The roles of testicular nuclear receptor 4 (TR4) in male fertility-priapism and sexual behavior defects in TR4 knockout mice.

BACKGROUND: Successful reproductive efforts require the establishment of a situation favorable for reproduction that requires integration of both behavior and internal physiological events. TR4 nuclear receptor is known to be involved in male fertility via controlling spermatogenesis, yet its roles in regulating other biological events related to reproduction have not been completely revealed. METHODS: Male TR4 knockout (TR4 -/-) and wild type mice were used for the sexual behavior and penile dysfunction studies. Mice were sacrificed for histological examination and corresponding genes profil

The CCN proteins: important signaling mediators in stem cell differentiation and tumorigenesis.

The CCN proteins contain six members, namely CCN1 to CCN6, which are small secreted cysteine-rich proteins. The CCN proteins are modular proteins, containing up to four functional domains. Many of the CCN members are induced by growth factors, cytokines, or cellular stress. The CCNs show a wide and highly variable expression pattern in adult and in embryonic tissues. The CCN proteins can integrate and modulate the signals of integrins, BMPs, VEGF, Wnts, and Notch. The involvement of integrins in mediating CCN signaling may provide diverse context-dependent responses in distinct cell types. CCN

Functional analysis of VopF activity required for colonization in Vibrio cholerae.

Vibrio cholerae, a Gram-negative facultative pathogen, is the etiologic agent for the diarrheal disease cholera. We previously characterized a clinical isolate, AM-19226, that translocates a type III secretion system (T3SS) effector protein with actin-nucleating activity, VopF, into the host cells. From comparative genomic studies, we identified a divergent T3SS island in additional isolates which possess a VopF homolog, VopN. Unlike the VopF-mediated protrusion formation, VopN localizes to stress fiber in host cells similarly to VopL, which is present in the pandemic strain of Vibrio parahaem

Mechanics of carotid arteries in a mouse model of Marfan Syndrome.

Mouse models of Marfan Syndrome (MFS) provide insight into the type and extent of vascular abnormalities manifested in this disease. Inclusion of the mgR mutation causes the otherwise normal extracellular matrix glycoprotein fibrillin-1 to be under-expressed at 15-25% of its normal level, a condition seen in MFS. Aortas in patients with MFS are generally less distensible and may experience dissecting aneurysms that lead to premature death, yet little is known about effects on other large arteries. In this study, common carotid arteries from mice heterozygous (R/+) and homozygous (R/R) for the

Metabolic profiling of kidney and urine in rats with lithium-induced nephrogenic diabetes insipidus by (1)H-NMR-based metabonomics.

Lithium (Li) treatment for bipolar affective disorders is associated with a variety of renal side effects. The metabolic response of the kidney to chronic Li treatment has rarely been studied. We applied a novel method of (1)H-nuclear magnetic resonance (NMR)-based metabonomics to integrate metabolic profiling and to identify the changes in the levels of metabolites in the kidney and urine from rats with Li-induced NDI. Metabolic profiles of urine and kidney homogenate [3 different zones (cortex, outer medulla, and inner medulla) or whole kidney] were investigated using high-resolution NMR spe

Animal models that elucidate basic principles of the developmental origins of adult diseases.

Human epidemiological and animal laboratory studies show that suboptimal environments in the womb and during early neonatal life alter development and predispose the individual to lifelong health problems. The concept of the developmental origins of adult diseases has become well accepted because of the compelling animal studies that have precisely defined the outcomes of specific exposures such as nutrient restriction, overfeeding during pregnancy, maternal stress, and exogenously administered glucocorticoids. This review focuses on the use of animal models to evaluate exposures, mechanisms,

High mobility group A2 potentiates genotoxic stress in part through the modulation of basal and DNA damage-dependent phosphatidylinositol 3-kinase-related protein kinase activation.

