Moyamoya disease (MMD) is a chronic cerebrovascular disorder characterized by progressive stenosis of the terminal portions of the bilateral internal carotid arteries and their major branches, accompanied by the development of an abnormal vascular network at the base of the brain. Although RNF213 is the major susceptibility gene, its relatively low penetrance in MMD suggests that genetic predisposition requires synergistic interaction with environmental stressors to induce the characteristic pathological vascular remodeling. Studies have demonstrated that patients with MMD exhibit mild stenosi

Lafora disease is a rare, autosomal recessive neurodegenerative disorder characterized by the progressive accumulation of abnormal, insoluble, and hyperphosphorylated forms of glycogen, known as Lafora bodies, in the brain and other tissues. The disease typically manifests during early adolescence with myoclonus, seizures, and rapidly progressive cognitive decline, ultimately leading to severe neurological deterioration and death within a decade of onset. Mutations in the EPM2A or EPM2B genes, which encode the proteins laforin and malin-key regulators of glycogen metabolism-are the underlying

Background: Ferroptosis is a nonapoptotic type of cell death characterized by an increase in lipid reactive oxygen species (ROS). Acute myeloid leukemia (AML)-derived bone marrow mesenchymal stem cells (AML-BMSCs) support the progression and drug resistance of AML by secreting various bioactive substances, including exosomes. However, the role of BMSCs in regulating lipid metabolism and ferroptosis in AML remains unexplored. Results: Exosomes secreted by AML-BMSCs increased the expression of miR-196a-5p in AML cells. MiR-196a-5p promoted the proliferation of AML cells, reduced lipid ROS and fe

Although immune checkpoint blockade (ICB) is an effective therapeutic strategy, tumor intrinsic resistance to ICB limits its benefits for a majority of cancer patients. Via multiple in vivo screens, we identified RNF19A as an immune suppressor. RNF19A suppresses ICB efficacy by ubiquitinating cGAS to inhibit the release of type I interferon. Surprisingly, UBE2L6, originally identified as a critical interferon-induced ISG15-conjugating E2 enzyme for ISGylation, functions as the major ubiquitin-conjugating E2 enzyme to enable RNF19A-driven cGAS ubiquitination. Further, RSK1 phosphorylates UBE2L6

Macrophages are crucial immune cells in periodontal tissues, which play key roles in both the destruction and repair of associated with periodontitis. Targeted modulation of macrophage function has emerged as a potentially effective approach to influence periodontitis progression. This study investigates the effects of methotrexate-loaded extracellular vesicles (MTX-EVs) on inflammatory macrophage polarization both in vivo and in vitro. In a murine periodontitis model, MTX-EVs inhibited alveolar bone resorption, suppressed pro-inflammatory macrophage activation, and promoted anti-inflammatory

Arsenic toxicity is a serious threat to human health, transmitted through many factors in the environment, especially water and contaminated food. Epidemiologic studies have reported that arsenite increases mortality and morbidity by causing cardiac damage, but the mechanism of action on cardiotoxicity remains to be elucidated. Zingerone (ZNG) obtained from ginger root is a monomer with pharmacological effects such as antioxidant, anti-inflammatory, and anticancer. This study was conducted to investigate the protective potential of zingerone against sodium arsenite-induced cardiac damage in ra

Inherited Parkinson's disease (PD) often involves missense mutations in the PRKN2 gene, encoding for Parkin protein. The PDR-1 protein is the C. elegans ortholog of human Parkin. Using a CRISPR/Cas9 genome editing approach, we generated the PDR-1C169Y point mutation on a conserved cysteine residue in the RING0 domain. This mutation in human Parkin, C212Y, has been identified in autosomal recessive juvenile Parkinsonism patients. The PDR-1C169Y homozygous mutant animals exhibited a shorter lifespan and decreased thrashing rate compared with wild-type or heterozygous animals. Unique mitochondria

Loss-of-function mutations in PINK1 and PRKN cause familial forms of Parkinson's disease (PD). In vitro studies have revealed incredible insights into the molecular and cell-biological function of these genes, which have focused predominantly on mitophagy - the autophagic degradation of damaged mitochondria. The mechanisms of PINK1/Parkin function ultimately require investigation in an in vivo context using classic genetic approaches in animal models. In this context, Drosophila models have proven to be remarkably informative, in part due to robust phenotypes arising from null mutations. They

