Clinical course of genetic diseases of the insulin receptor (type A and Rabson-Mendenhall syndromes): a 30-year prospective.

The interaction of insulin with its cell surface receptor is the first step in insulin action and the first identified target of insulin resistance. The insulin resistance in several syndromic forms of extreme insulin resistance has been shown to be caused by mutations in the receptor gene. We studied 8 female patients with the type A form of extreme insulin resistance and 3 patients (2 male and 1 female) with the Rabson-Mendenhall syndrome and followed the natural history of these patients for up to 30 years. The 11 patients ranged in age from 7 to 32 years at presentation. All 11 patients ha

Electrospray ionization mass spectrometric analyses of changes in tissue phospholipid molecular species during the evolution of hyperlipidemia and hyperglycemia in Zucker diabetic fatty rats.

The Zucker diabetic fatty (ZDF) rat is a genetic model of type II diabetes mellitus in which males homozygous for nonfunctional leptin receptors (fa/fa) develop obesity, hyperlipidemia, and hyperglycemia, but rats homozygous for normal receptors (+/+) remain lean and normoglycemic. Insulin resistance develops in young fa/fa rats and is followed by evolution of an insulin secretory defect that triggers hyperglycemia. Because insulin secretion and insulin sensitivity are affected by membrane phospholipid fatty acid composition, we have determined whether metabolic abnormalities in fa/fa rats are

The Pituitary Gland is a Novel Major Site of Action of Metformin in Non-Human Primates: a Potential Path to Expand and Integrate Its Metabolic Actions.

BACKGROUND/AIMS: Biguanides are anti-hyperglycaemic agents used to treat diabetes by acting primarily on the liver, inhibiting hepatic gluconeogenesis. However, biguanides may target other key metabolic tissues to exert beneficial actions. As the "master endocrine gland", the pituitary is a true homeostatic sensor that controls whole body homeostasis and metabolism by integrating central and peripheral signals. However, whether the pituitary is a primary site of biguanides action in normal adult humans/primates remains unknown. Therefore, we aimed to elucidate the direct effects of two biguani

LEP, LDLR and APOA4 gene polymorphisms and their relationship with the risk of overweight, obesity and chronic diseases in adults of the State of Sucre, Venezuela.

INTRODUCTION: Overweight, obesity and some chronic diseases have become more prevalent recently. It is well known that their causes may be genetic, epigenetic, environmental, or a mixture of these. OBJECTIVE: To analyze the relationship between nine single nucleotide polymorphisms of genes LEP (rs2167270), LDLR (rs885765, rs688, rs5925, rs55903358, rs5742911) and APOA4 (rs5095, rs675, rs5110) with obesity-related phenotypes and other comorbidities. MATERIAL AND METHODS: We recruited 144 adults (76 males and 68 females, with average ages of 29.93±8.29 and 32.49±11.15 years, respectively) in t

The role of adiponectin gene variants in glycemic control in patients with Type 2 diabetes.

The aim of this study was to investigate the association between glycemic control in Type 2 diabetes mellitus patients and common genetic variants of ADIPQO gene. A total of 427 Type 2 diabetes patients were recruited in the study and divided into two groups: 172 patients with good glycemic control and 249 with poor glycemic control. Genotyping of C11377G, G276T and T45G ADIPQO SNPs were carried out using restriction fragment length polymorphisms-polymerase chain reaction. The results showed that C11377G ADIPQO SNP is strongly associated with glycemic control in Type 2 diabetes patients. Patie

A phase plane graph based model of the ovulatory cycle lacking the "positive feedback" phenomenon.

When hormones during the ovulatory cycle are shown in phase plane graphs, reported FSH and estrogen values form a specific pattern that resembles the leaning "&" symbol, while LH and progesterone (Pg) values form a "boomerang" shape. Graphs in this paper were made using data reported by Stricker et al. [Clin Chem Lab Med 2006;44:883-887]. These patterns were used to construct a simplistic model of the ovulatory cycle without the conventional "positive feedback" phenomenon. The model is based on few well-established relations:hypothalamic GnRH secretion is increased under estrogen exposure duri

Long-term characterization of the diet-induced obese and diet-resistant rat model: a polygenetic rat model mimicking the human obesity syndrome.

The availability of useful animal models reflecting the human obesity syndrome is crucial in the search for novel compounds for the pharmacological treatment of obesity. In the current study, we have performed an extensive characterization of the obesity syndrome in a polygenetic animal model, namely the selectively bred diet-induced obese (DIO) and diet-resistant (DR) rat strains. We show that they constitute useful models of the human obesity syndrome. DIO and DR rats were fed either a high-energy (HE) or a standard chow (Chow) diet from weaning to 9 months of age. Metabolic characterization

Mineralocorticoid receptor blockade reverses obesity-related changes in expression of adiponectin, peroxisome proliferator-activated receptor-gamma, and proinflammatory adipokines.

BACKGROUND: In obesity, decreases in adiponectin and increases in proinflammatory adipokines are associated with heart disease. Because adipocytes express mineralocorticoid receptor (MR) and MR blockade reduces cardiovascular inflammation and injury, we tested the hypothesis that MR blockade reduces inflammation and expression of proinflammatory cytokines in adipose tissue and increases adiponectin expression in adipose tissue and hearts of obese mice. METHODS AND RESULTS: We determined the effect of MR blockade (eplerenone, 100 mg/kg per day for 16 weeks) on gene expression in retroperitonea

Mesenchymal stem cells from the outer ear: a novel adult stem cell model system for the study of adipogenesis.

