Regional Excitatory-Inhibitory Balance Relates to Self-Reference Effect on Recollection via the Precuneus/Posterior Cingulate Cortex-Medial Prefrontal Cortex Connectivity.

Self-related representation can enhance perception and memory-a phenomenon known as the self-referential effect (SRE). While SRE has been linked to the activation of the default mode network (DMN), including the precuneus/posterior cingulate cortex (Pcu/PCC) and the medial prefrontal cortex (mPFC), the underlying neurochemical processes of DMN activations remain unclear. The balance of excitation and inhibition (E/I balance) within brain circuits is crucial for cognition and may play a role in the SRE. We examine whether the ratio of glutamate/glutamine (Glx) to γ-aminobutyric acid (GABA) conc

Prenatal Valproate Exposure Affects Cortical Neurite Branching, GABAergic Markers, Motor Reflexes and Ultrasonic Vocalizations in the Male Rat Pups.

Autism spectrum disorder (ASD) is a complex neurodevelopmental condition, characterized by early-onset deficits in social interactions, communication, and repetitive behaviors. Dysfunction of gamma-aminobutyric acid (GABA) neurotransmission during neural development has been implicated as a potential mechanism, particularly within the frontal cortex. This study aimed to investigate the effects of prenatal valproate (VPA) exposure on the sensorimotor reflexes, ultrasonic vocalization (USV), and GABAergic markers in the frontal cortex of male offspring. The morphology of primary cortical neurons

Glutamatergic dysfunction of astrocytes in paraventricular nucleus of thalamus contributes to adult anxiety susceptibility in adolescent ethanol exposed mice.

Repeated ethanol exposure during adolescence increases adult anxiety risk, but the underlying mechanisms remain unclear. The paraventricular nucleus of the thalamus (PVT) has been considered a hub for controlling anxiety and is affected by experiences from early life. Thus, this study investigated how adolescent intermittent repeated ethanol exposure (AIE) affects the PVT activities and anxiety-related behaviors in adulthood. We found that AIE triggers anxiety-like behaviors and parallelly exhibited elevated firing rates and increased calcium signaling in the PVT neurons compared to control co

The periaqueductal gray in chronic low back pain: dysregulated neurotransmitters and function.

Mechanisms underlying chronic pain are insufficiently understood, hampering effective treatment approaches. Preclinical evidence suggests a potential contribution of decreased excitatory (glutamatergic) and increased inhibitory (γ-aminobutyric acid [GABA]ergic) neurotransmission in the periaqueductal gray (PAG), a key descending pain modulatory brainstem area. This magnetic resonance spectroscopy (MRS) study investigated (1) whether a lower excitatory/inhibitory balance is also observed in the PAG of patients with nonspecific chronic low back pain (CLBP) and (2) whether the excitatory/inhibito

A novel fungal alkaloid protects retinal ganglion cells against glutamate-induced excitotoxicity in vitro and in vivo via activation of GABA pathway.

Retinal ganglion cells (RGCs) degeneration drives irreversible vision loss in glaucoma, ischemic/hereditary optic neuropathies, and demyelinating disorders, yet current therapies inadequately prevent RGC death. While fungal-derived compounds have revolutionized drug discovery, their neuroprotective potential remains underexplored. This study investigates the soil-derived fungus Umbelopsis sp. DWS 372 for novel neuroprotective agents targeting RGC preservation. Ten previously uncharacterized alkaloids and polyketides (1-10), including a new natural product (10), were isolated from rice-fermente

Hippocampal synaptic and membrane function in the DBA/2J-mdx mouse model of Duchenne muscular dystrophy.

