In vivo and in vitro effects of doxycycline on leucocyte membrane receptors.

Tetracyclines, particularly doxycycline, have adverse effects on granulocyte function in vitro. We have examined the effects of doxycycline on membrane receptors for IgG (Fc gamma-R) and C3b (C3b-R) on granulocytes and lymphocytes, as well as on the sheep erythrocyte receptor (E-R) on T lymphocytes. Acne patients given doxycycline orally had a lower percentage of Fc gamma-R positive granulocytes (57%) than before treatment (80%) or compared to healthy controls (81%). Following in vitro doxycycline incubation, normal granulocytes showed decreased levels of Fc gamma-R positive cells. This effect

Changing patterns of hospital infections: implications for therapy. Changing mechanisms of bacterial resistance.

During the past decade there has been a marked increase in resistance of bacteria to antimicrobial agents. Microorganisms have developed the ability to make altered receptors for antimicrobial agents, have prevented agents from reaching their receptors within the bacterial cell, now have enzymes to destroy antibiotics, and have resistant metabolic pathways. Altered penicillin receptors, penicillin-binding proteins, have been found in Streptococcus pneumoniae, Streptococcus faecalis, Staphylococcus aureus, Neisseria gonorrhoeae, Clostridium, and Pseudomonas aeruginosa. Resistance based on decre

Mucopurulent cervicitis--the ignored counterpart in women of urethritis in men.

Among 100 randomly selected nonmenstruating women attending a clinic for sexually transmitted diseases, we assessed objective criteria for the clinical diagnosis of mucopurulent cervicitis. Visualization of yellow mucopurulent endocervical secretions on a white swab and the presence of 10 or more polymorphonuclear leukocytes per microscopical field (at a magnification of 1000) in satisfactory gram-stained endocervical smears were independently correlated with cervical Chlamydia trachomatis infection. Neither finding correlated with gonorrhea or genital herpes, although herpes caused characteri

Drug resistance in Shigella dysenteriae, S flexneri and S boydii in England and Wales: increasing incidence of resistance to trimethoprim.

A total of 2753 strains of shigella belonging to subgroups A, B, and C that were isolated from patients in England and Wales during the period from 1979 to mid-1983 were studied. Of these, 1690 (61%) were from patients recently returned from abroad or in contact with recent travellers, and 760 (45%) of these affected travellers from the Indian subcontinent. The number of strains resistant to sulphonamides and streptomycin remained at a high level throughout (average 76% and 72% respectively). Resistance to tetracyclines, ampicillin, and chloramphenicol rose, reaching 63%, 51%, and 48%, respect

Use of antibiotics in the outpatient setting.

Antibiotics with small but definable mortality, such as chloramphenicol, should not be used when safer drugs will suffice. Antibiotics with a low (1 to 5 per cent) morbidity should not be used when safer drugs are available. Therefore, cleocin, minocycline, or oral erythromycin estolate should rarely be used and regular erythromycin base is almost always preferable. Fever should not be treated with antibiotics since they are not antipyretics. "Colds" should not be treated with antibiotics, but antibiotics should be administered to patients with a history of chronic bronchitis, sinusitis, and r

Context-dependent sublethal hazard of glyphosate to non-target predators revealed by functional response analysis.

Glyphosate, one of the most widely used herbicides globally, is a significant environmental contaminant that may affect non-target organisms crucial to ecosystem services, including biological pest control. While glyphosate targets the shikimate pathway in plants, which is absent in animals, there is increasing evidence that glyphosate induces sublethal effects on arthropod predators. However, the factors modulating these effects remain unclear. Here, we investigated how physiological context, specifically pre-treatment with the broad-spectrum antibiotic tetracycline, commonly used to experime

Enhancing small-molecule-mediated translational control through multivalent RNA aptamers.

The precise translational control of gene expression by small molecules through RNA-based switches holds considerable promise for both research and therapeutic developments. However, current high-performance RNA switches remain limited in adaptability, with strong responses typically constrained to a narrow set of specific ligand-aptamer pairs. To address this limitation, we introduce a robust and generalizable RNA platform based on a multivalent aptamer design, which significantly enhances ligand-responsive protein expression through alternative splicing regulation. We have demonstrated that

Supramolecular DNA/amino acid-based oxidase-mimetic nanocatalysts exhibiting drug degradation capability.

Developing efficient and environmentally benign approaches for the remediation of antibiotic pollutants has become a paramount research imperative, since the extensive use of antibiotics has raised serious concerns due to their potential to induce antibiotic resistance and disrupt the ecological balance. In this work, we report the self-assembly of fluorenylmethyloxycarbonyl-lysine (Fmoc-K) aggregates with natural calf thymus DNA (CT-DNA) and Cu2+ to construct a catalyst that possesses copper-dependent active sites, mirroring the catalytic function of laccase, an oxidase known for its ability

Genomic characterization and diversity of tet(X4)-positive bacteria in poultry-associated fecal environments across multiple regions in China.

