Nature as blueprint: Global phenotype engineering of CHO production cells based on a multi-omics comparison with plasma cells.

Especially for the production of artificial, difficult to express molecules a further development of the CHO production cell line is required to keep pace with the continuously increasing demands. However, the identification of novel targets for cell line engineering to improve CHO cells is a time and cost intensive process. Since plasma cells are evolutionary optimized for a high antibody expression in mammals, we performed a comprehensive multi-omics comparison between CHO and plasma cells to exploit optimized cellular production traits. Comparing the transcriptome, proteome, miRNome, surfac…

TPST Gene Knock-Out Eliminates Tyrosine Sulfation on a Recombinant Antibody Produced in CHO Cells.

Tyrosine sulfation is a post-translational modification that has been reported to occur infrequently on recombinant monoclonal antibodies (mAbs). We recently demonstrated that tyrosine sulfation occurred on a bispecific antibody (bsAb) produced in Chinese hamster ovary (CHO) cells, using a multi-enzymatic approach in combination with intact mass and peptide-based mass spectrometry analysis supplemented with the use of synthetic peptides. Tyrosine sulfation needs to be controlled during the manufacturing process due to potential undesired effects, such as impact on potency and immunogenicity. H

MiRNA Chaining for Efficient Stable Overexpression to Improve Protein Quantity and Quality in CHO Cells.

MicroRNAs (miRNAs), small noncoding RNAs with a length of about 22 nucleotides, harbor the potential to be powerful tools for the genetic engineering of production cell lines like Chinese hamster ovary (CHO) cells. Their ability to regulate multiple targets at once and their potential to fine-tune effect strengths contrast with classical engineering approaches. However, most studies of miRNAs rely on transiently flooding the cells with miRNA mimics. Since this approach is not suitable for long-term cultivation in a bioprocess, stable overexpression of miRNAs becomes more and more important for

MAT2A Knockdown Enhances Recombinant Protein Expression in Transgenic CHO Cells Through Regulation of Cell Cycle.

Chinese hamster ovary (CHO) cells represent the most widely utilized host system for industrial production of high-quality recombinant prote … itumor activity. Notably, p21 induction has been shown to enhance recombinant protein expression in CHO cells by triggering cell cycle arrest. In this study, we identified MAT2A as a potential regulatory target, showing significant differential expression in transfected CHO cells with elevated versus diminished recombinant protein production. To investigate …

Evaluation of the toxic potency of selected cadmium compounds on A549 and CHO-9 cells.

Cytotoxicity of cadmium sulphide, oxide and chloride was tested using A549 and CHO-9 cells. Metabolic activity of cells (MTT test) and cell membrane permeability (NRU test) were used as cytotoxicity endpoints. The results revealed unexpectedly low toxicity of cadmium sulphide as compared to chloride and oxide. This preliminary report does not provide any explanation for this effect, but the result may nevertheless be interesting for future studies of toxicity mechanisms of cadmium compounds. First cadmium compounds caused damage or change in the permeability of cell membranes, then inhibition

Epigenetic regulation of histone methyltransferase SUV39H1 on the expression of recombinant protein in CHO cells.

Histone methylation-mediated epigenetic modification significantly influences gene transcription and expression regulation. This study examined the effects of histone 3 lysine 9 trimethylation (H3K9me3) methyltransferase SUV39H1 and its specific inhibitor chaetocin on recombinant protein expression in Chinese hamster ovary (CHO) cells. Results indicated that stable SUV39H1-knockdown CHO cells exhibited reduced H3K9me3 levels while showing increased expression of recombinant adalimumab (rADM) and human serum albumin (rHSA) by approximately 45% and 136%, respectively. Furthermore, treatment with…

Leveraging ANXA1 to enhance recombinant protein yields in CHO cells: A UPR-Mediated bioprocessing approach.

