The degree of interindividual and intraindividual variability in acetylator activity was investigated with caffeine used as a probe of enzyme activity. Acetylator phenotype and relative N-acetyltransferase activity were estimated in 46 subjects by measuring the urinary ratio of two metabolites, AFMU/1-MX, after a single 300 mg oral dose of caffeine on five separate occasions. Thirty homozygous slow (rr) and 15 heterozygous rapid (Rr) acetylators were identified. The degree of interindividual variability in acetylator activity was observed to be a mean of 32% (range 27% to 36%) and 20% (range 1

Acetyl-CoA participates in post-translational modification of proteins and in central carbon and lipid metabolism in several cell compartments. In mammals, acetyl-CoA transporter 1 (AT1, also known as SLC33A1) facilitates the flux of cytosolic acetyl-CoA into the endoplasmic reticulum (ER), enabling the acetylation of proteins of the secretory pathway, in concert with the activity of dedicated acetyltransferases such as NAT8. However, the involvement of the ER acetyl-CoA pool in acetylation of ER-transiting proteins in Apicomplexa is unknown. Here, we identified homologs of AT1 and NAT8 in Tox…

Salmonella are becoming increasingly resistant to fluoroquinolones (FQs), therefore determining the resistance mechanism is very important. Recent studies have shown that protein post-translational modifications (PTM) play a role in bacterial antibiotic resistance. One such type of PTM, lysine acetylation, is a reversible and highly regulated PTM which has been found to be associated with antibiotic resistance in Mycobacterium and Acinetobacter species. Salmonella Typhimurium are major zoonotic pathogens, which are becoming increasingly resistant to FQs, the antibiotics of choice where therapy…

In this study, we have constructed synthetic peptides which are identical to hyperacetylated amino termini of two Tetrahymena core histones … this variant. Immunofluorescent analyses with both antisera demonstrate that, as expected, histone acetylation is specific to macronuclei (or new macronuclei) at all stages of the life cycle except when micronuclei undergo periods of rapid replication and chromatin assembly. During this time micronuclear staining is also detected. Our results also suggest th …

Tabtoxin resistance protein (TTR) is an enzyme that renders tabtoxin-producing pathogens, such as Pseudomonas syringae, tolerant to their own phytotoxins. Here, we report the crystal structure of TTR complexed with its natural cofactor, acetyl coenzyme A (AcCoA), to 1.55A resolution. The binary complex forms a characteristic "V" shape for substrate binding and contains the four motifs conserved in the GCN5-related N-acetyltransferase (GNAT) superfamily, which also includes the histone acetyltransferases (HATs). A single-step mechanism is proposed to explain the function of three conserved resi

Eighteen healthy Caucasians were evaluated for the systemic acetylation of a caffeine metabolite using the urinary caffeine metabolite ratio 5-acetylamino-6-formylamino-3-methyluracil (AFMU) to 1-methylaxanthine (1X) and for N-acetyltransferase activity in peripheral blood mononuclear leukocytes (MNL) using p-aminobenzoic acid (PABA). These are markers for systemic NAT2 and NAT1 N-acetyltransferase activities, respectively. Fourteen slow acetylators and four fast acetylators (the NAT2 polymorphism) were identified by the caffeine metabolite ratio. In slow acetylators who have decreased levels …

The biochemical basis underlying the genetic polymorphism of drug N-acetylation was investigated using a combination of in vivo and in vitro assays for arylamine N-acetyltransferase (NAT) activity and content in human liver. The acetylator phenotype of 26 surgical patients was determined using caffeine as an innocuous probe drug by measurement of the 5-acetyl-amino-6-formylamino-3-methyluracil to 1-methylxanthine molar ratio in urine. Liver wedge biopsies from these patients and livers from 24 organ donors were then used for measurement of N-acetyltransferase activity with the substrate sulfam…

