Phosphocalmodulin has been shown to have a differential biological activity compared to nonphosphorylated calmodulin when assayed on a variety of calmodulin-dependent systems. However, the phosphocalmodulin preparations used so far in those experiments were not necessarily free of nonphosphorylated calmodulin. Therefore, the results obtained may not unquestionably show the real effect of pure phosphocalmodulin on the systems under study. To solve this problem, we describe here a method for the purification of phospho(Tyr)calmodulin free of nonphosphorylated calmodulin. The procedure consists o

A metastasis to the right liver lobe of an argyrophil/argentaffin midgut carcinoid tumour in a patient with the classical carcinoid syndrome was examined for the presence of tachykinins other than substance P, using a specific antiserum. The extract was initially purified using SepPak cartridges, and subsequently subjected to cation-exchange chromatography on SP Sephadex C-25 which separated the immunoreactive material into two main components (components I and II). Both were further purified by anion-exchange chromatography on DEAE-Sephadex A-25, and by reverse-phase fast protein liquid chrom

Previous study strongly suggests that silicotic fibrosis is mediated by macrophages and their soluble mediators. The biochemical properties of the mediators involved in silicotic fibrosis, however, are as yet ill defined. The current study, therefore, determined whether human monocyte-macrophages treated with fibrogenic silica dust released factors capable of activating fibroblasts as measured by an increase in fibroblast proliferation. Silica, but not nonfibrogenic diamond dust, stimulated the release of fibroblast proliferation factors. Moreover, the level of fibroblast proliferation activit

Factor VIII is a large protein molecule of molecular weight 2,000,000 or larger that elutes in the void volume on agarose gel chromatography. It has been shown previously that high concentrations of alkali halides and, more specifically, 0.25 M Ca(2+) dissociate the molecule into a large carrier protein and a small fragment that retains the factor VIII activity. Factor VIII was prepared from normal canine plasma collected in sodium oxalate and heparin and adsorbed with BaSO(4). Results with Ca(2+) dissociation were the same as those obtained with fraction prepared from canine plasma collected

The behavior of bioactive 1,3,4-thiadiazoles of anticancer potency and cholinesterases (ChEs) inhibitors was examined by high-performance thin-layer chromatography (HPTLC) in a reversed phase (RP) system. The chromatographic systems consisted of C-18 or C-8-bonded silica as the stationary phase and binary mixtures of water with methanol, acetonitrile, acetone, or dioxane as the mobile phase. The lipophilicity parameters RMw (the RM value of a solute extrapolated to pure water as the mobile phase) were determined based on the linear relationship between compound retention and water content in t

The report presents a new, robust, and reproducible liquid chromatography-tandem mass spectrometry (HPLC-MS/MS) and HPLC-fluorescence (FL) based methods for the determination of urinary homocysteine thiolactone (HTL). In particular, a versatile sample preparation procedure was designed to purify urine samples, involving chloroform liquid-liquid extraction (LLE) of HTL and its re-extraction (re-LLE) with formic acid, prior to chromatographic analysis. In relation to HPLC-FL assay, the quantification of HTL additionally uses o-phthaldialdehyde (OPA) as the on-column derivatization agent, while H

Knowledge of the radiochemical purity of radiopharmaceuticals is mandatory and can be evaluated by several methods and techniques. Planar chromatography is the technique normally employed in nuclear medicine since it is simple, rapid and usually of low cost. There is no standard system for the chromatographic technique, but price, separation efficiency and short time for execution must be considered. We have studied an alternative system using common chromatographic stationary phase and alcohol or alcohol:chloroform mixtures as the mobile phase, using the lipophilic radiopharmaceutical [(99m)T

The fumonisins are a recently identified group of fungal toxins, occurring worldwide in naturally contaminated maize, which have elicited considerable attention over the past decade due to their association with the animal disease syndromes, equine leukoencephalomalacia and porcine pulmonary oedema, and their reported association with oesophageal cancer in rural areas of Transkei, South Africa and Linxian County, China. This paper reviews the development of sensitive chromatographic analytical methods for the determination of these toxins in a range of mainly maize or maize-based food matrices

The two laboratory reactions focus on teaching several concepts associated with green chemistry. Each uses a commercial, nontoxic, and biodegradable surfactant, TPGS-750-M, to promote organic reactions within the lipophilic cores of nanoscale micelles in water. These experiments are based on work by K. Barry Sharpless (an azide-alkyne "click" reaction) and Robert Grubbs (an olefin cross-metathesis reaction); both are suitable for an undergraduate organic laboratory. The copper-catalyzed azide-alkyne [3+2] cycloaddition of benzyl azide and 4-tolylacetylene is very rapid: the triazole product is

Growth characteristics of the aerobic bacterial strain Arthrobacter citreus BI-100 in mineral salts medium with gamma-hexachlorocyclohexane (gamma-HCH) as the sole source of carbon and degradation of gamma-HCH by the strain are reported. The highest yield of the bacteria is observed at a gamma-HCH concentration of 100 mg/L. At this concentration, the bacteria entered the exponential phase of growth without any lag. At 8 h of growth, no residual HCH, but its metabolites, was detectable in the medium. The bacterium attained its stationary phase at 48 h and at 72 h; no metabolite of gamma-HCH cou

In investigations aimed at characterizing snake venom blood clot-dissolving enzymes, we have developed a rapid two-step high-performance chromatography method for the isolation of these fibrinolytic enzymes from the venoms of Agkistrodon contortrix contortrix and Agkistrodon piscivorus conanti. The first step consisted of hydrophobic interaction chromatography on a propyl-aspartamide column. Fractions containing the fibrinolytic activity were then concentrated and applied to a hydroxylapatite column. The resulting preparation, assessed for purity by reverse-phase chromatography and sodium dode

Human milk oligosaccharides (HMOs) are essential for babies' growth. The most commonly used method for HMOs analysis combines hydrophilic interaction liquid chromatography (HILIC) with ultra-performance liquid chromatography (UPLC). However, high-pressure operation and the cost of commercial HILIC columns limit progress in the HMOs research. In this study, we synthesized a novel mixed-mode adsorbent, PS-g-PAMPS, by grafting 2-acrylamido-2-methylpropane sulfonic acid (AMPS) onto a Merrifield resin (PS-g-Cl), and characterized it. The adsorbents were packed into a column (Mixed-HILIC column) for
