A FAP-α-activatable near-infrared theranostic probe for fluorescence imaging of pancreatic cancer and potent in vitro photodynamic activity.

Fibroblast activation protein-alpha (FAP-α), a highly specific biomarker in the tumor microenvironment of pancreatic cancer, is an exceptional target for precision medicine. However, creating a single molecule that integrates both diagnostic and therapeutic functions remains a significant challenge. Herein, we report a novel FAP-α-responsive near-infrared (NIR) theranostic probe, ZGP-LL-ICy. The probe was rationally designed for tumor imaging and phototherapy by conjugating a FAP-α-cleavable peptide to a NIR fluorophore-photosensitizer hybrid (ICyOH) via a self-immolative linker, which effecti

Telocytes in neuromuscular tissues: Integrating morphological and immunohistochemical evidence.

BACKGROUND: Telocytes are specialized stromal cells with roles in tissue organization, development, and regeneration. Their unique morphology (long telopodes expressing CD34/PDGFR-α) and 3D networking capabilities position them as potential key players in neuromuscular function and repair, though their mechanistic contributions remain underexplored. OBJECTIVE: This study investigates the structural and functional relationships between TCs and neuromuscular structures to evaluate their therapeutic potential for nerve injuries and degenerative disorders. METHODS: Histochemical analysis was per

Formulation and development of ACUPA-targeting PEGylated nanoliposomes for treatment of prostate cancer.

A promising strategy for improving the effectiveness, specificity and safety of cancer treatment is targeted medication delivery. Prostate-specific membrane antigen (PSMA) is an effective biomarker for tracking and treating prostate cancer. In this study, we developed a PSMA-targeted drug delivery system by modifying PEGylated liposomal doxorubicin (PLD) with 2-(3-((S)-5-amino-1-carboxypentyl) ureido) pentanedioic acid (ACUPA), a small-molecule PSMA inhibitor, to enhance tumor targeting and therapeutic outcomes. The physicochemical characterization of ACUPA-functionalized PLD (ACUPA-PLD) confi

Repairing osteoporotic bone defects under iron overload: A chitosan-hyaluronic acid hydrogel via synergistic iron chelation and ROS responsiveness.

Iron overload in the bone microenvironment elevates reactive oxygen species (ROS) and triggers osteoblast ferroptosis, exacerbating osteoporosis. We developed a genipin-crosslinked hydrogel composed of 3-aminophenylboronic acid-grafted hyaluronic acid and hydrocaffeic acid (DHCA)-grafted chitosan (CS-DHCA/HA-3-APBA) and loaded with chromium picolinate (CrPic). In the environment of osteoporosis, Fe3+ coordination compacted the network and increased compressive modulus by approximately twofold, while elevated ROS accelerated network degradation, supporting ROS-responsive CrPic release, whereas

Laurencia johnstonii extract reverses early lesions in the K14E7HPV16 murine cervical carcinogenesis model.

Persistent infection with high-risk human papillomavirus (HR-HPV) is a well-established risk factor to the development of cervical intraepithelial neoplasia (CIN), a condition that can progress to cervical cancer (CC) a major health problem worldwide. Recently, there has been growing interest in exploring alternative therapies utilizing natural products, among which is the algae species Laurencia johnstonii Setchell & Gardner, 1924 (L. johnstonii), proposed for the management of precancerous lesions. The aim of this work was to determine the effect of an organic extract from L. johnstonii (ELj

Annona muricata leaf water extract in combination with artemisinin-based combination therapy for increasing CCL19 levels in the treatment of severe Plasmodium berghei ANKA infection using swiss mice.

Cerebral malaria (CM) is a severe complication of Plasmodium falciparum infection, with resistance to antimalarial drugs, including artemisinin-based combination therapies(ACTs), posing a significant threat. CD4+ naive cells expressing CCR7 are known to play a protective role, as they readily migrate to secondary lymphoid tissues activated by CCL19 chemokines. In an effort to address this challenge, we investigated the impact of Annona muricata, an herbaceous and immunomodulatory plant, on CCL19 concentration. We conducted experiments on 24 mice, dividing them into four groups: one control gro

SIRT6 modulates lesion microenvironment in LPC induced demyelination by targeting astrocytic CHI3L1.

