In vitro toxic effects of metal compounds on kinetic traits and ultrastructure of rabbit spermatozoa.

Metal compounds have been associated with male reproductive toxicity in vivo. The aim of the present study was to investigate the in vitro effects of 20 metal compounds using rabbit ejaculated spermatozoa as a study model for spermiotoxicity. Five of the metals tested (arsenic, cadmium, chromium, mercury and vanadium) reduced sperm motility and curvilinear velocity. Ultrastructural analyses revealed three types of damage to sperm head membranes in relation to the metal used: acrosome breakage with formation of various sized microvesicles (arsenic, cadmium, mercury and platinum); a large round

Prebypass histological and ultrastructural evaluation of the long saphenous vein as a predictor of early graft failure.

BACKGROUND: Twenty percent of the long saphenous vein (LSV) grafts that are employed as coronary bypass conduits occlude during the first year after the operation. The aim of this study was to evaluate the morphological parameters of the LSV grafts before implantation as predictors for the early occlusion of the grafts. METHODS: Forty-two samples of LSV grafts were examined via light, transmission electron, and scanning electron microscopy and evaluated clinically and by angiography at 6 months and 2 years after the operation. Morphological parameters were statistically analyzed and examined

The venomous secrets of the web droplets from the viscid spiral of the orb-weaver spider Nephila clavipes (Araneae, Tetragnatidae).

The capture web of N. clavipes presents viscous droplets, which play important roles in web mechanics and prey capture. By using scanning and transmission electron microscopy, it was demonstrated that the web droplets are constituted of different chemical environments, provided by the existence both of an aqueous and a lipid layer, which, in turn, present a suspension of tenths of vesicles containing polypeptides and/or lipids. GC/EI-MS Analysis of the contents of these vesicles led to the identification of some saturated fatty acids, such as decanoic acid, undecanoic acid, dodecanoic acid, te

Nature of glomerular dysfunction in pre-eclampsia.

BACKGROUND: Pre-eclampsia is characterized by hypertension, proteinuria and edema. Simultaneous studies of kidney function and structure have not been reported. We wished to explore the degree and nature of glomerular dysfunction in pre-eclampsia. METHODS: Physiologic techniques were used to estimate glomerular filtration rate (GFR), renal plasma flow and afferent oncotic pressure immediately after delivery in consecutive patients with pre-eclampsia (PET; N = 13). Healthy mothers completing an uncomplicated pregnancy served as functional controls (N = 12). A morphometric analysis of glomeruli

An experimental uveitis induced by bovine serum albumin. A transmission and scanning electron microscopic study.

An acute experimental uveitis was produced by injecting bovine serum albumin (BSA) intravitreally into presensitized albino rabbits. Histopathological changes in the anterior uvea were followed during a period of 2 weeks using transmission and scanning electron microscopy. Both the ciliary body and the iris showed prominent ultrastructural changes. Based on the histopathological characteristics, the inflammatory reaction was divided into the 3 different phases: 1) a rapid and transient polymorphonuclear phase (3 h-day 1; 2) a slower and prolonged mononuclear phase (day 1-day 14) and 3) a phase

Otoconia in health and disease. A review.

Mammalian otoconia are made up of inorganic calcium carbonate crystallites and organic substance, which is thought to be sulfated glycosaminoglycans and glycoprotein. Mouse otoconia are formed in situ, and the primitive otoconia assume spindle, trigonal, dumbbell, or quadrilobed forms. As the otoconia mature, they assume the typical barrel-shaped body with pointed tips formed by three surfaces. A variety of otoconial disorders have been reported. Otoconial degeneration can result from ototoxic drugs, infection, trauma, and aging. Abnormal otoconia are also known to occur as a result of genetic

Scanning and transmission electron microscopic observations on the host-parasite relationship in intestinal cryptosporidiosis of neonatal calves.

The association of cryptosporidia with the intestinal epithelium of three neonatal calves was studied by scanning (SEM) and transmission (TEM) electron microscopy. Trophozoites and schizonts were observed embedded in the microvillous brush border of epithelial cells. Merozoites, released from schizonts, were seen free in the lumen and penetrating epithelial cells by SEM and TEM. Incorporation of microvilli into the parasitophorous envelope of trophozoites was seen by TEM. These findings indicate that cryptosporidia develop at an intracellular position in the apex of the epithelial cells follow

Chronic retinitis in rats infected as neonates with lymphocytic choriomeningitis virus: a clinical, histopathologic, and electroretinographic study.

The long-term sequelae to infection of neonatal rats with lymphocytic choriomeningitis virus were studied by a variety of approaches, including indirect ophthalmoscopic, electroretinographic, and histopathologic methods. Data from these studies demonstrated that a progressive chronic retinitis develops after the acute, virus-specific, immune-mediated retinopathy. This chronic inflammation eventually leads to a total destruction of the retinal architecture. An autoimmune reaction against normally sequestered retinal antigens, released during the acute state of necrotizing retinitis, is probably

Structure of the soft palate and composition of the oral mucous membrane in monkeys.

The soft palate of monkeys (M. irus, mulatta, fasciculata; 2.3 +/- 0.4 kg in weight) was studied morphologically and stereologically. After fixation by perfusion, the tissue was disected free, divided in halves and the halves into 5 segments each, and processed for light, transmission- and scanning electron microscopy. Morphometric techniques and a newly developed computer program for automatic data processing (Mumana) were employed in order to estimate, at the light microscopic level, the volumetric composition of the oral soft palate mucosa. The latter showed a different composition in anter

Venous endothelial damage produced by massive sticking and emigration of leukocytes.

