Evaluation of the biological responses of osteoblast-like UMR-106 cells to the engineered porous PHBV matrix.

Poly (3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV) has been investigated for biomedical applications due to its many biologically favorable properties. However, to explore its application in bone tissue engineering, the poorly bioactive surface property of PHBV must be improved. To engineer PHBV to achieve a biologically active surface, in this study each porous PHBV matrix was prepared by solute leaching of salt/PHBV cast film and was treated with ozone followed by dip coating with type I collagen. The biological responses of osteoblast-like UMR-106 cells after being grown on the engineered

The ability to form biofilm influences Mycobacterium avium invasion and translocation of bronchial epithelial cells.

Organisms of the Mycobacterium avium complex (MAC) are widely distributed in the environment, form biofilms in water pipes and potable water tanks, and cause chronic lung infections in patients with chronic obstructive pulmonary disease and cystic fibrosis. Pathological studies in patients with pulmonary MAC infection revealed granulomatous inflammation around bronchi and bronchioles. BEAS-2B human bronchial epithelial cell line was used to study MAC invasion. MAC strain A5 entered polarized BEAS-2B cells with an efficiency of 0.1 +/- 0.03% in 2 h and 11.3 +/- 4.0% in 24 h. In contrast, biofil

Reactivity of T cells from women with antibodies to the human platelet antigen (HPA)-1a to peptides encompassing the HPA-1 polymorphism.

The human platelet antigen-1a (HPA-1a) is the most common alloantigenic target in fetal and neonatal alloimmune thrombocytopenia (NAIT). Treatment currently depends on the outcome in previous pregnancies. HPA-1 specific T cell responses were determined in 14 HPA-1a alloimmunized women during or after pregnancies affected by NAIT. Peripheral blood mononuclear cells were incubated with peptides encompassing the Leu33Pro polymorphism (residues 20-39 and 24-45 in both Leu33 (HPA-1a) and Pro33 (HPA-1b) forms) or control recall antigens in the presence of autologous sera and T cell proliferation was

Abnormalities in CD4+ T lymphocyte subsets in patients with common variable immunodeficiency.

In order to investigate whether deficient immunoglobulin production in common variable immunodeficiency (CVI) patients was related to defective T cells functions, phenotype and proliferative responses to mitogen of peripheral blood mononuclear cells (PBMC) were investigated in 9 patients with CVI. The results were compared to those of 12 age- and sex-matched normal controls. The numbers of CD3+ and CD8+ T cells in the patients were not different from those in the control group, but the numbers of CD4 T cells were decreased (511 +/- 237 vs 844 +/- 247/mm3; P less than 0.01). The decrease in CD4

Targeting the EpCAM-AXL axis to overcome drug resistance in lung cancer.

Drug resistance significantly limits the long-term effectiveness of targeted therapies in lung cancer. AXL, a receptor tyrosine kinase (RTK), is a well-established driver of drug resistance in many cancers, including lung cancer. Epithelial cell adhesion molecule (EpCAM), which is commonly overexpressed in epithelial cancers, is also a critical signaling molecule. In this study, we elucidated the role of EpCAM in drug resistance and its mechanistic association with AXL. We found that soluble epithelial cell adhesion molecule (sEpCAM) expression robustly enhanced hallmark cancer phenotypes and

A multi-dataset single-cell meta-analysis of human keloid skin across Asian, Black and White populations reveals endothelial and mesenchymal programs linked to ethnic disparities.

INTRODUCTION: Keloid is a fibroproliferative scar with marked ethnic disparities, disproportionately affecting Asian and Black populations. Single-cell RNA sequencing (scRNA-seq) studies have mapped keloid pathology, but cross-study differences complicate generalization. We performed a meta-analysis to identify composition and transcriptional programs associated with keloid across ethnicities. METHODS: We aggregated human skin scRNA-seq datasets containing keloid and healthy samples with documented ethnicity (Asian, Black, White). After quality control and batch integration (scVI/scANVI), we

The potential mechanisms and regulatory roles of exosomal miRNA in neural repair after spinal cord injury.

Spinal cord injury (SCI) is a devastating disorder of the central nervous system. It is characterized by primary mechanical damage and secondary pathological cascades. These lead to persistent sensory and motor deficits, substantial socioeconomic burdens, and limited therapeutic efficacy. Exosomes are nanoscale vesicles secreted by various cells that serve as key mediators of intercellular communication by delivering bioactive molecules, particularly microRNAs (miRNAs), which regulate gene expression in target cells. This review explores how exosomal miRNAs contribute to neural repair in SCI.

Extracellular matrix stiffness in chronic liver disease: From mechanisms to treatments.

