Lipid injectable emulsions (ILE) are essential components of parenteral nutrition in neonatal intensive care units. Over the past two decades, the expanded availability of ILE has led to rapid changes in clinical practice, largely driven by concerns about intestinal failure–associated liver disease. Despite these evolving trends, the evidence supporting one ILE over another remains limited and nuanced. Importantly, no currently available ILE replicates the intrauterine supply of critical long-chain polyunsaturated fatty acids, particularly arachidonic acid and docosahexaenoic acid, both of whi

INTRODUCTION: Pancreatic ductal adenocarcinoma (PDAC) exhibits a profoundly immunosuppressive tumor microenvironment, with tumor-associated macrophages (TAMs) being the most abundant immune infiltrate. Among them, lipid-associated macrophages (LAMs) have emerged as a distinct subpopulation driving immune evasion through metabolic reprogramming.
METHODS: We conducted a narrative review by systematically searching PubMed, Web of Science, and Scopus for articles on LAMs in PDAC up to December 2023. Key themes were synthesized to cover defining markers, metabolic pathways, immunosuppressive funct

Increasing evidence highlights a tight interplay between lipid metabolism and mitochondrial homeostasis in neurons, with disruptions in either pathway amplifying cellular vulnerability. PTEN-induced kinase 1 (PINK1), a familial Parkinson's disease (PD)-related gene and a key regulator of mitochondrial quality control and homeostasis, emerges at the intersections of lipid metabolic pathways, influencing membrane composition, fatty acid utilization, and neuronal energy balance. Within this review, we discuss the role of mitochondria as hubs for lipid metabolism, the mechanisms and functional con

The brain is highly enriched in poly-unsaturated fatty acids (PUFAs) and their deficiency has been associated with several neuropsychiatric disorders. Here, we demonstrate that the Dopamine receptor D2 (D2R), a class A G protein coupled receptor (GPCR) which is a main target of antipsychotics, displays specific sensitivity to membrane PUFA composition. We found that membrane enrichment with either of two distinct PUFAs significantly impairs agonist-induced D2R endocytosis in HEK-293 cells and cortical neurons. This treatment does not affect clathrin-mediated endocytosis or the internalization

Lipid metabolism disorders and hepatic lipid deposition are major contributors to the development of metabolic diseases such as non-alcoholic fatty liver disease. Omega-3 fatty acids have been shown to exert beneficial effects on lipid homeostasis and liver function, but their molecular regulatory mechanisms remain incompletely understood. Here, we investigated the effects of omega-3 fatty acids on lipid metabolism and hepatic lipid deposition in addition to the modulatory role of miR-34a using an in vivo high-fat diet (HFD) model and an in vitro palmitic acid (PA)-induced HepG2 cell model. We

The role of lipid droplets (LDs), typically regarded as storage sites for neutral lipid, in plant defense remains poorly understood. In our study with barley stripe mosaic virus (BSMV), a (+)ssRNA virus, we found that infection significantly increases LD abundance in Nicotiana benthamiana and wheat. Using split-TurboID-based proximity labeling, we identified LD-associated proteins (LDAPs) at ER-chloroplast contact sites. Increasing LD formation by overexpressing mouse FIT2 or plant NbLDAP2.1 decreased viral accumulation, whereas impairing LD formation led to higher infection. Live cell imaging

Conformational variations in α-syn fibrils are thought to underlie the distinct clinical features of synucleinopathies, including Lewy body dementia (LBD), Parkinsons's disease (PD), and multiple system atrophy (MSA), suggesting that distinct fibril structures act as molecular fingerprints linked to disease phenotype. While the origins of these conformational variations remain unclear, increasing evidence points to membranes as key modulators of fibrils conformations. In this study, we investigated how age-related alterations in membrane composition and fluidity influence α-syn fibril formatio

BACKGROUND: The relationship between high-density lipoprotein cholesterol (HDL-C) concentrations and the risk of peritoneal dialysis-related peritonitis (PDRP) remains unclear. We aimed to examine the association between HDL-C concentrations and the risk of new-onset PDRP in patients undergoing maintenance peritoneal dialysis (PD), while investigating potential modifying factors that may influence this relationship. METHODS: This retrospective cohort study included 512 eligible patients with maintenance PD recruited from January 1, 2010 to December 31, 2020 at the Peritoneal Dialysis Center of