Demyelination occurs widely in the central nervous system (CNS) neurodegenerative diseases, especially the multiple sclerosis (MS), which with a complex and inflammatory lesion microenvironment inhibiting remyelination. Sirtuin6 (SIRT6), a histone/protein deacetylase is of interest for its promising effect in transcriptional regulation, cell cycling, inflammation, metabolism and longevity. Here we show that SIRT6 participates in the remyelination process in mice subjected to LPC-induced demyelination. Using pharmacological SIRT6 inhibitor or activator, we found that SIRT6 modulated LPC-induced

Epidermolysis Bullosa: A Review of the Tissue-Engineered Skin Substitutes Used to Treat Wounds.

Skin wound healing is a crucial process for regenerating healthy skin and avoiding the undesired consequences associated with open skin wounds. For epidermolysis bullosa (EB), a debilitating group of fragile skin disorders currently without a cure, skin blistering can often be severe and heal poorly, increasing susceptibility to life-threatening complications. To prevent these, investigational therapies have been exploring the use of tissue-engineered skin substitutes (TESSs) aimed at replacing damaged skin and promoting long-term wound closure. These products have either been developed in hou

Delivery of siRNA to Endothelial Cells In Vivo Using Lysine/Histidine Oligopeptide-Modified Poly(β-amino ester) Nanoparticles.

PURPOSE: Endothelial cell (EC) dysfunction underlies the pathology of multiple disease conditions including cardiovascular and pulmonary diseases. Dysfunctional ECs have a distinctive gene expression profile compared to healthy ECs. RNAi therapy is a powerful therapeutic approach that can be used to silence multiple genes of interests simultaneously. However, the delivery of RNAi to ECs in vivo continues to be a major challenge. Here, we optimized a polymer formulation based on poly(β-amino ester)s (pBAEs) to deliver siRNA to vascular ECs. METHODS: We developed a library of bioinspired oligop

Molecular Regulatory Mechanism and Toxicology of Neurodegenerative Processes in MPTP/Probenecid-Induced Progressive Parkinson's Disease Mice Model Revealed by Transcriptome.

Parkinson's disease (PD) is a neurodegenerative disease caused by a variety of unclear complex pathogenic factors. The 1-methyl-4-phenyl-1, 2, 3, 6-tetrahydropyridine/probenecid (MPTP/p)-induced progressive PD mice is a well-recognized classic model for studying PD, but the molecular toxicology of this model is still unclear. Here, for the first time, we report gradual neurodegenerative processes in MPTP/p-induced progressive PD mice model using RNA-seq. Transcriptional responses are orchestrated to regulate the expression of many genes in substantia nigra, such as Ntf3, Pitx3, Th, and Drd2, l

Responses to crizotinib and disease monitoring with circulating tumor cells in lung adenocarcinoma patient with MET exon 14 skipping mutation: A case report.

RATIONALE: Mesenchymal-to-epithelial transition (MET) exon 14 skipping mutation was a targetable alteration in nonsmall-cell lung cancer (NSCLC), and the MET inhibitor of crizotinib had the most efficacy among all the targeted drugs. Most of the cancer-related deaths are associated with metastasis. Circulating tumor cells (CTCs) have been a valuable biomarker in assessing metastasis. Recent experiences suggested that CTCs detection may help improve diagnosis and predict prognosis for patients with NSCLC. However, few literatures have reported the CTCs detection based on the (MET) exon 14 skipp

Theranostic Application of Mixed Gold and Superparamagnetic Iron Oxide Nanoparticle Micelles in Glioblastoma Multiforme.

The treatment of glioblastoma multiforme, the most prevalent and lethal form of brain cancer in humans, has been limited in part by poor delivery of drugs through the blood-brain barrier and by unclear delineation of the extent of infiltrating tumor margins. Nanoparticles, which selectively accumulate in tumor tissue due to their leaky vasculature and the enhanced permeability and retention effect, have shown promise as both therapeutic and diagnostic agents for brain tumors. In particular, superparamagnetic iron oxide nanoparticles (SPIONs) have been leveraged as T2-weighted MRI contrast agen

Vitamin D analogs: therapeutic applications and mechanisms for selectivity.

