Novel TRPM8 antagonist attenuates cold hypersensitivity after peripheral nerve injury in rats.

Abnormal cold sensitivity is a common feature of a range of neuropathies. In the murine somatosensory system, multiple aspects of cold sensitivity are dependent on TRPM8, both short term and in response to peripheral nerve injury. The specialized nature of cold-sensitive afferents and the restricted expression of TRPM8 render it an attractive target for the treatment of cold hypersensitivity. This current study examines the effect of a novel TRPM8 antagonist (M8-An) in naive and spinal nerve-ligated rats through behavioral and in vivo electrophysiological approaches. In vitro, M8-An inhibited

Cold ischemia/reperfusion injury in a mouse model of partial liver transplantation.

BACKGROUND: Injury to partial liver grafts as a result of ischemia reperfusion remains an important factor in the outcome after transplantation in rodents. Although well described in rats, extended preservation has not been studied in a mouse model of partial liver transplantation. The aim of this study was to develop a model of cold ischemia/reperfusion injury in mouse partial liver transplantation. MATERIALS AND METHODS: Partial liver transplantation was performed in C57BL/6 mice. The liver grafts were stored in 4 °C University of Wisconsin solution for different periods of preservation. Th

New preservation strategies for preventing liver grafts against cold ischemia reperfusion injury.

BACKGROUND: In spite of improvements in University of Wisconsin (UW) preservation solution, the injury from grafts during cold storage is an unresolved problem in liver transplantation. The aim of the present study was to evaluate the beneficial effect on ischemia-reperfusion injury associated with liver transplantation of the inversion of K(+) and Na(+) concentrations and the replacement of hydroxyethyl starch (HES) by polyethylene glycol (PEG) in UW preservation solution. METHODS: Using an orthotopic liver transplantation model, the effects on rat liver preservation of a modified preservati

Mitochondrial defects by intracellular calcium overload versus endothelial cold ischemia/reperfusion injury.

Questions as to the critical stress factor and primary targets of cold ischemia/reperfusion (CIR) injury were addressed by comparing mitochondrial defects caused by (1) CIR injury and (2) intracellular Ca2+ overload. CIR was simulated in transformed human umbilical vein endothelial cell cultures (tEC) by 8 h cold anoxia in University of Wisconsin solution and reoxygenation at 37 degrees C. Intracellular Ca2+ concentrations were changed by permeabilization of suspended cells with digitonin in culture medium (RPMI, 0.4 mM Ca2+). Binding of free Ca2+ by ethylene glycol-bis(beta-aminoethylether)-N

Inhibition of phosphodiesterase 10A mitigates neuronal injury by modulating apoptotic pathways in cold-induced traumatic brain injury.

Brain injury develops from a complex series of pathophysiological phases, resulting in acute necrotic or delayed apoptotic cell death after traumatic brain injury (TBI). Inhibition of apoptotic cell death is critical for the treatment of acute neurodegenerative disorders, such as TBI. Here, we investigated the role of phosphodiesterase 10A (PDE10A) in the development of neuronal injury, particularly in apoptotic cell death. Using the PDE10A inhibitor TAK-063, we found that PDE10A inhibition is associated with decreased brain injury, brain swelling, and blood brain barrier disruption 48 h after

Prolonged Cold Ischemic Time Before Normothermic Machine Perfusion Accentuates Postreperfusion Hepatocellular Injury.

BACKGROUND: Prolonged cold ischemic time (CIT) before liver transplantation with static cold storage preservation is associated with higher rates of ischemia-related complications. With the introduction of normothermic machine perfusion (NMP), the overall incidence of early allograft dysfunction and biliary complications is lower. NMP can be used as a device-to-donor approach (with short CIT) or back-to-base model which is typically associated with longer CIT. It is unknown if NMP can rehabilitate organs that have suffered from prolonged CIT. METHODS: Livers from a Yorkshire cross-bred pig do

Snow or ice-related injury claims in cold months during 2016-2021.

INTRODUCTION: Falls, a preventable worker-related injury, are major contributors to worker's disability and death. In spite of this, there is relatively little analysis on snow or ice- related slips, trips and falls among workers in the workers' compensation system. This analysis aimed to describe the frequency, characteristics, and occupational burden of snow or ice-related slip, trip, or fall (STF) claims in Wisconsin. METHODS: This study is a descriptive analysis of working age adults' snow or ice-related STF lost time claims during the months of October through April for the years 2016-20

Preliminary mechanism of inhibitor of SGLT2 in fatty liver cold ischemia injury.

With rapid development of liver transplantation technology, the demand for transplants have reached beyond the supply of organs, and thus development of effective strategies to reduce cold ischemia injury in fatty liver is important. Here, we explored the potential effect of SGLT-2 inhibitor in cold ischemia injury, fatty livers from 2 weeks methionine and choline deficient diet (MCD) rats were administered. After one week of intragastric administration of Sodium-dependent glucose transporters (SGLT-2) inhibitor empagliflozin (EMPA) or NaCI, liver were stored for 24 h. The results showed that

Self-inflicted finger cold injury leading to amputation: Report of a case.

