A National Catalog of Mapped Short-Form Six-Dimension Utility Scores for Chronic Conditions in the United States From 2010 to 2015.

OBJECTIVES: This study examined health preference utility weights and utility decrements associated with different types of chronic conditions in the United States. METHODS: We used the 2010-2015 Medical Expenditure Panel Survey data for persons aged ≥ 18 years with 12-Item Short-Form Survey Physical and Mental Component Summary scores. 12-Item Short-Form Survey scores were converted to Short-Form Six-Dimension (SF-6D) preference scores to measure utilities of different chronic diseases. We used the Clinical Classification Code to identify 30 chronic diseases from 12 categories, such as cardi

Common genetic substrates of alcohol and substance use disorder severity revealed by pleiotropy detection against GWAS catalog in two populations.

Alcohol and other substance use disorders (AUD and SUD) are complex diseases that are postulated to have a polygenic inheritance and are often comorbid with other disorders. The comorbidities may arise partially through genetic pleiotropy. Identification of specific gene variants accounting for large parts of the variance in these disorders has yet to be accomplished. We describe a flexible strategy that takes a variant-trait association database and determines if a subset of disease/straits are potentially pleiotropic with the disorder under study. We demonstrate its usage in a study of use d

Exploring various polygenic risk scores for skin cancer in the phenomes of the Michigan genomics initiative and the UK Biobank with a visual catalog: PRSWeb.

Polygenic risk scores (PRS) are designed to serve as single summary measures that are easy to construct, condensing information from a large number of genetic variants associated with a disease. They have been used for stratification and prediction of disease risk. The primary focus of this paper is to demonstrate how we can combine PRS and electronic health records data to better understand the shared and unique genetic architecture and etiology of disease subtypes that may be both related and heterogeneous. PRS construction strategies often depend on the purpose of the study, the available d

A catalog for the transcripts from the venomous structures of the caterpillar Lonomia obliqua: identification of the proteins potentially involved in the coagulation disorder and hemorrhagic syndrome.

Accidents with the caterpillar Lonomia obliqua are often associated with a coagulation disorder and hemorrhagic syndrome in humans. In the present study, we have constructed cDNA libraries from two venomous structures of the caterpillar, namely the tegument and the bristle. High-throughput sequencing and bioinformatics analyses were performed in parallel. Over one thousand cDNAs were obtained and clustered to produce a database of 538 contigs and singletons (clusters) for the tegument library and 368 for the bristle library. We have thus identified dozens of full-length cDNAs coding for protei

Limit-Cycle Proliferation Under Parametric Delayed Feedback in a Conductance-Based Neuron: Bifurcation Landscape, Orbit Catalog, and Capacity Analysis.

We show that a single Hodgkin-Huxley (HH) neuron with Pyragas-type delayed feedback control (DFC) can store multiple symbols as stable periodic orbits, where the specific orbit is selected by tuning the DFC gain K and time delay τ. Sweeping the (K,τ) parameter plane at fixed bias current Ibias = 10.0 μA/cm2 reveals 207 orbit types across 12 topological categories, with inter-spike interval (ISI) means from 5.9 to 56.9 ms. We establish: (i) a write protocol that reliably locks orbits with 13.9 ms median settling time; (ii) a novel Pattern-Oriented Limit-cycle Decoder (POLD) that reads orbits at

Evaluation of WHO catalog of mutations and five WGS analysis tools for drug resistance prediction of Mycobacterium tuberculosis isolates from China.

The continuous advancement of molecular diagnostic techniques, particularly whole-genome sequencing (WGS), has greatly facilitated the early diagnosis of drug-resistant tuberculosis patients. Nonetheless, the interpretation of results from various types of mutations in drug-resistant-associated genes has become the primary challenge in the field of molecular drug-resistance diagnostics. In this study, our primary objective is to evaluate the diagnosis accuracy of the World Health Organization (WHO) catalog of mutations and five WGS analysis tools (PhyResSE, Mykrobe, TB Profiler, Gen-TB, and SA

Comprehensive identification of pleiotropic loci for body fat distribution using the NHGRI-EBI Catalog of published genome-wide association studies.

We conducted a hypothesis-free cross-trait analysis for waist-to-hip ratio adjusted for body mass index (WHRadjBMI ) loci derived through genome-wide association studies (GWAS). Summary statistics from published GWAS were used to capture all WHRadjBMI single-nucleotide polymorphisms (SNPs), and their proxy SNPs were identified. These SNPs were used to extract cross-trait associations between WHRadjBMI SNPs and other traits through the NHGRI-EBI GWAS Catalog. Pathway analysis was conducted for pleiotropic WHRadjBMI SNPs. We found 160 WHRadjBMI SNPs and 3675 proxy SNPs. Cross-trait analysis iden

The Swiss Personalized Health Network Metadata Catalog: Platform for Health Data Discovery and Exploration Based on Findable, Accessible, Interoperable, and Reusable Principles.