N4-acetylcytidine (ac4C) is an epitranscriptomic regulator involved in diverse pathological processes. This study investigates the role of N-acetyltransferase 10 (NAT10), the primary enzyme responsible for ac4C deposition, in aortic valve calcification (AVC). NAT10 expression and global ac4C levels are upregulated in calcified aortic valves from human patients and murine models. Clinically, elevated NAT10 expression correlates with increased disease severity. Functionally, NAT10 promotes human valvular interstitial cells (hVICs) osteogenic differentiation. In high-cholesterol diet (HCD)-fed Ap

Autoimmune diseases arise from a breakdown of immune tolerance, leading to sustained inflammation and tissue damage. Increasing evidence indicates that epigenetic dysregulation contributes to this loss of tolerance. Among diverse epigenetic mechanisms, protein and RNA acetylation/deacetylation have emerged as pivotal modulators of immune cell activation, differentiation, and effector function. Aberrant acetylation of histone and non-histone proteins has been implicated in shaping pathogenic immune responses across multiple autoimmune disorders. Concurrently, a substantial body of research has …

Assigning causal function to post-translational modifications (PTMs) remains a central challenge in molecular biology, as most modification events cannot be readily interrogated in their native cellular context. Here, we present a generalizable chemical biology strategy for investigating the functional consequences of lysine acetylation through programmable induced proximity. By combining modular effector recruitment with chemically controlled proximity, this approach enables systematic elucidation of how enzyme identity shapes acetylation outcomes on target proteins in living cells. Across mu…

BACKGROUND: All organisms experience stress and must rapidly respond to changing conditions. Thus, cells have evolved sophisticated rapid-response mechanisms such as post-translational protein modification to rapidly and reversibly modulate protein activity. One such post-translational modification is reversible lysine acetylation, where proteomic studies have identified thousands of acetylated proteins across diverse organisms. While the sheer size of the 'acetylome' is striking, the function of acetylation for the vast majority of proteins remains largely obscure.
RESULTS: Here, we find tha…

Histone acetylation, catalyzed by histone acetyltransferases (HATs) and reversed by histone deacetylases (HDACs), is a central epigenetic mechanism that regulates gene expression in eukaryotes. Identifying and characterizing these enzymes has rapidly advanced our understanding of this dynamic modification. In phytopathogenic fungi, HATs and HDACs modulate several cellular processes, including development, pathogenicity, and secondary metabolism. The filamentous fungus Fusarium graminearum causes Fusarium head blight, a destructive disease that reduces wheat yield and contaminates grain with th…

Osteoarthritis (OA) is characterized as a chronic degenerative disorder affecting the joints. A growing body of evidence indicates that acetylation may play a role in the disease's pathogenesis. However, the underlying molecular mechanisms remain largely undefined. The objective of this study was to explore potential biomarkers linked to acetylation in OA through a comprehensive bioinformatics analysis. We utilized datasets GSE55235, GSE55457, and GSE12021 from the Gene Expression Omnibus (GEO) to identify differentially expressed genes (DEGs) by employing the limma package, followed by functi…

Manganese superoxide dismutase (MnSOD/SOD2) is an essential mitochondrial enzyme that detoxifies superoxide radicals generated during oxidative respiration. MnSOD/SOD2 lysine 68 acetylation (K68-Ac) is an important post-translational modification (PTM) that regulates enzymatic activity, responding to nutrient status or oxidative stress, and elevated levels have been associated with human illness. To determine the in vivo role of MnSOD-K68 in the heart, we used a whole-body non-acetylation mimic mutant (MnSODK68R) knock-in mouse. These mice exhibited several cardiovascular phenotypes, including…

Post-translational modifications (PTMs) are critical regulators of protein function and cellular signaling. While histone deacetylation by histone deacetylases (HDACs) is well established, the role of specific HDACs in modulating non-histone protein PTMs, particularly in an infectious context, is poorly understood. Here, we reveal a pivotal role for HDAC6 in orchestrating periodontal inflammation through its dual regulatory effects on FoxO1 acetylation and phosphorylation. Using Porphyromonas gingivalis , a key periodontal pathogen, as a model pathogen, we observed that infection induces HDAC6…
