Integrative single-cell, spatial, and bulk transcriptomics reveal an FMR1-FTO axis linked to the immune-excluded phenotype in gastric cancer.

BACKGROUND: Immune exclusion is a major barrier to immune checkpoint inhibitor (ICI) efficacy in gastric cancer, yet the spatial mechanisms by which m6A regulators drive this phenotype remain unclear. METHODS: We integrated single-cell RNA-seq, spatial transcriptomics, and functional assays to map the gastric cancer microenvironment and derived an m6A regulator score to quantify the spatial coupling between m6A patterns and immune exclusion. RESULTS: CAFs emerged as a central hub that excluded T cells via collagen-integrin interactions and MIF signaling, forming a CAF-defined collagen barrie

Coupling fibroblast mechanotransduction signaling to tissue growth in a multiscale model of skin expansion.

Skin growth and remodeling underlies health, disease, and treatments such as tissue expansion (TE). The mechanotransduction pathways in dermal fibroblasts are increasingly well characterized, and tissue-level growth has been described phenomenologically, but coupling between cell-level signaling and tissue-level growth remains poorly understood. We develop a dermal fibroblast signaling network through extensive literature data curation, comprising 151 reactions among 96 nodes. The inputs are mechanical stretch and eight ligands (TGF β , PDGF, FGF, IL1, IL6, TNF α , AngII, ET1); outputs of inte

LncRNA H19 drives atherosclerosis progression via the miR-let-7a/ITGB3 axis.

BACKGROUND: Atherosclerosis (AS) is a chronic inflammatory disease and a leading cause of global morbidity and mortality. Dysregulated expression of the long non-coding RNA (lncRNA) H19 has been implicated in AS progression. However, the underlying molecular mechanisms remain unclear. METHODS: To determine the role of H19 in AS, the AS mouse model was established using ApoE-/- mice fed a high-fat diet, and the AS cell model was generated by stimulating human umbilical vein endothelial cells (HUVECs) with oxidized low-density lipoprotein (oxLDL). H19 expression levels were subsequently measure

LncRNA FTX promotes myocardial fibrosis by sponging miR-335-3p to regulate TFEC/ILK signaling.

Myocardial fibrosis is a pivotal contributor to heart failure progression. This study aimed to investigate the molecular mechanisms underlying the role of transcription factor EC (TFEC) in myocardial fibrosis. Myocardial fibrosis was induced in mice via isoproterenol (ISO) administration, and human cardiac fibroblasts (CFs) were stimulated with TGF-β1. Gene overexpression or knockdown was achieved through transfection. Gene and protein expression levels were quantified using quantitative real-time PCR (qRT-PCR) and Western blotting. Fibrotic changes, including collagen deposition and the expre

Rare Hematopoietic Stem Cells resilient to infection-induced stress sense yet withstand inflammation.

Hematopoietic stem cells (HSCs) sustain lifelong hematopoiesis as their progeny differentiate into all blood cell lineages. Homeostatic HSCs are mostly quiescent and only rarely divide, however their proliferation and differentiation rates can be modulated by external factors. Acute and chronic infections from a wide range of pathogens are known to challenge HSCs at the population level, being forced to respond to inflammation-mediated organismal demand to replenish the myeloid cell pool. However, less is known about the degree of heterogeneity in the HSCs' response to inflammation at the sing

Targeting the ICAM-1/LFA-1 adhesion axis attenuates lethal Crimean-Congo hemorrhagic fever virus infection.

Crimean-Congo hemorrhagic fever virus (CCHFV) is a highly pathogenic tick-borne bunyavirus. Infection and dysfunction of the vascular endothelium are likely key factors in CCHFV pathogenesis, yet the underlying mechanisms remain poorly understood. Using a mouse model, we demonstrated that CCHFV directly infects vascular endothelium, leading to marked activation and damage, as evidenced by the upregulation of adhesion molecules. To further investigate this phenomenon, we performed a quantitative proteomics study on CCHFV-infected human umbilical vein endothelial cells (HUVECs), confirming a sig

cRGD-Functionalized Calcium Carbonate Nanoprobe for Enhanced NIR-II Fluorescence Imaging and Tumor-Targeted Navigation.

