Effect of long-term exposure to constant dim light on the circadian system of rats.

The newly discovered multi-oscillatory nature of the mammalian circadian clock system and the cloning of the genes involved in the molecular mechanism that generates circadian rhythmicity have opened new approaches for understanding how mammals are temporally organized and how the mammalian circadian system reacts to the lack of normal synchronization cues. In the present study we investigated the effects of long-term exposure to constant red dim light on the pattern of the expression of Period 1 in the suprachiasmatic nuclei of the hypothalamus and of Arylalkylamine N-acetyltransferase(Aa-nat

Bioptic significance of incarcerated contents at sclerotomy sites during vitrectomy.

PURPOSE: To study the clinical significance of incarcerated materials at sclerotomy sites during vitrectomy. METHODS: Fifty samples prolapsed from the entries to standard three-port pars plana vitrectomies were collected along the scleral surface. Samples from rhegmatogenous retinal detachments (RD) (n = 28), traumatic RD (n = 4), miscellaneous vitreous hemorrhages (n = 12), and intraocular foreign bodies (n = 6) were stained with hematoxylineosin and examined under light microscopy after being fixed in paraffin sections or smeared on slides. RESULTS: The specimens collected after sclerotomy

An immunohistochemical study of endothelin-1 converting enzyme in the human eye.

PURPOSE: The important role of ET-1 in vasoconstriction has been shown for the vasoregulation in the retina, choroid and optic nerve. ET-1 induced vaso-constriction, however, can be treated effectively at the level of endothelin-1 converting enzyme (ECE-1) by ECE-1 inhibitors because ECE-1 converts biologically almost inactive big endothelin to endothelin-1 (ET-1), the most potent vasoconstrictor known. The purpose of this study was to clarify the anatomical distribution of ECE-1 in the human eye. METHODS: 11 post-mortem eyes were fixed in 4% formalin and embedded in paraffin. 4 microm thin s

Molecular evidence that human ocular ciliary epithelium expresses components involved in phototransduction.

Here we report the expression, in the human ocular ciliary epithelium and in a human nonpigmented (NPE) ciliary epithelial cell line, of genes usually restricted to cone and rod photoreceptor cells of the retina. By RT-PCR and DNA sequencing we identified the expression of rhodopsin and components linked to its deactivation, including rhodopsin kinase, recoverin, and visual arrestin. We also detected the expression of transducin (T-alpha), phosphodiesterase (PDE-alpha), and cGMP-gated channel alpha-subunits. Cultured NPE cells responded to treatment with phorbol ester by enhancing the expressi

Brain monoamines and optokinetic performances in pigmented and albino rats.

The aim of this study was two-fold: 1) To provide in DA-HAN rats the basic brain monoamine data useful for later investigations of the neurochemical effects of sensory alterations and 2) to assess whether there is a relationship between the monoaminergic pattern in medial vestibular nuclei and optokinetic performances. We comparatively studied the regional brain monoamine distribution and the optokinetic performances in pigmented DA-HAN and albino Sprague-Dawley rats. As expected, the optokinetic responses and vestibulo-ocular reflex gain were by far more efficient in DA-HAN rats. Norepinephri

Optic nerve hypoplasia in fetal alcohol syndrome: an update.

Optic nerve hypoplasia was detected in up to one half of a group of Swedish children born to alcoholic mothers. Using an experimental model of pre- and postnatal alcohol exposure in rats fed a liquid diet, reduced optic nerve size from gestational day 21 (294 +/- 26 x 10(2) microns2 vs 502 +/- 16 x 10(2) microns2; n = 6; p < or = 0.001) to later in development was observed as a result of the daily mean blood alcohol levels achieved in dams and their offspring. Altered glial cells and degenerating and atrophic optic axons, myelin sheaths and ganglion cells were frequent in the alcohol-exposed o

[Intraocular involvement of Chagas disease (American trypanosomiasis). Studies in Paraguay/South America].

BACKGROUND: In Central and South America, Chagas' disease is of great epidemiologic importance. The epidemiologic agent is represented by Trypanosoma cruzi, a monocellular parasite, instrumental in human infection is the presence of vectors, which are various species of hematophagous bugs. The eye is one of the most important entrance sites of the parasite, and relatively little information about the relationship between Chagas' disease and eye complications is available. PATIENTS AND METHODS: We examined 79 chagasic patients in order to detect changes in the retina. As a control group, we ex

Post-traumatic macular break associated to congenital optic disc pit and pre-existing sensory macular detachment.

BACKGROUND: Optic disc pits represent a rare congenital abnormality frequently associated with macular detachment. Several theories on the pathogenesis of the detachment have been proposed, but the issue still remains unclear. Treatment remains speculative as well; advocated treatments include eye-patching, bed rest, steroids, laser photocoagulation, pars plana vitrectomy with fluid-air exchange and optic nerve sheath decompression. METHODS: The Authors report on the case of an optic pit with associated macular detachment which developed an inner layer macular hole and, after an apparently in

Management of traumatic rupture of the globe in aphakic patients.

We report the successful treatment of three cases of traumatic ruptures of the globe complicated by massive choroidal hemorrhage, uveal prolapse and retinal detachment. All three of the eyes were aphakic prior to injury and all patients were age 64 or older. The presenting visual acuity in all patients was light perception. The blunt injury in each case caused a wound dehiscence at the site of previous cataract extraction. All injuries were associated with uveal prolapse. Secondary surgical intervention was performed when the hemorrhagic choroidal detachments had decreased as demonstrated by e

[Results of vitrectomy for diabetic traction retinal detachment involving the macula; a comparison of six-month and three-year postoperative findings].