The emergence of tigecycline resistance genes, particularly tet(X) variants, has raised increasing concern as emerging antimicrobial resistance determinants in agro-environmental systems. Here, we conducted a large-scale environmental survey to characterize the burden and genomic features of tet(X)-positive bacteria in poultry-associated fecal matrices. A total of 514 chicken fecal samples were collected from fourteen poultry farms distributed across eight cities in five provinces of China, yielding 205 tet(X)-positive bacterial isolates. Escherichia coli was the predominant host species (72.2

Nitrogen-mediated engineering cobalt phosphide sites for enhanced peroxymonosulfate activation towards organic wastewater purification.

Introducing heteroatoms into cobalt phosphide-based materials is expected to enhance their effectiveness in wastewater purification through peroxymonosulfate (PMS) activation by altering the electronic structure and reconfiguring the catalytic sites. In this study, we describe an efficient one-step calcination method for fabricating CoP@NC/NF, wherein simultaneous phosphorization and nitrogen nitridation occur during the calcination process. The CoP@NC/NF/PMS system effectively activates PMS and achieves a tetracycline (TC) removal rate of 92.3 % within 30 min. The calculated rate constant ach

Dual S-scheme heterojunction via MOF-on-MOF strategy for efficient photoelectrocatalytic removal of organic contaminants: Detoxification and mechanism.

Accelerating the separation of carriers in the heterojunction plays vital role in the photoelectrocatalytic (PEC) process, yet it remains a challenging undertaking. Herein, a MOF-on-MOF based dual S-scheme heterojunction (BiVO4/NH2-MIL-125(Ti)/NH2-MIL-53(Fe), denoted as BVO/NM125/NM53) was rationally designed and prepared for PEC removing and detoxification of organic contaminants (phenol, tetracycline hydrochloride, ciprofloxacin and norfloxacin). The S-scheme heterojunction was double confirmed by DFT calculation and XPS analysis. The charge transfer resistance of BVO/NM125/NM53 photoanode d

Assessing interactions between antibiotics and triazine porous organic polymeric sorbents by photophysics.

This study purposes three triazine-based porous organic polymers (T-POPs 1-3) as advanced platforms for the early detection of antibiotic-polluted environments and effective water decontamination, in order to mitigate water pollution and antimicrobial resistance, which are two huge current problems damaging ecosystems and human health. T-POPs exhibited good performances as adsorbents for the removal of sulfamethazine (SMT) and tetracycline (TC) from water, with efficiencies up to 97% and 96%, and maximum adsorption capacities between (0.36-0.44) and (0.21-0.27) mmol g-1, respectively, which ar

Sex-dependent differences in behavioral and immunological responses to antibiotic and bacteriophage administration in mice.

INTRODUCTION: The problem of antibiotic resistance is a global one, involving many industries and entailing huge financial outlays. Therefore, the search for alternative methods to combat drug-resistant bacteria has a priority status. Great potential is seen in bacteriophages which have the natural ability to kill bacterial cells. Bacteriophages also have several advantages over antibiotics. Firstly, they are considered ecologically safe (harmless to humans, plants and animals). Secondly, bacteriophages preparations are readily producible and easy to apply. However, before bacteriophages can b

Combined antimicrobial effect of phage-derived endolysin and depolymerase against biofilm-forming Salmonella Typhimurium.

This study was designed to evaluate the antimicrobial activity of phage-derived endolysin (LysPB32) and depolymerase (DpolP22) against planktonic and biofilm cells of Salmonella Typhimurium (STKCCM). Compared to the control, the numbers of STKCCM were reduced by 4.3 and 5.9 log, respectively, at LysPB32 and LysPB32 + DpolP22 in the presence of polymyxin B (PMB) after 48-h incubation at 37 °C. LysPB32 + DpolP22 decreased the relative fitness (0.8) and the cross-resistance of STKCCM to chloramphenicol (CHL), cephalothin (CEP), ciprofloxacin (CIP), and tetracycline (TET) in the presence of PMB. Metaproteomic assessment of gut microbial and host functional perturbations in Helicobacter pylori-infected patients subjected to an antimicrobial protocol.

The impact of therapeutic interventions on the human gut microbiota (GM) is a clinical issue of paramount interest given the strong interconnection between microbial dynamics and human health. Orally administered antibiotics are known to reduce GM biomass and modify GM taxonomic profile. However, the impact of antimicrobial therapies on GM functions and biochemical pathways has scarcely been studied. Here, we characterized the fecal metaproteome of 10 Helicobacter pylori-infected patients before (T0) and after 10 days (T1) of a successful quadruple therapy (bismuth, tetracycline, metronidazole

Virulence Protein Pgp3 Is Insufficient To Mediate Plasmid-Dependent Infectivity of Chlamydia trachomatis.

Chlamydia trachomatis is the most common cause of infectious blindness and sexually transmitted bacterial infection globally. C. trachomatis contains a conserved chlamydial plasmid with eight coding sequences. Plasmid-cured Chlamydia strains are attenuated and display reduced infectivity in cell culture and in vivo genital infection of female mice. Mutants that do not express the plasmid-encoded proteins Pgp3, a secreted protein with unknown function, or Pgp4, a putative regulator of pgp3 and other chromosomal loci, display an infectivity defect similar to plasmid-deficient strains.
