In the occurrence and progression of breast cancer, tumor angiogenesis and metastasis play a central role. This indicates that anti-angiogenesis holds a significant position in anti-tumor therapy. Demethoxycurcumin is a bioactive diarylheptanoid compound extracted from Curcuma longa, a herb that serves both as food and medicine. As a member of the curcuminoid family, it exhibits anti-angiogenic and anti-tumor characteristics. But, the understanding of its potential mechanisms of action remains incomplete. Our experiments demonstrated that demethoxycurcumin significantly inhibited tumor growth

The tertiary C797S mutation in the EGFR tyrosine kinase domain disrupts osimertinib binding, driving resistance in NSCLC. We designed a series of novel diaminopyrimidine derivatives to establish the interaction with Ser797. H8b potently inhibited EGFRL858R/T790M/C797S kinase with an IC50 of 3.86 nM (30-fold lower than osimertinib) and suppressed both double mutant NCI-H1975 and triple mutant H1975-M3 cells with IC50s of 0.03 μM and 0.57 μM (14- and 11-fold reductions over osimertinib, respectively). Cocrystal X-ray analysis revealed that H8b forms a "two-point tethering" in the ATP pocket with

Pyruvate dehydrogenase kinase 1 (PDK1) is overexpressed in tumors, driving cancer progression by promoting the Warburg effect. Consequently, PDK1 inhibition represents a promising anticancer strategy, though potent low-toxicity inhibitors remain scarce. In this study, 20 new derivatives were designed and synthesized through further structural optimization of the dichloroacetate (DCA) derivatives, and their inhibitory effects on PDK1 and anticancer activities were systematically evaluated. Compound D16 emerged as the lead candidate, demonstrating potent PDK1 inhibition (53.20 % at 1 μM), signif

BACKGROUND: Liver fibrosis is a refractory disease whose persistence can eventually induce cirrhosis or even liver cancer. Early liver fibrosis is reversible by intervention. As a member of the transforming growth factor-beta (TGF-β) superfamily, bone morphogenetic protein 7 (BMP7) has anti-liver fibrosis functions. However, little is known about BMP7 expression changes and its potential regulatory mechanism as well as the relationship between BMP7 and TGF-β during liver fibrosis. In addition, the mechanism underlying the anti-liver fibrosis function of BMP7 needs to be further explored.

BACKGROUND: Non-alcoholic steatohepatitis-associated hepatocellular carcinoma (NASH-HCC) accounts for an increasing proportion of HCC cases. Currently, effective pharmacological options for treating both NASH and NASH-HCC remain limited, necessitating the identification of novel therapeutic agents. Our previous studies have demonstrated that ginger can ameliorate nonalcoholic fatty liver disease (NAFLD) and prevent the occurrence of NASH. The therapeutic effects and underlying mechanisms of NASH-HCC, however, remain poorly understood.
METHODS: Network pharmacology, bioinformatics, single-cell

BACKGROUND: Lipid metabolism dysregulation is a key characteristic of hepatocellular carcinoma (HCC) onset and progression. Elevated expression of immunoglobulin (Ig), especially the Igκ free light chain with a unique Vκ4-1/Jκ3 rearrangement in cancer cells, is linked to increased malignancy and has been implicated in colon cancer tumorigenesis. However, the role of Igκ in HCC carcinogenesis remains unclear. The aim of this study was to elucidate the pivotal roles of hepatocyte-derived Igκ in HCC development.
METHODS: The rearrangement sequence and expression level of hepatocyte-derived Igκ i

BACKGROUND AND PURPOSE: Carbon ion (C-ion) beams are concentrated to irradiate pancreatic carcinoma in the upper abdomen; however, this radiotherapy potentially causes adverse reactions in the gastrointestinal tract. FGF1 is a candidate radioprotector for radiation-induced intestinal damage, but may promote the malignancy of pancreatic cancer. An FGF1/CPP-C chimeric protein was created to enhance the intracellular signaling mode of FGF1 instead of FGFR signaling. The present study investigated the effects of FGF1/CPP-C on the intestinal adverse reactions of C-ion radiotherapy as well as its in

An inhibitor of the histone methyltransferase enhancer of zeste homologue 2 (EZH2), tazemetostat, has been developed for the treatment of B-cell lymphoma, but its mechanisms of action are not fully elucidated. We screened for genes targeted by tazemetostat in eleven B-cell lymphoma cell lines and found that tazemetostat significantly increased the expression of chemokine (C-C motif) ligand 17 (CCL17)/thymus- and activation-regulated chemokine (TARC) in all, which codes for a chemokine that is a hallmark of Hodgkin/Reed-Sternberg (H/RS) cells in Hodgkin lymphoma. Notably, gene set enrichment an

BACKGROUND: Cervical cancer (CC) remains a public health challenge in developing countries. Epidermal growth factor receptor (EGFR) is a receptor tyrosine kinase highly expressed in CC. Studies showed that PGE2 may transactivate EGFR in some models. However, understanding the interaction between the EGFR and PGE2 signaling pathways in CC is a key area for investigation.
METHODS: The Cancer Genome Atlas (TCGA) was used to evaluate gene expression correlation and clinical outcomes. Panitumumab and aspirin were employed to inhibit EGFR and COX-1/2, respectively. Cell migration was evaluated usin

BACKGROUND: Dysregulation of N6-methyladenosine (m6A) RNA methylation plays a crucial role in colorectal cancer (CRC) development. Despite the established oncogenic role of methyltransferase-like 3 (METTL3) in CRC, the mechanisms behind its tumor-promoting functions are not fully understood.
METHODS: Functional impacts were assessed by measuring cell viability, invasion, colony formation, migration, apoptosis, and M2 macrophage polarization in vitro, as well as in vivo tumorigenicity. The interaction between METTL3 and claudin 2 (CLDN2) was validated via RNA immunoprecipitation and luciferase