Adipocytes arise from multipotent stem cells of mesodermal origin, which also give rise to the muscle, bone, and cartilage lineages. However, signals and early molecular events that commit multipotent stem cells into the adipocyte lineage are not well established mainly due to lack of an adequate model system. We have identified a novel source of adult stem cells from the external murine ears referred to here as an ear mesenchymal stem cells (EMSC). EMSC have been isolated from several standard and mutant strains of mice. They are self-renewing, clonogenic, and multipotent, since they give ris

[Adipocytokins, obesity and development of type 2 diabetes].

Normal metabolic balance is maintained by a complex homeostatic system involving multiple tissues and organs. Acquired or inherited defects associated to environmental factors in any part of this system can lead to metabolic disorders such as the syndrome X which is presently a frequent syndrome in industrialized countries. It is characterized by a cluster of risk factors of atherosclerosis including insulin resistance, hyperinsulinemia, impaired glucose tolerance or type 2 diabetes, hypertension, dyslipidemia, and coagulation abnormalities. Its pathophysiology is likely to involve insulin res

Adrenomedullin and the blood-brain barrier.

Adrenomedullin (ADM) is present both in the periphery and brain. In addition to its peripheral effects, this peptide can exert central effects such as decreasing food ingestion. We used multiple-time regression analysis to determine that labeled ADM can cross from blood to brain with an apparent influx constant (K(I)) of 5.83 +/- 1.44 x 10(-4) ml/g-min, much faster than that of albumin, the vascular control. HPLC showed that almost all of the injected 125I-ADM in the brain was intact, and capillary depletion showed that it could reach the parenchyma of the brain. However, more 125I-ADM was rev

Early Time-Restricted Feeding Mitigates Insulin Resistance and Proteinuria in Prepubertal Obese Dahl Salt-Sensitive Rats.

BACKGROUND: Previous studies from our laboratory have shown that hyperinsulinemia and insulin resistance (IR) are elevated in obese Dahl salt-sensitive (SS) leptin receptor mutant (SSLepRmutant) rats compared to SS rats before puberty. Clinical studies suggest that time-restricted feeding improves insulin sensitivity and is considered beneficial in obese patients. Therefore, the current study examined whether time-restricted feeding improves IR and reduces renal injury in obese SSLepRmutant rats prior to puberty. METHODS: Four-week-old SS and SSLepRmutant rats were given either ad libitum acc

Exercise modalities and dosages on glycated haemoglobin and exerkines in type 2 diabetes: a systematic review with pairwise, network, and dose-response meta-analyses.

BACKGROUND: Exercise is a cornerstone of type 2 diabetes mellitus (T2DM) management; however, the comparative efficacy of specific modalities, optimal dosages, and associated exerkine responses remains incompletely defined. This study aimed to synthesise evidence on the effects of different exercise types and doses on glycated haemoglobin (HbA1c) levels and to explore modality-specific patterns of exerkine regulation in adults with T2DM. METHODS: A systematic review and Bayesian evidence synthesis of randomised controlled trials was conducted up to October 2025. Pairwise, network, and model-b

Comparative effects of different exercise types on inflammatory markers in type 2 diabetes mellitus patients with overweight and obesity: a systematic review and network meta-analysis.

BACKGROUND: For patients with type 2 diabetes mellitus (T2DM) living with overweight or obesity, systemic chronic low-grade inflammation induced by excessive fat accumulation not only exacerbates insulin resistance but also serves as a pivotal driver of disease progression and cardiovascular complications. As a vital component of non-pharmacological therapy, exercise has demonstrated the potential to modulate immune responses and improve metabolic status, holding positive significance for combating this inflammatory process and delaying disease progression. OBJECTIVE: This study aims to compar

Neuronal surface P antigen (NSPA) as a novel regulator of energy homeostasis and adipose tissue metabolism.

BACKGROUND: Regulation of body weight and glucose homeostasis includes the coordinated activity of hypothalamic neurons and adipocytes within a neuroendocrine network whose dysfunctions underlie obesity, insulin resistance, and type 2 diabetes (T2D). The neuronal surface P antigen (NSPA) is a plasma membrane protein with characteristics of an E3 ubiquitin ligase encoded by the unique gene Zzef1, which has been linked to T2D. NSPA's original discovery in neurons, as a cell-surface cross-reacting autoantigen of anti-P antibodies that associate with cognitive dysfunctions in patients with systemi

Prenatal inflammation exacerbates hyperoxia-induced neonatal brain injury.

BACKGROUND: Premature born infants are at high risk to develop white matter injury (WMI). Hyperoxia and perinatal inflammation are main risk factors for preterm birth and associated brain injury. To date the majority of experimental studies have focused on isolated insults. However, clinically, WMI injury is a multifactorial disorder caused by a variety of triggers. To establish a clinically relevant rodent model of WMI, we combined prenatal inflammation with postnatal hyperoxia to investigate individual, and additive or synergistic effects on inflammatory processes, myelination and grey matte

The Epidemiology of Slipped Capital Femoral Epiphysis in Children and Adolescents: A Systematic Review of Risk Factors and Incidence Across Populations.

BACKGROUND: Childhood obesity is a growing global health crisis with significant health and orthopedic complications such as slipped capital femoral epiphysis (SCFE), a hip disorder characterized by the displacement of the metaphysis relative to the epiphysis. SCFE always requires surgical intervention to prevent severe outcomes such as avascular necrosis, gait abnormalities, and lifelong disability and deformity. Obesity is a well-established risk factor for SCFE; however, emerging evidence suggests that elevated leptin levels may independently contribute to the development of SCFE, regardles