The high mobility group A2 (HMGA2) protein belongs to the architectural transcription factor HMGA family, playing a role in chromosomal organization and transcriptional regulation. We and others have previously reported that ectopic HMGA2 expression is associated with neoplastic transformation and anchorage-independent cell proliferation. Here, we reported a correlation between increased HMGA2 expression and enhanced chemosensitivity towards topoisomerase II inhibitor, doxorubicin, in breast cancer cells. Using cells exhibiting differential HMGA2 expression and small interfering RNA technique,

Redox-dependent effects of nitric oxide on microvascular integrity in oxygen-induced retinopathy.

Opposing effects have been ascribed to nitric oxide (NO) on retinal microvascular survival. We investigated whether changes in the redox state may contribute to explain apparent conflicting actions of NO in a model of oxygen-induced retinal vasoobliteration. Retinal microvascular obliteration was induced by exposing 7-day-old rat pups (P7) for 2 or 5 days to 80% O(2). The redox state of the retina was assessed by measuring reduced glutathione and oxidative and nitrosative products malondialdehyde and nitrotyrosine. The role of NO on vasoobliteration was evaluated by treating animals with nitri

Efficacy of a Self-Guided Transdiagnostic Intervention for Adults With Anxiety and Depression: Randomized Controlled Trial.

BACKGROUND: Anxiety and depressive disorders affect hundreds of millions globally, with substantial barriers limiting access to evidence-based treatments like cognitive behavioral therapy. Digital mental health interventions offer potential solutions to improve access to support. However, evidence of efficacy through randomized controlled trials is essential for clinical adoption. OBJECTIVE: This study evaluated the efficacy of the Headspace Anxiety and Depression Program, a digitally delivered 21-session intervention grounded in the Unified Protocol that integrates cognitive behavioral and m

[Optimizing the response to epidemics of meningococcal meningitis: report of a workshop of experts at CERMES (Niamey, Niger, 12th to 14th January 1998)].

Recent meningitis epidemics in West Africa have drawn attention to shortcomings in the response of the health services. The health ministries of the countries involved have identified particular requirements. Following WHO recommendations, OCCGE organized a meeting of experts at CERMES, Niamey, in January 1998. The aim of this workshop was to consider the problems common to these countries, identify their needs and to produce concrete recommendations defining the roles of OCCGE and CERMES. Difficulties in mobilization, as no procedure had been established, and a lack of resources limited the e

Changes in olfactory function in children and adolescents with obesity following an inpatient weight loss program: a longitudinal study.

BACKGROUND: Obesity is a major global public health burden, yet the mechanisms underlying interindividual variability in treatment response remain poorly understood. Olfactory function influences eating behavior and may differ in individuals with obesity. However, evidence in pediatric populations is limited, and their responsiveness to weight loss remains unclear. This study aimed to assess olfactory function in children and adolescents with obesity before and after an inpatient weight loss intervention. Specifically, we examined whether olfactory performance is within normative values and wh

Pharmacological, clinical, and translational evidence for N,N-dimethyltryptamine, 5-methoxy-N,N-dimethyltryptamine, and zalsupindole: a systematic review.

Short-acting tryptamine psychedelics are emerging as candidate psychiatric therapeutics with pharmacological profiles that may offer practical advantages over longer-acting psychedelics. N,N-dimethyltryptamine (N,N-DMT) and 5-methoxy-N,N-dimethyltryptamine (5-MeO-DMT) differ in pharmacokinetic, receptor, and phenomenological properties, while zalsupindole is a putatively non-hallucinogenic analogue. This systematic review aimed to synthesize pharmacological, clinical, safety, and preclinical behavioural evidence for these compounds. Ovid databases and PubMed were searched from inception to Dec

Naringenin-loaded liposomal nanoparticles attenuate chemotherapy-induced gastric mucositis: Histopathological and ultrastructural evidence with modulation of NRF2/HO-1, NF-κB/COX-2, and apoptotic pathways.

Chemotherapy-induced gastric mucositis is a frequent complication of 5-fluorouracil (5-FU) treatment, characterized by severe histopathological and ultrastructural injury accompanied by oxidative stress, inflammation, apoptosis, and disruption of the gastric mucosal barrier. The present study investigated the gastroprotective effect of naringenin-loaded liposomal nanoparticles (NAR-LNPs) against 5-FU-induced gastric injury in rats and compared their efficacy with free naringenin (NAR). NAR-LNPs were prepared using a modified thin-film hydration method and characterized before biological evalua

Beyond the germ: Rethinking implant-related infections as a host-microbiota-biomaterial ecosystem.