Indocyanine green (ICG) holds promise for near-infrared-II (NIR-II) fluorescence imaging but is hindered by poor photostability, rapid clearance, and lack of targeting specificity. To address these challenges, we engineered a tumor-targeted nanoprobe (cRGD@ICN) by encapsulating ICG within a calcium carbonate (CaCO3) core, followed by surface functionalization with phospholipids and cyclic RGD (cRGD) peptides. The CaCO3 encapsulation enhanced ICG photostability and amplified NIR-II emission by 9.21-fold relative to free ICG. Meanwhile, cRGD conjugation enabled active targeting of integrin αvβ3,

Neonatal Glanzmann's Thrombasthenia Presenting as Refractory Post-circumcision Hemorrhage in a Region of High Consanguinity: A Case Report.

Glanzmann's thrombasthenia (GT) is a rare autosomal recessive disorder characterized by deficient or dysfunctional platelet glycoprotein IIb/IIIa (αIIbβ3 integrin), the principal fibrinogen receptor. In neonates, GT is often clinically silent until unmasked by a hemostatic challenge such as surgery or trauma. Standard coagulation screens are normal, making timely diagnosis difficult and potentially leading to life-threatening complications. We report the case of a seven-day-old Saudi boy from Najran who developed severe, refractory hemorrhage following circumcision. Initial surgical hemostasis

Rh3R Attenuates RANKL-Induced Osteoclast Differentiation and F-Actin Ring Formation via the Suppression of c-Fos/NFATc1 Signaling in Primary Murine Cells.

The homeostatic balance of bone remodeling is governed by the precise coordination between bone-forming osteoblasts and bone-resorbing osteoclasts. In this study, we investigated the anti-resorptive properties of rhamnocitrin-3-rhamnoside (Rh3R), a flavonoid isolated from Loranthus tanakae, using primary bone marrow-derived macrophages (BMMs) and calvaria-derived osteogenic progenitor cells (COCs) to ensure biological relevance. Our findings demonstrate that Rh3R potently inhibits the RANKL-induced differentiation of BMMs into TRAP-positive multinucleated osteoclasts in a dose-dependent manner

Platelet FcγRIIA is dispensable for hemostasis and arterial thrombosis in the absence of FcγRIIA-specific extracellular stimuli.

BACKGROUND: FcγRIIA, a high-affinity receptor for immunoglobulin G immune complexes with no ortholog in mice, has a prominent role in host defense against pathogens via immune cell activation. A role for platelet FcγRIIA in enhancing thrombosis was suggested, involving interplay between αIIbβ3 integrin and FcγRIIA, without requiring FcγRIIA-specific extracellular stimuli. Recent data challenged this view, showing that FcγRIIA inhibition neither impaired platelet spreading on fibrinogen nor prevented thrombus growth and stability under flow. OBJECTIVES: This study assessed contribution of FcγR

Macrophage-fibroblast communication driven by biomechanical stress in the pulmonary profibrotic niche: From signalling transduction to clinical translation.

Pulmonary (interstitial) fibrosis, the terminal pathological stage of various interstitial lung diseases, remains a major therapeutic challenge. The three approved antifibrotic agents - pirfenidone, nintedanib, and nerandomilast - receive only moderate endorsements in current guidelines, as they decelerate rather than arrest or reverse fibrotic remodelling, and none directly engages the mechanical dysregulation of the extracellular matrix (ECM) that underlies fibrogenesis. Macrophages and fibroblasts, two highly plastic cell populations, are the principal orchestrators of the pulmonary profibr

Rat models of thrombotic thrombocytopenic purpura reveal crucial role of placental ADAMTS13 in perinatal survival.

Thrombotic thrombocytopenic purpura (TTP), a life-threatening thrombotic microangiopathy, is caused by severe deficiency of plasma ADAMTS13 (a disintegrin and metalloprotease with thrombospondin type 1 repeats, 13) activity. Pregnancy is found to be frequently associated with the onset of acute TTP. However, how pregnancy or postpartum affects the progression of TTP and how ADAMTS13 may play a role in perinatal outcome are not known. Using CRISPR/CRISPR-associated protein 9, we generated a novel rat model of TTP by deleting 13 nucleotides in the coding region for ADAMTS13 metalloprotease domai

Flow Cytometric Profiling Reveals Platelet Dysfunction and Impaired Immune Communication in Acute and Chronic Cerebrovascular Disease.

