Skeletal Muscle Index, Tumor and Systemic Immunity, and Survival in Advanced Pancreatic Ductal Adenocarcinoma.

BACKGROUND: This study investigated relationships between the skeletal muscle index (SMI) and the tumor immune microenvironment (TIME), peripheral immune phenotypes, and serum biomarkers in advanced pancreatic ductal adenocarcinoma (PDAC) and evaluated the prognostic significance of these parameters. METHODS: A retrospective analysis of 119 advanced PDAC patients was performed. Sarcopenia was diagnosed using sex-specific SMI cut-offs from baseline L3-level computed tomography (CT) images. The TIME was characterized by multiplex immunofluorescence (mIF; n = 31), peripheral immunity by flow cyt

In vitro and ex vivo immunomodulatory effects of human placental mesenchymal stem cells in hidradenitis suppurativa.

BACKGROUND: Hidradenitis suppurativa is a chronic inflammatory skin disease marked by immune dysregulation and elevated pro-inflammatory cytokines. While biologics like adalimumab target specific pathways, their limited efficacy highlights the need for broader immunomodulatory treatments. Mesenchymal stem/stromal cells (MSCs) have shown promise due to their immunosuppressive properties and ability to modulate both innate and adaptive immunity. This study investigates the effects of naïve (n-MSCs) and cytokine-preactivated (a-MSCs) placental MSCs on the immune responses in HS. METHODS: MSCs we

Clinicopathologic features of Good's syndrome: Two cases and literature review.

BACKGROUND: Good's syndrome (GS) is an immunodeficiency disease, causing thymoma, low or absent B-cells, hypogammaglobulinemia, and defects in cell-mediated immunity. The most common clinical presentation is recurrent infection, followed by refractory diarrhea, due to the immunodeficiency. However, there are only few reports on intestinal endoscopy and pathology. CASE SUMMARY: We report here two typical GS cases with diarrhea as the prominent manifestation. Both cases presented with thymoma combined with immunodeficiency, characterized by hypogammaglobulinemia, low or absent B lymphocytes, an

HTLV-1-Infected CD4+ T Cells Drive Spontaneous Lymphoproliferation and Virus-Specific CD8+ Cytotoxic T Lymphocyte Expansion in HAM/TSP.

BACKGROUND AND OBJECTIVES: Spontaneous lymphoproliferation (SP) is an ex vivo phenomenon where lymphocytes proliferate without exogenous stimulation in peripheral blood mononuclear cells (PBMCs) from patients with human T-cell leukemia virus type 1 (HTLV-1)-associated myelopathy/tropical spastic paraparesis (HAM/TSP). SP is thought to reflect spinal cord pathology in HAM/TSP, but its cellular mechanisms remain unclear. METHODS: PBMCs from 30 patients with HAM/TSP, 12 asymptomatic HTLV-1 carriers, and 8 healthy controls (HCs) were labeled with a proliferation tracer and cultured for 6 days wit

Complete molecular remission during biologic response modifier therapy for Sézary syndrome is associated with enhanced helper T type 1 cytokine production and natural killer cell activity.

BACKGROUND: Cutaneous T-cell lymphoma (CTCL) is a clonally derived, skin-invasive malignancy of CD4(+) T lymphocytes with the phenotype of mature helper T cells. Advancing stages of CTCL are associated with depressed cell-mediated immunity, increased production of T helper type 2 cytokines and decreased levels of T helper type 1 cytokines. OBJECTIVE: Our purpose was to evaluate the cytokine secretion pattern and cell-mediated cytotoxicity in peripheral blood mononuclear cells of patients with Sézary syndrome in relation to the presence of the malignant clone. METHODS: Serial polymerase chain

V(beta)8(+) T cells protect from demyelinating disease in a viral model of multiple sclerosis.

Previous studies illustrated the influence of T cell subsets on susceptibility or resistance to demyelination in the Theiler's murine encephalomyelitis virus (TMEV) model of multiple sclerosis. Genetic segregation analysis showed a correlation with disease phenotype in this model with particular V(beta) genes. In this study we investigated the contribution of specific V(beta) TCR to the pathogenesis of virus-induced demyelinating disease. Spectratype analysis of cells infiltrating the CNS early in infection demonstrated an over-representation of V(beta)8(+) T cells in mice expressing a suscept

Longevity of antibody responses is associated with distinct antigen-specific B cell subsets early after infection.

