Photochemical cross-linking of 125I-hydroxyphenylisopropyl adenosine to the A1 adenosine receptor of rat adipocytes.

Rat adipocyte plasma membranes were incubated with the A1 adenosine receptor agonist, 125I-hydroxyphenylisopropyl adenosine (1 nM) and then treated with the photoactive cross-linking agent, ANB-NOS. The membranes were solubilized and analyzed by SDS-PAGE and autoradiography. A single protein, Mr approx. 38,000, was specifically labeled. Reduction with 2-mercaptoethanol did not affect the apparent Mr of the labeled protein. Labeling was inhibited by the adenosine receptor agonists, HPIA, PIA and NECA, and by the antagonist, theophylline, but was unaffected by inosine. We conclude that the A1 ad…

Deficiency of Adenosine Deaminase 2.

Adenosine deaminase 2 (ADA2) deficiency is an autosomal recessively inherited autoinflammatory disorder caused by loss-of-function mutations in the ADA2 gene. Although the pathogenesis involves the triggering of a proinflammatory cascade due to increased production of inflammatory cytokines such as tumor necrosis factor (TNF)-α and dysregulation of neutrophil extracellular trap formation resulting from an excess accumulation of extracellular adenosine, the pathogenetic mechanism still needs further clarification due to the broad clinical spectrum. In addition to the initially described vasculi…

[Adenosine, its analogues and conjugates].

Adenosine is an endogenous purine nucleoside that plays an important role in many biochemical processes. It acts through the four types of adenosine receptors: A1, A2A, A2B, A3 which are located i.a. in the immune, nervous, circulatory, respiratory or urinary system. Adenosine is used as an antiarrhythmic agent in the treatment of paroxysmal supraventricular tachycardia. However, due to a very short blood half-time other clinical applications are limited. In order to overcome this obstacle many analogues and conjugates of adenosine with better pharmacokinetic properties have been synthesized. …

Tryptophan, adenosine, neurodegeneration and neuroprotection.

This review summarises the potential contributions of two groups of compounds to cerebral dysfunction and damage in metabolic disease. The k … d may contribute substantially to delayed neuronal damage following an infarct or metabolic insult. Adenosine is an ubiquitous purine that can protect neurons by suppressing excitatory neurotransmitter release, reducing calcium fluxes and inhibiting NMDA receptors. The extent of brain injury is critically dependent on the balance between the two opposing fo …

Adenosine in burn blister fluid.

BACKGROUND: Burn wound blister fluid is known to sustain suppressive effects on various components of the immune system. Damaged tissues cause an increase of adenosine concentrations. Since adenosine is a potent anti-inflammatory agent we hypothesized that burn blister fluid contains high concentrations of this nucleoside. METHODS: Burn blister fluid was drawn from eleven patients who suffered a second degree burn injury. Adenosine concentrations were determined using high performance liquid chromatography (HPLC). RESULTS: Elevated adenosine levels were detected in 6 of the 11 patients (54.5…

Adenosine deaminase deficiency in adults.

Adenosine deaminase (ADA) deficiency typically causes severe combined immunodeficiency (SCID) in infants. We report metabolic, immunologic, and genetic findings in two ADA-deficient adults with distinct phenotypes. Patient no. 1 (39 years of age) had combined immunodeficiency. She had frequent infections, lymphopenia, and recurrent hepatitis as a child but did relatively well in her second and third decades. Then she developed chronic sinopulmonary infections, including tuberculosis, and hepatobiliary disease; she died of viral leukoencephalopathy at 40 years of age. Patient no. 2, a healthy 2…

Adenosine in treating cardiac arrhythmias.

1. Adenosine is an endogenous nucleoside which causes a brief blockade of the AV nodal conduction pathway following intravenous administration. 2. Such a brief AV block can be used therapeutically for reliable termination of AV nodal re-entry tachycardia and WPW re-entry tachycardia. It can also be used for demasking atrial activity in rapid suspected supraventricular tachycardia with a broad QRS complex or a Delta wave, not present during sinus rhythm with normal AV node conduction, indicating the presence of a hidden WPW syndrome. 3. Side effects after adenosine application are frequent, but…

Bronchospasm induced by intravenous adenosine.

1. Adenosine is well known to cause bronchoconstriction when inhaled, but to date, there has been only one report following the intravenous route. 2. Adenosine caused bronchoconstriction in two patients (the only two with asthma) out of 26 treated with the drug. In one subject the duration of response was much longer than previously reported following inhalation. 3. We suggest that adenosine may aggravate bronchospasm in some asthmatics and alternative treatments for supraventricular tachycardia should be considered in this group.

Adenosine administration for neonatal SVT.

Adenosine is a purine nucleoside that impairs conduction through the AV node and is thus effective in terminating tachycardias involving the AV node. Gaining acceptance as the drug of choice for neonatal supraventricular tachycardia (SVT), it is given IV as a rapid bolus with an initial dose of 0.05 mg/kg and can be increased in increments of 0.05 mg/kg every one to two minutes until termination of SVT (to a maximum of 0.25 mg/kg). Because of its half-life of 0.6 to 10 seconds, adenosine will not prevent reinitiation of SVT, therefore other medications should be considered if prophylaxis is re…

Adenosine for paroxysmal supraventricular tachycardia.

Adenosine is the most recent drug approved for the treatment of paroxysmal supraventricular tachycardia. Its advantage is that it is just as effective as verapamil and is far less toxic. The lack of toxicity is due to the extremely short half-life (one to seven seconds). The mechanism of action of adenosine is different from that of other antiarrhythmic agents. For the specific purpose of ameliorating paroxysmal supraventricular tachycardia, adenosine appears to be an ideal antiarrhythmic drug.

Adenosine cycle in African trypanosomes.

African trypanosomes can convert adenosine to adenosine monophosphate. However, in Trypanosoma brucei, as in T. vivax and T. congolense, most of the adenosine is broken down to adenine before conversion to the nucleotide by adenine phosphoribosyltransferase. Trypanosoma brucei and T. vivax use the purine nucleoside hydrolase for adenosine cleavage while T. congolense uses purine nucleoside phosphorylase for the nucleoside cleavage. Trypanosoma vivax also deaminates adenine to hypoxanthine before its conversion to adenosine monophosphate by way of inosine monophosphate. All African trypanosomes…

Adenosine receptor interactions and anxiolytics.

[3H]-N6-cyclohexyladenosine and [3H]-1,3-diethyl-8-phenylxanthine label the A1 subtype of adenosine receptor in brain membranes. The affinities of methylxanthines in competing for A1 adenosine receptors parallel their potencies as locomotor stimulants. The adenosine agonist N6-(phenylisopropyl) adenosine is a potent locomotor depressant. Both diazepam and N6-(L-phenylisopropyl)adenosine cause locomotor stimulation in a narrow range of subdepressant doses. Combined stimulant doses of the two agents depress motor activity, as do larger doses of either one, given separately. Evidence supporting a…

Adenosine A2A signals and dystonia.

Dystonia is a movement disorder characterized by sustained or intermittent involuntary muscle contractions, which is also seen in an advanced stage of Parkinson's disease (PD) as camptocormia, torticollis, and Pisa syndrome. Istradefylline, an adenosine A2A receptor antagonist, can be used for the treatment of PD to reduce 'off'-time period, and several clinical studies demonstrated the improvement of camptocormia, which have many similar features to dopa-responsive/non-responsive dystonia. Many animal models of dystonia showed that adenosine A2A receptor colocalized with dopamine D2 positive …
