Inflammation and its discontents: the role of cytokines in the pathophysiology of major depression.

Recognition that inflammation may represent a common mechanism of disease has been extended to include neuropsychiatric disorders including … inflammatory signaling molecules, including nuclear factor kappa B, in part, through activation of sympathetic nervous system outflow pathways. Interestingly, depressed patients with increased inflammatory biomarkers have been found to be more likely to exhibit treatment resistance, and in several studies, antidepressant therapy has been associated with decreased inflammat …

Blockade of adrenoreceptors inhibits the splenic response to stroke.

Recent studies have highlighted the involvement of the peripheral immune system in delayed cellular degeneration after stroke. In the perman … nstrating the detrimental role of this organ in stroke-induced neurodegeneration. Activation of the sympathetic nervous system after MCAO results in elevated catecholamine levels both at the level of the spleen, through direct splenic innervation, and throughout the systemic circulation upon release from the adrenal medulla. These catecholamines bind to splenic alpha and be …

Fluid retention in cirrhosis: pathophysiology and management.

Accumulation of fluid as ascites is the most common complication of cirrhosis. This is occurring in about 50% of patients within 10 years of the diagnosis of cirrhosis. It is a prognostic sign with 1-year and 5-year survival of 85% and 56%, respectively. The most acceptable theory for ascites formation is peripheral arterial vasodilation leading to underfilling of circulatory volume. This triggers the baroreceptor-mediated activation of renin-angiotensin-aldosterone system, sympathetic nervous system and nonosmotic release of vasopressin to restore circulatory integrity. The result is an avid …

Integrative control of the lower urinary tract: preclinical perspective.

Storage and periodic expulsion of urine is regulated by a neural control system in the brain and spinal cord that coordinates the reciprocal activity of two functional units in the lower urinary tract (LUT): (a) a reservoir (the urinary bladder) and (b) an outlet (bladder neck, urethra and striated muscles of the urethral sphincter). Control of the bladder and urethral outlet is dependent on three sets of peripheral nerves: parasympathetic, sympathetic and somatic nerves that contain afferent as well as efferent pathways. Afferent neurons innervating the bladder have A-delta or C-fibre axons.

Hypertension and obesity after the menopause.

It is well known that in pre-menopausal women the incidence of cardiovascular events is lower than in men of the same age and that after the … of obesity-related hypertension recognizes a multifactorial mechanism including overactivity of the sympathetic nervous system, insulin resistance, leptin resistance, overactivity of the renin-angiotensin-aldosterone system and, finally, a blunted biological activity of the natriuretic peptides. The latter mechanism has been investigated by our group in recent years. Adipos …

Time course of direct cardiotoxic effects of high cocaine concentration in isolated rabbit heart.

Studies of acute cocaine cardiotoxicity are often confounded by the effects of cocaine on peripheral hemodynamics and an intact sympathetic nervous system. To study the direct effects of an acute dose of cocaine on heart, we used Langendorff-perfused isolated rabbit hearts. The hearts were attached to a Langendorff perfusion system through the aorta and were perfused with Krebs-Henseleit buffer. In nonpaced hearts, cocaine in concentrations of 10, 25, and 50 microM (reported to be in the range of drug concentrations lethal for humans) caused dose-dependent deterioration in heart contractility …

Comparison of hemodynamic and volume responses to different levels of lower body suction and head-up tilt.

Orthostatic challenge such as lower body suction (LBNP) or head-up tilt (HUT) induces marked shifts of blood from the cardiopulmonary into lower extremity vascular compartments. As a reaction to central hypovolemia, counterregulating mechanisms take place immediately, including central/peripheral circulatory responses and neurohumoral responses. Compensatory answers are exerted via autonomic reflexes, ie, cardiopulmonary and arterial baroreceptor mechanisms. The sympathetic nervous system increases heart rate and peripheral resistance to adjust the cardiovascular system to the stress situation…

Absence of fetal heart rate cycling on the intrapartum cardiotocograph (CTG) is associated with intrapartum pyrexia and lower Apgar scores.

BACKGROUND: Cycling consists of alternating periods of reduced and normal fetal heart variability, reflecting changes in fetal behavioral states. Occurrence of active and quiet sleep cycles is considered to be a hallmark of fetal autonomic nervous system integrity, demonstrating healthy interaction between the parasympathetic and sympathetic nervous systems. Cycling is an overlooked feature in most international cardiotocography (CTG) guidelines. The authors tested the hypothesis that fetuses showing no cycling in the intrapartum period have poorer outcomes. AIM: To investigate whether the ab…

There is a dearth of literature exploring the effect of concussion on HRV in youth athletes, and developmental differences preclude the application of adult findings to a pediatric population. Further, the current state of HRV methodology has prim …

Involvement of cyclic GMP and protein kinase G in the regulation of apoptosis and survival in neural cells.

