Gastrointestinal Stromal Tumor: History, Molecular Subtypes, and Risk Stratification.

The gastrointestinal stromal tumor (GIST) is one of the most common mesenchymal tumors of the gastrointestinal tract. Between the 1990s and early 2000s, GIST was identified as a tumor characterized by KIT or PDGFRA mutations, resulting in imatinib being established as an effective targeted therapy. However, with advances in molecular diagnostics, approximately 10%-15% of GISTs have been reported to harbor alternative mutations, such as those in the succinate dehydrogenase subunit genes and BRAF, leading to the development of additional targeted therapies. GISTs exhibit a wide spectrum of clini

Discovery of Cbl-b Targeting Cyclic Peptide Ligands by Phage Display.

Cbl-b is a RING-type E3 ubiquitin ligase and intracellular immune checkpoint that negatively regulates immune-cell activation. However, ligand discovery for Cbl-b remains limited, largely because the RING-type E3 ubiquitin ligase is regulated by shallow and extended protein-protein interaction interfaces that are difficult to target with conventional small molecules. Here, we employed a genetically encoded, macrocyclization-enabled phage-display library incorporating the cysteine-reactive noncanonical amino acid O2beY to identify cyclic peptide ligands for Cbl-b. Affinity selection followed by

The role of the locus coeruleus in giant migrating contractions of colonic motility in Suncus murinus.

Colonic contraction motility, composed of colonic motor complexes (CMCs) and giant migrating contractions (GMCs), is regulated through the coordinated integration of hormonal signals, the nervous system and intraluminal cues. Abnormalities in colonic function and motility can result in conditions such as constipation or diarrhea. It is believed that brain regions involved in regulating colonic motility play a role in colonic dysfunction observed under stress conditions and in Parkinson's disease. However, the detailed central mechanisms underlying colonic motility regulation remain unclear. Th

Transcriptional regulation of ventral hippocampus-nucleus accumbens circuit excitability drives cocaine seeking.

Ventral hippocampus (vHPC) CA1 pyramidal neurons send glutamatergic projections to nucleus accumbens (NAc), and this vHPC-NAc circuit mediates cocaine seeking and reward, but it is unclear whether vHPC-NAc neuron properties are modulated by cocaine exposure to drive subsequent behavior. The immediate early gene transcription factor FosB/ΔFosB is induced throughout the brain by cocaine and is critical for cocaine seeking, but its function in vHPC-NAc neurons is not understood. We now show that circuit-specific knockout of FosB/ΔFosB in vHPC-NAc neurons impaired cocaine reward expression and for

PRAME-Positive Eruptive Melanocytic Nevi in the Setting of BRAF-Inhibitors.

Preferentially Expressed Antigen in Melanoma (PRAME) is an immunohistochemical marker utilized in the diagnosis of melanocytic neoplasms. While still considered highly specific, both diffuse and variable PRAME immunoreactivity have been demonstrated in benign melanocytic lesions. This phenomenon is widely documented, as well as the phenomenon of eruptive melanocytic nevi (EMN) arising in the context of BRAF-inhibition. We report here a patient on encorafenib who developed numerous new pigmented lesions within 3 weeks of therapy. Initial biopsy suggested evolving melanoma in situ with 70%-75% i

Mapping Fos-immunoreactive neurons activated by intra-oral infusion of quinine, sucrose or water throughout the brain of B6 mice.

Fos immunohistochemistry was used to identify neurons in taste-related brain areas throughout the B6 mouse brain activated by intra-oral (IO) infusion of 3.0 mM quinine hydrochloride (Q), 1.0 M sucrose (S) or filtered water (W). IO infusion of Q and S elicited more Fos-immunoreactive (Fos-IR) neurons than W in the central medial (CM) and dorsomedial (DM) subareas of the parabrachial nucleus (PBN) and the central medial (CeM) amygdala (ps < 0.05). Infusion of Q led to more Fos expression than W in the central lateral (CL) PBN and the parvocellular reticular formation (PCRT; ps < 0.05). The only

Mutant KRAS in brain endothelial cells promotes vascular inflammation and impairs vascular integrity in brain arteriovenous malformation.