A cold injury can result in devastating outcomes, leading to significant morbidity and loss of distal extremities. Amputations are common after severe frostbite injuries with delayed presentation, often mediated by post-injury arterial thrombosis. Ischemic injuries are managed according to the ischemia time. The most controversial aspect of treating a salvage injury is the time of surgical intervention, which used to be based on the previous management dogma freeze in January, amputate in July. Recently, the paradigm has shifted to early surgical management if the level of viability of the dee

Neuroprotective effect of lithium in cold- induced traumatic brain injury in mice.

Apart from its well-established therapeutic activity on bipolar disorder and depression, lithium exerts neuroprotective activity upon neurodegenerative disorders, such as traumatic brain injury (TBI). However, the cellular signaling mechanisms mediating lithium's neuroprotective activity and long-term dose- and time-dependent effects on close and remote proximity are largely unknown. Herein, we tested prophylactic and acute effects of lithium (2 mmol/kg) after cold- induced TBI. In both conditions, treatments with lithium resulted in reduced infarct volume and apoptosis. Its acute treatment re

Blocking cold-inducible RNA-binding protein protects liver from ischemia-reperfusion injury.

Cold-inducible RNA-binding protein (CIRP) is a nuclear protein that has been recently identified as a novel inflammatory mediator in hemorrhagic shock and sepsis. We hypothesized that CIRP acts as a potent inflammatory mediator in hepatic ischemia-reperfusion (I/R), and thus blocking CIRP protects against I/R-induced liver injury. Male C57BL/6 mice were subjected to 70% hepatic ischemia by microvascular clamping of the hilum of the left and median liver lobes for 60 min, followed by reperfusion. Anti-CIRP antibody (1 mg/kg body weight) or vehicle (normal saline) in 0.2 mL was injected via the

Hypothermia and cold injuries - rare but severe manifestations of child abuse and neglect.

BACKGROUND: While thermal injuries caused by heat are well-recognized in cases of child maltreatment, injuries resulting from cold exposure (hypothermia and local cold injury) appear to be rare in this context. Although hypothermia and cold injuries related to child abuse and neglect pose a significant threat to affected children, systematic data on this topic are currently lacking. The goal of this article was to better understand the frequency, forms and contexts of child maltreatment involving cold exposure. METHODS: We conducted a narrative review with systematic elements based on a PubMe

Diagnostic difficulties in a patient with paroxysmal cold haemoglobinuria and acute kidney injury.

Paroxysmal cold haemoglobinuria (PCH) is a form of autoimmune haemolytic anaemia (AIHA) characterised by a sudden onset of haemoglobinuria, either spontaneously or following exposure to cold. In children, it is commonly seen following a viral illness or after immunisation. Diagnosis of PCH is confirmed by a positive Donath Landsteiner (DL) test in which biphasic haemolysins are detected. However, in a real clinical setting, the serological diagnosis of PCH is not always easy. PCH can cause tubular renal injury, which in turn can lead to renal impairment. We describe a case of a two-year-old bo

The effect of cold stress on UVB injury in mouse skin and cultured keratinocytes.

The effect of cold stress on skin damage caused by UVB irradiation was investigated both in vivo and in vitro. Ear skin of mice that had been exposed to cold stress at 0 degree C for 20 min and at 5 degrees C for 24 h was exposed to UVB radiation. Sunburn cell production was less in mice exposed to the lower temperature. In addition, the effect of cold stress on the survival rate of UVB-irradiated rat keratinocytes was examined in a cytotoxicity test, with the results showing that keratinocytes exposed to cold stress of 0 degree C had a higher survival rate than control cells. A simplified and reproducible ex vivo model of cold and ischemia-reperfusion injury.

Both cold stress and ischemia-reperfusion injury significantly contribute to poor prognosis after liver transplantation (LT). However, limited animal models incorporating both stimuli hinder the advancement of transplant-related research. Here, a simplified and reproducible isolated perfused liver model is established to simulate the stresses experienced by livers maximally during transplantation. We provide a detailed protocol for a straightforward technique that requires 20-30 min for harvesting, 24-48 h for static cold storage (SCS), and 2 h for normothermic machine perfusion (NMP) to induc

The cold truth: the role of cryotherapy in the treatment of injury and recovery from exercise.

Cryotherapy is utilized as a physical intervention in the treatment of injury and exercise recovery. Traditionally, ice is used in the treatment of musculoskeletal injury while cold water immersion or whole-body cryotherapy is used for recovery from exercise. In humans, the primary benefit of traditional cryotherapy is reduced pain following injury or soreness following exercise. Cryotherapy-induced reductions in metabolism, inflammation, and tissue damage have been demonstrated in animal models of muscle injury; however, comparable evidence in humans is lacking. This absence is likely due to