Our current understanding of nitric oxide (NO), cyclic GMP (cGMP) and protein kinase G (PKG) signaling pathways in the nervous systems has its origins in the early studies conducted on vascular tissues during the late 1970s and early to mid-1980s. The pioneering research into the NO/cGMP/PKG pathway in blood vessels conducted by the laboratories of Drs. Ferid Murad, Louis Ignarro and Robert Furchgott ultimately led to the awarding of the 1998 Nobel Prize in Physiology or Medicine to these three scientists. On the basis of further pioneering studies by Drs. John Garthwaite, Solomon Snyder, Stev

Experimental study to localize the neurons projecting to the lamb retractor penis muscle.

Aim of the present study was to locate the neurons projecting to the lamb retractor penis muscle, a smooth muscle associated to the penis. The retrograde neuronal tracer Fast Blue was injected into the bulbopenile portion of the left retractor penis muscle. Labelled cells were found bilaterally in the S2-S4 spinal ganglia, from the last two lumbar (L5-L6 or L6-L7) to S4 sympathetic trunk ganglia and in the hypogastric and pelvic plexuses. Fast blue-positive (FB+) neurons were also found in the intermediate gray substance in the S1-S4 segments of the spinal cord. Our research enables us to desc

Differential effects of central angiotensin II and substance P on sympathetic nerve activity in conscious rats. Implications for cardiovascular adaptation to behavioral responses.

The centrally induced effects of angiotensin II and substance P on the cardiovascular system and on neuronal efferent activity of the splanc … pressor effects of angiotensin II are initiated by vasopressin release but become dependent on the sympathetic nervous system when vasopressin is absent or not effective. When rats were allowed to drink in response to angiotensin II, a further sharp rise in blood pressure occurred, together with increases in heart rate and splanchnic nerve activity. The results demonstrate …

Spinal cord injury reduces the efficacy of pseudorabies virus labeling of sympathetic preganglionic neurons.

The retrograde transsynaptic tracer pseudorabies virus (PRV) is used as a marker for synaptic connectivity in the spinal cord. Using PRV, we sought to document putative synaptic plasticity below a high thoracic (T) spinal cord transection. This lesion has been linked to the development of a number of debilitating conditions, including autonomic dysreflexia. Two weeks after injury, complete T4-transected and/or T4-hemisected and sham rats were injected with PRV-expressing enhanced green fluorescent protein (EGFP) or monomeric red fluorescent protein (mRFP1) into the kidneys. We expected greater

Kv1.3 channels in postganglionic sympathetic neurons: expression, function, and modulation.

Kv1.3 channels are known to modulate many aspects of neuronal function. We tested the hypothesis that Kv1.3 modulates the function of postganglionic sympathetic neurons. RT-PCR, immunoblot, and immunohistochemical analyses indicated that Kv1.3 channels were expressed in these neurons. Immunohistochemical analyses indicated that Kv1.3 protein was localized to neuronal cell bodies, processes, and nerve fibers at sympathetic neurovascular junctions. Margatoxin (MgTX), a specific inhibitor of Kv1.3, was used to assess the function of the channel. Electrophysiological analyses indicated that MgTX s

The role of Tc-99m bone imaging in the management of pain after complicated total hip replacement.

A case of reflex sympathetic dystrophy (RSD) after total hip replacement (THR) is reported. It is a very unusual cause of pain after any arthroplasty procedure. Three-phase bone imaging diagnosed the condition and sympathetic blockade was used for confirming and treating the condition. The role of the bone scan in differentiating RSD from other causes of pain after THR is highlighted.

Alpha 2A adrenergic receptor antagonism reduces fibrosis, inflammation and portal hypertension in models of chronic liver disease.

BACKGROUND & AIMS: Sympathetic nervous system over-activity is associated with liver disease progression and development of portal hypertension, influencing systemic inflammation. This study examined the role of the sympathetic alpha-2A adrenergic receptor (ADRA2a) and its antagonism in hepatic stellate cell (HSC) activation, a key event in fibrosis progression, in experimental metabolic dysfunction-associated steatohepatitis (MASH) and in portal pressure elevation in a cirrhotic-rat model. METHODS: An established bile duct ligation (BDL)-induced cirrhosis model (n = 14) was used to assess AD…
