Targeting intratumoral bacteria reverses MHC-I suppression and enhances nano-based chemoimmunotherapy in colorectal cancer.

BACKGROUND: Emerging evidence indicates that diverse bacteria colonize tumors and facilitate oncogenesis through multifaceted mechanisms, predominantly through immunosuppression of the tumor microenvironment (TME). Fusobacterium nucleatum (F.n) exemplifies this phenomenon in colorectal cancer (CRC), where it mediates immune evasion via tumor cell autophagy and upregulation of programmed death-ligand 1 (PD-L1). Effective strategies for combining precise elimination of intratumoral F.n with chemotherapy remain lacking. METHODS: A targeted multimodal nano-based chemoimmunotherapy was engineered

Exploration of experience and sustained engagement with a web-based cognitive rehabilitation intervention amongst patients with aggressive lymphoma: a qualitative sub-study.

BACKGROUND: Cancer-related cognitive impairment (CRCI), a common side effect of cancer and its treatment, is characterised by difficulties in memory, attention and executive function. This qualitative sub-study was part of a single-site, parallel-group, pilot randomised controlled trial in which recruitment and retention exceeded our expectations. The aim was to explore participants' experience and motivation for sustained engagement with a web-based cognitive rehabilitation (eReCog) intervention amongst people with aggressive lymphoma who were self-reporting cognitive decline. METHODS: We us

Enhanced detection of network intrusions and anomalies in internet of things applications using a hybrid artificial intelligence model combining CNN and LSTM.

The rapid development of the internet of things (IoT) has made an increasing number of large-scale, interconnected networks more vulnerable to advanced attacks. There are increased risks of hidden intrusions and compromised security in IoT environments, resulting from class imbalance in network traffic datasets, high-dimensional feature spaces, and the requirement for spatial and temporal anomaly detection. Traditional intrusion detection systems (IDS) fail to address these problems. This research work presents IaADS, an intrusion and anomaly detection system integrating 3 balancing components

Modelling of hybrid deep ensemble learning based skin lesion detection using FCNN denoising and inception-dilated ResNetV2.

Specific metabolic and genetic abnormalities can cause skin lesions with cancerous potential. Cancerous cells might be spreading to every part of the body, whereas cancer is critical. Skin cancer is one of the prevalent cancers, and its global incidence continues to rise. Skin lesion classification is a significant stage in computer-aided diagnosis (CAD) for automatic analysis of melanoma. Recently, increased focus has been given to the deep learning (DL) methods applied for image analysis owing to their capability to utilise machine learning (ML) methods to convert input data into higher-leve

Nanovaccines with ferroptosis, necroptosis and STING-activation for synergistic immunotherapy.

Nanovaccines based on immunogenic cell death (ICD) present a promising strategy in tumor immunotherapy, serving as a bridge connecting tumor cell death to the induction of anti-tumor immunity. However, their efficacy is severely limited by the immunologically silent nature of apoptosis-dependent ICD and the immunosuppressive tumor microenvironment (TME). In this work, we designed a metal‒polyphenol network nanovaccine (SRF@FeShik-cGAMP/HA). Upon reaching the tumor tissue through specific targeting, the nanovaccines disassemble within the tumor cells, releasing the Fenton reaction catalyst ferr

Peptidyl arginine deiminase 2 (PAD2) inhibition is associated with acute and long-term benefits in a murine model of traumatic brain injury.

BACKGROUND: Traumatic brain injury (TBI) is a major contributor to trauma-related deaths and disability worldwide. Management of TBI is currently limited to supportive care. We have previously shown that global knockout of PAD2 confers neuroprotection, but its therapeutic potential was unclear. This study was designed to test the hypothesis that selective inhibition of PAD2 following TBI would improve neurological outcomes. METHODS: Male mice (c57bl6/j, 11-14 wk) were subjected to controlled cortical impact-induced TBI, and 5 to 10 minutes later, randomly given either PAD2 inhibitor ("loading

A Comparative Analysis of Pig-to-Rhesus Corneal Xenotransplantation with Various Immunosuppressive Regimens.

PURPOSE: Previous studies had made efforts to find the optimal regimens to reduce xenogeneic rejection in corneal xenotransplantation. However, it had not been fully determined which immunosuppressive choice would be optimal for long-term graft survival. We aimed to compare clinical results in wild-type pig-to-rhesus corneal xenotransplantation with various immunosuppressive regimens. METHODS: Medical records of 23 Chinese rhesus macaques who had received full-thickness corneal transplantation with wildtype pigs were retrospectively analyzed. They were administered various immunosuppressants

Stereo-electroencephalography-guided cross-electrode radiofrequency thermocoagulation in focal epilepsy: A review of current methodologies and outcomes.

Advances in stereo-electroencephalography-guided radiofrequency thermocoagulation (SEEG-guided RFTC) have led to the development of cross-electrode RFTC, which has been shown to result in significantly larger lesions and higher seizure-freedom rates compared to standard RFTC methods. Given the novelty of cross-electrode RFTC, herein we review the reported surgical techniques and treatment-related outcomes to date to identify current research gaps and future directions. Ten articles were identified for review including 187 participants who underwent cross-electrode RFTC. Hypothalamic hamartoma

Effects of chronic levodopa in the paraquat and lectin rat model of the "body-first" subtype of Parkinson's disease.

