Piezo1 channels mediate vasorelaxation of uterine arteries from pseudopregnant rats.

Introduction: There is a great increase in uterine arterial blood flow during normal pregnancy, which is a result of the cardiovascular changes that occur in pregnancy to adapt the maternal vascular system to meet the increased metabolic needs of both the mother and the fetus. The cardiovascular changes include an increase in cardiac output and more importantly, dilation of the maternal uterine arteries. However, the exact mechanism for the vasodilation is not fully known. Piezo1 mechanosensitive channels are highly expressed in endothelial and vascular smooth muscle cells of small-diameter ar

Perivascular Adipose Tissue as a Relevant Fat Depot for Cardiovascular Risk in Obesity.

Obesity is associated with increased risk of premature death, morbidity, and mortality from several cardiovascular diseases (CVDs), including stroke, coronary heart disease (CHD), myocardial infarction, and congestive heart failure. However, this is not a straightforward relationship. Although several studies have substantiated that obesity confers an independent and additive risk of all-cause and cardiovascular death, there is significant variability in these associations, with some lean individuals developing diseases and others remaining healthy despite severe obesity, the so-called metabol

Vascular restoration: Is there a window of opportunity?

The usage of drug eluting stents (DES) has markedly reduced the rates of coronary revascularization procedures compared with bare metal stents (BMS). However, this technology still faces challenges in terms of the prevention of late stent thrombosis, major adverse cardiac events (MACE) progression, and the catch-up phenomenon of restenosis. Restoration of endothelial function upon after stenting, therefore, is the key to mitigating the risk of these toxicities and determining the level of the efficacy and safety of an implant. Review of the clinical studies of multiple DES, has suggested that

Transgenic mice expressing mutant Notch3 develop vascular alterations characteristic of cerebral autosomal dominant arteriopathy with subcortical infarcts and leukoencephalopathy.

Cerebral autosomal dominant arteriopathy with subcortical infarcts and leukoencephalopathy (CADASIL) is an increasingly recognized adult-onset autosomal dominant vascular dementia, caused by highly stereotyped mutations in the Notch3 receptor. CADASIL is a widespread angiopathy characterized by a degeneration of vascular smooth muscle cells (VSMCs) and the abnormal accumulation of electron-dense granular material called GOM and Notch3 protein, because of an impaired clearance. Evidence that VSMCs are the primary target of the pathogenic process is supported by the restricted expression of Notc

Effect of hydrogen ion concentration on in vitro pulmonary vascular reactivity.

The present investigation studied the effects of hydrogen ion concentration on pulmonary vascular reactivity to the biogenic amines in vitro. Pulmonary arterial segments from cats were dissected free of surrounding tissue and endothelium and isometrically suspended in tissue baths. Cumulative dose-response curves were constructed to norepinephrine, histamine, serotonin, and potassium chloride, and the protocol was designed such that only one agent and hydrogen ion concentration was studied in each vessel segment. Alkalosis ([H+] less than or equal to 28 nmol/l) produced enhanced maximum respon

Pharmacological Profile of Purinergic P2Y Receptors Involved in Neurogenic Cardiovascular Modulation Induced by ADP and Related Compounds in Rats.

Cardiovascular modulation mediated by adenine-based purines is a very complicated and dynamic process driven by adenosine, adenosine diphosphate (ADP), and adenosine triphosphate (ATP). It is orchestrated through the interaction of these molecules with specific purinergic P1, P2Y, and P2X receptors, located throughout the central and peripheral nervous systems. This, along with the constant enzymatic interconversion of these compounds by ectonucleotidases, results in overlapping and highly variable cardiovascular responses. Activation of G protein-coupled purinergic P2Y receptors plays a funda

Inhibition of Hsp110-STAT3 interaction in endothelial cells alleviates vascular remodeling in hypoxic pulmonary arterial Hypertension model.

