Allosteric regulation is a central mechanism for modulating biological functions and offers an attractive strategy in drug discovery, particularly for targets considered challenging or "undruggable." However, the discovery of allosteric inhibitors is hindered by poorly defined binding sites and the lack of effective screening approaches. Here, we present a dual DNA-encoded library (DEL) screening strategy that integrates reversible DEL and covalent DEL (CoDEL) technologies to identify novel allosteric inhibitors of the SARS-CoV-2 RNA-dependent RNA polymerase (RdRp). Using this approach, we dis…

INTRODUCTION/OBJECTIVE: Quantitative Structure-Activity Relationship (QSAR) modeling is a critical computational strategy in drug discovery; however, data heterogeneity and experimental noise frequently compromise model reliability. In this study, QSAR models were developed to predict the inhibitory activity of 9,10-dihydrophenanthrene derivatives against the SARS-CoV-2 3CLpro enzyme using a novel fuzzy Support Vector Regression (FSVR) framework. The proposed approach incorporates a fuzzy membership function to weight activity data according to their reliability and employs both the full molec…

UNLABELLED: Inhibiting the catalytic activity of 3CLpro is a mainstream strategy to block coronavirus replication. However, the appearance of SARS-CoV-2 3CLpro resistance to protease inhibitors raises concerns for effective therapies. In this work, we first investigated the resistance profile of simnotrelvir, an approved anti-SARS-CoV-2 drug that targets 3CLpro. We found that the T21I/E166A mutations in 3CLpro equally emerged when SARS-CoV-2 was passaged in the HEK293T-hACE2 cells with increasing concentrations of simnotrelvir. The SARS-CoV-2 isolate carrying 3CLproT21I/E166A (SARS2-T21I/E166A…

The post-acute sequelae of SARS-CoV-2 infection (PASC), or long COVID, represent a multifaceted challenge in pediatric populations, characterized by symptoms persisting beyond the acute phase. In Taiwan, where early public health measures initially contained the pandemic, the 2022 Omicron surge prompted focused investigation into pediatric PASC, highlighting the critical need for longitudinal data in this specific demographic. To address this challenge, the Diagnosis and Support for COVID Children to Enhance Recovery (DISCOVER) study was established as a prospective, multidisciplinary cohort. …

Vaccines capable of eliciting broadly neutralizing antibodies (bnAbs) are a major goal for pandemic preparedness. A challenge across vaccine fields is how to deliberately recruit the rare B cell clones that recognize conserved epitopes shared across diverse viral variants. BnAbs have been known to frequently emerge through extensive somatic hypermutation during affinity maturation, here we describe an alternative mechanism for bnAb selection. We designed an mRNA vaccine in which two antigenically distinct (severe acute respiratory syndrome coronavirus 2) SARS-CoV-2 variant's (Omicron and Delta…

BACKGROUND AND OBJECTIVES: Older adults (≥65 years) face a disproportionate burden of COVID-19 mortality yet remain underrepresented in clinical trials despite this group accounting for 76% of COVID-19 deaths. Hundreds of SARS-CoV-2 vaccine clinical trials have been registered on ClinicalTrials.gov; however, the extent of older adult representation remains unclear. Our objective was to evaluate the representation of older adults in SARS-CoV-2 vaccine clinical trials registered on ClinicalTrials.gov and identify enrollment criteria that disproportionately limit older adult participation.

RESEARCH QUESTION: With the nationwide relaxation of controls for the COVID-19 epidemic from December 2022 in China, fertility and in vitro fertilization (IVF) centers reported increasing numbers of infected patients in a short time. However, there is a lack of information regarding the effects of SARS-CoV-2 infection on pregnancy outcomes after IVF–embryo transfer (ET)/intracytoplasmic sperm injection (ICSI)–ET/frozen–thawed embryo transfer (FET), especially the live birth rate. DESIGN: This multicenter retrospective study was conducted at three public IVF centers in Jiangsu province of China…

INTRODUCTION: Longitudinal assessments of SARS-CoV-2 seroprevalence among a triad of parents, teachers and children in low-middle income countries are scarce. CoKiDSS assessed changes in seroprevalence among asymptomatic primary school learners, their parents, and teachers in a semi-rural South African school setting.
METHODS: SARS-CoV-2 seroprevalence was estimated at baseline (May-August 2023) and follow-up (October 2023) using a point-of-care COVID-19 IgG/IgM test. Results were adjusted for test sensitivity and specificity. Between-group differences and within-participant serostatus change…

