Architectural and biochemical advances in understanding microbial CO2 concentrating mechanisms.

CO2 concentrating mechanisms (CCMs) are ubiquitous in phototrophs. They are found in the micro as well as the macro world. Different domains of life have developed strikingly different approaches to concentrate inorganic carbon around Rubisco, the most prevalent enzyme on earth. While prokaryotes have developed carboxysomes, icosahedral shell protein cages, microbial eukaryotic phototrophs such as green algae, on the other hand, have developed pyrenoids, liquid-liquid phase separated organelles. In contrast to carboxysomes and pyrenoids, which represent biophysical CCMs, some vascular plants (

Comparative Analysis of Carbohydrate-Active Enzyme (CAZyme) Diversity Across Diverse Algal Lineages.

Algae are characterized by lineage-specific storage and structural carbohydrates; however, the metabolic pathways governing these compounds remain largely uncharacterized despite their high commercial value in the food, pharmaceutical, and nutraceutical sectors. This study addresses this knowledge gap through a comprehensive genome-wide analysis of Carbohydrate-Active Enzymes (CAZymes) across green, red, and brown algae. By utilizing functional homology searches, the CAZyme repertoires of these lineages were identified. Systematic comparative profiling revealed both distinct lineage-specific "

Taxonomic update for the giant algal chloroviruses of the family Phycodnaviridae.

Chloroviruses were first described in the late 1970s, and this opened a new area in virology. These giant DNA viruses, capable of infecting unicellular eukaryotic algae, are ubiquitous in inland waters and have been isolated from various locations worldwide, revealing extensive and complex genetic diversity. These viruses belong to the genus Chlorovirus, family Phycodnaviridae, order Algavirales. Currently, only six species are officially recognized by the International Committee on Taxonomy of Viruses (ICTV), but recent data from our group suggest the existence of many others. In this work, w

Individual and mixture effect of selected high-hazard pharmaceuticals on aquatic primary producers.

The extensive use of pharmaceuticals has led to their occurrence in surface waters due to insufficient treatment processes for their removal. Their environmental impact remains largely unexplored for certain trophic levels, particularly plants and algae. Pharmaceuticals often occur in mixtures with other pollutants, highlighting the need for comprehensive toxicological assessments that evaluate their combined interactions. This study evaluated the acute toxicity of four high-hazard pharmaceuticals -diphenhydramine, fluoxetine, ketoprofen, and trimethoprim- and their binary mixtures, on the gre

Semisynthesis of new sulfated heterorhamnan derivatives obtained from green seaweed Gayralia brasiliensis and evaluation of their anticoagulant activity.

Marine green algae produce sulfated polysaccharides with diverse structures and a wide range of biological activities. This study aimed to enhance the biotechnological potential of sulfated heterorhamnan (Gb1) from Gayralia brasiliensis by chemically modifying it for improved or new biological functions. Using controlled Smith Degradation (GBS) and O-alkylation with 3-chloropropylamine, we synthesized partially water-soluble amine derivatives. GBS modification increase sulfate groups (29.3 to 37.5 %) and α-l-rhamnose units (69.9 to 81.2 mol%), reducing xylose and glucose, compared to Gb1. The

Structure and anticoagulant activity of a galactoarabinan sulfate polysaccharide and its oligosaccharide from the green algae, Codium fragile.

A polysaccharide, CZS-0-1, was obtained from the marine green algae Codium fragile using ion-exchange and size-exclusion chromatography. Composition and characteristics analyses showed CZS-0-1 was a sulfated galactoarabinan consisting of arabinose, galactose and a small amount of glucose in a ratio of 9:2:1 with 21% sulfate content and a molecular weight of 810 kDa. Structural properties were determined using desulfation and methylation analyses combined with instrument analysis. The results showed that the backbone of CZS-0-1 was (1 → 3)-β-L-Arap. Its O-4 and/or O-2 positions showed sulfate m

A pyruvylated and sulfated galactan from the green alga Dictyosphaeria cavernosa: Structure, anticoagulant property and inhibitory effect on zebrafish thrombosis.

A pyruvylated and sulfated galactan from the green alga Dictyosphaeria cavernosa, designated PSG, was obtained by dilute alkali extraction, ion-exchange and gel filtration chromatography. The backbone of PSG was composed of 3-linked β-d-Galp units with partial sulfation on C-4 and C-6. Pyruvate ketals were linked to O-3 and O-4 of nonreducing terminal β-d-Galp, as well as O-4 and O-6 of 3-linked β-d-Galp. The branches consisting of 6-linked β-d-Galp(4SO4) and β-d-Galp(3,4-Pyr)-(1→ units were located at C-6 of 3-linked β-d-Galp unit. PSG possessed obvious anticoagulant effect in vitro as assess

Combined toxicity of fluorescent silica nanoparticles with cadmium in Ceriodaphnia dubia: Interactive effects of natural organic matter and green algae feeding.

