Intermediate-dose busulfan and cyclophosphamide as a conditioning regimen for bone marrow transplantation in a case of Fanconi anemia in myelodysplastic transformation.

We report an 11-year old female with myelodysplastic (refractory anemia with excess of blasts) presentation of Fanconi anemia. After failure of initial chemotherapy with low doses of 6-mercaptopurine and prednisolone she underwent allogeneic bone marrow transplantation (BMT) from her HLA-matched sibling. Busulfan 8 mg/kg and cyclophosphamide 40 mg/kg were used as conditioning. The post-transplant course was uneventful with fast trilineage engraftment and mild cutaneous acute GVHD. She is alive 17 months after BMT with full hematological reconstitution without evidence of MDS.

Porphyria cutanea tarda and chronic lymphoid leukemia.

A case of a familial porphyria cutanea tarda (PCT-II) is reported in which the clinically overt form of PCT was provoked by factors relating to chronic lymphoid leukemia (CLL). Typical lesions of PCT developed on a 55-year-old woman after several blood transfusions and chlorambucil treatment. Besides these provoking factors, cytomegalovirus (CMV) infection was diagnosed. Erythrocyte uroporphyrinogen decarboxylase activity was about 50% of normal in the patient and in her two children. This case supports the suggestion that development of PCT in patients with hematological disorders is more tha

Relationship between DNA cross-links, cell cycle, and apoptosis in Burkitt's lymphoma cell lines differing in sensitivity to nitrogen mustard.

We surveyed 11 Burkitt's lymphoma cell lines for chemosensitivity to nitrogen mustard (HN2) in order to determine whether any simple correlates to cytotoxic response might be revealed. The lines tested varied over a 5-fold range in concentration of HN2 required to inhibit tumor cell growth by 50%. Drug sensitivity correlated neither with continental origin of tumor, growth fraction, presence of Epstein-Barr virus, nor with the precise locations of (8;14) translocation breakpoints. Furthermore, contrary to experience with other cell lines, no simple correlation was found between the HN2 sensiti

Abnormal regulation of DNA replication and increased lethality in ataxia telangiectasia cells exposed to carcinogenic agents.

The effect of different carcinogenic agents on the rate of semiconservative DNA replication in normal and ataxia telangiectasis (AT) cells was investigated. The rate of DNA synthesis in all AT cell strains tested was depressed to a significantly lesser extent than in normal cells after exposure to X-rays under oxia or hypoxia or to bleomycin, agents to which AT cells are hypersensitive. In contrast, inhibition of DNA replication in normal human and AT cells was similar after treatment with some DNA-methylating agents or mitomycin C. Colony-forming ability of AT cells treated with these agents

Impact of Ether Side-Chain Length and Anion Structure on the Physicochemical and Electrochemical Properties of Tetrahydrothiophenium Ionic Liquids.

A series of ionic liquids (ILs) based on tetrahydrothiophenium (Tht) cations bearing alkyl or ether side chains (containing one to four oxygen atoms), paired with bis(fluorosulfonyl)amide (FSA) or bis(trifluoromethanesulfonyl)amide (TFSA) anions, was synthesized to examine relationships among ether side-chain length, anion structure, and physicochemical properties. All samples showed glass transitions, with glass transition temperature increasing as the ether chain lengthened. Crystallization and melting behavior depended strongly on side-chain length, anion species, and cooling rate. ILs with

Parkinson's disease linked LRRK2 G2019S drives oxidative nuclear DNA damage and PARP1 hyperactive signaling.

LRRK2 mutations are the most common cause of autosomal-dominant Parkinson's disease (PD), with G2019S linked to both familial and sporadic PD. Although LRRK2-mediated mitochondrial DNA damage is implicated in PD, the contribution of nuclear DNA damage is less understood. Using CRISPR/Cas9-generated LRRK2G2019S/G2019S knock-in cells, we discovered increased sensitivity to oxidative and alkylating DNA-damaging agents compared to wild-type, consistent with compromised tolerance/repair of lesions processed by base excision repair (BER). The oxRADD assay revealed elevated endogenous oxidative nucle

Fisetin attenuates oxidative and haemolytic damage induced by quinuclidine derivatives in human erythrocytes.

