The CYP152-family P450 enzyme CypC of Bacillus subtilis converts non-natural substrates in plasma-driven biocatalysis.

Plasma-driven biocatalysis utilizes in situ H2O2 production by atmospheric pressure plasmas to drive H2O2-dependent enzymatic reactions. Having previously established plasma-driven biocatalysis using recombinant unspecific peroxygenase from Agrocybe aegerita (rAaeUPO) to produce (R)-1-phenylethanol from ethylbenzene, we here employed CypC from Bacillus subtilis 168 (synonyms: YbdT, P450BSβ), an integral enzyme of surfactin and fengycin biosynthesis. CypC naturally hydroxylates medium and long-chain carboxylic acids. With short-chain carboxylic acids as decoy molecules, it also converts non-nat

Supramolecular assembly strategy of modified starch chains for achieving recyclable emulsion biocatalysis within a narrow pH range.

Stimuli-responsive Pickering emulsions are promising in biocatalysis for their ease of product separation and emulsifier recovery. However, pH responsiveness, though simple and cost-effective, faces challenges in precise control and narrow transition ranges, limiting its use in enzymatic catalysis. Herein we introduced amorphous octenyl succinic anhydride-modified debranched starch chains (Am-OSA-St) to control emulsion properties within a pH range suitable for enzymatic catalysis. By adjusting the OSA group density and molecular weight, Am-OSA-St allowed emulsions to transition reversibly bet

A Combination of Biocatalysis and Fenton-Like Reaction Induced OH• Burst for Cascade Amplification of Cancer Chemodynamic Therapy.

Chemodynamic therapy (CDT) is a novel antitumor strategy that employs Fenton or Fenton-like reactions to generate highly toxic hydroxyl radical (OH•) from hydrogen peroxide (H2O2) for inducing tumor cell death. However, the antitumor efficacy of the CDT strategy is harshly limited by the redox homeostasis of tumor cells; especially the OH • is easily scavenged by glutathione (GSH) and the intracellular H2O2 level is insufficient in the tumor cells. Herein, we propose the Mn2+-menadione (also known as vitamin K3, MK3) cascade biocatalysis strategy to disrupt the redox homeostasis of tumor cells

Recent advances in AI-driven deep eutectic solvent design for biocatalysis: Co-design of extremozyme and reaction medium.

Deep eutectic solvents (DES) have emerged as sustainable, highly tunable alternatives to conventional organic media for non-aqueous biocatalysis. DES dynamically interact with enzymes to shift reaction equilibria, enhance thermostability and tailor stereoselectivity. However, the vast combinatorial space and complex physicochemical properties of DES make traditional trial-and-error solvent engineering highly inefficient. This review systematically examines recent advancements in DES-driven biocatalysis and highlights the role of artificial intelligence (AI) for rational DES design. Specificall

A multi-dimensional engineering strategy integrating computational design, self-cyclization and whole cell biocatalysis for efficient epilactose production.

Epilactose is a promising functional disaccharide, but its biomanufacturing is limited by insufficient enzyme activity, poor thermostability, and costly catalyst preparation. We first used the REME platform to computationally evaluate candidate enzymes. Among them, cellobiose 2-epimerase from Caldicellulosiruptor saccharolyticus (CsCE) showed the highest epilactose synthesis activity. We therefore developed an integrated strategy combining computational design, SpyTag/SpyCatcher-mediated cyclization, and ethanol-permeabilized whole-cell catalysis. By combining enzyme ligand binding energy anal

Do We Still Need Chemistry for Water-Soluble Prodrugs? What Biocatalysis Already Does, and What It Does Not.

Poor aqueous solubility limits roughly 40% of marketed oral drugs and up to ~90% of development candidates. One established remedy is the prodrug strategy: the covalent attachment of a bioreversible promoiety that regenerates the parent drug in vivo. The same conjugation can move a molecule in either direction along the hydrophilic-lipophilic axis: lipophilization raises membrane and oil solubility for permeation-limited or topical uses, whereas hydrophilization raises aqueous solubility. This review addresses hydrophilization and asks whether enzymatic (biocatalytic) synthesis can replace con

Novel organic solvent-responsive expression vectors for biocatalysis: application for development of an organic solvent-tolerant biodesulfurizing strain.