Chronic liver disease (CLD) is a major global health threat and has emerged as a leading cause of human death. CLD essence lies in the remodeling of liver tissue due to excessive accumulation of collagen and extracellular matrix (ECM), which can largely progress to cirrhosis and even lead to end-stage liver diseases. Notably, the progressive elastic forces exerted by the extracellular matrix on cells, known as matrix stiffness, are among the most important mechanical stimuli. Recently, the role of matrix stiffness in the progression of chronic liver disease has been extensively studied. Numero

Novel bioactive compounds in the treatment of osteoporosis: chemistry-driven discovery from preclinical validation to clinical translation.

Osteoporosis affects an estimated 200 million people worldwide. Fragility fractures of the hip, spine, and wrist carry high mortality and impose a heavy socioeconomic burden. Bisphosphonates, denosumab, selective estrogen receptor modulators (SERMs), teriparatide, abaloparatide, and the recent sclerostin antibody romosozumab have improved clinical outcomes, but unmet needs remain. Long-term antiresorptive use is complicated by atypical femoral fractures and osteonecrosis of the jaw; denosumab discontinuation causes rebound bone loss; romosozumab carries a cardiovascular caution. Persistence on

Chronic Stress Destabilizes Ectoplasmic Specialization Dynamics in the Rat Testis via Downregulating the Focal Adhesion Kinase-Associated Protein Complex.

Prolonged stress adversely affects male reproductive health, with infertility affecting millions of couples worldwide due to lifestyle and mental health factors. Chronic unpredictable stress (CUS) has been shown to induce testicular dysfunction via blood-testis barrier (BTB) disruption. The BTB, maintained by ectoplasmic specialisations (ES), is regulated by β1-integrin and phosphorylated focal adhesion kinase (FAK)-associated proteins. Still, the impact of prolonged stress on these regulators has not been fully explored. This study investigated CUS-induced testicular damage over extended dura

PCB118 Is Associated with Impaired Decidualization and Angiogenesis Through miR-542-3p-Mediated Regulation of ILK Signaling.

2,3',4,4',5-Pentachlorobiphenyl (PCB118) is a persistent environmental pollutant associated with adverse female reproductive outcomes; however, its effects on uterine function and epigenetic regulation remain incompletely understood. This study investigated whether PCB118 disrupts uterine decidualization and angiogenesis through miRNA-mediated regulatory pathways. Human endometrial stromal cells (HESCs) and human umbilical vein endothelial cells (HUVECs) were exposed to an environmentally relevant, non-cytotoxic concentration of PCB118. Decidualization and angiogenesis were evaluated in vitro,

Resilience to Diabetic Retinopathy (RDR) Is Associated with a Pre-Retinopathy Transcriptional Program Induced by Diabetes.

The purpose of this project was to define gene expression changes associated with the acquisition and loss of resilience to diabetic retinopathy (RDR) in individual retinal cell types. A non-immune form of type 1 diabetes mellitus (DM) was induced by injecting male C57Bl6J mice with streptozotocin. Single-cell RNA sequencing was performed on retinas from mice that experienced DM for 5 or 15 days, along with retinas from age-matched, non-DM mice. The resulting data sets were analyzed to identify DM-associated differentially expressed genes and pathway enrichments after each duration of DM. We o

Epithelial stem cell-derived chemokines track clinical remission in inflammatory bowel disease in a disease-specific manner.

BACKGROUND AND AIMS: Stem cell-derived organoids are promising platforms for therapeutic screening in inflammatory bowel disease (IBD), but identifying functional organoid readouts with translational utility is challenging. Colon epithelial organoids from patients with ulcerative colitis (UC) overexpress chemokines CXCL1, CXCL11, CCL2, and CCL28, yet whether these inflammatory signatures correlate with disease activity and treatment response is unknown. This short report investigates whether organoid-retained chemokines correlate with disease activity and therapeutic outcomes. METHODS: We int

The janus-faced extracellular matrix remodeling in acute lung injury: a gatekeeper of macrophage localization and phenotype.

Although the extracellular matrix (ECM) is a highly dynamic partner of the immune system, its immunoregulatory role during the early phase of sepsis-induced acute lung injury (ALI) remains poorly defined. Using a murine cecal ligation and puncture (CLP) model, we observed dynamic ECM remodeling characterized by rapid collagen loss within 24 h, followed by a pathological fibroproliferation phase. Proteomic profiling of the degraded ECM demonstrated its immunoregulatory potential. In vitro studies revealed that annexin A1 (ANXA1), an upregulated ECM component, promoted anti-inflammatory macropha

Use of plant-derived compounds for chilled gamete storage in felids and canids.

Wild felids and canids play important roles in maintaining ecosystem stability and biodiversity, yet many species are threatened by habitat loss, climate change, urbanization, and poaching. Assisted reproductive technologies (ARTs) have therefore become valuable tools for wildlife conservation and genetic resource banking. However, prolonged storage and post-mortem deterioration remain major limitations affecting gamete quality and fertilizing ability. Oxidative stress is a major cause of gamete deterioration during chilled storage. During chilled storage, excessive reactive oxygen species (RO
