Among the now pandemic sexually transmitted infections (STIs), Chlamydia trachomatis (C. trachomatis) is the predominant bacterial pathogen and human immunodeficiency virus type 1 (HIV-1) is the most lethal of the viral pathogens. The female genital tract is the primary site for heterosexual transmission of both C. trachomatis and HIV-1. Infection with C. trachomatis, and with a variety of other STIs, increases the risk for transmission of HIV-1, although the mechanisms for this finding remain unclear. We have used in vitro modeling to assess the mechanisms by which infection with genital C. t

The nociceptive nature of spinal dorsal horn neurons expressing NK1 and gamma-aminobutyric acid (GABA)(B) receptors was evaluated in the rat. Immunodetection of the Fos protein, induced by noxious mechanical stimulation of the skin, was combined with immunocytochemistry for NK1 or GABA(B) receptors (double-immunostaining study) or both receptors (triple-immunostaining study). Neurons double-labeled for Fos and for each receptor largely prevailed in lamina I. The proportions of Fos-positive cells immunostained for NK1 or GABA(B) receptors were higher in lamina I than in the remaining spinal lam

The events responsible for repair of DNA interstrand cross-links in mammalian cells, the proteins involved and their interactions with each other are poorly understood. The present study demonstrates that the structural protein nonerythroid alpha spectrin (alphaSpIISigma*), present in normal human cell nuclei, plays an important role in repair of DNA interstrand cross-links. These results show that alphaSpIISigma* relocalizes to nuclear foci after damage of normal human cells with the DNA interstrand cross-linking agent 8-methoxypsoralen plus ultraviolet A (UVA) light and that FANCA and the kn

This study examines the contribution of anion transporters to the swelling and intracellular acidification of glial cells from an extracellular lactacidosis, a condition well-known to accompany cerebral ischemia and traumatic brain injury. Suspended C6 glioma cells were exposed to lactacidosis in physiological or anion-depleted media, and different anion transport inhibitors were applied. Changes in cell volume and intracellular pH (pH(i)) were simultaneously quantified by flow cytometry. Extracellular lactacidosis (pH 6.2) led to an increase in cell volume to 125.1 +/- 2.5% of baseline within

The neonatal opossum (Monodelphis domestica) was used to assess how different populations of cells are generated in the olfactory region, and how they migrate along pathways to the central nervous system. Developing nerve cells were immunocytochemically labelled using antisera directed against two specific markers of olfactory receptor neurones: olfactory marker protein (OMP) and the dipeptide carnosine. In new-born opossums both carnosine and OMP are already co-expressed in primary olfactory neurones and in those axons that extend towards the olfactory bulb. Expression of these markers in olf

ON direction-selective (DS) ganglion cells were identified by electrophysiological recordings in DAPI labeled, isolated rabbit retinas. Their responses to a flashing spot were sustained. Their responses to moving stimuli were strong in the preferred direction and weak in the null direction. Injection of the recorded cells with Lucifer yellow revealed that the cells had a distinct dendritic morphology, consistent with that described previously (Buhl & Peichl, 1986; Amthor et al., 1989; Famiglietti, 1992a). When neighboring cells were injected, an extensive dendritic co-fasciculation was observe

This study investigated the effects of the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) spike protein (S protein) on hepatitis B virus (HBV) replication and immune function in patients with chronic hepatitis B (CHB), aiming to elucidate the underlying mechanisms of S protein-HBV interaction. Through in vitro cell experiments and clinical sample analyses, S protein expression was induced in HepG2.2.15 cells, leading to enhanced HBV replication; this effect was reduced by autophagy inhibitors, confirming the critical mediating role of autophagy. Furthermore, peripheral blood mono

Spatial transcriptomics captures context-dependent gene expression; however, existing workflows do not consistently account for measurement scale and often rely on a user-defined spatial neighbor graph, making results sensitive to this choice and limiting cross-study comparability. We present geneSCOPE (gene Spatial Co-Occurrence of Pairwise Expression), a framework that integrates ecology-inspired spatial statistics with network analysis to explicitly capture measurement scale and spatial information. In this framework, molecules are binned on a grid with a width selected near the mode of the

