Antibodies to centromere antigens measured by an automated enzyme immunoassay.

BACKGROUND: Anticentromere antibodies (ACA) are frequently observed in patients with Raynaud's phenomenon and in the CREST syndrome, a subclass of systemic sclerosis. Likewise, ACA are also found in other autoimmune and non-autoimmune diseases. The objective of the present study was to evaluate the clinical utility of the measurement of antibodies to the best characterized centromere antigen (CENP-B) protein by an enzyme-linked immunosorbent assay (ELISA) that uses human recombinant CENP-B antigen and compare it with indirect immunofluorescence assay (IFA) on HEp-2 cells. METHODS: We have ana…

Complete chromosome 21 centromere sequencing of families with Down syndrome.

Down syndrome, the most common form of human intellectual disability, results from nondisjunction and an extra copy of chromosome 21 (chr21), also known as trisomy 21 (T21). Small centromeres have been hypothesized to contribute to its etiology, and studies on mice suggest that larger centromeres are more efficiently transmitted, yet complete sequencing of chr21 centromeres has been particularly challenging due to their repetitive nature and homology to chromosome 13. Using long-read sequencing, we sequenced and assembled the centromeres from eight families with a child with T21 (one parent-ch…

[Filgotinib treatment of calcinosis in centromere-antibody-positive systemic sclerosis].

Centromere-antibody-positive limited cutaneous systemic sclerosis (LCSSc) is a common disease. It generally takes a benign course, but can still considerably reduce quality of life. Our patient-a woman born in 1987 who was first diagnosis 2017-suffered from pain and declining hand function due to sclerodactyly and calcinosis. Methotrexate (MTX) and analgesics were not effective. After commencing off-label treatment with filgotinib, the patient reported rapid improvement. An X-ray after 8 months showed a reduction in calcinosis load. Filgotinib is well tolerated. Sclerodactyly has resolved, and…

Centromere localization in medaka fish based on half-tetrad analysis.

Gene-centromere (G-C) mapping provides insight into vertebrate genome composition, structure and evolution. Although medaka fish are important experimental animals, no genome-wide G-C map of medaka has been constructed. In this study, we used 112 interspecific triploid hybrids and 152 DNA markers to make G-C maps of all 24 linkage groups (LGs). Under the assumption of 50% interference, 24 centromeres were localized onto all corresponding medaka LGs. Comparison with 21 centromere positions deduced from putative centromeric repeats revealed that 19 were localized inside the centromeric regions o…

Dicentric chromosome formation and epigenetics of centromere formation in plants.

Plant centromeres are generally composed of tandem arrays of simple repeats that form a complex chromosome locus where the kinetochore forms and microtubules attach during mitosis and meiosis. Each chromosome has one centromere region, which is essential for accurate division of the genetic material. Recently, chromosomes containing two centromere regions (called dicentric chromosomes) have been found in maize and wheat. Interestingly, some dicentric chromosomes are stable because only one centromere is active and the other one is inactivated. Because such arrays maintain their typical structu…

Dicentric chromosomes: unique models to study centromere function and inactivation.

Dicentric chromosomes are products of genome rearrangement that place two centromeres on the same chromosome. Depending on the organism, dicentric stability varies after formation. In humans, dicentrics occur naturally in a substantial portion of the population and usually segregate successfully in mitosis and meiosis. Their stability has been attributed to inactivation of one of the two centromeres, creating a functionally monocentric chromosome that can segregate normally during cell division. The molecular basis for centromere inactivation is not well understood, although studies in model o…

A dicentric chromosome of Drosophila melanogaster showing alternate centromere inactivation.

Dicentric chromosomes are rarely found, because they interfere with normal cell division causing chromosome instability. By in situ hybridization of region-specific heterochromatic yeast artificial chromosomes we have found that the artificially generated C(1)A chromosome of Drosophila melanogaster has two potential centromeres: one carries all the sequences of the centromere of the Y chromosome and the other carries only a part of the Y centromeric region that is rich in telomere-related sequences. Immunostaining with anti-Bub1 (a kinetochore-specific marker) shows that, in spite of the diffe…

Single Cd band in dicentric translocations with one suppressed centromere.

Six human dicentric translocations involving the presence of a suppressed centromere were studied: one case of t dic(9;22), one of t dic(13;18), one of t dic(14;15), and three of t dic(13;14). All exhibited chromatid separation at an acrocentric centromere and were demonstrated to have two regions of centromeric constitutive heterochromatin. The four that were available for retrospective study showed a single Cd band, with absence of the Cd band at the suppressed centromere.

