Molecular survey and risk factors of Trypanosoma pestanai in Eurasian badgers from the northern Iberian Peninsula.

Trypanosoma (Megatrypanum) pestanai is a parasite of the Eurasian badger (Meles meles), reported in various European countries. However, its presence in the Iberian Peninsula had not been previously investigated. To address this knowledge gap and to assess its occurrence and potential risk factors associated with infection, we analyzed DNA from the spleens of 145 badgers sampled across 4 autonomous regions in northern Spain. Two real-time PCR assays using a reference 18S rRNA partial sequence of T. pestanai (92 bp) were developed: one based on SYBR Green chemistry and the other employing a Taq

Self-Powered Microfluidic Device with Laser-Converted Graphene Electrodes for Immobilization-Free Electrochemical Detection of MPOX Virus DNA via Mismatch-Driven Nanocomplexes.

The re-emergence of monkeypox virus (MPXV) underscores the urgent need for rapid and decentralized diagnostic tools. Herein, we present an innovative, immobilization-free, and label-free paper-based electrochemical fast-flow microfluidic device (eFMD) integrated with near-field communication (NFC) technology for point-of-care detection of MPXV. The platform utilizes a C─C mismatch-mediated Ag+ intercalation mechanism, generating distinct current shifts detectable via differential pulse voltammetry (DPV). A microfluidic device with optimized serpentine mixing channels ensures efficient Ag+ inte

Radiosensitizing in situ cancer vaccines combined with immune checkpoint blockade to overcome resistance to radioimmunotherapy.

Radiotherapy is a widely used cancer treatment; however, it is often limited by adverse effects on normal tissues, treatment-resistant tumor microenvironment (TME), and low efficacy in distant tumors. To address these limitations, TME-regulating Fe3O4 radiosensitizer was coated with immunogenic mesoporous silica (MS) to create Fe3O4@MS-NH2, which serves as both radiosensitizer and immune sensitizing adjuvant. Tumor cells cultured with Fe3O4 nanoparticles exhibited up to 2.8-fold increased DNA damage and 1.6-fold higher levels of ROS, alongside an oxygen probe concentration as low as 55 % and a

Space-Confined Hydrogel Particle-Based Biosensor for Early Warning of Aflatoxin B1 via Rapid Label-Free Fluorescence Detection of the aflD Gene.

Aflatoxin B1 (AFB1), produced by the Aspergillus species, is one of the most potent carcinogens, posing significant health risks through contamination of food supplies. Monitoring an aflatoxin biosynthesis gene such as the aflD gene provides an early warning mechanism to prevent contamination. However, traditional methods for gene detection have limitations, including the need for sophisticated equipment and slow reaction kinetics, making them unsuitable for rapid and on-site testing. To address these challenges, we developed a novel biosensing platform based on space-confinement mechanisms us

Celiac disease risk HLA-DQ haplotypes in an Egyptian cohort using multiplex ligation-dependent probe amplification.

Celiac disease (CD) is one of the most common autoimmune disorders. It is triggered by exposure to dietary gluten proteins resulting in small intestine mucosal injury. Previous studies showed that CD is highly associated with human leukocyte antigens (HLA) class II DQ heterodimers, mainly HLA-DQ2.5 and HLA-DQ8. The aim of the work was to evaluate the distribution of the CD associated risk HLA-DQ haplotypes in CD patients, CD patients with Type 1 diabetes mellitus (T1DM) comorbidity, in at-risk and healthy individuals in an Egyptian cohort. The study included 124 individuals, divided into 4 gro

A "signal-on" photoelectrochemical biosensor utilizing in-situ synthesis of SnS2/MgIn2S4 heterostructures and enzyme-assisted target cycling amplification for the sensitive detection of the mecA gene.