Recent studies have identified a "body-first" subtype of Parkinson's disease (PD) that is associated with more rapid progression to Parkinson's disease dementia (PDD). It is currently unknown whether chronic levodopa, the primary treatment in PD, contributes to the progression of disease in the "body-first" subtype of PD. We investigated whether chronic levodopa affects motor or cognitive function in the paraquat and lectin (P + L) rat model of PD, a model which recapitulates the "body-first" subtype of PD. Rats (n = 9) were administered 7 days of P + L (1 mg/kg P + 0.05% L in 1% sucrose solut

Mechanical History and Substrate Stiffness Shape Integrin-Mediated Endothelial Cell Behavior on Bioactive Hydrogels.

Synthetic blood contacting devices frequently fail due to the lack of requisite biochemical and biomechanical cues needed to support transanastamotic endothelialization. During transanastomotic endothelialization, endothelial cells experience dynamic changes in extracellular mechanical cues as they migrate from compliant native vessels onto stiffer blood contacting device surfaces. However, how substrate stiffness and mechanical memory from prior mechanical environments influence temporal integrin remodeling and downstream endothelialization processes necessary to establish a stable endothelia

Case Report: Probable CLIPPERS in a patient with cutaneous T-cell lymphoma in remission: 5-year follow-up.

Chronic lymphocytic inflammation with pontine perivascular enhancement responsive to steroids (CLIPPERS) may be associated with lymphoma, although the underlying relationship between these entities remains unclear. While the diagnosis is centered on specific imaging characteristics and can be difficult to establish, CLIPPERS typically responds robustly to corticosteroid therapy. We report a 60-year-old woman who presented with memory and cognitive impairment 4 years after achieving complete remission from cutaneous T-cell lymphoma. Brain MRI revealed atypical CLIPPERS features, including scatt

Melatonin alleviates retinal injury induced by vigabatrin and partially enhances its antiepileptic effects.

Structural optimization and combinational use with additive agents might help expand the clinical use of vigabatrin (VGB). Aiming to investigate the combinational effect of melatonin (MLT) on the antiepileptic action and retinal damage of VGB, a rat epilepsy model was induced by intraperitoneal injection of kainic acid (KA) and evaluated via both Racine grading and electroencephalogram (EEG). MLT (5, 10, 20 mg/kg) and VGB (50, 150 mg/kg) were administered intragastrically for 4 weeks, alone or together. The behavioral performance was measured during the remission of seizures via a series of ta

A feature fusion network with spatial-temporal-enhanced strategy for the motor imagery of force intensity variation.

INTRODUCTION: Motor imagery (MI)-based brain-computer interfaces (BCI) offers promising applications in rehabilitation. Traditional force-based MI-BCI paradigms generally require subjects to imagine constant force during static or dynamic state. It is challenging to meet the demands of dynamic interaction with force intensity variation in MI-BCI systems. METHODS: To address this gap, we designed a novel MI paradigm inspired by daily life, where subjects imagined variations in force intensity during dynamic unilateral upper-limb movements. In a single trial, the subjects were required to compl

Proposed SmartBarrel System for Monitoring and Assessment of Wine Fermentation Processes Using IoT Nose and Tongue Devices.

This paper introduces SmartBarrel, an innovative IoT-based sensory system that monitors and forecasts wine fermentation processes. At the core of SmartBarrel are two compact, attachable devices-the probing nose (E-nose) and the probing tongue (E-tongue), which mount directly onto stainless steel wine tanks. These devices periodically measure key fermentation parameters: the nose monitors gas emissions, while the tongue captures acidity, residual sugar, and color changes. Both utilize low-cost, low-power sensors validated through small-scale fermentation experiments. Beyond the sensory hardware

Decoding the Variable Velocity of Lower-Limb Stepping Movements From EEG.

Accurate decoding of lower-limb movement from electroencephalography (EEG) is essential for developing brain-computer interface (BCI) controlled exoskeletons in neurorehabilitation. This study investigates 3D velocity decoding at three fibular anatomical markers during overground stepping in healthy participants ( ${N}={9}$ ), using two approaches: (1) linear regression (LR) and (2) a deep learning (DL) framework combining convolutional neural networks (CNNs) and long short-term memory (LSTM) units. Participants were divided into two groups: G1 ( ${n}={5}$ ) performed cued forward and self-pac

Shedding new light on BACE1-mediated modulation of IL-6 signaling: Implications for neural activity and synaptic plasticity in mice.

The pleiotropic cytokine IL-6 regulates numerous processes in the body, including neuronal functions. IL-6 either binds to membrane-bound receptor (mIL-6R) and triggers signaling via heteromerization with the signal transducer gp130 (classical signaling), or binds to its soluble form (sIL-6R) to act on cells that do not express mIL-6R (trans-signaling). The ß-secretase BACE1 can cleave gp130 as well as IL-6R and we hypothesized that BACE1 may alter neuron activity and synaptic transmission via modulation of IL-6 signaling. We used multielectrode array (MEA) recordings to monitor electrical act
