AIM: To study the antagonistic effect of oleanolic acid (OA) on anaphylactic shock in guinea-pigs sensitized with 6-aminopenicillanic acid (APA)-protein.
METHODS: The anaphylactic shock was achieved by i.p. of APA-protein in guinea pigs. The serum antibody titers of guinea pigs were measured with the passive cutaneous anaphylaxis test, and the pulmonary histamine by the method of fluorometric determination. The degranulation of mast cells was examined by rat calvarian periosteum method.

The undecapeptide substance P (SP) is both a potent stimulant of intestinal (guinea pig ileum), vascular venous (rabbit mesenteric vein) and tracheal (guinea pig trachea) smooth muscles, and a relaxant of large arteries (dog common carotid artery, rabbit aorta). SP also decreases the peripheral vascular resistance in the coronary vessels of rabbit isolated hearts. The contractions of the guinea pig ileum in response to SP are partially antagonised by atropine and significantly depressed by indomethacin, while the contractions of the guinea pig trachea are strongly potentiated by indomethacin.

A single intradermal vaccination with an antibiotic-less version of BCGΔBCG1419c given to guinea pigs conferred a significant improvement in outcome following a low dose aerosol exposure to M. tuberculosis compared to that provided by a single dose of BCG Pasteur. Guinea pigs were fed L. pneumophila through an orogastric tube. Gastric acid was rapidly cidal to the organisms. Serial necropsies demonstrated the organisms in the colon and blood stream at 1 hr. Guinea pigs are the most frequently used animals in phototoxicity studies. However, general toxicity studies most often use Sprague-Dawley (SD) rats. To reduce the number of animals needed for drug development, we examined whether skin phototoxicity studies could be performed using SD rats. We have examined the capacity of the major cytoplasmic membrane protein (MCMP) of Legionella pneumophila, a genus common antigen and member of the hsp 60 family of heat shock proteins, to induce protective immunity in a guinea pig model of Legionnaires' disease.