Somatic KRAS (KRASG12V) mutation in endothelial cells (EC) induces brain arteriovenous malformation (bAVM) that could lead to vascular instability and ultimately bleeding. However, the causes of bAVM instability remain unclear. Here we demonstrate that KRASG12V expressing cultured ECs (KRAS-G12V-EC) have increased expression of pro-inflammatory mediators and reduced expression of blood-brain-barrier (BBB) junction constituents. The conditioned medium from KRAS-G12V-EC can activate BV2-microglia (BV2-MG) and conditioned media from this primed BV2-MG can compromise the expression of EC-junction

Proteomic Signatures of Vemurafenib Resistance in Canine Urothelial Carcinoma Harboring the BRAFV595E Mutation.

Naturally occurring canine invasive urothelial carcinoma (InvUC) often harbors a BRAFV595E mutation, analogous to human BRAFV600E found across multiple cancer types, and has been used to investigate the effects of BRAF-targeted therapy. To investigate proteomic and phosphoproteomic changes during therapy, we analyzed 42 samples from 14 dogs with BRAFV595E-positive InvUC treated with vemurafenib. Tumors were collected via cystoscopy before treatment (Pre-Vem), 1 month into treatment (Vem-1-month), and at the time of progressive disease (Vem-PD). We observed consistent early suppression of prote

19q13 amplification with AKT2 and ERCC2 gains in sarcomatoid carcinoma of the urinary bladder.

BACKGROUND: Sarcomatoid carcinoma of the bladder is a rare and aggressive biphasic malignant neoplasm, representing approximately 0.1-0.3% of all bladder cancers. It is characterized by the coexistence of malignant epithelial and mesenchymal components. Its poor prognosis and limited systemic treatment options underscore the need for improved molecular characterization. CASE PRESENTATION: A 75-year-old male with chronic cystitis and squamous metaplasia of the bladder presented with acute urinary obstruction caused by a large bladder mass. Histopathological evaluation revealed a biphasic tumor

SHED-Exo delivery of miR-708-5p activates PI3K/AKT signaling pathway to alleviate glucocorticoid-induced osteogenic inhibition in BMSCs.

Steroid-induced osteonecrosis of the femoral head (SONFH) is the most prevalent type of non-traumatic femoral head necrosis. Exosomes (Exo) secreted from stem cells of human exfoliated deciduous teeth (SHED) play crucial roles in biological processes, particularly in tissue regeneration and repair. Previous studies have demonstrated that exosomes from SHED (SHED-Exo) can alleviate dexamethasone (Dex)-induced osteogenic inhibition in bone marrow mesenchymal stem cells (BMSCs). This study aims to further investigate the molecular mechanism by which SHED-Exo rescues BMSCs osteogenic inhibition. D

A Rare Case of Cutaneous Crystal-Storing Histiocytosis With Kappa Light Chain Restriction and Unusual BCL6 Expression.

Crystal-storing histiocytosis (CSH) is a rare histopathologic phenomenon characterized by the accumulation of crystalline material within histiocytes, most often associated with lymphoplasmacytic or plasma cell neoplasms. Cutaneous involvement is uncommon and may present diagnostic challenges. We report a 63-year-old woman with indurated plaques involving the face, chest, and upper extremities. Skin biopsies revealed aggregates of epithelioid histiocytes containing intracytoplasmic crystalline material, admixed with a lymphoplasmacytic infiltrate. The histiocytic cells were positive for CD68 a

Endothelial Klf9 fine-tunes Akt signaling to act as a transcriptional brake restraining retinal angiogenesis.

Retinal angiogenesis requires precise transcriptional regulation. Krüppel-like factor 9 (Klf9) has been implicated in various biological processes; however, its specific role in retinal vascular development and ocular neovascular disease remains unclear. In this study, we identified Klf9 as a critical transcriptional regulator of retinal vascular homeostasis. Spatiotemporal transcriptomic and single-cell RNA sequencing analyses revealed that Klf9 was highly enriched in retinal endothelial cells and upregulated during vascular maturation. Using genetic mouse models, we demonstrated that endothe

Spitz Nevi With Novel BRAF Fusions: A Report of Two Cases With Striking Morphologic Features.

