Male infertility affects about 7% of the general male population, and it is a multifactorial, polygenic pathological condition. Known genetic factors, accounting for about 20-25% of male factor infertility, are present in each etiological category: i) hypothalamic-pituitary axis dysfunction; ii) quantitative and qualitative alterations of spermatogenesis; iii) ductal obstruction/dysfunction. Areas covered: All routinely available genetic tests are described. Indication for testing for chromosomal anomalies and Y chromosome microdeletions is based on sperm count (severe oligozoospermia/azoosper

BACKGROUND: Osteoarthritis (OA) is a multifactorial disease with strong genetic and occupational components. Although published studies have described several risk factors for OA, very few studies have investigated the occupational and genetic factors that contribute to this debilitating condition.
OBJECTIVE: To describe occupational and genetic factors that may contribute to the risk of developing (OA).
METHODS: A literature search was conducted in PubMed using the search terms osteoarthritis, occupation, work, and genetics.
RESULTS: Heavy physical work load was the most common occupationa

Subarachnoid hemorrhage (SAH) due to rupture of an intracranial aneurysm (IA) is a devastating condition with high mortality and morbidity. Genetic as well as environment factors play important roles in the pathogenesis of SAH and IAs. We review the present knowledge on the genetic factors responsible for SAH or IAs. Linkage analysis and association study are used for genetic dissection. Genome-wide linkage analyses have specified several genetic loci for IAs and 6 loci (1p34-36, 7q11, 11q24-25, 14q22-31, 19q13, and Xp22) have been replicated in different populations. A complete understanding of the genetic architecture of eating disorders requires adequately large sample sizes from individuals of all ancestries. Failure to include representative samples truncates understanding and may even exacerbate health disparities. Several countries in Asia have made rich contributions in psychiatric genetics; however, the eating disorders field requires concerted global efforts to increase representation from Asian ancestry populations to ensure that our global efforts accurately reflect the true distribution of eating disorders around the world and across ancestries

Celiac disease is an autoimmune disease affecting the small intestine, triggered by gluten sensitization in genetically susceptible individuals worldwide. Celiac disease development is strongly linked to the presence of HLA-DQ2 and/or DQ8, which present the immunogenic gluten peptides and trigger the immune response leading to pathogenesis. Because of the variability of clinical symptoms, the disease is often underdiagnosed. Intestinal biopsy and the presence of antibodies to deamidated gliadin and tissue transglutaminase are recommended diagnostic tools. Genetic testing for HLA DQ2 and DQ8 ca

Myoclonus-dystonia (M-D) is an autosomal-dominant movement disorder with onset in the first two decades of life. Mutations in the epsilon-sarcoglycan gene (SGCE, DYT11) on chromosome 7q21-q31 represent the major genetic cause of M-D in some populations. The syndrome was related with mutations in two other genes (DRD2 and DYT1). A second locus has been reported in one large M-D family (DYT15, 18p11), but no gene has been identified yet. In this review, we discuss genetic aspects of myoclonus-dystonia.

Autosomal recessive familial amyotrophic lateral sclerosis (RFALS) is a rare form of ALS that usually presents at an early age with slow progression of symptoms. RFALS is clinically and genetically heterogeneous and the locus of RFALS type 3 was mapped to 2q33 (ALS2) in a single family. We now report linkage of a more-common form of RFALS to chromosome 15q15-q22 markers (ALS5) and show further genetic locus heterogeneity in RFALS. ALS5 is the locus for most families with RFALS and appears to be present in both North African and European populations.

Metabolism of serotonin was studied in pineal gland of Wistar rats with low and high rate of audio excitability as well as in the Krushinsky-Molodkina rat population genetically predisposed to audio-dependent convulsions. Intensive turnover of serotonin via N-acetyl serotonin into melatonin was found in pineal gland of rats insensitive to sensory stimulus, whereas the rate of this response was decreased in the gland tissue of animals sensitive to audio effects as well as inhibition of serotonin turnover into melatonin was noticed in pineal gland of rats predisposed genetically to audio-depende

The predisposition to autoimmune disease has been termed an autoimmune diathesis. The genetic basis for this predisposition has been analyzed, using three different models: juvenile thyroiditis in humans, spontaneous thyroiditis in chickens, and experimental thyroiditis in mice. At least three different categories of genetic factors contribute to the development of autoimmunity. They include an increased immune response due to immunoregulatory genes associated with the major histocompatibility complex, the balance of thymus-derived lymphocyte population with helper/inducer vs suppressor proper

Alpacas (Vicugna pacos) are of increasing economic importance, mainly due to their high-quality fiber. This study aimed to develop and validate molecular tools for sex identification, individual identification and parentage testing, and to characterize genetic diversity and population structure in Huacaya alpacas bred in Poland. Forty-five previously described microsatellites and one newly developed SRY-based marker were organized into three multiplex panels containing sixteen, seventeen and thirteen markers. The SRY marker was validated in 17 reference animals of known sex (5 males, 12 female