The vitamin D endocrine system plays a central role in mineral ion homeostasis through the actions of the vitamin D hormone, 1,25-dihydroxyvitamin D(3) [1,25(OH)(2)D(3)], on the intestine, bone, parathyroid gland, and kidney. The main function of 1,25(OH)(2)D(3) is to promote the dietary absorption of calcium and phosphate, but effects on bone, kidney and the parathyroids fine-tune the mineral levels. In addition to these classical actions, 1,25(OH)(2)D(3) exerts pleiotropic effects in a wide variety of target tissues and cell types, often in an autocrine/paracrine fashion. These biological ac

The effects of glutathione glycoside in methyl mercury poisoning.

The use of glutathione glycoside (GSH-glyc), a compound newly synthesized in our laboratory, was highly effective in raising cellular GSH levels both in vitro and in vivo. Based on mass spectrometry and 1H NMR data, the structure of GSH-glyc was determined to be that of an S-glycoside. Rapid GSH uptake was observed by confocal microscopy in both A549 cells and mouse astrocytes following incubation with GSH-glyc. Intraperitoneal (i.p.) and per oral administration of GSH-glyc in C57BL/6J mice raised GSH concentrations in brain and liver to significantly higher levels than normal. Methyl mercury

New drugs, new toxicities: severe side effects of modern targeted and immunotherapy of cancer and their management.

Pharmacological and cellular treatment of cancer is changing dramatically with benefits for patient outcome and comfort, but also with new toxicity profiles. The majority of adverse events can be classified as mild or moderate, but severe and life-threatening complications requiring ICU admission also occur. This review will focus on pathophysiology, symptoms, and management of these events based on the available literature. While standard antineoplastic therapy is associated with immunosuppression and infections, some of the recent approaches induce overwhelming inflammation and autoimmunity.

Long-term outcome of olfactory ensheathing cell therapy for patients with complete chronic spinal cord injury.

The neurorestorative effect of the parenchymal transplantation of olfactory ensheathing cells (OECs) for cord trauma remains clinically controversial. The aim of this article is to study the long-term result of OECs for patients with complete chronic spinal cord injury (SCI). One hundred and eight patients suffered from complete chronic SCI were followed up successfully within the period of 3.47 ± 1.12 years after OEC therapy. They were divided into two groups based on the quality and quantity of their rehabilitative training: group A (n = 79) in sufficient rehabilitation (or active movement-t

Flow-induced, inflammation-mediated arterial wall remodeling in the formation and progression of intracranial aneurysms.

OBJECTIVE: Unruptured intracranial aneurysms (UIAs) are relatively common lesions that may cause devastating intracranial hemorrhage, thus producing considerable suffering and anxiety in those affected by the disease or an increased likelihood of developing it. Advances in the knowledge of the pathobiology behind intracranial aneurysm (IA) formation, progression, and rupture have led to preclinical testing of drug therapies that would prevent IA formation or progression. In parallel, novel biologically based diagnostic tools to estimate rupture risk are approaching clinical use. Arterial wall

Ketohexokinase inhibition improves NASH by reducing fructose-induced steatosis and fibrogenesis.

BACKGROUND & AIMS: Increasing evidence highlights dietary fructose as a major driver of non-alcoholic fatty liver disease (NAFLD) pathogenesis, the majority of which is cleared on first pass through the hepatic circulation by enzymatic phosphorylation to fructose-1-phosphate via the ketohexokinase (KHK) enzyme. Without a current approved therapy, disease management emphasises lifestyle interventions, but few patients adhere to such strategies. New targeted therapies are urgently required. METHODS: We have used a unique combination of human liver specimens, a murine dietary model of NAFLD and

A new mouse model of radiation-induced liver disease reveals mitochondrial dysfunction as an underlying fibrotic stimulus.

BACKGROUND & AIMS: High-dose irradiation is an essential tool to help control the growth of hepatic tumors, but it can cause radiation-induced liver disease (RILD). This life-threatening complication manifests itself months following radiation therapy and is characterized by fibrosis of the pericentral sinusoids. In this study, we aimed to establish a mouse model of RILD to investigate the underlying mechanism of radiation-induced liver fibrosis. METHODS: Using a small animal image-guided radiation therapy platform, an irradiation scheme delivering 50 Gy as a single dose to a focal point in m
