Escherichia coli uropathogenic-specific protein, Usp, is a bacteriocin-like genotoxin.

BACKGROUND: Bacterial genotoxins provoke DNA damage and carcinogenesis. The Escherichia coli uropathogenic-specific protein gene, usp, and its linked genes, imu1-3, are associated with strains from pyelonephritis, prostatitis, and bacteremia of urinary tract origin. While the Usp C-terminal domain exhibits similarity with DNase-like colicins and pyocins, its role and mechanisms of action, as well as those of the 3 associated proteins, is unknown. METHODS: We isolated Usp and Imu1-3 and examined their activity on plasmid DNA, human umbilical vein endothelial cells, and human embryonic kidney c

Production of a bacteriophage, a phage tail-like bacteriocin and an antibiotic by Bacillus azotofixans.

Twenty-two nitrogen-fixing Bacillus azotofixans strains were shown to produce an inhibition zone against themselves in plate assays. The B. azotofixans type strain P3L-5, chosen for further studies, produced inhibition zones against various Bacillus strains and other bacterial genera. This antibacterial substance was also produced in liquid medium and its production was enhanced in semisolid medium (0.4% agar) after 3 to 5 days of incubation. The substance was suggested to be an antibiotic and its preliminary characterization showed resistance to heat (100 degrees C, 15 minutes), to trypsin, p

Studies of the relationship between the production of bacteriocines by group A streptococci and acute glomerulonephritis.

The previously reported relationship between nephritogenicity and bacteriocine production among Group A streptococci has been examined. Using techniques comparable to the earlier study, the present study did not reveal any such relationship. Among 73 strains tested, 53% produced bacteriocines; no significant difference was noted between bacteriocines from strains isolated from cases of nephritis and those from a group of strains recovered from patients with other conditions. In addition, no correlation between type and bacteriocine production was observed.

Serotype-independent inhibition of S. pneumoniae by SMiTE, a commensal-derived bacteriocin.

Streptococcus pneumoniae remains a leading cause of disease despite widespread vaccination, highlighting the need for serotype-independent strategies. We recently identified commensal streptococci that produce bacteriocins with anti-pneumococcal activity. Here, we evaluate these bacteriocins as candidates for pneumococcal control. Using cell-free protein synthesis, we screened 58 bacteriocins, the majority of which absent in available pneumococcal genomes, and identified SMiTE as the most potent. Purified SMiTE disrupted pneumococcal membrane integrity, as shown by confocal, transmission, and …

Bacteriocin production facilitates nosocomial emergence of vancomycin-resistant Enterococcus faecium.

Gastrointestinal colonization by the nosocomial pathogen vancomycin-resistant Enterococcus faecium (VREfm) can lead to bloodstream infections with high mortality rates. Shifts in VREfm lineages found within healthcare settings occur, but reasons underlying these changes are not understood. Here we sequenced 710 VREfm clinical isolates collected between 2017 and 2022 from a large tertiary care centre. Genomic analyses revealed a polyclonal VREfm population, although 46% of isolates formed genetically related clusters, suggesting a high transmission rate. Comparing these data to a global collect

Interactions between Lipopolysaccharide and Peptide Bacteriocin BacSp222 Influence Their Biological Activities.

This study describes the interactions between two different pro-inflammatory factors produced by bacteria, lipopolysaccharide (LPS) from Gram-negative bacteria and the peptide BacSp222 produced by a Gram-positive zoonotic strain, Staphylococcus pseudintermedius 222. We demonstrate that the mentioned molecules interact, forming a complex, and this phenomenon selectively reduces their biological activities in vitro and in vivo. Specifically, the levels of tumor necrosis factor (TNF) and nitric oxide (NO) produced by monocyte-macrophage cells were lower in samples treated with both LPS and BacSp2

Lactic Acid Bacteria-derived Bacteriocins: A Promising Antimicrobial Strategy against Multidrug-resistant for Neonatal Sepsis Pathogens.

Neonatal sepsis continues to pose rising critical challenges to global health, particularly in low and middle-income countries, where it acc … t is growing to develop the microbial-derived peptides as novel antimicrobial agents, particularly "bacteriocins." In comparison to traditional antibiotics, many bacteriocins exhibit narrow-spectrum action, enabling them to inhibit specific pathogens without disrupting the host microbiota. Recent studies have highlighted the remarkable potential of lactic acid …

Response of sensitive and resistant Listeria monocytogenes strains against bacteriocins produced by different Enterococcus spp. strains.

Listeria monocytogenes is a relevant foodborne pathogen causing invasive listeriosis in humans, a disease with high mortality rates. Its ubiquity and growth characteristics enable this pathogen to survive harsh food processing environments. The addition of bacteriocins, antimicrobial peptides ribosomally synthesized by certain bacteria, appears as a natural alternative to control this pathogen in food. However, the emergence of L. monocytogenes strains resistant to the inhibitory action of bacteriocins has been detected. In order to analyse the development of this resistance, different propert…

Split Intein-Mediated Engineering of Bacteriocins Through Backbone Circularization, Modular Assembly of Chimeras, and Inducible Circularization.

The increasing prevalence of antimicrobial resistance has renewed interest in bacteriocins and other antimicrobial peptides as promising alternatives to conventional antibiotics. In this context, in vitro cell-free protein synthesis (IV-CFPS) provides a rapid and versatile platform for the production and functional evaluation of bacteriocins, enabling antimicrobial screening without the limitations associated with cellular expression, low production yields, or peptide toxicity. In previous studies, we demonstrated the utility of IV-CFPS for the production and characterization of a broad range …

Demystifying Bacteriocins of Human Microbiota by Genome Guided Prospects: An Impetus to Rekindle the Antimicrobial Research.

The human microbiome is a reservoir of potential bacteriocins that can counteract multidrug resistant bacterial pathogens. Unlike antibiotics, bacteriocins selectively inhibit a spectrum of competent bacteria and are said to safeguard gut commensals, reducing the chance of dysbiosis. Bacteriocinogenic probiotics or bacteriocins of human origin will be more pertinent in human physiological conditions for therapeutic applications to act against invading pathogens. Recent advancement in the omics approach enables the mining of diverse and novel bacteriocins by identifying biosynthetic gene cluste…

Increasing prevalence of bacteriocin carriage in a 6-year hospital cohort of E. faecium.

Vancomycin-resistant enterococci (VRE) are important pathogens in hospitalized patients; however, the factors involved in VRE colonization of hospitalized patients are not well characterized. Bacteriocins provide a competitive advantage to enterococci in experimental models of colonization, but little is known about bacteriocin content in samples derived from humans and even less is known about their dynamics in the clinical setting. To identify bacteriocins which may be relevant in the transmission of VRE, we present a systematic analysis of bacteriocin content in the genomes of 2,248 patient…