BACKGROUND: Pulmonary arterial hypertension (PAH) is a progressive and devastating disease characterized by pulmonary vascular remodeling which is associated with the malignant phenotypes of pulmonary vascular cells. Recently, the effects of heat shock protein 110 (Hsp110) in human arterial smooth muscle cells were reported. However, the underlying roles and mechanisms of Hsp110 in human pulmonary arterial endothelial cells (HPAECs) that was disordered firstly at the early stage of PAH remain unknown. METHODS: In this research, the expression of Hsp110 in PAH human patients and rat models was

Male and female high-fat diet-fed Dahl SS rats are largely protected from vascular dysfunctions: PVAT contributions reveal sex differences.

Vascular dysfunctions are observed in the arteries from hypertensive subjects. The establishment of the Dahl salt-sensitive (SS) male and female rat models to develop a reproducible hypertension with high-fat (HF) diet feeding from weaning allows addressing the question of whether HF diet-associated hypertension results in vascular dysfunction similar to that of essential hypertension in both sexes. We hypothesized that dysfunction of three distinct vascular layers, i.e., endothelial, smooth muscle, and perivascular adipose tissue (PVAT), would be present in the aorta from HF diet-fed versus c

Correlation of tissue drug concentrations with in vivo magnetic resonance images of polymer drug depot around arteriovenous graft.

Sustained delivery of anti-proliferative drugs to the perivascular area using an injectable polymeric platform is a strategy to inhibit vascular hyperplasia and stenosis. In this study, the concentrations of sirolimus in vascular tissues were evaluated after delivery using an injectable platform made of poly(lactic-co-glycolic acid)-polyethylene glycol-poly(lactic-co-glycolic acid) (PLGA-PEG-PLGA). In order to optimize the drug release profile, the effect of two solvents or solid loading of the sirolimus into the polymer gel was first examined in vitro. The early release was slower with loadin

Smooth muscle LRRC8A knockout reduces O 2 ·- influx, inflammation, senescence and atherosclerosis.

BACKGROUND: Leucine Rich Repeat Containing 8A (LRRC8A) anion channels (VRACs) associate with NADPH oxidase 1 (Nox1) and support extracellular superoxide (O 2 · - ) production, inflammation, and contractility in vascular smooth muscle cells (VSMCs). We proposed previously that VRACs also support influx of O 2 · - to localize cytoplasmic redox signals. METHODS: We assayed O 2 · - influx, assessed changes in mRNA expression (RNAseq), and tested multiple phenotypes of cultured LRRC8A knockout (KO) VSMCs. Aortic atherosclerotic burden and inflammation, and mesenteric vascular reactivity were compa

Cardiovascular responses produced by 5-hydroxytriptamine:a pharmacological update on the receptors/mechanisms involved and therapeutic implications.

The complexity of cardiovascular responses produced by 5-hydroxytryptamine (5-HT, serotonin), including bradycardia or tachycardia, hypotension or hypertension, and vasodilatation or vasoconstriction, has been explained by the capability of this monoamine to interact with different receptors in the central nervous system (CNS), on the autonomic ganglia and postganglionic nerve endings, on vascular smooth muscle and endothelium, and on the cardiac tissue. Depending, among other factors, on the species, the vascular bed under study, and the experimental conditions, these responses are mainly med

Uterine natural killer cell-mediated immune imbalance impairs uterine vascular remodeling and contributes to adverse pregnancy outcomes in dengue virus-infected mice.

Recent studies have shown that dengue virus (DENV) infection during pregnancy is linked to adverse pregnancy outcomes, presenting an emerging clinical challenge with poorly understood pathogenic mechanisms. Uterine vascular remodeling, a critical determinant of successful pregnancy, is predominantly regulated by uterine natural killer (uNK) cells. In this study, we explored the effect of uNK cells on uterine injury in a DENV-induced adverse pregnancy mouse model. Transcriptomic analysis revealed significant dysregulation of uterine vascular remodeling and immune response-related gene expressio

Cellular senescence at the bidirectional interface between ageing and coronary artery disease.

Coronary artery disease (CAD) is a major global cause of morbidity and mortality, and population ageing is reshaping its clinical phenotype, disease trajectory, and therapeutic complexity. Chronological age alone cannot explain the heterogeneity of CAD outcomes in older individuals, highlighting the need to integrate biological ageing into cardiovascular disease research and management. Cellular senescence, characterised by stable cell-cycle arrest, metabolic reprogramming, and acquisition of a senescence-associated secretory phenotype (SASP), provides a mechanistic bridge between ageing and C

Pirfenidone Attenuates Fibrosis and Neovascularization in 3D Spheroid-Laden Hydrogel Culture.