Alcohol use disorder (AUD) is associated with widespread epigenetic alterations, including changes in DNA methylation (DNAm). This multi-cohort study validated and extended previous findings on DNAm of HECW2 and GDAP1 in AUD, assessed sex differences, and explored DNAm in blood and brain tissue in humans and rats. DNAm was measured via pyrosequencing in human blood (NCtrl = 341, NAUD = 258), postmortem frontal cortex (Brodmann area 9; discovery cohort: NCtrl = 10, NAUD = 13, replication cohort: NCtrl = 64, NAUD = 55) and rat blood and medial prefrontal cortex (NCtrl = 16, NAUD = 15). Telomeric loops (t-loops) are thought to protect chromosome ends, with their stabilization generally requiring the shelterin protein TRF2. However, the mechanisms operating in pluripotent cells remain unknown. Here, we identify TELS1 as a TRF2-independent t-loop stabilizer in pluripotent cells. TELS1 binds single-stranded, G-rich telomeric DNA tracts likely present within duplex telomeric regions and promotes strand invasion in vitro, consistent with a direct role in t-loop formation. When targeted to telomeres in differentiated cells, TELS1 is able to substitute for TRF2 in making the t-loop,

Angelman syndrome and Fragile X syndrome are neurodevelopmental disorders (NDDs) caused by mutations in the UBE3A and FMR1 genes, respectively. However, they share common features such as cognitive and motor deficits, anxiety, and impaired social behavior. In this study, we utilized zebrafish as an animal model to investigate the anxiety-like effects of mutations in these genes across larval and adult stages, employing two widely used behavioral assays: the light-dark test (LDT) and the novel tank diving test (NTT). We conducted detailed analyses of anxiety-like behaviors and exploration patte

Angelman syndrome (AS) is a rare neurodevelopmental disorder caused by loss of expression of the maternal UBE3A allele and is characterized by a constellation of impactful neurologic symptoms. While previous work has uncovered outsized contributions of GABAergic neuron-selective Ube3a deletion to seizure susceptibility and electroencephalography (EEG) phenotypes in a mouse model of AS, the neuronal populations governing a broader range of behaviors have not been studied. Here, we used male and female mice to test the consequences of Ube3a deletion from GABAergic or glutamatergic neurons across

Angelman syndrome is a rare neurodevelopmental disorder caused by the loss of function of the maternally inherited UBE3A gene within the chr15q11-q13 region. This gene is subjected to a tissue-specific form of genomic imprinting leading to the silencing of the paternal allele in neurons. Angelman syndrome can result from various (epi)genetic mechanisms, with paternal uniparental disomy of chromosome 15 (patUPD15) being one of the rarest and least studied due to the absence of suitable models. To address this gap, we generated three independent induced pluripotent stem cell (iPSC) lines from in

Osteopetrosis is a group of genetically and clinically diverse inherited disorders characterized by an increase in bone density. The main known cause is an abnormality in the development or function of osteoclasts. Hence, the process of bone resorption is impaired, resulting in: 1- a reduction in bone marrow volume and, subsequently, a decrement in the hematopoietic capacity of bone marrow, which leads to anemia and compromised immunological function; 2- improper bone development, which leads to pressure on peripheral nerves, causing auditory, visual, and movement impairments; and 3- disturban

Oncolytic adenoviruses (OAVs) engineered to carry antitumor or immune-activating transgenes are being actively explored for cancer immunotherapy. While many efforts have focused on modifying OAVs to counteract the immunosuppressive tumor microenvironment, comparatively less attention has been given to enhancing OAV replication by targeting host factors to increase oncolytic activity. Here, we report that TRIM56 is strongly upregulated during HAdV-C5 infection, with its expression correlating with increasing levels of the viral E1A protein, ultimately promoting HAdV-C5 replication. Neisseria gonorrhoeae, the causative agent of the human disease gonorrhea, is the second most common sexually transmitted pathogen in the United States. Gonorrhea has a significantly high morbidity rate due to the ability of N. gonorrhoeae to rapidly develop antibiotic resistance. In this paper, crystal structures of tryptophanyl-tRNA synthetase (TrpRS) from N. gonorrhoeae (NgTrpRS) were determined in both its apo form and in complex with tryptophan. The structures reveal conserved HIGH and KMSKS motifs critical for ATP binding and catalysis, and highlight conformational changes in the active

Endometrial receptivity relies on the functional and morphological change of endometrium stromal cells (EnSCs) and epithelial cells in the secretory phase. Decidualization of ESCs and transitions in endometrium epithelial cells are crucial for successful uterine implantation and maintaining pregnancy. Accumulated data have demonstrated that decidualization is tightly coordinated by lipid metabolism. However, the lipidomic change and regulatory mechanism in uterine decidualization are still unknown. Our study showed that endometrium stromal cells and decidual stromal cells had different lipidom