The environmental risks of silica nanoparticles (SiNP) reported in the literature are contradictory and bring into question its safety for use in consumer applications. Organisms are never exposed to NPs alone in the real environment, while studies of the combined toxicity of SiNP are limited. To address this, we compared the acute toxicity of fluorescent core-shell SiNPs alone and in mixtures with Cd2+ to Ceriodaphnia dubia in the absence and presence of NOM. We identified biodistribution and feeding behaviour in addition to the traditional endpoints. NOM increased the colloidal stability of

Biotransformation of macrolide antibiotics using enriched activated sludge culture: Kinetics, transformation routes and ecotoxicological evaluation.

The biotransformation of three prominent macrolide antibiotics (azithromycin, clarithromycin and erythromycin) by an activated sludge culture, which was adapted to high concentrations of azithromycin (10 mg/L) was investigated. The study included determination of removal kinetics of the parent compounds, identification of their major biotransformation products (TPs) and assessment of ecotoxicological effects of biotransformation. The chemical analyses were performed by ultra-performance liquid chromatography/quadrupole-time-of-flight mass spectrometry, which enabled a tentative identification

Biosynthesis, purification and characterization of polyhydroxybutyrate from Botryococcus braunii kütz.

Polyhydroxybutyrate (PHB) is completely biodegradable which is metabolised by microorganisms in the soil as their sole food source in few years. The level of PHB up to 10.6% of algal dry weight is of great potential of the eco-friendly nature. Botryococcus braunii is mainly used for the production of biodiesel and is also capable of producing biopolymer polyhydroxy butyrate (PHB). In this study, Botryococcus braunii is used which generally produce PHB to around 20% of the dry weight. Three different microalgae were isolated from the fresh water of Kolavoi lake of Tamil Nadu. Effect of manganese and calcium deficiency on the growth and oxygen exchange of Scenedesmus intermedius cultured for successive generations.

The green alga Scenedesmus intermedius was grown in synchronous culture under manganese or calcium deficiency for six successive generations. The growth rate, pigment and protein contents gradually decreased in comparison with the control. In Mn-deficient cells, the rate of oxygen evolution was sharply decreased. This inhibition was restored to normal in less than 1 h (40-60 min) by adding Mn salt to the suspension medium. In Ca-deficient cells, the inhibition of photosynthesis appears to be irreversible.

High variability of the electron carrier plastocyanin in microalgae.

1. The plastocyanin content of Scenedesmus can be dramatically varied with the copper content of the culture medium. Figures as high as 7 mmol plastocyanin/mol chlorophyll are possible. Electron paramagnetic resonance (EPR) spectroscopy has been used to determine this physiological response quantitatively in intact cells having different amounts of plastocyanin. The results obtained by the EPR technique were compared with data on isolated plastocyanin determined either by spectrophotometry or immunoelectrophoresis. Agreement was found for the amount of plastocyanin detected by the first two me

[Effect of the ionic composition of the environment on the intracellular potential of the alga Acetabularia].

Concentration of K+-, Na+-, Mg++-, Ca++-, Cl-ions defined in Acetabularia crenulata cell was the same as that of Acetabularia mediterranea, both were cultivated under the same conditions. At the variations of the ionic composition of the external medium it has been shown that the potential of Acetabularia (A.) cell depends on K+-, Cl-ions and alga A. can accumulate K+- Cl-ions and lose Na+, Mg++ ones when there is a deficiency of these ions in the medium. Na+-, Mg++-ions participation in the generation of the rest potential of Alga A. is insignificant. K+- ion is able to change the ionic perme

Comparative genomics of nitrogen acquisition and assimilation gene-family copy-number architecture across chlorophytes.

Nitrogen acquisition and assimilation determine nitrogen availability for algal growth and metabolism; however, their gene-family organization across chlorophytes remains poorly resolved. We examined 10 gene families (NiR, NR, NRT, AMT, GS, GOGAT, GDH, ASN, CPS-L, and CPS-S) in 30 chlorophyte genome assemblies using family-specific sequence-based curation. The assemblies represented 26 biological species after taxonomic standardization and pseudoreplication control. One representative assembly per species defined the primary dataset (D1; n = 26), with BUSCO C ≥ 80% (D2; n = 22) and C ≥ 90% (D3

Extraction of astaxanthin from fresh Haematococcus pluvialis biomass: a sustainable approach using vegetable oils.

In this study, we developed an optimized process targeting the key challenges in extracting astaxanthin from fresh H. pluvialis biomass. First, we compared the effectiveness of several commonly used cell disruption methods (acidic and alkaline hydrolysis, ultrasound, enzymatic treatment, and freeze-thaw cycles) on astaxanthin yield and quality. We then evaluated the effects of polar (alcohols: methanol, ethanol, and glycerol) and non-polar solvents (argan, cannabis, and olive oils) on astaxanthin extraction under different conditions (temperature and extraction time). Alkaline treatment with 1

Eluates from façades at the beginning of their service time affect aquatic and sediment organisms.

Biocides are used in building materials to prevent microbial growth during storage (in-can preservatives) as well as after application (film preservatives). These compounds can leach out from the material into the environment and harm non-target organisms. In this study, the ecotoxicological effect of leachates at the beginning of a façade lifetime, on sediment and aquatic organisms was examined. For this purpose, leaching tests were carried out in the setting of a natural weathering experiment and a laboratory immersion with façade samples consisting of render/paint systems. The leaching expe
