Cu-Mediated Amination of (Hetero)Aryl C-H bonds with NH Azaheterocycles.

Direct synthesis of N-(hetero)arylated heteroarenes has been realized through Cu-mediated C-N coupling of NH azaheterocycles with aryl C-H bonds under aerobic conditions. This method features a broad scope of both heterocyclic arenes (pyridine, quinoline, pyrazole, imidazole, furan, thiophene, benzofuran, and indole) and NH azaheterocycles (imidazole, pyrazole, indole, azindole, purine, indazole, benzimidazole, pyridone, carbazole), providing a versatile method for the synthesis of pharmaceutically important N-(hetero)arylated heteroarenes. The versatility of this reaction was further demonstr

Theoretical studies of iron(III)-catalyzed intramolecular C-H amination of azides.

Density functional theory calculations have been carried out to study the reaction mechanism of the [Fe(III)(F20TPP)Cl] catalyzed C-H amination reaction. The calculations show that the classical three-step mechanism for other metals (Ru, Rh, Ir and Zn), including N2 liberation, C-N bond formation and 1,2-hydrogen shift, does not fit the iron(III)-catalyzed system. After N2 liberation, the favorable reaction pathway for the iron(iii)-catalyzed system is a 1,2-hydrogen shift preceding C-N bond formation, i.e., a H-abstraction/radical rebound mechanism.

Decarboxylative carbonyl alkylative amination: A general and practical strategy for aliphatic amine synthesis.

Aliphatic amines are ubiquitous core structural motifs in bioactive molecules including natural products, pharmaceuticals, and agrochemicals, driving ongoing demand for versatile, green, and practical synthetic strategies to construct complex aliphatic amines from inexpensive, readily available, structurally diverse feedstocks. Here, we report an iron-catalyzed decarboxylative carbonyl alkylative amination of aldehydes, amines, and carboxylic acids in HFIP under mild light irradiation at room temperature, with no additional additives and only CO2 and H2O as environmentally benign by-products. …

Catalytic Enantioselective Nucleophilic Amination of α-Halo Carbonyl Compounds with Free Amines.

Catalytic enantioselective substitution of the readily available racemic α-halo carbonyl compounds by nitrogen nucleophiles represents one of the most convenient and direct approaches to access enantioenriched α-amino carbonyl compounds. Distinct from the two available strategies involving radicals and enolate ions, herein we have developed a new protocol featuring an electronically opposite way to weaken/cleave the carbon-halogen bond. A suitable chiral anion-based catalyst enables effective asymmetric control over the key positively charged intermediates. This protocol not only allows free a

Regioselective copper-catalyzed amination of bromobenzoic acids using aliphatic and aromatic amines.

A chemo- and regioselective copper-catalyzed cross-coupling procedure for amination of 2-bromobenzoic acids is described. The method eliminates the need for acid protection and produces N-aryl and N-alkyl anthranilic acid derivatives in up to 99% yield. N-(1-Pyrene)anthranilic acid has been employed in metal ion-selective fluorosensing. Titration experiments showed that this pyrene-derived amino acid forms an equimolar complex with Hg(II) in water resulting in selective fluorescence quenching even in the presence of other metal ions such as Zn(II) and Cd(II).

Optical resolution of H/D isotopic chirality via asymmetric C-H amination.

Isotopic chirality arising from hydrogen/deuterium substitution has long resisted optical resolution because isotopic substitution introduces only minimal stereochemical differences, rendering such molecules structurally pseudo-achiral. Recognizing that the small yet distinct difference in C-H/C-D bond dissociation energies (BDEs) creates an energetic asymmetry, we envisioned that an enantioselective C-H functionalization exploiting this disparity could achieve the kinetic resolution of isotopically chiral molecules. Here, we report a catalytic method for the optical resolution of benzylic and

C-H Functionalization and Allylic Amination for Post-Polymerization Modification of Polynorbornenes.