Biodesulfurization is an attractive alternative to hydrodesulfurization for lowering the sulfur content of petroleum products. However, the fuel oils are toxic to microorganisms, which have seriously hindered the application of biodesulfurization. Here, a solvent-tolerant desulfurizing bacterium, Pseudomonas putida DS23, was developed using one of the organic solvent-responsive expression vectors newly constructed for biocatalysis, in which gene expression could be regulated in an organic solvent-dependent fashion. The biodesulfurizing activity of P. putida DS23 could be induced by all the org

A Food Protein-Based CO2/N2-Responsive Magnetic Platform for Accelerated Pickering Interfacial Biocatalysis in Flavor Ester Synthesis.

The enzymatic synthesis of flavor esters is often limited by poor interfacial mass transfer and water-induced equilibrium constraints in conventional systems. Herein, we report a food-grade magnetic Pickering interfacial biocatalysis (PIB) platform based on sodium caseinate (NaCas)-Fe2O3 nanoparticles for accelerated flavor ester synthesis. The nanoparticles, synthesized via a one-pot coprecipitation method, exhibit excellent colloidal stability, CO2/N2-responsive behavior, and magnetic responsiveness. Under an external magnetic field, magnetic actuation induces localized interfacial perturbat

Gelatin-Stabilized Water-in-Water Emulsions for Biocatalysis: Interfacial Enzyme Confinement, In Situ Product Extraction, and Cryogenic Recyclability.

Water-in-water (W/W) emulsions offer high biocompatibility for enzymatic processes but are difficult to stabilize with conventional oil-water surfactants due to their ultralow interfacial tension. Here, we design a gelatin-stabilized PEG/dextran (PEG/Dex) emulsion platform enabling integrated "reaction-separation-recovery" biocatalysis. By tuning the pH and polymer concentration, programmable microarchitectures such as core-shell, Janus, or triple-phase droplets were achieved. In cascade oxidation of guaiacol, the compartmentalized system reached an 86% yield of tetraguaiacol─50% higher than i

Usnea Acid-Incorporated Ca2+ /Mn2+ Ions Reservoirs for Elevated Ion-Interference Therapy through Synergetic Biocatalysis and Osmolarity Imbalance.

Ion-interference therapy (IIT) utilizes ions to disturb intracellular biological processes and has been received increasing attention in tumor treatments recently. However, the low therapeutic efficiency still hinders its further biological applications. Herein, via a simple and one-pot gas diffusion process, polyethylene glycol (PEG)-modified Mn2+ ions and usnic acid (UA)-incorporated CaCO3 nanomaterials (PEG CaMnUA) as Ca2+ /Mn2+ ions reservoirs are prepared for magnetic resonance imaging (MRI)-guided UA-elevated IIT. Biocatalysis-mediated MOF-to-prussian blue transformation enabling sensitive detection of NSCLC-associated miRNAs with dual-readout signals.

Sensitive and accurate miRNAs assay is critical for early diagnosis of non-small-cell lung carcinomas (NSCLC). Herein, we demonstrate a photothermal and electrochemical dual-readout assay method for miRNA detection based on a novel biocatalysis-mediated MOF-to-prussian blue (PB) transformation (BMMPT) strategy and the catalytic hairpin assembly (CHA) amplification strategy. It is found that the Fe2+-based MOF (MOF-Fe2+) can act as the Fe2+ source to react with K3[Fe(CN)6], leading to the in-situ formation of prussian blue (PB) on MOF-Fe2+. Due the inherent near-infrared (NIR) photothermal conv

Multi-lab, multi-enzyme study demonstrates the versatility of bacterial microcompartment shells as a modular platform for confined biocatalysis.

Bacterial microcompartments (BMCs) are proteinaceous organelles that spatially organize metabolic reactions in bacteria and represent an attractive scaffold for pathway engineering. Here, we present a proof-of-concept in vitro study demonstrating a simple, scalable, and modular BMC shell-based platform for enzyme encapsulation using the SpyCatcher-SpyTag (SC-ST) covalent conjugation system. To evaluate the generality of this approach, 16 dehydrogenases were selected, of which 13 were successfully expressed and purified as SC-tagged enzymes in E. coli by five research groups working in parallel

Genomic prospecting and biochemical characterization of a novel thermostable 3-quinuclidinone reductase from hot spring metagenomes for efficient biocatalysis.

