Motor deficits in the McGill-R-Thy1-APP transgenic rat model of Alzheimer's disease.

The McGill-R-Thy1-APP rat is a transgenic model of Alzheimer's disease (AD) which expresses APP with two mutations found in cases of familial AD, resulting in the development of amyloid pathology and cognitive deficits. Motor deficits are common symptoms of AD, emerging early in the disease, and are correlated with AD neuropathology and cognitive symptoms. This study evaluated hemizygous and homozygous McGill-R-Thy1-APP rats and their wildtype littermates for spontaneous alternation and locomotion in the T and Y mazes, and motor behaviour on an accelerating rotarod at 12-13 months of age. We f

Immunomodulatory effects of mesenchymal stem cell-derived extracellular vesicles on NLRP3 inflammasome activation in the adult mouse brain after chronic ethanol exposure.

Background NOD-like receptors (NLRs) and inflammasome complexes play critical roles in the neuroinflammatory responses triggered by chronic ethanol exposure. We previously demonstrated that mesenchymal stem cell-derived extracellular vesicles (MSC-EVs) attenuated binge-like alcohol consumption-induced NLRP3 inflammasome activation in the adolescent hippocampus; however, their broader effects on additional NLR pathways and brain regions remained unclear. Methods and Results This study investigates the therapeutic potential of intravenously administered adipose-derived MSC-EVs (20 µg/dose every

Retrotransposition Events Shape the Evolution of the Ataxin-3 Gene Family in Primates.

Evolutionary studies of disease-associated genes provide crucial insights into pathological mechanisms and potential therapeutic targets. Polyglutamine spinocerebellar ataxias (SCAs) are human neurodegenerative diseases caused by toxic expanded CAG repeats. Studies on SCA1 have shown that a paralog of the causing-gene can partially rescue protein function and alleviate the neuropathology. The most common SCA, Machado-Joseph disease (MJD/SCA3), caused by mutated ataxin-3 gene (ATXN3), has no treatment currently available. Its paralog ataxin-3 like (ATXN3L) remains largely unexplored. Here, we i

1-deoxysphinganine promoted microglial glycolytic reprogramming and neuroinflammation in alzheimer's disease.

Recent evidence suggests that microglial activation, driven by a metabolic shift towards glycolysis, was involved in the pathogenesis of Alzheimer's disease (AD). Although sphingolipid (SL) dysregulation has been linked to AD, the role of 1-deoxysphinganine (deoxySO), an atypical and neurotoxic SL, on microglial glycolytic reprogramming remains unclear. We measured serum deoxySO levels in AD patients and evaluated their association with cognitive performance. In APP/PS1 mice, we examined cerebral deoxySO level and the effects of deoxySO supplementation on cognitive function, neuropathology, an

Value and Use of Pediatric Postmortem Magnetic Resonance Imaging.

BACKGROUND: The gold standard for postmortem investigations of fetal and pediatric deaths involving neurological pathologies is a traditional autopsy; however, acquiring all diagnostic information from an autopsy alone can be challenging. In addition, there are substantial obstacles to performing traditional autopsies on the pediatric population such as parental opinion, religious beliefs, and cultural backgrounds. Postmortem computed tomography, sonography, and magnetic resonance (MR) imaging are less invasive alternative approaches for investigating central nervous system-related deaths. Alt

Synergistic-redundant dysfunction in autism spectrum disorder: Heterogeneity and molecular mechanisms.

Delineating the systematic functional integration and regional functional robustness in shared and individual-specific subspaces is critical to elucidating the neural basis underlying social and cognitive deficits in autism spectrum disorder (ASD). Here, integrated information decomposition and common orthogonal basis extraction approaches were combined to map individual synergistic and redundant interactions in shared and individual-specific subspaces based on functional connectivity networks derived from 842 individuals with ASD and 933 typically developing controls (TC), examined across age

Cytokines and related signaling pathways in traumatic brain injury.