INTRODUCTION: Upon infection, T cell-driven B cell responses in GC reactions induce memory B cells and antibody-secreting cells that secrete protective antibodies. How formation of specifically long-lived plasma cells is regulated via the interplay between specific B and CD4+ T cells is not well understood. Generally, antibody levels decline over time after clearance of the primary infection. METHOD: In this study, convalescent individuals with stable RBD antibody levels (n=14, "sustainers") were compared with donors (n=13) with the greatest antibody decline from a cohort of 132. To investiga

Natural killer T cells recognize diacylglycerol antigens from pathogenic bacteria.

Natural killer T (NKT) cells recognize glycosphingolipids presented by CD1d molecules and have been linked to defense against microbial infections. Previously defined foreign glycosphingolipids recognized by NKT cells are uniquely found in nonpathogenic sphingomonas bacteria. Here we show that mouse and human NKT cells also recognized glycolipids, specifically a diacylglycerol, from Borrelia burgdorferi, which causes Lyme disease. The B. burgdorferi-derived, glycolipid-induced NKT cell proliferation and cytokine production and the antigenic potency of this glycolipid was dependent on acyl chai

CD1d1-dependent control of the magnitude of an acute antiviral immune response.

CD1d1-restricted NK T (NKT) cells rapidly secrete both Th1 and Th2 cytokines upon activation and are therefore thought to play a regulatory role during an immune response. In this study we examined the role of CD1d1 molecules and NKT cells in regulating virus-induced cytokine production. CD1d1-deficient (CD1KO) mice, which lack NKT cells, were infected with lymphocytic choriomeningitis virus, and spontaneous cytokine release from splenocytes was measured. We found that CD1KO mice produce significantly higher amounts of IL-2, IL-4, and IFN-gamma compared with wild-type controls postinfection. D

Interleukin-15 up-regulates the expression of CD154 on synovial fluid T cells.

To investigate the role of the CD40-CD154 interaction in rheumatoid arthritis (RA), we analysed the expression of CD154 on CD3+ and CD4+ T cells in synovial fluid (SF) from patients with RA and in peripheral blood (PB) from patients and normal controls. As interleukin (IL)-15 is a potent activator of synovial T cells we wanted to study whether IL-15 also regulated the expression of CD154 on these T cells. Freshly isolated synovial T cells did not express significant levels of CD154, as evaluated using flow cytometry, whereas the expression of CD86 and human leucocyte antigen (HLA)-DR was signi

Natural killer (NK) and lymphokine-activated killer (LAK) activities in a patient who recovered from cancer, and the characteristics of LAK cells generated from CD4-CD8- and CD8+ peripheral blood lymphocytes.

The natural killer (NK) and lymphokine-activated killer (LAK) activities of a patient who recovered from bladder transitional cell carcinoma were determined. The patient was diagnosed in 1980 and repeated several therapies until 1986. Since 1987, he has been in a tumor-free state. Recently, very high NK and LAK activities were found. The NK activity was recognized not only against the NK-sensitive cell line K562, but also against NK-unsensitive Raji and Daudi cell lines and against autotumor cell line H41. The percentage of CD16+ in peripheral blood lymphocytes (PBL) was 52.9%, the CD8+CD16+ w

Atypical memory B cells acquire Breg phenotypes in hepatocellular carcinoma.

The functional plasticity of tumor-infiltrating lymphocyte B-cells (TIL-B) spans from antitumor responses to noncanonical immune suppression. Yet, how the tumor microenvironment (TME) influences TIL-B development is still underappreciated. Our current study integrated single-cell transcriptomics and B cell receptor (BCR) sequencing to profile TIL-B phenotypes and clonalities in hepatocellular carcinoma (HCC). Using trajectory and gene regulatory network analysis, we were able to characterize plasma cells and memory and naive B cells within the HCC TME and further revealed a downregulation of B

CD56+CD57+ infiltrates as the most predominant subset of intragraft natural killer cells in renal transplant biopsies with antibody-mediated rejection.