Chinese hamster ovary (CHO) cells undergo endoplasmic reticulum stress (ERS) during intensive recombinant protein production, triggering the unfolded protein response (UPR) to balance cell survival and protein output. Nevertheless, key regulatory components of this process remain incompletely characterized. In this study, we demonstrate that Annexin A1 (ANXA1) functions as a UPR suppressor in CHO cells. Employing the PiggyBac transposon system, we generated a stable ANXA1-knockdown cell line exhibiting a 4.5-fold increase in recombinant antibody expression and a 4.2-fold increase in specific p…

P-glycoprotein modulates the fluidity gradient of the plasma membrane of multidrug resistant CHO cells.

Cryo-electron microscopy has yielded high-resolution structural data of the multidrug efflux transporter P-glycoprotein (ABCB1), but its direct and indirect interactions within the native membrane environment have remained largely unexplored. Here, we compared the fluidity gradients of plasma membranes of the drug-sensitive CHO cell line AuxB1 and its P-glycoprotein overexpressing derivative B30 by fluorescence anisotropy of embedded n-(9-anthroyloxy) fatty acid probes (n = 2, 7, 9, 12, 16) in the temperature range of 10-50 °C. The shape of the temperature profiles of probe mobility was compar

Elevated endoplasmic reticulum pH is associated with high growth and bisAb aggregation in CHO cells.

Chinese hamster ovary (CHO) bioprocesses, the dominant platform for therapeutic protein production, are increasingly used to produce complex multispecific proteins. Product quantity and quality are affected by intracellular conditions, but these are challenging to measure and often overlooked during process optimization studies. pH is known to impact quality attributes like protein aggregation across upstream and downstream processes, yet the effects of intracellular pH on cell culture performance are largely unknown. Recently, advances in protein biosensors have enabled investigations of intr

Effect of different oxidation states of chromium in causing chromosome alterations in cultured CHO cells.

Four chromium salts with different oxidation states were tested for their influence in causing chromosome aberrations and sister-chromatid exchange in Chinese hamster ovary cells in vitro. Cell cultures were treated with CrO3, K2Cr2O7, CrCl2 and Cr(NO3)3.9H2O at concentrations of 10(-7), 10(-6), 10(-5) and 10(-4) M for the aberration assay, and 10(-8), 10(-7), 10(-6) and 10(-5) M for the sister-chromatid exchange assay. It was noticed that Cr (VI) compounds--CrO3 and K2Cr2O7--considerably enhanced the frequencies of aberrations and sister-chromatid exchanges compared to the control cultures. C

Anthrax protective antigen interacts with a specific receptor on the surface of CHO-K1 cells.

The interaction of protective antigen (PA), a component of the anthrax toxin, with receptors on the Chinese hamster ovary cell line CHO-K1 was characterized. Protective antigen binding at 4 degrees C is highly specific, concentration dependent, saturable (Kd = 0.9 nM), and reversible. Scatchard analysis indicates the presence of a single class of PA binding sites at a concentration of 10,000 +/- 2,000 per cell. Pretreatment of cells with a number of different proteases strongly inhibits PA binding, suggesting that the receptor may be at least partially proteinaceous. Direct chemical cross-link

A cell line development vector strategy for improved expression of a trispecific T-cell engager in CHO.

Recent advances in trispecific antibody (trisAb) engineering offer great therapeutic potential, but achieving high product yield and quality in cell line development remains a challenge due to complex chain pairing requirements in production cell lines. In this study, three distinct expression vector configurations were evaluated for their ability to support robust, high-level expression of a structurally complex, synapse-gated trisAb T-cell engager (TriMab) in stable Chinese hamster ovary cells. Initial configurations using conventional dual heavy chain (HC) and triple light chain (LC) vector

The role of sodium pyruvate in mitigating the cytotoxic effects of vanadium on CHO-K1 cells.

Vanadium is a hazardous, pro-oxidant element that contributes to environmental pollution and has been reported as a risk factor for human health through occupational or environmental exposure. Pyruvate, on the other hand, is a natural alpha-keto acid with exceptional antioxidant and cytoprotective properties. Therefore, the aim of this study was to evaluate the mitigating effect of exogenous pyruvate against vanadium-induced toxicity in cultured Chinese hamster ovary (CHO)-K1 cells. To this end, CHO-K1 cells were exposed to 100 μM vanadyl sulfate (VOSO4) for 24 h in the presence of 4.5 and 8 m