Dystrophin deficiency is associated with alterations in cell physiology. The functional consequences of dystrophin deficiency are particularly severe for muscle physiology, as observed in Duchenne muscle dystrophy (DMD). DMD is caused by the absence of a 427 kDa isoform of dystrophin. However, in addition to muscular dystrophy symptoms, DMD is frequently associated with memory and attention deficits and epilepsy. While this may be associated with a role for dystrophin in neuronal physiology, it is not clear what neuronal alterations are linked with DMD. Our work shows that CA1 pyramidal neuron

The Relationship between the Level of Anterior Cingulate Cortex Metabolites, Brain-Periphery Redox Imbalance, and the Clinical State of Patients with Schizophrenia and Personality Disorders.

Schizophrenia is a complex mental disorder whose course varies with periods of deterioration and symptomatic improvement without diagnosis and treatment specific for the disease. So far, it has not been possible to clearly define what kinds of functional and structural changes are responsible for the onset or recurrence of acute psychotic decompensation in the course of schizophrenia, and to what extent personality disorders may precede the appearance of the appropriate symptoms. The work combines magnetic resonance spectroscopy imaging with clinical evaluation and laboratory tests to determin

Down-expressed GLT-1 in PSD astrocytes inhibits synaptic formation of NSC-derived neurons in vitro.

Little is known about the effect of astroglial GLT-1 of post-stroke depression (PSD) rat model on the function of neural stem cells (NSCs). This study aimed to investigate whether astroglial GLT-1 of PSD rats affect differentiation of NSCs from neonatal rat hippocampus and synaptic formation of NSC-derived neurons. Astrocytes were isolated from the left hippocampus of normal adult SD rats and PSD rats. A lentiviral vector was used to silence the expression of GLT-1 in astrocytes of PSD rats. NSCs were respectively co-cultured with normal (control), PSD, and GLT-1 silenced astrocytes for 7 days

Perinatal thiamine restriction affects central GABA and glutamate concentrations and motor behavior of adult rat offspring.

The purposes of the present study were to investigate the effects of perinatal thiamine deficiency, from the 11th day of gestation until the 5th day of lactation, on motor behavior and neurochemical parameters in adult rat offspring, using 3-month-old, adult, male Wistar rats. All rats were submitted to motor tests, using the rotarod and paw print tasks. After behavioral tests, their thalamus, cerebellum and spinal cord were dissected for glutamate and GABA quantifications by high performance liquid chromatography. The thiamine-restricted mothers (RM) group showed a significant reduction of ti

The role of mitochondrial function in glutamate-dependent metabolism in neuronal cells.

Glutamate is an important neurotransmitter in neurons and glial cells and it is one of the keys to the neuron-glial interaction in the brain. Glutamate transmission is strongly dependent on calcium homeostasis and on mitochondrial function. In the present work we presented several aspects related to the role of mitochondria in glutamate signaling and in brain diseases. We focused on glutamateinduced calcium signaling and its relation to the organelle dysfunction with cell death processes. In addition, we have discussed how alterations in this pathway may lead or aggravate a variety of neurodeg

Clinical characteristics and pathophysiological mechanisms of focal and diffuse traumatic brain injury.

Traumatic brain injury (TBI) is a frequent and clinically highly heterogeneous neurological disorder with large socioeconomic consequences. TBI severity classification, based on the hospital admission Glasgow Coma Scale (GCS) score, ranges from mild (GCS 13-15) and moderate (GCS 9-12) to severe (GCS ≤ 8). The GCS reflects the risk of dying from TBI, which is low after mild (∼1%), intermediate after moderate (up to 15%) and high (up to 40%) after severe TBI. Intracranial damage can be focal, such as epidural and subdural haematomas and parenchymal contusions, or diffuse, for example traumatic a

Long-term enhancement of synchronized oscillations by adrenergic receptor activation in the olfactory bulb.

The noradrenergic system is widely thought to be important for associative learning in the olfactory system through actions in the first processing structure, the main olfactory bulb (MOB). Here, we used extracellular local field potential (LFP) and patch-clamp recordings in rat MOB slices to examine norepinephrine (NE)-induced long-term changes in circuit properties that might underlie learning. During responses to patterned olfactory nerve stimulation mimicking the breathing cycle, NE induced a long-term increase in gamma frequency (30-70 Hz) synchronized oscillations. The enhancement persis

Bronchial epilepsy or broncho-pulmonary hyper-excitability as a model of asthma pathogenesis.