Spitz neoplasms comprise a spectrum of melanocytic lesions with overlapping clinical and histologic features. BRAF fusions account for only 5%-6% of Spitz neoplasms with a limited number of fusion partners being characterized. We report 2 Spitz nevi with novel BRAF fusions and reproducible morphologic findings. The first case, a 35-year-old man with a finger mass, morphologically demonstrated intraepidermal and dermal proliferation of enlarged epithelioid melanocytes without maturation, and a striking pattern of neurotropism and angiotropism. Immunohistochemistry revealed <30% PRAME expression

Down-regulation of Laminin and its Correlated Significance to Interstitial Cells of Cajal in Hirschsprung's Disease.

INTRODUCTION: Hirschsprung's Disease (HSCR) is characterized by aganglionosis in the distal gut, but the role of Extracellular Matrix (ECM) components in its pathogenesis remains unclear. This study investigated the relationship between laminin, a key ECM protein, and Interstitial Cells of Cajal (ICC) in HSCR. METHODS: Immunofluorescence staining was used to analyze the expression and localization of laminin and ICC in paraffin-embedded colon sections from HSCR patients. Whole-mount preparations and confocal microscopy were employed to visualize the ICC network. Laminin and c-Kit expression l

Splenic B-Cell Lymphoma/Leukemia With TCL1 Gene Rearrangement.

BACKGROUND: Splenic B-cell lymphoma/leukemia includes a group of indolent small B-cell lymphomas/leukemias that share overlapping morphological and immunophenotypic features, making accurate diagnosis challenging. Specific gene rearrangements have not been commonly associated with splenic B-cell lymphomas/leukemias, and the TCL1 gene rearrangement-characteristic of T-cell prolymphocytic leukemia-has not been previously reported in B-cell lymphomas/leukemias. Here, we report two unique cases of splenic B-cell lymphoma/leukemia harboring an IGH::TCL1 rearrangement. CASE PRESENTATION: Patient #1

Protective effect of Lactococcus lactis NZ9000 secreting Bcl2 against Trichinella spiralis infection in mice.

BACKGROUND: Trichinellosis, a globally distributed zoonosis, threatens both human health and livestock productivity. The limited efficacy of existing anthelmintics underscores the need for innovative strategies to control Trichinella spiralis infection. RESULTS: A recombinant food-grade Lactococcus lactis strain (NZ-Bcl2) was engineered to secrete the anti-apoptotic protein Bcl2 by fusing the Bcl2 gene to the Usp45 signal peptide. Western blotting confirmed successful expression and secretion. NZ-Bcl2 strains exhibited stable growth, maintained plasmid inheritance over 24 generations without

Brucella proline racemase protein A targets Tpl2 to promote IL-10 secretion for establishment of chronic infection.

IL-10, an anti-inflammatory cytokine, plays a crucial role in limiting immune responses to pathogens, preventing host damage. However, the mechanisms underlying Brucella-mediated IL-10 production remain incompletely understood. In this study, we demonstrate that the proline racemase protein A (PrpA) of Brucella melitensis M5-90 induces macrophages to secrete IL-10 by activating the Tpl2-ERK signaling pathway, thereby promoting chronic infection. Moreover, Tpl2 deletion impairs macrophage bactericidal ability, accompanied by reduced TNF-α, IL-1β and IL-10 levels. Additionally, Trp309, Glu103, a

The ubiquitin ligase CBL and Fas-associated factor 2 cooperate to regulate the innate immune response to M. tuberculosis.

As a first line of host defense, macrophages must be able to effectively sense and respond to diverse types of pathogens, and while a particular response may be beneficial in some circumstances, it can be detrimental in others. Upon infection, Mycobacterium tuberculosis (Mtb) induces proinflammatory cytokines and activates antibacterial responses. Surprisingly, Mtb also triggers antiviral responses that actually hinder the ability of macrophages to restrict Mtb growth. In Mtb-infected macrophages, the ubiquitin ligase CBL suppresses antiviral responses and preserves the antibacterial capacity