Traumatic brain injury (TBI) represents a critical public health challenge with profound consequences for patients' neurological function and quality of life. The delayed secondary injuries following TBI can lead to devastating long-term sequelae encompassing cognitive deficits, emotional disturbances, post-traumatic epilepsy, and neurodegeneration manifested as Alzheimer's disease or Chronic Traumatic Encephalopathy (CTE). Emerging evidence highlights neuroinflammation as a pivotal mechanism driving secondary injury progression, establishing it as a prime therapeutic target in TBI management.

Tau Pathology in Chronic Traumatic Encephalopathy: Mechanisms and Diagnostic Advances.

Chronic Traumatic Encephalopathy (CTE) is a progressive neurodegenerative tauopathy associated with repetitive head impacts (RHI), yet it remains diagnosable only at post-mortem. Tau, a microtubule-associated protein, normally stabilizes neuronal microtubules and regulates cytoskeletal dynamics. Mechanical strain from RHI is thought to disrupt calcium homeostasis and kinase-phosphatase balance, driving hyperphosphorylation and phosphorylated-tau (p-tau) formation. This results in detachment from microtubules and subsequent p-tau aggregation. These mechanically-induced biochemical changes produ

Revisiting the cognitive potential of lithium in alzheimer's disease: the role of cholinergic system and brain connectivity.

Recent evidence demonstrating a marked reduction in endogenous brain lithium levels in mild cognitive impairment and Alzheimer's disease (AD) has renewed interest in lithium as a physiological modulator of brain aging and neurodegeneration. Experimental data indicate that lithium deficiency accelerates amyloid-β deposition, tau pathology, synaptic and myelin loss, whereas physiological lithium replacement mitigates neuropathology and cognitive decline. Here, we argue that lithium's cognitive potential in AD has been underestimated due to the historical neglect of two interrelated mechanisms: m

Cracking a New Era of Therapeutic Development in ALS.

Recent advances in genetics, RNA biology, disease modeling, biomarkers, and clinical trial methodology have fundamentally transformed therapeutic development in amyotrophic lateral sclerosis (ALS). Recent clinical successes, including edaravone, ultrahigh-dose methylcobalamin, and antisense oligonucleotide (ASO) therapies such as tofersen, have demonstrated that therapeutic success depends not only on effective drugs but also on improved therapeutic development strategies. These advances have highlighted several fundamental principles, including patient enrichment, intervention during appropri

MALDI Mass Spectrometry Imaging in Alzheimer's Disease Lipidomics: Matrix Selection, Spatial Lipid Pathology and Emerging Analytical Strategies.

Alzheimer's disease (AD) involves not only amyloid-β and tau pathology but also extensive disturbances in lipid metabolism, membrane organization, neuroinflammatory signaling, and tissue homeostasis. Conventional lipidomics has identified changes in phospholipids, sphingolipids, sulfatides, ceramides, gangliosides, and cholesterol-related pathways, but tissue homogenization removes their anatomical context. The aim of this review is to critically assess how matrix selection, sample preparation, ionization polarity, and emerging analytical strategies influence the detection and interpretation o

Glioneuronal Tumour With Neurocytic Differentiation (GNTN): a Molecularly Defined Tumour Type Formerly Referred to as Extraventricular Neurocytoma.

Tumours historically classified as extraventricular neurocytoma (EVN), initially in comparison with central neurocytoma, are defined by location and morphology; however, emerging molecular data demonstrate that this term is outdated. We report 14 previously unpublished tumours with a robust EVN DNA methylation profile, independent of original histopathologic diagnosis or anatomic location. Integrated molecular analyses identified FGFR1 alterations, particularly FGFR1::TACC1 fusions, as the predominant molecular driver of this tumour class. A smaller number of tumours harboured FGFR1 hotspot mu

Microglial efferocytosis dysfunction contributes to depression-like behaviors and cognitive impairment in aged mice following chronic sleep deprivation.