Three-year follow-up examinations were performed on 45 cases that underwent pars plana vitrectomies for diabetic retinopathy complications from 1982 to 1983. All of these cases had traction retinal detachment involving the macula. Visual improvement was noted in 27 eyes (60%) and 11 eyes (24%) had worse visual acuity at 6 months after surgery. Final visual improvement was noted in 21 eyes (47%) at the there-year follow-up examinations. Five (11%) of the 45 cases had better visual acuity at the three-year examination than at six months, 18 (40%) had the same visual acuity, but 22 (49%) had poor

Retinal vasculitis.

Evidence is now accumulating on both clinical and experimental grounds that the retina is an a priori source of inflammatory activity. Reactive inflammation in the retina may produce many of the clinical signs previously ascribed to uveal inflammation. Autoimmune mechanisms are probably responsible for the majority of cases of retinal vasculitis. Autoimmune retinal vasculitis occurs without other classical signs of inflammatory response in any other parts of the body. When associated systemic manifestations occur they may reflect different underlying immunopathogenic abnormalities. Thus in dis

First pilot study of intravenous rAAV-PAX6 gene therapy increases retinal-ganglion-cell-layer thickness and Notch1 transcription in a mouse model of aniridia.

Aniridia is a rare congenital vision-loss disorder that is caused primarily by heterozygous loss-of-function variants in the PAX6 gene. There is currently no curative treatment. Gene therapy has emerged as a powerful strategy for treating inherited retinal diseases. Here, we aim to establish a systemic PAX6 gene-augmentation therapy capable of addressing retinal pathologic phenotypes in a preclinical Sey mouse model of aniridia. We evaluated the intravenous delivery of FLAG-tagged PAX6 packaged in the AAV-PHP.eB capsid. Treatment was administered on postnatal day 21 to mimic the average age at

Head-mounted surgical robots are an enabling technology for subretinal injections.

Therapeutic protocols involving subretinal injection, which hold the promise of saving or restoring sight, are challenging for surgeons because they are at the limits of human motor and perceptual abilities. Excessive or insufficient indentation of the injection cannula into the retina or motion of the cannula with respect to the retina can result in retinal trauma or incorrect placement of the therapeutic product. Robotic assistance can potentially enable the surgeon to more precisely position the injection cannula and maintain its position for a prolonged period of time. However, head motion

Cryptochrome 4b protein is probably irrelevant for radical pair-based magnetoreception in the European robin.

Avian cryptochrome 4 (Cry4) protein is a putative magnetosensitive molecule facilitating precise long-distance navigation in migratory birds. Two splice variants of Cry4 have been reported in the European robin (Erithacus rubecula), namely ErCry4a and ErCry4b. ErCry4a protein is known to generate magnetically sensitive radical pairs for magnetoreception through electron transfer between the flavin adenine dinucleotide (FAD) cofactor and tryptophan residues within the protein. However, little is known about the ErCry4b isoform. We therefore characterize the properties of ErCry4b to determine wh

Bioengineering Approaches for the 3D Blood-Retinal Barrier: Disease Modeling and Drug Development.

The integrity and functionality of the blood-retina barrier (BRB) are essential for maintaining homeostasis of the retina. Disruption of the BRB is a key feature in different stages of retinal diseases including diabetic retinopathy, age-related macular degeneration (AMD), and inherited retinal diseases such as retinitis pigmentosa. In order to understand and recapitulate various retinal diseases, there has been significant interest in the progress of in vitro 3D-BRB models as they may address limitations associated with traditional 2D cultures and xenogeneic animal models. Therefore, it is cr

miRNAs, piRNAs, and lncRNAs: A triad of non-coding RNAs regulating the neurovascular unit in diabetic retinopathy and their therapeutic potentials.

Diabetic Retinopathy (DR), a leading complication of diabetes mellitus, has long been considered as a microvascular disease of the retina. However, recent evidence suggests that DR is a neurovascular disease, characterized by the degeneration of retinal neural tissue and microvascular abnormalities encompassing ischemia, neovascularization, and blood-retinal barrier breakdown, ultimately leading to blindness. The intricate relationship between the retina and vascular cells constitutes a neurovascular unit, a multi-cellular framework of retinal neurons, glial cells, immune cells, and vascular c

Visual Function in Alzheimer's Disease: Current Understanding and Potential Mechanisms Behind Visual Impairment.

Alzheimer's disease (AD) is the leading cause of dementia worldwide and is becoming one of the most morbid diseases of this century. Recently, ocular research in AD has gained significance, as the eye, due to its close relationship with the brain, can reflect the presence of neurological disorders. Several studies have reported alterations in various ocular structures in AD, ranging from tear fluid to the retina. These changes, particularly in the retina and the optic nerve, along with cerebral atrophy affecting visual brain areas, may lead to visual dysfunctions. This narrative review summari

Retinal Phosphenes Induced by Transorbital Electrical Stimulation: Influence of Light Adaptation, Electrode Montage, and View Direction.

In this study, the perception of phosphenes was used as a surrogate identifier for stimulation sites for use in retinal-degenerative diseases. We aimed to investigate the influence of adaptation, electrode montage, and direction of view on electrically induced phosphenes. We developed a practical methodology to assess non-invasive ocular electrical stimulation, addressing specific areas in the retina. Phosphene thresholds were identified under light and dark adaptation for non-invasive transorbital electrical stimulation. The location and extent characterized the morphology of electrically ind