Implant-related infections remain one of the most challenging complications in modern medicine, often leading to implant failure, revision surgery, significant patient morbidity and high rate of patient mortality. Traditionally framed within the germ theory paradigm, their pathogenesis has been attributed primarily to microbial contamination and biofilm formation. However, growing evidence reveals a far more complex picture in which infection susceptibility emerges from the dynamic interplay between host biology, microbiota composition, biomaterial properties, and environmental influences . Orchestrating the gut microbiota-mitochondrial-immune axis in gynecological diseases: mechanisms and dual-targeting therapeutic strategies.

The "gut microbiota-mitochondria axis" has become the core hub connecting the metabolism, immunity, and endocrine regulation of gynecological diseases. In this review, the hierarchical regulation mechanism of this axis is systematically combed: at the upstream level, intestinal short-chain fatty acids (SCFAs), bile acids (BAs), tryptophan derivatives, and other metabolites can activate AMPK/PGC-1α, FXR/TGR5, and AhR-mediated energy sensing and receptor signaling pathways; On the functional level, bacterial lipopolysaccharide-TLR4 signal and cGAS-STING/NLRP3 inflammasome pathway activated by cy

Synergistic Effects of Herbal Bioactive Compounds and Growth Factors on Osteogenic Differentiation of Dental Derived Mesenchymal Stem Cells.

Repairing large bone defects is still not an easy task and is considered an important challenge in regenerative medicine and maxillofacial surgery. The methods that currently exist each have their own limitations. Bone grafting from the patient's own body is known as the best method, but it is not always possible. This is because the donor site may get damaged, the amount of bone that can be taken is limited, and the surgery itself is complicated. In recent years, stem cells that are isolated from dental tissues have received a lot of attention. These cells proliferate quickly, have the abilit

Metal-dependent regulated cell death: Molecular architecture and translational frontiers.

Intracellular metal dyshomeostasis has emerged as a key regulator of specialized regulated cell death (RCD) programs, challenging classical views that regard necrosis as entirely accidental. This review systematically delineates the molecular architecture and translational trajectories underlying metal-dependent RCD, including iron-driven ferroptosis, copper-mediated cuproptosis, and additional emerging modalities such as calcicoptosis, necrosis by sodium overload (NECSO), and the newly designated zincoptosis, mnoptosis, and coptosis. We examined distinct execution mechanisms, ranging from mem

When water turns toxic: how climate change drives cyanotoxin biosynthesis-A mechanistic review.

Cyanobacterial harmful algal blooms (cHABs) represent an escalating global threat due to their ability to produce a wide range of potent cyanotoxins that threaten aquatic ecosystems, drinking water safety and public health. While eutrophication has long been recognized as a primary driver of bloom formation, increasing evidence suggests that climate change acts as a critical catalyst influencing not only bloom frequency and intensity but also toxin diversity and regulation. However, the mechanistic pathways through which climate-associated abiotic stressors regulate cyanotoxin biosynthesis rem

Adipose Mesenchymal Stem Cells-Derived Microvesicles Ameliorate Cyclophosphamide-Induced Cystitis via Inhibiting Pyroptosis and Ferroptosis in Rats.

AIMS: Cyclophosphamide (CYP) may through its toxic metabolite, acrolein, induce hemorrhagic cystitis and bladder hyperactivity. Previous studies demonstrated intra-iliac arterial administration of adipose derived mesenchymal stem cells (ADSC)-derived microvesicles with less immune response and adverse effects than ADSC itself may confer anti-oxidative stress and anti-inflammatory potential to improve bladder dysfunction. We explored whether ADSC-derived microvesicles may prevent CYP-induced bladder cystitis and overactivity. METHODS: Female Wistar rats were divided into control (Con), CYP (Cy
