Are families of children with reading difficulties at risk for immune disorders and nonrighthandedness?

This study used questionnaire data to examine immune disorders and nonrighthandedness in the families of children enrolled in a learning disabilities school and children attending regular classrooms in public schools. Groups were organized according to their performance on a standardized test of reading comprehension to avoid overlap. In total, 468 questionnaires were returned, from which we were able to derive a final sample of carefully matched subjects: 55 subjects undergoing remediation for reading problems and 55 age- and sex-matched control subjects. The results indicated that children w

Effects of computerized cognitive training as add-on treatment to stimulants in ADHD: a pilot fMRI study.

The neurofunctional effects of Cognitive training (CT) are poorly understood. Our main objective was to assess fMRI brain activation patterns in children with ADHD who received CT as an add-on treatment to stimulant medication. We included twenty children with ADHD from a clinical trial of stimulant medication and CT (10 in medication + CT and 10 in medication + non-active training). Between-group differences were assessed in performance and in brain activation during 3 fMRI paradigms of working memory (N-back: 0-back, 1-back, 2-back, 3-back), sustained attention (Sustained Attention Task - SA

The effect of methylphenidate on prefrontal cognitive functioning, inattention, and hyperactivity in velocardiofacial syndrome.

OBJECTIVE: Methylphenidate (MPH) is commonly used to treat attention-deficit/hyperactivity disorder (ADHD) in all children, including those with velocardiofacial syndrome (VCFS). Yet concerns have been raised regarding its safety and efficacy in VCFS. The goal of this study was to examine the safety and efficacy of MPH in children with VCFS. METHODS: Thirty-four children and adolescents with VCFS and ADHD participated in a randomized, controlled trial with a 2:1 ratio of MPH versus placebo. All subjects underwent a cardiological evaluation before and after MPH administration. The primary outc

Aberrant recursive splicing in a human disease locus.

Recursive splice sites are rare motifs postulated to facilitate splicing across massive introns and shape isoform diversity. The necessity of recursive splicing (RS) and its role in human disease are unsubstantiated. Here, we use CRISPR to model intronic deletions eliminating an RS site (RS1) in CADM2 and conferring risk for attention deficit hyperactivity disorder and other neurobehavioral traits. Ablating RS1 in human induced neurons alters the gradient of RNA abundance across CADM2's first intron, decreases CADM2 expression, and impacts transcript usage. Decreased CADM2 expression is reflec

Recurrence of exostosis as a result of medication-induced bruxism: case study.

INTRODUCTION: Alveolar oral exostosis is a common, benign condition routinely found in dentistry. Clinical problems associated with exostoses are the maintenance of oral hygiene as well as the fabrication of prosthodontic appliances. Over time, exostoses may contribute to irritation and periodontal disease. CASE DESCRIPTION: The patient in this case study had a recurrence of exostoses and was bothered by consistent and prominent pain. She reported being a bruxer; her bruxism was exacerbated due to attention-deficit hyperactivity disorder and antidepressant medications. DISCUSSION: The etiolo

IL-4 shapes microglia-dependent pruning of the cerebellum during postnatal development.

Interleukin-4 (IL-4) is a type 2 cytokine with pleiotropic functions in adaptive immunity, allergies, and cognitive processes. Here, we show that low levels of IL-4 in the early postnatal stage delineate a critical period in which microglia extensively prune cerebellar neurons. Elevating the levels of this cytokine via peripheral injection, or using a mouse model of allergic asthma, leads to defective pruning, permanent increase in cerebellar granule cells, and circuit alterations. These animals also show a hyperkinetic and impulsive-like phenotype, reminiscent of attention-deficit hyperactivi

Self-Strangulation Through A Sphygmomanometer: An Uncommon Suicide.

Suicide by asphyxia is quite a common event in forensic practice and may be implemented in different ways. The authors report a unique case of a 16-year-old youth who committed suicide by means of a standard mercury sphygmomanometer. This manner of suicide has never been described in the literature reviewed. A complete forensic investigation led to the conclusion that the cause of death was mechanical asphyxia, ascribed to self-strangulation by means of an atypical item. The victim suffered from attention-deficit/hyperactivity disorder (ADHD) syndrome and was assisted by support teachers. He h

Magnesium in Prevention and Therapy.

