Characterisation of oral and i.v. glucose handling in truncally vagotomised subjects with pyloroplasty.

OBJECTIVE: Glucagon-like peptide 1 (GLP1) is rapidly inactivated by dipeptidyl peptidase 4 (DPP4), but may interact with vagal neurons at it … in vagotomised subjects. Despite this, the incretin effect was equal in both groups. DPP4i enhanced insulin secretion in controls, but had no effect in the vagotomised subjects. Controls suppressed glucagon concentrations similarly, irrespective of the route of glucose administration, whereas vagotomised subjects showed suppression only during IIGI and exhibited hy …

[A case of recessive dystrophic epidermolysis bullosa associated with dwarfism with special reference to pathophysiological role of growth hormone].

Epidermolysis bullosa is a group of disorders whose common primary feature is the formation of blisters following trivial trauma. Recessive dystrophic epidermolysis bullosa (RDEB), a subtype of epidermolysis bullosa, is frequently associated with growth retardation. This growth retardation has been reported to be caused by trophopathy following protein loss through skin lesions. Endocrine disorders as the cause of growth retardation in RDEB have not been clearly described. An 11-year-old female had a typical RDEB with dwarfism. Her height was 125 cm and weight was 21 kg, both of which were 2.5

Oxidized metabolites of linoleic acid as biomarkers of pancreatic beta-cell function in the first-onset type 2 diabetic patients with obesity.

OBJECTIVES: The role of oxylipins in type 2 diabetes (T2D) with obesity remains poorly understood. This study aimed to characterize oxylipin profiles in T2D patients with obesity, identify associations with pancreatic beta-cell function, and suggest potential therapeutic targets. METHODS: We recruited first-onset T2D patients with obesity (OB group, n = 15) and normal-weight T2D patients (NW group, n = 15), matched by gender, age, and blood lipid levels. Serum samples were collected in a fasting state before medication. Oxylipin profiling was analyzed using liquid chromatography-tandem mass sp

Clinical significance of miR-506-5p in type 2 diabetes mellitus and its regulatory effect on pancreatic β-cell proliferation.

INTRODUCTION: One of the core pathological features of type 2 diabetes mellitus (T2DM) is pancreatic β-cell dysfunction. MicroRNAs, askey re … verexpression of miR-506-5p suppressed pancreatic β-cell viability, promoted apoptosis, and reduced insulin secretion, whereas knockdown reversed these effects. miR-506-5p could directly target PAK1 and negatively regulate its expression, and knockdown of PAK1 could reverse the protective effect of miR-506-5p inhibitor. CONCLUSIONS: miR-506-5p is highly expressed …

NAMPT activity plays a key role in driving autoimmune processes that mediate beta-cell death and type 1 diabetes development in mice.

Type 1 diabetes (T1D) is characterised by destruction of pancreatic beta-cells by islet-infiltrating cytotoxic lymphocytes and elevated intra-islet secretion of pro-inflammatory cytokines. However, the underlying pathophysiological mechanisms remain incompletely understood. We hypothesised that abnormal elevation of islet NAD, via activation of NAMPT, plays a key role in driving islet autoimmune processes, leading to beta-cell death in T1D. Here, we report that NAMPT inhibition protects against pro-inflammatory cytokine (IL-1β, TNFα and IFNγ) mediated beta-cell dysfunction and apoptosis in iso

Early postpartum dysglycemia after gestational diabetes: β-cell dysfunction and risk stratification in a multiethnic cohort.

INTRODUCTION: Postpartum oral glucose tolerance test (OGTT) screening is recommended after gestational diabetes (GDM), but attendance is often incomplete and testing may not capture metabolic heterogeneity. Western-derived risk models may be less applicable to Asians, in whom dysglycemia may occur at lower body mass index (BMI) and reflect greater β-cell vulnerability. We examined the prevalence, clinical phenotype and predictors of early postpartum dysglycemia in women with GDM. METHODS: We retrospectively studied 575 women with GDM managed at a tertiary center in Singapore between 01/10/202

Contribution of cytokeratin 19-expressing cells towards islet regeneration induced by multipotent stromal cell secreted proteins.

Residual beta cell function has been documented in "medalist" patients who have lived with Type 1 diabetes (T1D) for >50 years. In addition, endocrine cell neogenesis first occurs in the developing human embryo from progenitor cells derived from pancreatic ductal epithelial structure. Thus, beta cell conversion from a dormant epithelial precursor remains a promising approach to regenerate islets during T1D. We have previously shown that intra-pancreatic (iPan) injection of Wnt pathway-stimulated conditioned media (Wnt+ CdM) generated from human bone marrow-derived multipotent stromal cells (MS

Declining FXR expression coordinates neonatal beta cell mass development with microbial bile acid metabolism maturation in mice.

AIMS/HYPOTHESIS: Diet switch during weaning induces gut microbiome maturation, accompanied by the formation of adequate functional beta cell mass. Bile acid (BA), an essential microbial metabolite, regulates host glucose homeostasis by binding to its main receptor, farnesoid X receptor (FXR, encoded by NR1H4). However, the precise roles of microbial BA metabolism and FXR signalling in neonatal beta cell development are still unclear. METHODS: Islet FXR levels were determined at different perinatal stages. Postnatal changes in gut microbiome and BA profiles were examined in mice, with changes

Islet regeneration protein Reg3g promotes macrophage clearance of β cell-derived dysfunctional mitochondria-rich vesicles to mitigate T2DM.