Our understanding of the influence of ancestral background on genetically determined expression remains limited, especially when gene expression models are applied to studies from different or multiple populations. We perform transcriptome-wide association studies of 6 psychiatric conditions, leveraging gene expression models trained in cohorts with different proportions of African, European, and Indigenous American genetic ancestries. For comparison, we repeat each transcriptome-wide association study using a model trained in individuals of predominantly European ancestry. We identify 1416 st

Whole-genome duplication (WGD) is often considered as either an adaptive dead-end or a facilitator of rapid adaptation under changing environments. How diploids vs tetraploids out of WGD respond to selection remains, however, poorly understood. Using whole-genome sequence data and phenotypic evidence from transplant experiments of diploid and autotetraploid populations of Biscutella laevigata (Brassicaceae) sampled at low and high elevation in a factorial design, we here assessed how genomic and phenotypic variation responded to selection across elevation in each ploidal level. While all popul

The Government of India has launched the National Sickle Cell Anaemia Elimination Mission, a significant public health initiative targeting a genetic disorder that disproportionately affects the nation’s tribal populations. Sickle cell disease causes severe lifelong complications, and this mission aims to eliminate it as a public health problem by 2047. The primary objectives are to provide accessible, high-quality care to all patients and to reduce the disease’s prevalence through targeted screening and awareness. The mission’s comprehensive strategy is structured around three pillars: Health

The arrival of the Neolithic way of life in Europe is associated with the migration of Anatolian farmers and their admixture with local hunter-gatherers. In the Paris Basin (Northern France), the Middle Neolithic Cerny culture (4700-4300 bce) combines agricultural practices with funerary items linked to hunting, suggesting a particular connection with the local descendants of Mesolithic hunter-gatherers. The monumental burials gathering only a handful of deceased indicate the development of an elite. To better understand Cerny social structures, we generated genome-wide data for 28 individuals

There are well-known associations between various types of adult-onset gynecological cancers and tumor predisposition syndromes, but less is known about gynecological tumor predisposition syndromes in the pediatric and adolescent population. With advancing genetic technology and more extensive testing, we have come to learn that ∼10%-20%, if not more, of benign and malignant pediatric neoplasms result from a germline variant in a tumor predisposition gene. Gynecological neoplasms found in this population can be inherited, but they can also arise from a de novo gene variant. So too, some syndro

Nakaseomyces glabrata (former name: Candida glabrata) is the second most common cause of vulvovaginal candidiasis (VVC), but its molecular epidemiology and antifungal resistance in China remain poorly understood. This study analysed 204 N. glabrata isolates from VVC patients in Suzhou, Eastern China, using multi-locus sequence typing (MLST) and microsatellite genotyping, alongside antifungal susceptibility testing. A total of 46 sequence types (STs) were identified by MLST, as well as 146 genotypes (GTs) revealed by microsatellite. According to MLST, ST7 was the predominant ST in vaginal N. gl

Background: Dyggve-Melchior-Clausen syndrome (DMC) is a rare autosomal recessive skeletal dysplasia characterized by dwarfism, coarse facial features, and intellectual disability. Caused by loss-of-function variants in the DYM gene, which encodes dymeclin, DMC is predominantly reported in consanguineous populations but remains poorly studied in South America. Methods: We report a 21-year-old Ecuadorian male with clinical features suggestive of DMC. Comprehensive clinical, radiological, and genetic evaluations were conducted, including clinical exome sequencing and Sanger sequencing, followed b

Skin, hair, and eye (oculocutaneous) color is due to melanin, a pigment produced by melanocytes. This review considers processes required for pigmentation and the complex genetic network that regulates them. The first requisite is migration of neural crest-derived melanoblasts, which populate various embryonic sites, then differentiate into melanocytes or seed stem cell niches. Differentiation is marked by expression of genes essential for melanogenesis, which takes place in melanosomes and involves conversion of tyrosine into melanin. Melanosome biogenesis requires premelanosome maturation th

Intrahepatic Cholangiocarcinoma (iCCA) with FGFR alterations is relatively rare, and its identification is important in the era of targeted therapy. We collected a large series of FGFR-altered cases in the Chinese population and characterized their clinicopathological and genetic features. Among the 18 FGFR-altered cases out of 260 iCCAs, 10 were males and 8 were females, ranging in age from 35 to 74 years (mean, 57.3 years; median, 58 years). Pathologically, they include 9 cases of large duct (LD, 50 %) and small duct (SD, 50 %) types each. All of them (100 %, 18/18) showed microsatellite sta