Quinuclidine derivatives are potent bioactive compounds investigated as cholinesterase inhibitors with potential intravenous therapeutic application. However, the cytotoxic properties of some, particularly those linked to oxidative stress, pose significant safety concerns. This study aimed to evaluate if flavonoid fisetin could protect cells from such toxic effects. Erythrocytes were chosen due to high susceptibility to reactive oxygen species (ROS) and well-characterised endogenous antioxidant system, making them a sensitive and physiologically relevant model. Over 24 h, human erythrocytes we

Impact of ionic liquids on protein fibrillation: A comprehensive review.

Ionic liquids (ILs) have emerged as a versatile class of liquids with applications across diverse fields. Their unique properties arising from a diverse combination of ions allow the tuning of structure to serve a desired purpose. Beyond their broad applications, ILs have attracted increasing interest in protein science because of their ability to modulate protein structure, stability, and aggregation. This review provides an overview of how ILs influence protein amyloid formation, either inhibiting or promoting it depending on factors such as the type of cation and anion, alkyl chain length,

Noninvasive 11.7-T Magnetic Resonance Spectroscopy and Imaging Reveals Retinal Metabolic Alterations Induced by Blue Light Exposure.

The eye is a complex structure, with multiple systems involved in focusing and detecting light. Among them, the retina, an integral component of the central nervous system, is considered the most vital and exhibits the highest metabolic activity among all tissues in the human body. It interacts with light, and excessive exposure, especially to blue light, is prone to produce degeneration, mainly through oxidative reactions. This mechanism is involved in age-related macular degeneration or diabetic retinopathy. Animal research is important, considering the high prevalence of these diseases; yet

Homologous Self-Assembled Oligomers as Dual Anode/Cathode Interface Layers for Efficient Organic Photovoltaics.

Engineered interfacial layers that tune electrode energetics, suppress recombination, and enforce carrier selectivity are critical for high-efficiency organic photovoltaics (OPVs), yet current anode and cathode interface counterparts rely on distinct chemistries, increasing synthesis complexity and hindering scale-up. Here, we report a versatile molecular platform that generates both anode and cathode interface layers (AILs and CILs) from the homologous donor-acceptor-donor (carbazole-benzothiadiazole-carbazole) oligomer. Using a concise three-step route, we synthesized phosphonate-functionali

Nanogravure Printing for PFAS-Free Water-Repellent Textiles with Hierarchical Roughness.

The development of durable efficient water repellent textiles without perfluoroalkyl and polyfluoroalkyl substances (PFAS) is critical for sustainability. However, alternative alkyl-based coating solutions are generally less efficient. Here, we show how the hydrophobicity of a typical biobased fluorine-free coating (Lamoral A5) can be boosted by the multiscale design of the topographic spectrum of polyester fabrics from the submicrometer to the millimeter range. Using nanogravure printing, we fabricated mechanically robust textiles exhibiting nanopillared arrays superimposed on the intrinsic f

Control of Conformation and Spin for Programmable Singlet-Triplet Exciton Balance in Through-Space Charge-Transfer Emitters.

Achieving continuous and predictable control over singlet-triplet exciton partitioning in solution remains challenging for organic molecules, partly because electronic structure, conformational dynamics, and nonradiative decay are intertwined. Here we introduce a dual-key, coordination-activated through-space charge-transfer (TSCT) platform that enables programmable modulation of spin-dependent excited-state pathways in solution. Flexible donor-σ-acceptor ligands Cz-nC-TPP comprising a carbazole donor (Cz) and a terpyridine acceptor (TPP) linked by alkyl spacers (n = 2, 4, 6, 8) predominantly

Spontaneous Galvanic Electron Injection for Programmable Functionalization and Patterning of Monolayer Graphene.

Achieving high-dose, spatially programmable covalent functionalization of monolayer graphene has so far relied on externally powered activation─imposed electrochemical bias, highly reducing alkali metals, or laser/plasma inputs─approaches that are often aggressive and tie chemical dose to hardware-defined fields, limiting orthogonal control. Here, we introduce galvanic electron injection (GEI), a self-powered strategy that leverages the spontaneous interfacial potential at a metal-graphene junction to achieve programmable covalent functionalization under ambient conditions. The process is gove

Platinum-based therapeutics as emerging multi-modal radiosensitizers in glioblastoma treatment.

