed …

Association between antihypertensive medication use and kidney cancer risk: a meta-analysis accounting for hypertension.

BACKGROUND: Evidence that antihypertensive medication (AHTN) use is associated with an increased risk of kidney cancer (KC) is emerging. However, limited evidence is available on disentangling the effects of AHTN use on KC from hypertension, which is a risk factor for KC. We aimed to identify pooled estimates for the associations between AHTN use and KC risk, independent of hypertension. METHODS: We searched for observational studies that investigated the associations between AHTN use and KC through January 2025. To identify the independent effects of AHTN from hypertension, we conducted stra

Trends of antihypertensive use among patients with cancer and hypertension in the United States 2002-2019.

BACKGROUND: Hypertension (HTN) is the most frequently reported comorbidity in patients with malignancy. This study was conducted to assess the trend of different antihypertensive (AHT) medications used in cancer patients. METHODS: We used the Medical Expenditure Panel Survey (MEPS) database from 2002 to 2019 to identify adult (age >18 years) cancer patients with HTN using appropriate International Classification of Disease (ICD)-9 and ICD-10 codes. Benign and uncertain neoplasms were excluded. P-trend values were calculated using weighted logistic regression with "year" as the predictor varia

Patterns of anti-hypertensive therapy in diabetic patients with and without reduced renal function.

Renal function deterioration is a common complication in patients with diabetes mellitus and hypertension. Appropriate use of anti-hypertens … verting enzyme inhibitors (ACEI)/angiotensin receptor blocker (ARB), followed by diuretics (40.8%), calcium channel blockers (25.1%) and (beta-blockers (12.5%). The majority of patients (> 55%) were either on mono or no drug therapy. Patients on monotherapy were mostly receiving ACEI/ARB (60%). In patients with reduced renal function, use of diuretics, but not ACEI/ARB or …

[Effect of statin therapy on mortality in patients with non-ischemic dilated cardiomyopathy].

OBJECTIVE: To explore the effect of early statin therapy (atorvastatin and simvastatin) on mortality in patients with non-ischemic dilated cardiomyopathy. METHODS: A retrospective study was conducted at a single center. A total of 315 patients with non-ischemic dilated cardiomyopathy, admitted into our hospital from January 2002 to December 2008, were enrolled. The association of statin therapy at the initial hospitalization with all-cause mortality was evaluated. The median follow-up period was 45.1 months. RESULTS: By the single-factor analysis, we found that the follow-up mortality was 17

Single-cell RT-PCR and functional characterization of Ca2+ channels in motoneurons of the rat facial nucleus.

Voltage-dependent Ca2+ channels are a major pathway for Ca2+ entry in neurons. We have studied the electrophysiological, pharmacological, and molecular properties of voltage-gated Ca2+ channels in motoneurons of the rat facial nucleus in slices of the brainstem. Most facial motoneurons express both low voltage-activated (LVA) and high voltage-activated (HVA) Ca2+ channel currents. The HVA current is composed of a number of pharmacologically separable components, including 30% of N-type and approximately 5% of L-type. Despite the dominating role of P-type Ca2+ channels in transmitter release at

Secretagogue-induced RVD in HSY cells is due to K+ channels activated by Ca2+ and protein kinase C.

To maintain cell volume, absorptive epithelia must coordinate ion fluxes associated with transcellular transport with those required for volume regulation. K+ channels are thought to play a central regulatory role in this process. Electronic cell sizing was used to study the regulatory volume decrease (RVD) response of the human salivary ductal cell line HSY. Following a hypotonic challenge, RVD was markedly enhanced by the muscarinic agonist carbachol (half-maximal effect approximately 1.4 microM). Carbachol-induced RVD was mimicked by the K+ ionophore valinomycin and inhibited by K+ channel

Upregulated expression of STIM2, TRPC6, and Orai2 contributes to the transition of pulmonary arterial smooth muscle cells from a contractile to proliferative phenotype.

Pulmonary arterial hypertension (PAH) is a progressive disease that, if left untreated, eventually leads to right heart failure and death. Elevated pulmonary arterial pressure (PAP) in patients with PAH is mainly caused by an increase in pulmonary vascular resistance (PVR). Sustained vasoconstriction and excessive pulmonary vascular remodeling are two major causes for elevated PVR in patients with PAH. Excessive pulmonary vascular remodeling is mediated by increased proliferation of pulmonary arterial smooth muscle cells (PASMC) due to PASMC dedifferentiation from a contractile or quiescent ph

Systemic gene expression mapping of voltage-gated calcium channel α1-subunits in cats.

BACKGROUND: In veterinary medicine, voltage-gated calcium channels (VGCCs) are key drug targets. Calcium channel blockers (CCBs) are the standard treatment for common feline diseases, such as systemic hypertension and hypertrophic cardiomyopathy. However, therapeutic strategies are limited due to the adverse effects of existing drugs like amlodipine, such as activation of the renin-angiotensin-aldosterone system. A critical lack of feline-specific data on the tissue distribution of VGCC subtypes fundamentally hinders the safe and effective application of CCBs in cats. This is because knowledge…

Nitric oxide inhibits L-type Ca2+ current in glomus cells of the rabbit carotid body via a cGMP-independent mechanism.

Previous studies have shown that nitric oxide (NO) inhibits carotid body sensory activity. To begin to understand the cellular mechanisms associated with the actions of NO in the carotid body, we monitored the effects of NO donors on the macroscopic Ca2+ current in glomus cells isolated from rabbit carotid bodies. Experiments were performed on freshly dissociated glomus cells from adult rabbit carotid bodies using the whole cell configuration of the patch-clamp technique. The NO donors sodium nitroprusside (SNP; 600 microM, n = 7) and spermine nitric oxide (SNO; 100 microM, n = 7) inhibited th

Anesthesia-triggered dopaminergic bursts actively induce forgetting: a paradigm shift in understanding cold-shock amnesia.

Retrograde memory loss, the inability to recall events preceding amnesia onset, is a well-documented consequence of anesthesia. Recent discoveries, pioneered by Drosophila research, have challenged the view that forgetting is a passive process, and have demonstrated that rather it is a highly active, well-regulated biological process. This work builds on this and makes a remarkable and unexpected discovery that anesthesia itself triggers a robust, widespread burst of activity in dopaminergic neurons. Here, we report that both cold-shock and CO 2 anesthesia elicits strong activation in PAM and

Targeting Vascular Calcification: Novel Insights into Molecular Pathways and Clinical Interventions.

Vascular calcification (VC), a pathological hallmark of advanced atherosclerosis, exerts a profound impact on arterial stiffness and the incidence of cardiovascular disease. Its development involves not only actively regulated biological processes but also passive mineral deposition and is particularly prevalent in patients with diabetes, chronic kidney disease, and endocrine disorders. Although conventional therapies-such as phosphate binders, calcium channel blockers, bisphosphonates, and endovascular interventions-form the current clinical foundation, they remain inadequate for preventing e…