Post-polymerization modification (PPM) via direct C-H functionalization is a powerful synthetic strategy to convert polymer feed-stocks into value-added products. We found that a metal-free, Se-catalyzed allylic C-H amination provided an efficient method for PPM of polynorbornenes (PNBs) produced via ring-opening metathesis polymerization. Inherent to the mechanism of the allylic amination, PPM on PNBs preserved the alkene functional groups along the polymer backbone, while also avoiding transposition of the double bonds. Amination using a series of aryl sulfonamides led to good control over t…

Photocatalysed direct amination of benzene and ammonia over Ti-V-MCM-41.

Aniline is one of the important organic chemical raw materials and is widely used in the chemical production industry, including dyes, pharmaceuticals, pesticides, explosives, spices, etc. At present, the yield and selectivity of aniline synthesis by direct amination are relatively low, and how to improve the catalytic efficiency has become an urgent problem to be solved. A series of Ti-V-MCM-41 catalysts with different silicon-titanium ratios was prepared by the hydrothermal synthesis method. According to the analysis of XRD, FT-IR, UV-vis DRS, BET, and SEM characterization of Ti-V-MCM-41, it…

Copper-Catalyzed Amination of C(sp3)-H bonds: From Anilides to Indolines.

Copper-catalyzed oxidative intramolecular cyclization of o-alkylated anilines via cleavage of C(sp3)-H and N-H bonds for the production of indolines is described. This approach provides a straightforward strategy for the synthesis of nitrogen-containing heterocyclic compounds through the functionalization of unactivated C(sp3)-H bonds with high site selectivity. The present catalytic system shows high preference for functionalization of unactivated C(sp3)-H bonds over C(sp2)-H bonds, leading to C-N bond formation.

Mild and efficient copper-catalyzed amination of aryl bromides with primary alkylamines.

[reaction: see text] An efficient copper-catalyzed amination of aryl bromides with primary alkylamines was developed that uses commercially available diethylsalicylamide as the ligand. This amination reaction can be performed at 90 degrees C in good yield. A variety of functional groups are compatible with these reaction conditions. Preliminary results show that this reaction can be carried out under solvent-free conditions with comparable yields.

Palladium-Catalyzed Amination of Protected Phenylalanine Iodides Using (Hetero)aryl Amines as Nucleophiles.

We report a robust palladium-catalyzed C-N cross-coupling method optimized for selective (hetero)arylation of derivatives containing p-iodophenylalanine (p-I-Phe). The utility of this approach is demonstrated by its use in late-stage modification of clinically relevant peptides in solution-phase. The method preserves the stereochemical integrity of the peptide backbone and exhibits compatibility with side-chain protecting groups. This provides a streamlined synthetic gateway to p-amino-substituted phenylalanines and peptides, expanding the chemical space accessible for peptide-based drug desig

Direct C1-H Amination of Free NH Carbazoles Enabled by the Catellani Reaction.

Carbazoles are valuable building blocks for emerging pharmaceuticals and bioactive compounds. However, methods to access C1-H-aminated carbazoles remain underdeveloped. Current approaches often require multistep syntheses or the use of toxic metals. Herein, we report the first Pd/NBE-catalyzed C1-H amination of NH-free carbazoles. The method introduces a broad range of cyclic amines onto carbazole scaffold and tolerates several functional groups, including acyl, pivaloyl, and halogen substituents. This approach expands the scope of the Catellani reactions to the direct amination of unprotected…

Double-Volcano Catalytic Response in Furfural Reductive Amination Over Inverse Ni─Al Catalysts.

Reductive amination of aldehydes and ketones to primary amines is an important route for biomass valorization, yet achieving efficient synergy between substrate and hydrogen activation in cascade reactions remains challenging. Herein, we construct a compositionally tunable Ni─Al catalyst system whose architecture evolves from conventional Ni/Al2O3 to inverse Al2O3/Ni. This structural evolution gives rise to an unusual double-volcano catalytic behavior in furfural reductive amination. Two activity maxima are identified over 30Ni/Al2O3 and inverse Al2O3/90Ni, respectively, with the optimal Al2O3…