Glioblastoma (GBM) is a highly lethal brain cancer with poor patient prognosis and limited treatment options. Standard-of-care therapy involves maximally safe surgical resection followed by radiation therapy (RT) in combination with the alkylating agent temozolomide (TMZ) to treat the residual and brain-invading components of the disease. While RT remains a central part of definitive treatment, its efficacy is limited by intrinsic and acquired radiation resistance, making long-term tumor control and survival rare. While TMZ provides modest anti-tumor benefits, tumor resistance to TMZ and lack

DES-based eutectogel as a smart sorbent for selective microextraction of phthalates in physiological saline prior to GC-MS analysis.

In this study, a novel deep eutectic solvent (DES)-based eutectogel was developed as a functional and smart sorbent for the selective extraction and determination of phthalates in aqueous matrices using gas chromatography-mass spectrometry (GC-MS). Unlike conventional approaches, where DESs serve mainly as environmentally friendly extraction media, the proposed system exploits their role as active components incorporated into a polymeric matrix, enabling enhanced analyte-sorbent interactions and improved extraction selectivity. The developed method exhibited satisfactory analytical performance

Phosphatidylcholine and CHPT1 as Central Drivers of Chemoresistance in Colorectal Cancer: Lipidomic and Functional Insights.

Chemoresistance remains a major barrier to effective colorectal cancer (CRC) therapy, yet its metabolic underpinnings are poorly defined. Here, we integrate lipidomic profiling, enzymatic analysis, and functional perturbation approaches to elucidate the contribution of phosphatidylcholine (PC) metabolism and its biosynthetic regulator Choline Phosphotransferase 1 (CHPT1) to drug response. Comparative analysis of chemosensitive and chemoresistant CRC cell lines revealed that resistant HT29 cells exhibited significantly higher PC content and altered PC/lysophosphatidylcholine (LPC)ratios relativ

Precise End-Group Conjugated-Site Isomerization of Wing-Typed Dimeric Acceptors Enables 20.32% Efficiency Organic Solar Cells.

Molecular isomerization has been proven as an effective strategy to optimize the backbone geometry of photovoltaic materials and improve power-conversion-efficiency (PCE) of organic solar cells (OSCs). However, the precise isomerization of dimeric acceptors and their effect on optoelectronic and photovoltaic properties have been rarely studied. Herein, four isomeric wing-typed dimeric acceptors (named Dimer-1, Dimer-2, Dimer-3, and Dimer-4) with an alkylated linker are developed to investigate how different conjugated-sites of end-groups (methoxy-substituted EG1 and methoxy-free EG2) manipulat

Stability of Sodium Dodecyl Sulfate Micelles in Reline/Water Mixtures: A Molecular Dynamics Study.

Deep eutectic solvents of type III, such as reline, a mixture of choline chloride and urea, are a relatively new class of green solvents that display physical properties intermediate between conventional salt solutions and ionic liquids. Well-established models used to explain colloidal stability and macromolecule conformation in usual solvents, including the hydrophobic effect and the formation of the electrical double layer, are not applicable to these solvents and their mixtures with water due to strong nonideal behavior and high ionic strength of those solvents. Hence, in this work, molecu

Cosurfactant-Mediated Interfacial Flexibility: A Key to Controlling the Morphology of Anisotropic Nanostructures in Cetyltrimethylammonium Bromide Reverse Micelles.

Reverse micelles (RMs) have emerged as versatile soft templates for engineering anisotropic nanostructures. However, insights into how cosurfactant-mediated interfacial flexibility governs morphology remain limited, even though this parameter is critical for the rational design of anisotropic nanomaterials. In this study, we examine how cosurfactant-mediated interfacial dynamics in cetyltrimethylammonium bromide (CTAB)-based RMs direct the growth of copper oxalate (CuOx) nanostructures. By employing two distinct cosurfactants-1-butanol and 1-octanol-we demonstrate a morphology switch from nano