BACKGROUND: Chronic sleep deprivation (CSD), commonly observed in the elderly, is associated with an increased risk of neuroinflammation, depression, and cognitive impairment. Microglial efferocytosis plays a vital role in maintaining neural homeostasis and tissue repair. However, the mechanistic role of microglial efferocytosis in linking CSD to neurobehavioral impairments in aging remains largely undefined. METHODS: A 21-day CSD model was established in aged mice, and depression-like behaviors and cognitive function were evaluated. Hippocampal neuronal injury and neuroinflammation were eval

Neurosteroid-Mediated Neuroprotection via mTORC1/AMPK/BDNF Signaling Pathway in Alzheimer's Disease.

Alzheimer's disease (AD) remains the leading cause of dementia worldwide. AD is a neurodegenerative disease associated with progressive synaptic dysfunction, neuronal loss, and cognitive impairment. Current pharmacological therapies for AD offer only symptomatic relief and fail to halt or reverse the progression of the disease. It has been shown that neurosteroids (NSs), the endogenous modulators synthesized within the central nervous system (CNS), have emerged as promising therapeutic candidates in the management of AD by regulating of neuronal function, synaptic plasticity, and neurogenesis.

Antibiotic treatment reveals the contributions of the gut microbiome to CLN2 disease in the central and enteric nervous system.

The Neuronal Ceroid Lipofuscinoses (NCLs) are fatal inherited lysosomal storage diseases, with pronounced neuron loss in the central nervous system (CNS). Gastrointestinal issues are frequently reported by people with NCLs, although mechanisms underlying these symptoms are poorly understood. We recently demonstrated degeneration occurs within the enteric nervous system (ENS) in several NCLs. Given that the gut microbiome has been shown to be altered a CLN2 mouse model (Tpp1R207X/R207X) and may potentially influence both CNS and ENS pathology, we investigated the long-term impact of modulating

Polyamine metabolic enzyme SAT1 remodels the neuronal transcriptome and rescues α-synuclein toxicity in Drosophila.

Polyamine homeostasis is tightly regulated by interconversion and catabolic pathways and has been increasingly implicated in neurodegenerative disorders, including Parkinson's disease (PD), where accumulation of α-synuclein (α-Syn) perturbs neuronal homeostasis. Spermidine/spermine N¹-acetyltransferase 1 (SAT1) occupies a central position in polyamine interconversion, and alterations in SAT1 activity have been linked to α-Syn toxicity and PD-related neuropathology. To investigate how SAT1 activity influences α-Syn-associated neurodegeneration, we employed a Drosophila model of neuronal α-Syn e

NTN-1 attenuates amyloid-β-mediated microglial neuroinflammation and memory impairment via the NF-κB pathway and NLRP3 inflammasome in a rat model of Alzheimer's disease.

INTRODUCTION: Neuroinflammation driven by microglial activation represents a pivotal pathological mechanism underlying brain injury in Alzheimer's disease (AD), with NLRP3 inflammasome activation being a hallmark feature of this process. Netrin-1 (NTN-1) was recently shown to have potent anti-inflammatory and anti-apoptotic properties in a range of inflammatory diseases; however, its potential effect on neuroinflammation in AD treatment has not been well examined. Accordingly, this study aimed to investigate the effects of NTN-1 on cognitive impairment and to explore the anti-inflammatory prop

In Situ Activatable Hydrogen-Bonded Organic Framework-Based Nanozyme as NADH Peroxidase Mimic Restoring Homeostasis for Treatment of Alzheimer's Disease.

Nicotinamide adenine dinucleotide (NAD) serves as a vital regulator in metabolic networks, and the decrease in the level of its oxidized form, NAD+, is closely related to Alzheimer's disease (AD). However, unsatisfactory efficacy, non-targeted and imprecise treatment, along with interconnected pathogenic factors in AD, constrain the therapeutic efficacy of current AD therapeutic strategies. Herein, in lieu of complex multi-module treatments for pathogenesis, a bespoke in situ activatable hydrogen-bonded organic framework (HOF)-based nanozyme (denoted as NADH@Pre-Cu-HOF@KD8) has been constructe