INTRODUCTION: Beyond hemostasis and thrombosis, platelets are increasingly recognized as multifunctional cells involved in inflammation, vascular remodeling, and neurological diseases. Chronic cerebrovascular diseases impair cerebral blood flow and oxygen supply, leading to cognitive and motor dysfunction, where platelets may act as peripheral sensors reflecting neurovascular stress. This study investigated platelet activation profiles and platelet-leukocyte interactions in acute intracerebral hemorrhage (aICH), subacute intracerebral hemorrhage (sICH), and chronic cerebrovascular diseases (CC

Proteomic and Functional Comparison of Extracellular Vesicles from Wild-Type and Lyn-Deficient Stromal Cells.

Chronic lymphocytic leukemia (CLL) progression critically depends on bidirectional communication between malignant B cells and non-malignant bystander cells within the tumor microenvironment, including stromal cells, macrophages, and T lymphocytes. Among the key regulators of this cross talk is the Src-family kinase Lyn, whose activity in stromal cells controls extracellular matrix remodeling, fibrotic niche formation, and the release of tumor-supportive extracellular vesicles (EVs). This chapter presents a comprehensive proteomics-based strategy to identify Lyn-dependent effector molecules co

Grass carp reovirus activates fish Ppp1r3g to suppress innate immunity and exacerbate hemorrhage via the 14-3-3ζ/Itgb3 signaling axis.

Protein phosphatase 1 regulatory subunit 3G (PPP1R3G) is one of the regulatory subunits of protein phosphatase 1 (PP1). Current research on PPP1R3G is primarily focused on glycogen synthesis and lipid metabolism. However, the role of ppp1r3g in the innate immune response of fish has not been completely elucidated. This study demonstrated that ppp1r3g regulates RLR signaling pathway through interactions with Rig-I, Mda5, and Traf6. In parallel, Ppp1r3g inhibits platelet activation by regulating 14-3-3 zeta (14-3-3ζ) and consequently modulating the formation of the 14-3-3ζ-integrin-β3 (Itgb3) co

Trio-based whole-exome sequencing identifies convergent epithelial junction-related pathways in syndromic hidradenitis suppurativa.

INTRODUCTION: Hidradenitis suppurativa (HS)-related autoinflammatory syndromes, simply termed as syndromic HS (sHS), represent a group of rare immune-mediated inflammatory disorders in which HS coexists with systemic or cutaneous autoinflammatory features like PASH (pyoderma gangrenosum-PG-, acne and HS), PAPASH (PASH, pyogenic arthritis), PASS (PG, acne, HS, and ankylosing spondylitis), and SAPHO syndrome (synovitis, acne, pustulosis, hyperostosis, and osteitis). In recent years, genetic studies identified several novel pathogenic variants underlying sHS; however, most investigations rely exc

The role of von Willebrand factor in gastrointestinal angiodysplasia and obscure gi bleeding: a narrative review.

OBJECTIVE: Gastrointestinal bleeding (GIB) is a major cause of morbidity in von Willebrand disease (vWD), most commonly resulting from angiodysplasia. Current obscure gastrointestinal bleeding (OGIB) algorithms are primarily anatomy-based and often overlook underlying hemostatic disorders, delaying diagnosis and promoting recurrent bleeding. This review summarizes current evidence on the molecular basis, diagnosis, and management of vWD-associated GIB and proposes a mechanism-oriented diagnostic framework. METHODS: A comprehensive narrative review of experimental, translational, and clinical

Irisin-BDNF axis mediates muscle-brain communication: a complete molecular cascade and potential bidirectional feedback from peripheral activation to central protection.

Alzheimer's disease (AD) is a neurodegenerative disorder primarily characterized by cognitive decline, with core pathological mechanisms including β-amyloid (Aβ) deposition, tau protein hyperphosphorylation, neuroinflammation, and impaired synaptic plasticity. Although exercise has neuroprotective effects, the molecular mechanisms by which it mediates peripheral-central communication remain unclear. The concept of the 'muscle-brain dialogue' offers a new perspective on this process. irisin, secreted by skeletal muscle in response to exercise, forms a molecular link between peripheral exercise
