Gene knockdown studies revealed CCDC50 as a candidate gene in mantle cell lymphoma and chronic lymphocytic leukemia.

The two B-cell non-Hodgkin's lymphoma entities, chronic lymphocytic leukemia (CLL) and mantle cell lymphoma (MCL), show recurrent chromosomal gains of 3q25-q29, 12q13-q14 and 18q21-q22. The pathomechanisms affected by these aberrations are not understood. The aim of this study was to identify genes, located within these gained regions, which control cell death and cell survival of MCL and CLL cancer cells. Blood samples collected from 18 patients with CLL and 6 patients with MCL, as well as 6 cell lines representing both malignancies were analyzed by gene expression profiling. By a comparison

Specific patterns of oncogene activation in transplacentally induced tumors.

Transplacental exposure of rats to a single dose of the direct acting carcinogen methylnitrosourea (MNU) results in the induction of a variety of neoplasias of neuroectodermal, epithelial, and mesenchymal origin. Molecular analysis of the oncogenes present in these tumors revealed a striking degree of tissue specificity. neu oncogenes were found to be reproducibly activated in tumors derived from the peripheral nervous system (PNS), but not in those arising from the central nervous system (CNS). No ras oncogenes were found in either PNS- or CNS-derived tumors. However, Ha-ras oncogenes were de

Enhancer collapse dictates PKHD1 silencing and centrosome-mediated chromosomal instability and metastasis in clear cell renal cell carcinoma.

Chromosomal instability (CIN) drives clear cell renal cell carcinoma (ccRCC) progression, yet its upstream triggers remain elusive. To identify key drivers governing renal genomic integrity, we integrated machine learning-driven screening with Mendelian randomization analysis to prioritize candidates associated with both CIN and clinical outcomes. This computational pipeline identified Polycystic Kidney and Hepatic Disease 1 (PKHD1) as a top candidate and a crucial guardian of the renal genome. Through single-cell and spatial transcriptomics, combined with functional assays in ccRCC cell lines

Identification of LncRNAs scaffolding a chromatin-associated gene regulatory complex in breast cancer cells comprising ERα, DOT1L and menin.

In Estrogen Receptor (ER)-positive Breast Cancer (BC), the development of resistance to endocrine therapies remains a significant clinical challenge, often linked to transcriptional and epigenetic deregulations. The main hallmark in this neoplasm, the nuclear receptor ERα, drives oncogenesis by recruiting multiple coregulators, including the histone methyltransferase DOT1L and the scaffold protein menin, forming a pro-tumorigenic machinery. However, the precise mechanism of this complex assembly is not fully understood. Our study investigates the molecular basis of ERα-DOT1L-menin functional a

CRISPR/Cas9-mediated miR-21 editing in high-grade urothelial carcinoma cells and its biological effects.

BACKGROUND: Urothelial carcinoma, the predominant form of bladder cancer, represents a global public health challenge due to its high rates of recurrence and progression. At the molecular level, microRNA-21 (miR-21) has been characterized as an "oncomir" because of its ability to negatively regulate tumor suppressor genes, thereby promoting tumor survival and progression. In this context, the CRISPR/Cas9 system has emerged as a precise genome-editing tool. OBJECTIVE: To investigate the biological effects of miR-21 modulation using CRISPR/Cas9-mediated genome editing in the T24 high-grade inva

Dual role and ceRNA networks of miR-146a in lung cancer: regulating proliferation, metastasis, and chemoresistance.

Lung cancer persists as the predominant cause of cancer-related mortality globally. MicroRNAs, a class of short non-coding RNAs, are recognized as critical regulators in the pathogenesis of lung cancer, functioning either as oncogenes or tumor suppressors. Among these, miR-146a has been identified as a significant modulator in non-small-cell lung cancer, exerting influence over cellular proliferation, resistance to chemotherapy, apoptosis, and metastatic potential. Notably, miR-146a displays a dualistic role, acting as either a tumor suppressor or an oncogene depending on the molecular mechani

A fishnet between nasopharyngeal carcinoma and resistance: the competing endogenous RNA network.

Chemotherapy and radiotherapy (chemo-/radiotherapy) have advanced as the main treatment modalities for nasopharyngeal carcinoma (NPC), improving patient survival rates. However, chemo-/radiotherapy resistance in NPC cells has emerged as a key factor contributing to poor prognosis. Recently, competing endogenous RNA networks (ceRNETs) have garnered attention for their potential clinical value in studying chemo-/radiotherapy resistance. In this review, we aimed to explore the molecular mechanisms of ceRNA-related molecules, including circular RNA (circRNA), long non-coding RNA (lncRNA), microRNA

Endoplasmic reticulum stress-dependent regulation of carboxypeptidase E expression in glioblastoma cells.

Objective. Carboxypeptidase E (CPE) plays an important role in the biosynthesis of neurotransmitters and peptide hormones including insulin. It also promotes cell proliferation, survival, and invasion of tumor cells. The endoplasmic reticulum stress, hypoxia, and nutrient supply are significant factors of malignant tumor growth including glioblastoma. There are data indicating that the knockdown of the endoplasmic reticulum to nucleus signaling 1 (ERN1) suppressed glioblastoma cell proliferation and increased invasiveness of these cells. The present study aims to investigate the regulation of

Significance of CD10 protein expression in the diagnostics of follicular lymphoma: A comparison of conventional immunohistochemistry with flow cytometry supported by the establishment of BCL2 and BCL6 rearrangements.

INTRODUCTION: Histopathological examination and immunohistochemistry (IHC) with a crucial role of CD10 expression remain a standard diagnostic tool in follicular lymphoma (FL). The results of IHC CD10 detection with different primary antibodies are not fully reproducible, but some reports show that flow cytometry (FCM) can be a reliable method of CD10 identification. METHODS: The aim of the study was to compare results of CD10 expression in FL by IHC and FCM including immunophenotypic features in the context of the BCL2 and BCL6 alterations. RESULTS: Out of 76 histopathologically diagnosed F

Context-dependent activation of SIRT3 is necessary for anchorage-independent survival and metastasis of ovarian cancer cells.

Tumor cells must alter their antioxidant capacity for maximal metastatic potential. Yet the antioxidant adaptations required for ovarian cancer transcoelomic metastasis, which is the passive dissemination of cells in the peritoneal cavity, remain largely unexplored. Somewhat contradicting the need for oxidant scavenging are previous observations that expression of SIRT3, a nutrient stress sensor and regulator of mitochondrial antioxidant defenses, is often suppressed in many primary tumors. We have discovered that this mitochondrial deacetylase is specifically upregulated in a context-dependen