A scanning and transmission electron microscope study of canine jugular and femoral veins revealed that large numbers of white cells adhered to the vessel walls, passed through the endothelial intercellular junctions and accumulated in pockets between the endothelium and basement membrane. This led to extensive separation and desquamation of endothelial cells with exposure of subendothelial structures in many areas. The white cell invasion was caused by surgical trauma to adjacent tissues (rather than to the vessel itself) followed by 1 or 7 minutes occlusion by pressure applied externally jus

Selective adhesion, lipid exchange and membrane-fusion processes between vesicles of various sizes bearing complementary molecular recognition groups.

Equimolar mixtures of large unilamellar vesicles (LUVs) obtained from mixtures of egg lecithin and lipids containing complementary hydrogen bonding head groups (barbituric acid (BAR) and 2,4,6-triaminopyrimidine (TAP)) were shown to aggregate and fuse. These events have been studied in detail using electron microscopy and dynamic light scattering, and by fluorimetry using membrane or water-soluble fluorescence probes. It was shown that aggregation was followed by two competitive processes: a) lipid mixing leading to redispersion of the vesicles; b) fusion events generating much larger vesicles

Detection of alpha-synuclein seeding activity in tear fluid in patients with Parkinson's disease.

Detection of alpha-synuclein (α-Syn) seeding activity in tear fluid (TF) offers a promising non-invasive biomarker for Parkinson's disease (PD). α-Syn seeding amplification assay (αSynSAA) detected seeding activity in 67% of PD-TF, while non-synucleinopathy samples remained negative. Electron microscopy of seeding-positive end products revealed fibrillar structures morphologically consistent with results of αSynSAA. αSynSAA effectively distinguished PD from controls and prion diseases based on seeding activity in TF.

[features of ultrastructural and molecular-cellular manifestations of pyelo-nephritis].

Three clinical observations in patients with various manifestations of pyelonephritis are presented. Using 18F-FDG PET/CT and transmission electron microscopy of the renal parenchyma at 10,000x magnification, the ultrastructural and molecular-cellular features of acute purulent pyelonephritis, chronic pyelonephritis with neurogenic hypertension, and chronic pyelonephritis resulting in secondary contracted kidney are identified. The clinical significance of the identified changes, the reliability of the obtained results, and the prospects for their further application are discussed.

A case of HIV associated cryptococcal nephritis: Ultrastructural findings and literature review.

Acute renal failure in the setting of infection with human immunodeficiency virus can be due to various causes including pre-renal causes such as hypotension, sepsis, and nephrotoxic agents; thrombotic microangiopathy; or direct renal parenchymal infections by opportunistic organisms. We present a case of cryptococcal nephritis in a patient with systemic cryptococcosis and discuss the clinical findings as well as the histological, immunofluorescent, and transmission electron microscopy findings in the renal biopsy.

Toxoplasmosis in a bar-shouldered dove (Geopelia humeralis) from the Zoo of Clères, France.

Toxoplasmosis causes mortality in several avian species, especially passerine birds. Toxoplasmosis was diagnosed in a bar-shouldered dove (Geopelia humeralis) found dead at the zoo of Clères (France). The bird had necrotizing pneumonia and nephritis with intralesional tachyzoites of Toxoplasma gondii. The diagnosis was confirmed by immunostaining with polyclonal rabbit T. gondii antibodies and by transmission electron microscopy. To our knowledge, the bar-shouldered dove is a new host record for T. gondii.

Structural perspective on the formation of ribonucleoprotein complex in negative-sense single-stranded RNA viruses.

Negative-sense single-stranded RNA viruses (NSRVs) possess a ribonucleoprotein (RNP) complex composed of viral polymerase and genomic RNA surrounded by viral nucleoprotein. The RNP facilitates virus replication, transcription, and assembly. To date, a large body of structural work, through crystallography and electron microscopy (EM) analysis, has been performed to aid understanding the molecular mechanism of RNP formation in NSRVs, and provides great potential for the discovery of antiviral agents targeting viral RNP formation.

Autophagy in Gaucher disease due to saposin C deficiency.

Gaucher disease, due to a deficit of glucosylceramidase or, rarely, of its activator saposin C, is characterized by accumulation of glucosylceramide in the lysosomes of monocyte/macrophage lineage. In our study we demonstrate that saposin C deficiency due to mutations involving a cysteine residue results in increased autophagy. Autophagy was monitored by LC3 analysis and confirmed by electron microscopy; we observed a correlation among saposin C mutation, Gaucher phenotype and increased autophagy.

Unusual manifestation of cutaneous toxoplasmosis in a HIV-positive patient.

We report a case of unusual cutaneous toxoplasmosis manifestation in a HIV-positive patient. He presented with hard and painful nodular lesions on the arms, hands and chest. Serology tests for anti-Toxoplasma antibody were negative. However, histopathologic examination of the lesion revealed foci of macrophages containing crescent-shaped organisms resembling the zoites of the protozoan parasite Toxoplasma gondii. Ultrastructure examination under electron microscopy and PCR confirmed the organism as T. gondii.