Functional domain structure of human centromere protein B. Implication of the internal and C-terminal self-association domains in centromeric heterochromatin condensation.

Centromere protein B (CENP-B) is a common centromere DNA-binding protein among mammalian centromeres. CENP-B possesses the specific DNA binding activity to the 17-base pair sequence dispersed in centromeric repetitive DNA sequences. In the previous study, we have shown that its DNA-binding domain exists within the N-terminal 134 amino acid residues. Here, to clarify the whole domain structure, another functional unit required for CENP-B self-association was examined. Recombinant CENP-B was expressed in Escherichia coli. First, a chemical cross-linking reagent, disuccinimidyl suberate, was used…

Assessing the concordance between centromere AC-3 immunofluorescence pattern and anti-centromere protein-B antibody, and analyzing clinical correlates of dual positivity: A retrospective study.

This study evaluates the consistency between the centromere AC-3 immunofluorescence pattern and the anti-centromere protein B (anti-CENP-B) antibody test results and analyzes differences in the positive rates of potential related diseases with anti-centromere antibody (ACA) positivity. A retrospective collection of antinuclear antibody fluorescent patterns and anti-CENP-B test data from inpatients at Maanshan People's Hospital between February 2022 and February 2024 was performed. The consistency of different methods was evaluated using the kappa test and overall consistency calculation. Subse…

Pan centromeric FISH enhances precision in radiation biodosimetry.

Accurate biodosimetry is critical for assessing radiation exposure in radiological emergencies, occupational monitoring, and clinical management, where precise dose estimation informs life-saving decisions and regulatory compliance. Current gold-standard cytogenetic methods face limitations in sensitivity and reproducibility, especially at low doses (< 0.5 Gy) (practical limitations at low doses, including very low dicentric yields, higher statistical noise, and greater scoring uncertainty as aberration frequencies near background levels). This study presents a systematic comparison of pan-cen

Dicentric chromosomes are resolved through breakage and repair at their centromeres.

Chromosomes with two centromeres provide a unique opportunity to study chromosome breakage and DNA repair using completely endogenous cellular machinery. Using a conditional transcriptional promoter to control the second centromere, we are able to activate the dicentric chromosome and follow the appearance of DNA repair products. We find that the rate of appearance of DNA repair products resulting from homology-based mechanisms exceeds the expected rate based on their limited centromere homology (340 bp) and distance from one another (up to 46.3 kb). In order to identify whether DNA breaks ori…

[Study on chromosomal centromeric dots in couples with spontaneous abortion].

Chromosomal centromeric dots (Cd) of peripheral blood in 20 couples with spontaneous abortion and 10 normal couples were studied by Cd-banding technique. The frequencies of Cd loss and centromeric dots-nucleolar organizer region (Cd-NOR) fusion were 0.36 +/- 0.15/cell and 0.31 +/- 0.20/cell in the couples with abortion, 0.22 +/- 0.13/cell and 0.20 +/- 0.19/cell in normal couples respectively. These results showed that frequencies of Cd loss and Cd-NOR fusion in couples of spontaneous abortion were significantly higher than those in the normal couples. We suggested that the occurrence of sponta

Sequence of centromere separation: a mechanism for orderly separation of dicentrics.

Stable dicentric chromosomes from three mouse cell lines (viz., SEWA Rec4, brain tumor, and L-cells), as well as a human t(9;11) line were analyzed for the sequence in which the two centromeres separate. At prometaphase, as well as in many cells at midmetaphase, the dicentrics express the two centromeres in the form of two primary constrictions. As the cell advances to late metaphase, one of the constrictions loosens the two chromatids so that eventually there is no connection between them. The other centromere stays intact during this period and separates into two units at the metaanaphase ju…

Anti-kinetochore antibodies: use as probes for inactive centromeres.

Application of a modified immunofluorescence technique using an anti-kinetochore serum enables cytogeneticists to obtain quality metaphase spreads and to localize kinetochores. In a patient with a 45, XX, -9, -11, tdic (9p;11p) constitution, we found that the dicentric marker chromosome has an intensely fluorescent kinetochore (no. 11), the functional centromere, and a less intensely fluorescent kinetochore (no. 9), the inactive centromere. The data suggest that in the process of tandem fusion (telomere-telomere between 11p and 9p), the centromere of chromosome 9 was not deleted, but, rather, …