Fibrosis and angiogenesis are key contributors to synovial inflammation in both the early and progressive stages of rheumatoid arthritis (RA) and osteoarthritis (OA), making them important therapeutic targets to mitigate joint tissue damage. In vitro drug screening, particularly for antifibrotic and antiangiogenic efficacy, is a standard method for evaluating therapeutic candidates prior to in vivo testing. Traditionally, most studies have relied on two-dimensional (2D) monolayer cell cultures, which lack physiologically relevant cell-matrix and cell-cell interactions. Substantial evidence now

Ferroptosis in arterial atherosclerosis: mechanistic hypotheses, cell type specific vulnerabilities, translational biomarkers, and therapeutic opportunities.

Despite significant progress in lipid-lowering and anti-thrombotic therapies, atherosclerosis remains a leading cause of myocardial infarctions and ischemic strokes. Residual risk persists, often linked to sustained vascular inflammation, oxidative stress, and plaque instability-processes that converge on lipid peroxidation pathways within the arterial wall. Ferroptosis, an iron-dependent form of regulated cell death, occurs when antioxidant defenses fail, leading to toxic accumulation of phospholipid peroxides. Atherosclerotic plaques provide a permissive microenvironment rich in oxidized lip

Erectile dysfunction as an early biomarker of cardiovascular toxicity: Mechanisms and clinical implications.

Erectile dysfunction (ED) has been traditionally regarded as a quality-of-life disorder; however, accumulating evidence positions it as an early and clinically actionable biomarker of cardiovascular toxicity. ED and cardiovascular disease share fundamental pathophysiological mechanisms including endothelial dysfunction, reduced nitric oxide bioavailability, oxidative stress, systemic inflammation, autonomic imbalance, and vascular smooth muscle dysfunction. Because penile cavernosal arteries (~1-2 mm diameter) are substantially smaller than coronary arteries (~3-4 mm), the penile vascular bed

Transcriptome Insights into Protective Mechanisms of Ferroptosis Inhibition in Aortic Dissection.

Aortic dissection (AD) is a life-threatening vascular condition with limited pharmacological options, and shared risk factors with cardiac disease include hypertension, atherosclerosis, smoking, and dyslipidemia. This study investigated Ferrostatin-1 (Fer-1), a ferroptosis inhibitor, in a BAPN/Ang-II-induced mouse model of AD, revealing significant therapeutic potential. Fer-1 significantly reduced AD incidence and mortality by preserving aortic wall integrity. RNA sequencing identified 922 differentially expressed genes, with 416 upregulated and 506 downregulated. Non-canonical PKG1 regulation in cardiovascular health and disease.

It is well established that the cyclic GMP-dependent protein kinase I (PKG1) is canonically activated by cyclic guanosine monophosphate (cGMP), enabling its regulation of vascular tone, cardiac function and smooth muscle homeostasis. However, diverse non-canonical stimuli of PKG1 have also been identified. This includes oxidants, the immune-derived cyclic nucleotide 2'3'-cyclic-GMP-AMP (cGAMP), cross-activation by cyclic adenosine 3',5'-monophosphate (cAMP) and small molecular activators. These alternative regulatory mechanisms allow fine tuning of PKG1 activity and enable the regulation of di

[Role of the TGF-β1 signaling pathway in modulating in vivo adaptation of vascular grafts].

Intimal hyperplasia and pseudoaneurysm formation after vascular graft implantation are key factors affecting long-term graft patency. The transforming growth factor-beta 1 (TGF-β1) signaling pathway, through Smad-dependent and non-Smad pathways, differentially regulates smooth muscle cell phenotypic switching and proliferation, shapes the immune-inflammatory microenvironment, influences the quality of the re-endothelialization process, and maintains the balance between extracellular matrix synthesis and degradation, thereby playing a central role in the occurrence and progression of graft sten