Under metabolic stress in type 2 diabetes mellitus (T2DM), β cells accumulate damaged mitochondria, and proinflammatory macrophages infiltrate pancreatic islets. In several tissues, mitochondrial transfer between macrophages and parenchymal cells has been shown to alleviate inflammation and sustain cellular function reponse to stress. However, whether a similar process occurs between pancreatic β cells and macrophages remains unclear. Here, we identified a form of intercellular communication mediated by damaged mitochondrial-rich extracellular vesicles (mEVs) from β cells to macrophages within

3D Imaging Resolves Human Pancreatic Duct-β-Cell Clusters During Cystic Change.

Pancreatic cystic changes in adults are increasingly identified through advanced cross-sectional imaging. However, the impact of initial/intralobular epithelial remodeling on the local β-cell population remains unclear. In this study, we examined 10 human cadaveric donor pancreases (tail and body regions) via integration of stereomicroscopy, clinical hematoxylin and eosin histology, and three-dimensional (3D) immunohistochemistry, identifying 36 microcysts (size: 1.22 ± 0.56 mm) alongside 54 low-grade pancreatic intraepithelial neoplasias (positive control of epithelial remodeling; size: 2.42

Proinflammatory Cytokines Mediate Pancreatic β-Cell-Specific Alterations to Golgi Integrity via iNOS-Dependent Mitochondrial Inhibition.

UNLABELLED: Type 1 diabetes (T1D) is caused by the selective autoimmune ablation of pancreatic β-cells. Emerging evidence reveals β-cell secretory dysfunction arises early in T1D development and may contribute to diseases etiology; however, the underlying mechanisms are not well understood. Our data reveal that proinflammatory cytokines elicit a complex change in the β-cell's Golgi structure and function. The structural modifications include Golgi compaction and loss of the interconnecting ribbon resulting in Golgi fragmentation. We further show that Golgi structural alterations coincide with

Regenerative medicine approaches for treating diabetes: Current advances and future directions.

Diabetes mellitus, a chronic metabolic disorder characterized by hyperglycemia, has become a global health issue. The disease primarily presents as type 1 diabetes (T1D), caused by autoimmune β-cell destruction; type 2 diabetes (T2D), driven by insulin resistance and β-cell dysfunction; and monogenic diabetes, resulting from single-gene mutations affecting insulin production or β-cell function. Chronic hyperglycemia increases the risk of severe complications, including cardiovascular disease, neuropathy, and kidney failure. Despite advancements in insulin therapy and glucose monitoring, no cur

Searching for protein partners of short-chain 3-hydroxyacyl-CoA dehydrogenase (SCHAD) reveals keratin 8 as a novel candidate for interaction in pancreatic β-cells.

BACKGROUND: Short-chain 3-hydroxyacyl-CoA dehydrogenase (SCHAD) is a ubiquitously expressed mitochondrial enzyme with a role in the degradation of fatty acids. Because the protein also is a negative regulator of insulin secretion in pancreatic β-cells, inactivating mutations in the SCHAD gene (HADH) cause congenital hyperinsulinism of infancy (CHI) and severe hypoglycemia. Here we sought to identify novel interaction partners of SCHAD that might be particularly relevant for the endocrine pancreas. RESULTS: Employing the SCHAD protein as bait, we performed yeast 2-hybrid screening of a cDNA li…

Serotonin transporter imaging agent as a probe for β-cells of pancreas.

OBJECTIVE: Diabetes mellitus (DM) is one of the major diseases in the world. Nuclear medicine imaging may be able to detect functional status of pancreatic β cells in vivo, which might elucidate the pathological mechanisms of diabetes and develop individualized treatment plans. In this study, we evaluated the ability of [125I]ADAM, a serotonin transporter (SERT) imaging agent, as a probe for detecting pancreatic β-cell mass (BCM). METHODS: In vitro cell studies were evaluated in INS-1 cells (rat islet β cell line). Biodistribution studies were performed in male normal Sprague-Dawley rats and

Anti-Diabetic Potentials of Lactobacillus Strains by Modulating Gut Microbiota Structure and β-Cells Regeneration in the Pancreatic Islets of Alloxan-Induced Diabetic Rats.

Diabetes mellitus, a most common endocrine disorder of glucose metabolism, has become a global epidemic and poses a serious public health threat with an increased socio-economic burden. Escalating incidence of diabetes is correlated with changes in lifestyle and food habits that cause gut microbiome dysbiosis and β-cells damage, which can be addressed with dietary interventions containing probiotics. Hence, the search for probiotics of human origin with anti-diabetic, anti-AGE, and anti-ACE potentials has gained renewed interest for the effective management of diabetes and its associated compl

UTRs and Ago-2/miR-335 Complex Restricts Amylin Translation in Insulinoma and Human Pancreatic β-Cells.

Amylin promoter and transcriptional factors are well-established, inducible factors in the production of the main amyloidogenic pancreatic hormone, human islet amyloid peptide (hIAPP) or amylin. However, posttranscriptional mechanisms driving hIAPP expression in pancreas remain enigmatic, and hence were explored here. The translational assay revealed that both 5' and 3' untranslated regions (UTRs) of hIAPP restricted expression of the luciferase constructs only in constructs driven by the hIAPP promoter. Bioinformatics analysis revealed several putative seed sequences for a dozen micro RNAs (m
