GM1 gangliosidosis is one of the inherited metabolic lysosomal storage disorders characterized by neurological symptoms caused by beta-galactosidase deficiency and consequent accumulation of GM1 ganglioside in neuronal cells. Shiba dogs affected with GM1 gangliosidosis have been found to suffer from corneal opacity. In our morphological analysis, keratocyte enlargement was induced by abnormal intracellular accumulation of neutral carbohydrates, resulting in the loss of normal arrangement of collagen fibrils in the opaque cornea was found to be associated with the disorder. We therefore conclud…

Corneal opacification associated with glycosaminoglycan (GAG) deposition occurs in canine mucopolysaccharidosis I (MPS I), a deficiency of the lysosomal enzyme alpha-L-iduronidase. In affected dogs corneal lesions appear similar to those in children with the same disease. Transplantation of bone marrow from unaffected littermates was performed in 5 MPS I affected dogs at 5 months of age. In three recipients that became long-term survivors corneal clouding was largely alleviated compared to affected control dogs. In no case, however, did the corneas remain totally clear throughout the course of

Corneal opacity resulting from corneal injury is a leading cause of blindness. The interaction of extracellular matrix (ECM) proteins, cytokines, and immune cells induces corneal opacity after corneal injury. Periostin, which is secreted into the ECM, is involved in wound healing and is associated with immune cell infiltration. The function of periostin in corneal wound healing and in the development of corneal opacity was investigated. Wild-type (WT) and Postn knockout (KO) mice underwent central corneal incision. Periostin expression level was significantly increased after the incision in WT…

AIM: To assess the utility and efficiency of endoscopy-assisted vitrectomy (EAV) for the treatment of corneal opacity in severe ocular trauma.
METHODS: Patients who underwent fundus examination using a preoperative slit lamp and intraoperative endoscopy, followed by EAV and additional surgery were retrospectively recruited. Silicone oil removal and penetrating keratoplasty were used in selected eyes at postoperative follow-ups. Outcome measurements included the best corrected visual acuity (BCVA), intraocular pressure (IOP), findings of endoscopic fundus examination, and postoperative complic…

BACKGROUND: Trachoma is the leading infectious cause of blindness. Patients with trachomatous corneal opacity (TCO) are traditionally considered high-risk cases for graft failure. However, anecdotal evidence suggests that corneal transplantation may restore vision in such individuals. We wanted to review the available evidence for keratoplasty outcomes in TCO.
METHODS: A literature search of PubMed, MEDLINE, and Web of Science was performed using the search terms "trachoma* AND (keratoplasty OR cornea* transplant*)". The search was restricted to studies published between 1 January 1992 and 12…

PURPOSE: To investigate whether three consanguineous families from the Punjab province of Pakistan, with affected members with recessively inherited congenital cataract microcornea with corneal opacity, are genetically homogeneous.
METHODS: An ophthalmic examination was performed on each family member to establish the diagnosis. The two largest families were analyzed by homozygosity mapping using SNP arrays. Linkage was confirmed using polymorphic microsatellite markers, and logarithm of odds (LOD) scores were calculated. Candidate genes were prioritized using the ENDEAVOUR program.

PURPOSE: This is a case report of a 31-year-old male patient presenting progressive and markedly asymmetric keratoconus treated with sequential intracorneal ring segment (ICRS) implantation followed by accelerated corneal cross-linking (CXL).
OBSERVATIONS: The follow-up after the last procedure revealed a thin, opacified cornea with an unexpected massive flattening of up to 20.3 D. The central flattening attributed to the individual effect of CXL (post-ICRS-implant vs. post-CXL) was 19.1 D.
CONCLUSIONS AND IMPORTANCE: This original case reports how CXL followed by ICRS implant may result in

Artificial intelligence (AI) has emerged as a transformative tool in ophthalmology for disease diagnosis and prognosis. However, use of AI for assessing corneal damage due to chemical injury in live rabbits remains lacking. This study aimed to develop an AI-derived clinical classification model for an objective grading of corneal injury and opacity levels in live rabbits following ocular exposure of sulfur mustard (SM). An automated method to grade corneal injury minimizes diagnostic errors and enhances translational application of preclinical research in better human eyecare. SM induced corne

Anterior segment dysgenesis describes a group of heterogeneous developmental disorders that affect the anterior chamber of the eye and are associated with an increased risk of glaucoma. Here, we report homozygous mutations in peroxidasin (PXDN) in two consanguineous Pakistani families with congenital cataract-microcornea with mild to moderate corneal opacity and in a consanguineous Cambodian family with developmental glaucoma and severe corneal opacification. These results highlight the diverse ocular phenotypes caused by PXDN mutations, which are likely due to differences in genetic backgroun…

PURPOSE: To report successful vision restoration after lamellar keratectomy using a femtosecond (FS) laser in a patient with superficial corneal opacity that developed after an episode of epidemic keratoconjunctivitis.
METHODS: A 19-year-old girl with corneal opacity in the left eye was referred to our department for treatment. The opacity was principally located within the superficial anterior cornea. To remove the opacity with minimization of change in the keratometric value, we performed lamellar keratectomy with the aim of creating a 100-μm-thick flap, using an FS laser (IntraLase FS; AMO…

Previously undescribed ocular and dermatologic complications of prolonged chlorpromazine therapy have been noted in 70 patients of a series of many thousands receiving similar therapy. All affected patients were women who had been receiving high doses of chlorpromazine, averaging 500 to 1500 mg. daily, for at least three to five years before the complications became apparent. Skin manifestations consisted of a peculiar purplish pigmentation of the skin of exposed areas of the face, neck and hands, characterized histologically by deposition of material with the staining properties of melanin in