Little is known about the characteristics and clinical implications of specific subsets of intragraft natural killer (NK) cells in kidney transplant recipients. We analyzed 39 for-cause renal transplant biopsies performed at our center from May 2015 to July 2017. According to histopathologic reports, 8 patients (20.5%) had no rejection (NR), 11 (28.2%) had T cell-mediated rejections (TCMR) only, and 20 (51.3%) had antibody-mediated rejection (ABMR). NK cells were defined as CD3-CD56+ lymphocytes that are positive for CD57, CD49b, NKG2A, or KIR. The density of NK cells was significantly higher

Integrative Phosphoproteomics Links IL-23R Signaling with Metabolic Adaptation in Lymphocytes.

Interleukin (IL)-23 mediated signal transduction represents a major molecular mechanism underlying the pathology of inflammatory bowel disease, Crohn's disease and ulcerative colitis. In addition, emerging evidence supports the role of IL-23-driven Th17 cells in inflammation. Components of the IL-23 signaling pathway, such as IL-23R, JAK2 and STAT3, have been characterized, but elements unique to this network as compared to other interleukins have not been readily explored. In this study, we have undertaken an integrative phosphoproteomics approach to better characterise downstream signaling e

Mitogenic response of neoplastic B cells: comparison of reactivity to Staphylococcus aureus Cowan I and anti-immunoglobulin antibodies.

Neoplastic B cells from two patients with hyperleucocytic hairy cell leukaemia (HCL) and 19 patients with chronic lymphocytic leukaemia of B cell Type (B-CLL) were investigated to examine the mitogenic responses to the F(ab')2 fraction of anti-human immunoglobulins (anti-Igs) and Staphylococcus aureus Cowan I (STA). Neoplastic cells from both HCL patients lacked surface Tac antigen. Mononuclear cells from the two HCL patients strongly responded to both anti-Igs and STA as measured by 3H-thymidine incorporation in vitro. Although the mononuclear cells from two patients with B-CLL showed high re

MAIT Cells Maintain Intestinal Homeostasis Through IL-22 in an Experimental Colitis.

OBJECTIVE: Mucosa-associated invariant T (MAIT) cells are a unique subset of innate-like lymphocytes abundant in the gastrointestinal tract, yet their role in maintaining intestinal homeostasis remains incompletely understood. We investigated how MAIT cells contribute to epithelial barrier integrity and the molecular mechanisms underlying this process during experimental colitis. METHODS: Acute colitis was induced by dextran sulfate sodium (DSS) in wild-type (WT) and MR1-deficient (MR1-/-) mice. Disease severity was quantified via weight loss, colon length, and histopathology. Barrier functio

Adoptive cell therapy in lung cancer: progress, challenges, and prospects.

Lung cancer is the leading cause of cancer-related deaths worldwide. Over the past decade, immunotherapy, represented by immune checkpoint inhibitors (ICIs) targeting programmed cell death protein-1/programmed cell death protein ligand-1 (PD-1/PD-L1) or cytotoxic T lymphocyte associated antigen-4 (CTLA-4), has revolutionized the treatment paradigm of lung cancer, reducing the risk of death by approximately 30%. However, approximately 70% of patients with advanced non-small cell lung cancer are either non-responsive to ICIs or initially respond to treatment but eventually experience disease pro

Increased density of CD103-positive tissue-resident memory T cells predicts favorable response to immunotherapy in oral cancers.

Immune checkpoint inhibitors (ICIs) have become an important therapeutic option for oral cancers; however, reliable biomarkers predicting treatment efficacy remain limited. In this study, we investigated the association between intratumoral immune cell subsets, regional lymph node (RLN) immune status, and clinical responses to ICIs in patients with recurrent oral cancer. Pretreatment tumor tissues and RLNs were analyzed by immunohistochemistry to evaluate tumor-infiltrating lymphocytes, including CD8+, CD103+, CD3+, ICOS+, and CD163+ cells, as well as programmed death-ligand 1 (PD-L1) expressi

Development of anti-CD4-1/CD8β monoclonal antibodies and spatiotemporal immune profiling of response to live attenuated IHNV in rainbow trout (Oncorhynchus mykiss).

Understanding the cellular mechanisms of live attenuated vaccines against infectious hematopoietic necrosis virus (IHNV) has long been hindered by the lack of species-specific immunological reagents in rainbow trout (Oncorhynchus mykiss). To bridge this gap and elucidate the protective basis of a high-efficacy mutant candidate (rIHNVMut), this study first established a core technical platform by developing and validating monoclonal antibodies (mAbs) targeting the T-cell co-receptors CD4-1 and CD8β. Validation via Western blot and immunohistochemistry confirmed the high specificity of these mAb