The incorporation of plant-derived fibers as co-encapsulating agents has emerged as a strategy to improve probiotic stability and functionality. This study evaluated grape pomace flour (GF) as a co-encapsulant in sodium alginate (SA) based microcapsules containing Lacticaseibacillus rhamnosus GG, focusing on structural properties, thermal resistance, antioxidant activity, gastrointestinal survival, and viability in orange juice. High encapsulation efficiencies were obtained for alginate (94.80%) and alginate-GF systems (91.13%), with viable counts above 8.3 log CFU/mL. FTIR, DSC, and TGA analy

INTRODUCTION: Galectin-9 (Gal-9) is a multifunctional protein belonging to the β-galactoside-binding lectin family playing crucial roles in immune regulation of tumor and pregnancy. However, its roles in placental pathophysiology such as preeclampsia have not been well understood. In this study the involvement of Gal-9 in trophoblast functioning and sustained immune balance were explored.
METHODS: Immortalized human trophoblast cells (HTR-8/SVneo) were subjected to chemical hypoxia (induced by CoCl2) and oxidative stress (induced by HMGB1), then treated with HIF-1α knockdown or specific inhib

Supercritical CO₂ extraction was applied to Heliotropium arbainense to evaluate how extraction pressure influences phytochemical composition and biological activity. Extractions of phenolic and flavonoid were performed at two pressures (200 and 550 bar), by using Supercritical CO₂. Phenolic and flavonoid compounds, were determined by HPLC analysis. Higher pressure extraction (550 bar) resulted in a greater extraction yield and a marked enrichment of phenolic and flavonoid compounds. Gallic acid (1496.01 µg/g) was the most prevalent component under SFE 1, followed by catechin (859.51 µg/g), rut

The Hsp70 molecular chaperone system is the front line of defense in maintaining cellular proteostasis. In eukaryotes, ATP/ADP nucleotide exchange in the Hsp70 chaperone cycle is stimulated by Hsp110, a divergent member of the Hsp70 chaperone superfamily and co-chaperone of Hsp70. Hsp110 is also a known modifier of neurodegenerative and other protein misfolding-related disorders. Biochemical aspects of Hsp110 chaperone functions have been characterized in vitro, and pathway interactions have been extensively characterized genetically in yeast model systems; however, a detailed understanding of

Immune checkpoints act like dimmer switches that keep immune responses in balance. In autoimmune disease, problems usually fall into four overlapping types: weak inhibitory signals, too much co-stimulation, metabolic or epigenetic rewiring of checkpoint pathways, and a mismatch between tissue and blood findings. This narrative review focused on human and translational studies from PubMed/Medical Literature Analysis and Retrieval System Online (MEDLINE), Excerpta Medica database (Embase), Web of Science, and Scopus (to 2025), prioritizing tissue-based data, trials of checkpoint agonists or co-s

Vehicle engine exhausts contain complex mixtures of gaseous and particulate pollutants, which are known to affect lung functions adversely. Many in vitro studies have shown that exposure to engine exhaust can induce oxidative stress in lung cells, leading to cellular inflammation and cytotoxicity. However, it remains challenging to identify key harmful components and their specific adverse effects via traditional toxicological assessments. Machine learning (ML) methods offer new ways of analyzing such complex datasets and have gained attention in predicting toxicity outcomes and identifying ke

Acyl CoA binding protein encoded by diazepam binding inhibitor (ACBP/DBI) is a tissue hormone that stimulates lipo-anabolic responses and inhibits autophagy, thus contributing to aging and age-related diseases. Protein expression profiling of ACBP/DBI was performed on mouse tissues to identify organs in which this major tissue hormone is expressed. Transcriptomic and proteomic data bases corroborated a high level of human-mouse interspecies conservation of ACBP/DBI expression in different organs. Single-cell RNA-seq data confirmed that ACBP/DBI was strongly expressed by parenchymatous cells fr