To address the urgent need for rapid detection of drug resistance biomarkers for methicillin-resistant Staphylococcus aureus (MRSA), particularly the mecA gene that encods the low-affinity penicillin-binding protein PBP2a, this study developed a dual-amplification photoelectrochemical biosensing platform utilizing in-situ synthesized SnS2/MgIn2S4 heterojunction photoanodes. This system synergistically combines the co-sensitization effect of AgInS2 nanoparticles with Exonuclease III-driven target circular amplification: a type II heterojunction structure was constructed through the low-temperat

Generating Whole Bacterial Genomes from Clinical Samples using a Target Enrichment Workflow.

Obtaining complete genomes of bacterial sexually transmitted infections (STIs) directly from clinical samples is challenging due to the presence of human DNA, microbiota, and very low bacterial pathogen loads. Culture is often not an option, as these species are fastidious, and most samples are taken in transport media that lyse microorganisms, rendering them non-viable for culture. To address these issues, we used a probe panel designed across four species to generate whole-genome sequences of bacterial STIs and performed target enrichment, a two- to three-day hybridization procedure prior to

Cell-free DNA next-generation sequencing for Mycobacterium tuberculosis obtained from plasma of children with active tuberculosis.

BACKGROUND: Difficulties in microbiologically confirming childhood tuberculosis (TB) can result in delayed treatment and increased disease severity. METHODS: In this study, we for the first time used whole genome next-generation sequencing (NGS) to detect cell-free DNA (cfDNA) from Mycobacterium tuberculosis (MTB) in plasma from children. RESULTS: We enrolled 94 children with active TB and 32 children with other respiratory infections. Combining NGS with probe capture enrichment (targeted cfNGS) showed higher coverage and detecting capability than did NGS alone. The targeted cfNGS showed sli

Portable Saliva Sensor Based on Dual Recognition Elements for Detection of Caries Pathogenic Bacteria.

Dental caries is one of the most common diseases affecting more than 2 billion people's health worldwide. In a clinical setting, it is challenging to predict and proactively guard against dental cavities prior to receiving a confirmed diagnosis. Streptococcus mutans (S. mutans) in saliva has been recognized as the main causative bacterial agent that causes dental caries. High sensitivity, good selectivity, and a wide detection range are incredibly important factors to affect S. mutans detection in practical applications. In this study, we present a portable saliva biosensor designed for the ea

CpG Methylation of Receptor Activator NF-κB (RANK) Gene Promoter Region Delineates Senescence-Related Decrease of RANK Gene Expression.

While the rapid decrease in estrogen is well known as the main cause of postmenopausal osteoporosis in women, the precise pathogenesis of senile osteoporosis in the elderly regardless of gender is largely unknown. The age-related epigenetic regulation of receptor activator NF-κB (RANK) gene expression was investigated with the use of a high-passaged mouse osteoclast progenitor cell line, RAW264.7, as an in vitro model of aging. In the RAW264.7 cells after repeated passages, receptor RANK expression was downregulated, resulting in decreased soluble RANK ligand (sRANKL)-induced osteoclastogenesi

Novel 4.18 Mb deletion resulting in 2q37 microdeletion syndrome combined with PTH resistance found in one Chinese patient.

BACKGROUND: 2q37 microdeletion syndrome is a rare clinical condition characterized by a series of physical abnormalities. Its Albright hereditary osteodystrophy (AHO)-like manifestations and possible complication of biochemical abnormalities indicating PTH resistance greatly increased the likelihood of misdiagnosis with classic pseudohypoparathyroidism (PHP) caused by GNAS mutation or methylation alteration, even though there have only been six reports of such clinical occasions. PURPOSE: to investigate the underlying genetic defect in a male patient presenting hypocalcemia, elevated PTH and

Fungal cell barriers and organelles are disrupted by polyhexamethylene biguanide (PHMB).

The similarities between fungal and mammalian cells pose inherent challenges for the development of treatments for fungal infections, due to drug crossover recognition of host drug targets by antifungal agents. Thus, there are a limited number of drug classes available for treatment. Treatment is further limited by the acquisition and dissemination of antifungal resistance which contributes to the urgent need of new therapies. Polyhexamethylene biguanide (PHMB) is a cationic antimicrobial polymer with bactericidal, parasiticidal and fungicidal activities. The antifungal mechanism of action app