Over the last 20 years, the prevalence of asthma has nearly doubled in industrialized countries. A similar increase has been predicted for the next two decades. Asthma is major illness in terms of morbidity and suffering, asthma is the leading cause of hospitalizations in children under 15 years of age. According to many top experts, asthma is correctly characterized as a syndrome rather than disease. This lack of definition for asthma makes the search for a cause, prevention and potential cure elusive. Episodic airway obstruction and reversible bronchial hyperresponsiveness to non-specific ir

The involvement of glutamatergic transmission in the mechanism of movement disorders induced by reversive rotation of white mice.

The ability of the selective non-competitive NMDA receptor blocker MK-801 and a series of new glutamate antagonists --the adamantane derivatives IEM-1754 and IEM-1857 and phencyclidine (IEM-1925)--to prevent movement disorders induced by reversive rotation in mice was studied. l.p. MK-801 at a dose of 0.15 ml and IEM-1754 at a dose of 5.0 mg/kg prevented the development of akinesia in response to reversive rotation as effectively as scopolamine, a known agent which provides effective prophylaxis for movement diseases. IEM-1857, the quaternary analog of IEM-1754, was not effective. IEM-1925 sig

Hypoxia-tolerant neonatal CA1 neurons: relationship of survival to evoked glutamate release and glutamate receptor-mediated calcium changes in hippocampal slices.

Neurons in the neonatal mammalian brain survive greater degrees of hypoxic stress than those in the mature brain. To investigate how developmental changes in glutamate receptor-mediated neurotoxicity contribute to this difference, we measured hypoxia-evoked glutamate release, glutamate receptor contribution to hypoxia-evoked intracellular calcium changes, and survival of hypoxia-/ischemia-sensitive CA1 neurons in rat hippocampus. Glutamate release was measured by a fluorescence assay, calcium changes in CA1 neurons with fura-2, and cell viability using Nissl and fluorescence staining with calc

Distribution of perlecan in mouse hippocampus following intracerebroventricular kainate injections.

The distribution of the heparan sulphate proteoglycan (HSPG), perlecan, was studied by immunocytochemistry in the normal mouse hippocampus after intracerebroventricular injections of the potent convulsant and neurotoxin, kainate. A light staining to perlecan was observed in neurons in the normal hippocampus. Following kainate injection, an increase in perlecan immunoreactivity was observed in degenerating neurons from one to three post-injection days, followed by glial cells from 5 days to 4 weeks post-injection. The latter were found at electron microscopy to contain light cytoplasm and dense

The structure of the superantigen exfoliative toxin A suggests a novel regulation as a serine protease.

Exfoliative toxin A (ETA) causes staphylococcal scalded skin syndrome which is characterized by a specific intraepidermal separation of layers of the skin. The mechanism by which ETA causes skin separation is unknown although protease or superantigen activity has been implicated. The X-ray crystal structure of ETA has been solved in two crystal forms to 2.1 and 2.3 A resolution and R-factors of 17% and 19%, respectively. The structures indicate that ETA belongs to the chymotrypsin-like family of serine proteases and cleaves substrates after acidic residues. The conformation of a loop adjacent

Neurotoxic effect of intranigral injection of 1-methyl-4-phenylpyridinium on GABA-containing neurons and its relation to circling behavior.

The ionic species 1-methyl-4-phenylpyridinium (MPP+) seems to be the metabolite responsible for the damage to dopaminergic neurons occurring after administration of the parkinsonian drug 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine. In the present study we show that the unilateral stereotaxic microinjection of MPP+ into the substantia nigra pars reticulata in rats produces immediately intense and long-lasting (up to 96 h) contralateral turning behavior in a dose-dependent manner. This behavioral effect was correlated with a dose- and time-dependent decrease (up to 90%) of glutamate decarboxyla
