Suitability of TBA method for the evaluation of the oxidative effect of non-water-soluble and water-soluble rosemary extracts.

The antioxidative effects of rosemary and grape-seed extracts spiked in human plasma were examined using the thiobarbituric acid (TBA) method. The TBA values of plasma spiked with reagents to generate reactive oxygen species, such as singlet oxygen ((1)O(2)), hydroxyl radicals ((·)OH), peroxynitrite (ONOO(-)), and superoxide anions (O(2)(·-)), were measured by a flow injection analysis method with fluorescence (FL) detection. TBA values obtained by the addition of 50 mg/mL of rosemary extracts for (1)O(2), (·)OH, ONOO(-), and O(2)(·-) increased to 964 ± 65%, 1063 ± 61%, 758 ± 78%, and 698 ± 41

Studies on possibility for alleviation of lifestyle diseases by low-dose irradiation or radon inhalation.

Our previous studies showed the possibility that activation of the antioxidative function alleviates various oxidative damages, which are related to lifestyle diseases. Results showed that, low-dose X-ray irradiation activated superoxide dismutase and inhibits oedema following ischaemia-reperfusion. To alleviate ischaemia-reperfusion injury with transplantation, the changes of the antioxidative function in liver graft using low-dose X-ray irradiation immediately after exenteration were examined. Results showed that liver grafts activate the antioxidative function as a result of irradiation. In

Anti-ulcer role of herbomineral Siddha drug - Thamira parpam on experimentally induced gastric mucosal damage in rats.

Anti-ulcerogenic activity of Thamira parpam (TP) was investigated in two ulcer models (aspirin + pylorus ligation and HCl-ethanol). Aspirin-pylorus ligation (Asp 200 mg/kg-7 days + PL-4 hours) and HCl-ethanol (150 mM HCl in 70% ethanol) induction in rat resulted in elevation of thiobarbituric acid reactive substances (TBARS) and depletion of antioxidants (superoxide dismutase [SOD], glutathione [GSH], glutathione peroxidase [GPx]) with high ulcer scores (p < .01). In Asp + PL model, TP treatment showed mild inhibition on ulcer scores, changes in pH, gastric volume, total and free acidity, and

Investigation of supplemental ultraviolet-B-induced changes in antioxidative defense system and leaf proteome in radish (Raphanus sativus L. cv Truthful): an insight to plant response under high oxidative stress.

Impact of supplemental UV-B (sUV-B) has been investigated on photosynthetic pigments, antioxidative enzymes, metabolites, and protein profiling of radish plants under realistic field conditions. Exposure of sUV-B leads to oxidative damage in plants. However, plants possess a number of UV-protection mechanisms including a stimulation of antioxidant defense system. It caused alteration in reactive oxygen species metabolism primarily by decreasing catalase activity vis-à-vis enhanced activities of other enzymatic (superoxide dismutase, ascorbate peroxidase, and glutathione reductase) and non-enzy

Radiation-induced delayed bystander-type effects mediated by hemopoietic cells.

Genetic lesions and cell death associated with exposure to ionizing radiation have generally been attributed to DNA damage arising as a consequence of deposition of energy in the cell nucleus. However, reports of radiation-induced bystander effects, in which DNA damage is produced in nonirradiated cells as a consequence of communication with irradiated cells, indicate additional mechanisms. At present, most information has been obtained using in vitro systems, and the in vivo significance of bystander factors is not clear. In this study we show that signals generated in vivo in the bone marrow

Methionine-induced hyperhomocysteinemia modulates lipoprotein profile and oxidative stress but not progression of atherosclerosis in aged apolipoprotein E knockout mice.

It is documented that hyperhomocysteinemia (HHcy) is an independent risk factor for atherosclerosis, but whether elevated plasma homocysteine contributes to the progression of atherosclerosis in aged animals with hypercholesterolemia is still unknown. HHcy was induced in apolipoprotein E (ApoE) knockout mice (male, 32 weeks old) by feeding 2% methionine/low folate (1 mg/kg) diet for 20 weeks. HHcy induced by methionine feeding significantly increased oxidative stress, as measured by thiobarbituric-reactive substances in livers (P < .05) and genetic expression of Cu,Zn-superoxide dismutase, in

Oxidative stress in experimental vitiligo C57BL/6 mice.

Источник пометил публикацию как отозванную

AIM: To evaluate whether oxidative stress is implicated in melanocyte damage in vitiligo. BACKGROUND: Vitiligo is a complex disorder characterized by gradually enlarging areas of depigmentation. A new unifying hypothesis for the etiology of this pigment disorder is proposed, in which we postulate that the final destruction of melanocytes in vitiligo results from a cascade of reactions initiated by a disregulation of melanogenesis, as the result of a breakdown in free radical defense. METHODS: We evaluated 18 vitiligo mice and 12 controls that were age matched. Parameters of oxidative stress

A role for hydrogen peroxide in the pro-apoptotic effects of photodynamic therapy.

Although the first reactive oxygen species (ROS) formed during irradiation of photosensitized cells is almost invariably singlet molecular oxygen (1O2), other ROS have been implicated in the phototoxic effects of photodynamic therapy (PDT). Among these are superoxide anion radical (*O2(-)), hydrogen peroxide (H2O2) and hydroxyl radical (*OH). In this study, we investigated the role of H2O2 in the pro-apoptotic response to PDT in murine leukemia P388 cells. A primary route for detoxification of cellular H2O2 involves the peroxisomal enzyme catalase. Inhibition of catalase activity by 3-amino-1,

In ovo exposure of a Fusarium mycotoxin butenolide induces hepatic and renal oxidative damage in chick embryos, and antioxidants provide protections.

Butenolide is a mycotoxin produced by several toxigenic Fusarium species. It frequently invades many important grains, and evokes a broad spectrum of toxic effects. For these reasons, butenolide poses a health risk to both humans and animals. However, many toxicology issues of butenolide including targets and mechanisms of toxicity remain to be elucidated so far. The present study therefore attempts to reveal the toxic profile of butenolide from a viewpoint of oxidative damage, using chick embryos as an in vitro model. A single in ovo injection of butenolide resulted in significant oxidative i

NADPH oxidase-derived reactive oxygen species in the regulation of endothelial phenotype.

Endothelial dysfunction comprising impairment of endothelium-dependent vasodilator function and increased endothelial activation contributes to the pathophysiology of cardiovascular diseases such as atherosclerosis, diabetic vasculopathy, heart failure and hypertension. The changes in endothelial phenotype in these conditions occur in response to diverse stimuli including inflammatory cytokines, activation of renin-angiotensin-aldosterone system, hyperlipidaemia, hyperglycemia, ischemia-reperfusion and mechanical forces. An increased production of reactive oxygen species (ROS), such as superox

Antioxidant therapy for prevention of inflammation, ischemic reperfusion injuries and allograft rejection.

Oxidative stress results from an imbalance, an excess of oxidants, depletion of antioxidants or failure to repair oxidative damage induced by reactive oxygen species. A vast amount of evidence implicates oxygen-derived free radicals and high-energy oxidants as mediators in many pathological conditions of inflammation, shock, and organ responses to ischemia/reperfusion, which arise during a number of clinical surgical interventions, including transplant graft rejection and coronary bypass surgery, and in such diseases as, diabetes, atherosclerosis, hypertension, organ ischemia/reperfusion, card