Magnesium is the fourth most abundant mineral in the body. It has been recognized as a cofactor for more than 300 enzymatic reactions, where it is crucial for adenosine triphosphate (ATP) metabolism. Magnesium is required for DNA and RNA synthesis, reproduction, and protein synthesis. Moreover, magnesium is essential for the regulation of muscular contraction, blood pressure, insulin metabolism, cardiac excitability, vasomotor tone, nerve transmission and neuromuscular conduction. Imbalances in magnesium status-primarily hypomagnesemia as it is seen more common than hypermagnesemia-might resul

Selective enhancement of cost sensitivity by methylphenidate in adult ADHD: a randomized placebo-controlled trial.

BACKGROUND: Attention-deficit hyperactivity disorder (ADHD) is associated with impairments in real-life decisions, many involving balancing costs and benefits. Increasing dopamine and norepinephrine with stimulant medication improves decision-making in ADHD. However, it is unclear whether stimulant medication affects sensitivity to costs and/or benefits or alters the speed-accuracy tradeoff. Here, we applied the drift diffusion model (DDM) to assess how the stimulant medication methylphenidate (MPH) affects subprocesses of cost-benefit decision-making in ADHD. METHODS: Sixty-one adult partici

Group dialectical behavior therapy skills training versus group cognitive behavioral therapy for adults with ADHD: a randomized controlled trial.

BACKGROUND: Attention-Deficit/Hyperactivity Disorder (ADHD) is a neurodevelopmental disorder characterized by inattention, hyperactivity, impulsivity, emotion dysregulation, and executive dysfunction. Current treatment guidelines recommend a multimodal approach. Cognitive Behavioral Therapy (CBT) has proven effective for alleviating core symptoms, improving executive functioning, and reducing emotional comorbidities, with benefits sustained for up to one year. Dialectical Behavior Therapy (DBT), an adaptation of CBT that integrates mindfulness and emotion regulation components, has also shown

Polygenic architecture of brain structure and function, behaviors, and psychopathologies in children.

Childhood brain development involves dynamic remodeling shaped by genetic influences contributing to long-term neurodevelopmental variation. Here we report integrative analyses of over 8600 children, using 7 brain imaging-derived phenotype (IDP) modalities, polygenic scores (PGS) of 33 complex traits, and 266 cognitive and psychological phenotypes. Most brain IDPs show low-to-moderate SNP-based heritability, lower than typically observed in adults. Sparse generalized canonical correlation analysis reveals positive associations between cognitive-related PGS and structural MRI features (e.g., to

Enhancing Goal Achievement in Adults With ADHD: A Participant-Centered Evaluation of Transcranial Direct Current Stimulation From the TUNED Trial.

OBJECTIVE: Few trials in ADHD incorporate participant-centered outcomes that evaluate the impact of interventions on meaningful life activities. Additionally, in psychiatry, the translation of changes in symptom rating scales into meaningful impacts on participants' lives has been questioned. The Transcranial Direct Current Stimulation (tDCS) for the Treatment of Inattention Symptoms in Adult Patients with ADHD (TUNED) trial demonstrated improved inattention symptoms, assessed using a clinician-administrated scale, after a 4-week treatment with daily home-based tDCS in adults with ADHD. Here,

The effects of transcranial random noise stimulation on excitation/inhibition balance in ADHD.

BACKGROUND: Children with attention-deficit/hyperactivity disorder (ADHD) often show aberrant neural activity, including excitation/inhibition (E/I) imbalances, atypical event-related potentials (ERPs), and neural network dysfunction. Transcranial Random Noise Stimulation (tRNS) has shown promise in modulating neural activity in ADHD. METHODS: The current study examined differences in behavioral and EEG signals recorded during an inhibitory control task in children with (N = 23) and without (N = 33) ADHD. Changes in these signals were further assessed following a combined tRNS and cognitive t

Altered Morphological Characteristics Associated With Spatial Working Memory Performance in Children With ADHD.

BACKGROUND: Spatial working memory (SWM) deficit is a common problem in attention deficit disorder with hyperactivity (ADHD), often correlating with the severity of ADHD symptoms and academic difficulties. Although previous studies have broadly described abnormal brain structural changes in ADHD, the potential changes in brain morphology in children with ADHD with SWM dysfunction are still uncharacterized. This cross-sectional study was used to elucidate the brain morphological alterations associated with SWM performance in boys with ADHD. METHODS: Data for this investigation were retrieved f…