This study presents the discovery and characterization of a novel thermophilic 3-quinuclidinone reductase (ScQR) identified through metagenomic mining of hot spring environments. ScQR, a member of the short-chain dehydrogenase/reductase (SDR) superfamily, was heterologously expressed in Escherichia coli, and its catalytic properties were systematically characterized. The enzyme demonstrates exceptional thermal stability, retaining 86% of its activity after 48 hours at 70°C. Furthermore, K+ and Mg²+ ions significantly enhanced ScQR's activity at specific concentrations. Structural analysis reve

Substrate Channeling on Bienzyme Assembly Significantly Enhances Cascade Biocatalysis of Ene Reductase and Aldehyde Dehydrogenase at High Substrate Concentrations.

Multienzyme cascades enable efficient synthesis of value-added chemicals by coupling sequential biotransformations. In the ene reductase (OYE)-aldehyde dehydrogenase (ALDH) cascade for chiral α-substituted carboxylic acids, accumulation of the saturated aldehyde intermediate inhibits OYE and thereby limits cascade performance, particularly at high substrate concentrations. Here, complementary RIDD/RIAD (DD/AD) peptides were used to organize OYE and ALDH into an activity-matched 2:1 assembly. Linker optimization identified the DD-(EAAAK)4-OYE fusion (DD-EK4OYE), which retained 86.4% of free OYE

Coupling bacterial nitrogen fixation to yeast whole-cell biocatalysis enables amine production using air as the sole nitrogen source.

BACKGROUND: Alkylamine production depends on ammonium produced via the energy-intensive, fossil-based Haber-Bosch process. Diazotrophic bacteria could offer a biological alternative by supplying nitrogen directly from air to heterotrophic whole-cell biocatalysts for alkylamine production. However, their integration with yeast production systems remains largely unexplored. RESULTS: Here, we evaluated the use of an ammonium-excreting strain of Azotobacter vinelandii (AZBB664) to supply nitrogen to an engineered Saccharomyces cerevisiae biocatalyst for vanillylamine production using air as the s

Multidimensional signal amplification architectures in electrochemical immunosensing integrate porous nanomaterials, biocatalysis, and nucleic acid circuits to achieve attomolar detection.

Electrochemical immunosensors have significantly advanced point-of-care diagnostics and environmental monitoring, owing to their high specificity, portability, and compatibility with miniaturized systems. Nevertheless, their detection sensitivity remains limited by the ultralow concentrations of target analytes (such as disease biomarkers, pathogens, or environmental contaminants), creating an urgent need for innovative signal amplification strategies to meet practical and regulatory demands. This review presents a systematic overview of emerging signal amplification strategies, placing a dedi

Hydroxynitrile lyases from cyanogenic millipedes: molecular cloning, heterologous expression, and whole-cell biocatalysis for the production of (R)-mandelonitrile.

Hydroxynitrile lyases (HNLs), which are key enzymes in cyanogenesis, catalyze the cleavage of cyanohydrins into carbonyl compounds and hydrogen cyanide. Since HNLs also catalyze the reverse reaction, they are used industrially for the asymmetric synthesis of cyanohydrins, which are valuable building blocks of pharmaceuticals and fine chemicals. HNLs have been isolated from cyanogenic plants and bacteria. Recently, an HNL from the cyanogenic millipede Chamberlinius hualienensis was shown to have the highest specific activity for (R)-mandelonitrile synthesis, along with high stability and enanti

Electrochemically driven biocatalysis of the oxygenase domain of neuronal nitric oxide synthase in indium tin oxide nanoparticles/polyvinyl alcohol nanocomposite.

Nitric oxide synthase (NOS) plays a critical role in a number of key physiological and pathological processes. Investigation of electron-transfer reactions in NOS would contribute to a better understanding of the nitric oxide (NO) synthesis mechanism. Herein, we describe an electrochemically driven catalytic strategy, using a nanocomposite that consisted of the oxygenase domain of neuronal NOS (D290nNOSoxy), indium tin oxide (ITO) nanoparticles and polyvinyl alcohol (PVA). Fast direct electron transfer between electrodes and D290nNOSoxy was observed with the heterogeneous electron transfer rat