BACKGROUND: The Swiss Personalized Health Network (SPHN) facilitates the interoperability and secure sharing of health-related data for research in Switzerland, in line with the findable, accessible, interoperable, and reusable (FAIR) principles. Since medical datasets can be highly sensitive, access is often governed by complex legal and regulatory requirements. Enabling researchers to discover, understand, and evaluate datasets through rich, well-structured metadata is therefore essential to support informed decisions about data suitability and reuse. OBJECTIVE: This study describes the des

An endothelial RNA splicing atlas catalogs effects of IL-1β and identifies an alternative PROCR isoform with genetic links to pleiotropic vascular disease.

Alternative splicing (AS) alters the sequence and dynamics of mRNA but has not been comprehensively annotated in human endothelial cells (ECs). EC dysfunction is a hallmark of complex inflammatory diseases, including cancer and atherosclerosis. Therefore, we modeled inflammation in vitro using 53 genetically distinct human aortic EC lines exposed to interleukin-1β (IL-1β) or control media. This identified 1,224 differentially spliced transcripts (DSTs). DSTs were enriched for alternative first (AF) exon usage, including isoforms of several disease-associated and metabolic genes. To confirm tha

Japanese Population Norms of EQ-5D-5L and Health Utilities Index Mark 3: Disutility Catalog by Disease and Symptom in Community Settings.

OBJECTIVES: This study aimed to establish the Japanese population norms of the EQ-5D-5L and Health Utilities Index Mark 3 (HUI3) and estimate the disutility associated with diseases and symptoms. METHODS: We performed a door-to-door survey of the general population by random sampling. The planned sample size was 10 000 residents (age ≥16 years) of 334 districts in Japan. In addition to the EQ-5D-5L and HUI3 questionnaires, questions regarding demographic factors and self-reported main diseases and symptoms were asked. The EQ-5D-5L and HUI3 responses were converted to index values on the basis

Integrated AHP-Delphi and Mini-HTA for selection of metabolic drugs in China's national medical insurance catalog: a framework towards quantitative dynamic adjustment.

BACKGROUND: China's high burden of metabolic disease requires more systematic approaches to selecting medicines for inclusion in the National Medical Insurance Catalog. Existing processes face difficulties in quantitatively balancing clinical value, target population needs, pharmacoeconomic evidence, and resource constraints, and may therefore rely heavily on subjective judgment. METHODS: We developed a decision framework integrating the analytic hierarchy process (AHP), the Delphi method, and mini-health technology assessment (mini-HTA) principles. Candidate criteria were identified from the

3-generation family histories of mental, neurologic, cardiometabolic, birth defect, asthma, allergy, and autoimmune conditions associated with autism: An open-source catalog of findings.

The relatively few conditions and family member types (e.g., sibling, parent) considered in investigations of family health history in autism spectrum disorder (ASD, or autism) limits understanding of the role of family history in autism etiology. For more comprehensive understanding and hypothesis-generation, we produced an open-source catalog of autism associations with family histories of mental, neurologic, cardiometabolic, birth defect, asthma, allergy, and autoimmune conditions. All live births in Denmark, 1980-2012, of Denmark-born parents (1,697,231 births), and their 3-generation fami

Diagnostic Performance of Large Language Models for Orthopedic-Related Rare Diseases and Their Impact on Physicians' Diagnostic Accuracy: 2-Stage Comparative Evaluation Study Based on the Chinese Rare Disease Catalog.

BACKGROUND: Orthopedic-related rare diseases are difficult to diagnose because of their low prevalence, heterogeneous phenotypes, and fragmented knowledge. Large language models (LLMs) can serve as dynamic knowledge-support tools, but their diagnostic performance and effect on physicians' decision-making remain unclear. OBJECTIVE: This study aims to compare the diagnostic performance of advanced LLMs for orthopedic-related rare diseases and to evaluate the effect of a 2-stage LLM-assisted diagnostic workflow on physicians' diagnostic accuracy and subjective acceptance. METHODS: We selected 4

The fleas (Siphonaptera) of South Carolina with an assessment of their vectorial importance.

We document 25 species of fleas from South Carolina including new state records for two species, the ctenophthalmids Epitedia cavernicola and Rhadinopsylla orama. Host and other collection data, by county, are provided, including many new records amassed from 1992-1998 and some older records gleaned from the Clemson University Arthropod Collection. One flea species, the rhopalopsyllid Polygenis gwyni, is especially common and widespread in South Carolina, particularly on the cotton rat (Sigmodon hispidus) in coastal plain habitats. The largest number of flea species (8) from a single host spec

Between Centralization and Fragmentation: The Past, Present, and Future of Phage Collections.

Despite over a century of collecting bacteriophages, there has been a persistent lack of interest in systematically cataloging resulting phage banks. The result was a situation in which the ongoing growth of phage infrastructures was paralleled by an increasing fragmentation of knowledge about collections' contents and existence. Over the last two decades, renewed interest in phage therapy and phage biology has further exacerbated confusion amid a rapid increase in the number of large and small phage collections and an ongoing dearth of coordination and standardized cataloging. Whatever the mo…