The most commonly accepted scenario of early Earth includes: creation of the universe around 13.8 Ga (Giga-annus; or 109 years ago); establishment of our solar system ~ 4.60 Ga; and formation of Earth ~ 4.54 Ga. The earliest life forms on our planet so far observed to have existed, are microbes that left signals of their presence in rocks ~ 3.6 Ga - suggesting that Life forms existed within the first 940 million years after Earth's formation. However, an intriguing recent publication [1] infers that the last universal common ancestor (LUCA) likely existed by 4.2 Ga, and that the inferred LUCA

The interaction of bioactive compounds with lipid membranes strongly influences their biological activity and selectivity. In this study, the interfacial behavior of an isovanillin derivative (1) was investigated using Langmuir monolayers of DPPC and DPPS as models of zwitterionic non-tumorigenic and anionic tumor-associated membranes, respectively. Surface pressure-area isotherms, surface compressional modulus, relaxation assays, PM-IRRAS, and Brewster angle microscopy (BAM) were employed to evaluate the effects of the compound on monolayer organization and mechanics. (1) incorporated into bo

BACKGROUND AND PURPOSE: Achieving herd immunity via mass vaccination is recognized as the most effective strategy for curbing the transmission of contagious diseases. Among the various types of vaccines, subunit protein vaccines stand out for their favourable safety profile, proven efficacy and straightforward production processes. The development of next-generation vaccines is currently hindered by the scarcity of adjuvants capable of simultaneously amplifying both humoral and cellular immune responses.
EXPERIMENTAL APPROACH: This study identified the physical characteristics of empty lipid

The gut microbiota influences systemic immunity and cancer through inter-organ communication, but OMV-mediated mechanisms remain unclear. Here, we uncover a previously unrecognized role of Bacteroides intestinalis in restraining extra-intestinal tumor growth via OMVs enriched in sphingosine (SP), a bioactive lipid that directly binds to ATP5F1A-a subunit of the mitochondrial ATP synthase-to enhance NK cell function. This microbial lipid-ATP synthase interaction augments mitochondrial efficiency, reduces reactive oxygen species (ROS) production, and potently upregulates IFN-γsecretion in NK cel

The availability of T-cell therapies is limited in part due to their complex and costly biomanufacturing. Genetic engineering of T cells, such as to express chimeric antigen receptor (CAR), is commonly achieved using viral vectors, that are costly to produce and add to safety concerns. Non-viral alternatives have advantages, but a major barrier is their far lower efficiency of gene delivery. This has motivated the development of new strategies to increase nonviral gene delivery efficiencies, including using lipid nanoparticles (LNPs). We investigated the transfection of human T and NK cells wi

Using a combination of spectroscopy and micromanipulation techniques, we reveal that lipids in SLBs are strongly attached to the support. These SLBs are intrinsically stretched and under tension, causing membrane rupture, and form stable ∼2 nm aqueous pores that allow the permeation of small hydrophilic molecules. Despite strong adhesion, …

Numerous metabolic enzymes translocate from the endoplasmic reticulum (ER) membrane bilayer to the lipid droplet (LD) monolayer, where they perform essential functions. Mislocalization of certain LD-targeted membrane proteins, including HSD17B13 and PNPLA3, is implicated in metabolic dysfunction-associated steatotic liver disease. However, the mechanisms governing the trafficking and accumulation of ER proteins on LDs remain poorly understood. Here using minimal fluorescence photon fluxes nanoscopy and highly inclined and laminated optical single-molecule tracking combined with machine learnin

Electron paramagnetic resonance (EPR) spectroscopy of spin-labeled membrane proteins is a valuable biophysical technique to study structural details and conformational transitions of proteins close to their physiological environment, e.g., in liposomes, membrane bilayers, and nanodiscs. Unlike in nuclear magnetic resonance (NMR) spectroscopy, having only one or a few specific side chains labeled at a time with paramagnetic probes makes the size of the object under investigation irrelevant in terms of technique sensitivity. As a drawback, extensive site-directed mutagenesis is required in order

Biomolecular condensates formed via liquid-liquid phase separation often contain multiple molecular components whose collective interactions determine their physical properties. However, how different molecular species cooperatively regulate condensate mechanics remains poorly understood. In this study, we integrated Raman-Brillouin imaging with fluorescence recovery after photobleaching (FRAP) analysis to quantify the molecular composition, mobility, and high-frequency viscoelastic responses of multicomponent condensates composed of heterochromatin protein 1α (HP1α), DNA, and lipids. DNA inco…