Background: Sex-related differences in SARS-CoV-2 viral susceptibility, severity, and recovery are prevalent. Factors such as age and biological variation provide some explanation for immune response, yet the use of biological sex as a variable may limit understanding of underlying individual mechanisms related to endocrine-immune function involved in long-term immune memory. Endogenous sex hormone concentrations provide a means to assess the magnitude or durability of antibody persistence and disease recovery. Objective: This study examined salivary concentrations of 17β-estradiol and testost…

Nanodecoys that competitively inhibit viral entry represent a promising antiviral strategy, yet the influence of their physical properties on performance remains underexplored. In this work, we systematically investigate the size-dependent antiviral activity of ACE2-conjugated nanodecoys against a safe SARS-CoV-2 mimic. Our results demonstrate that the inhibition of mimic cellular uptake is highly size-dependent. The 10 nm nanodecoys exhibited the most potent neutralization, followed by the comparable 5 and 20 nm, while the 50 and 100 nm nanodecoys demonstrated similar, lower efficacies. This …

SARS-CoV-2 main protease (MPro) is a proven target for drug discovery of small-molecule antiviral agents due to its crucial role in viral polyprotein processing, high structural conservation across numerous divergent variants, and the lack of similar human enzymes. Unlike covalent compounds, noncovalent inhibitors of MPro do not modify the enzyme's active site, and may offer improved safety profiles, greater chemical tractability, and better oral bioavailability without the need for pharmacokinetic enhancement. In this study, we designed, synthesized and characterized thirteen noncovalent nonp…

Free-ranging white-tailed deer (WTD) are established SARS-CoV-2 reservoirs, but the susceptibility of other cervid species remains unclear. Here we integrate receptor analysis, structural modeling, and field surveillance to assess SARS-CoV-2 susceptibility across North American cervids. We identify species- and variant-specific differences in ACE2-spike compatibility. Elk ACE2 exhibits weak binding to the ancestral strain (Wuhan-Hu-1) and Delta spike receptor-binding domains (RBDs), likely due to a unique K31N substitution. In contrast, it shows stronger binding to Alpha, Beta, Gamma, and Omic…

Two sarbecoviruses, SARS-CoV-1 and SARS-CoV-2 that engage ACE2 through their receptor-binding domains, have caused major human outbreaks. The pandemic potential of sarbecoviruses has prompted the discovery and classification of bat and other zoonotic sarbecoviruses that are also able to use human ACE2 or ACE2 ortholog receptors for infection. However, the current human immunological landscape reactive to these SARS-CoV-2-related viruses is not well profiled. Using a panel of pseudotyped lentiviruses expressing only spike proteins, we assess serum neutralization activity against clade 3 and 4 (…

BACKGROUND: Respiratory tract infections are a leading cause of hospitalization in patients under 18 years of age and remain a major contributor to pediatric morbidity worldwide. Understanding the epidemiology of viral pathogens—including their seasonal trends and response to global events like pandemics—is essential for public health planning, surveillance strategies, and timely clinical interventions. METHODS: This retrospective study evaluated the epidemiology and dynamics of respiratory virus detection in hospitalized patients younger than 18 years before and during the SARS-CoV-2 pandemic…

SARS-CoV-2 papain-like protease (PLpro) is a key antiviral target as it plays a dual role in viral replication and modulation of innate immune responses by deubiquitinating or deISGylating host proteins. Therefore, therapeutically targeting PLpro may serve as a two-pronged approach to mitigate SARS-CoV-2 infection. Interestingly, PLpro shares structural and functional similarities with cellular deubiquitinating enzymes (DUBs), and this fact was leveraged in our study to identify DUB inhibitors that target the ubiquitin/ISG15-binding site and the known substrate-binding pocket of PLpro. Among t…

Developing covalent inhibitors of the SARS-CoV-2 papain-like protease (PLpro) represents a promising therapeutic strategy, but translation of this approach into in vivo antiviral efficacy remains limited. A major challenge is the shallow and minimally druggable binding environment surrounding the catalytic cysteine, which reflects the enzyme's stringent requirement for P1 and P2 Gly-Gly recognition. In addition, many previously reported covalent PLpro inhibitors have relied on warheads or linker designs with limited metabolic stability or nonspecific thiol reactivity, restricting their progres…
